Membrane Potential Dynamics

Membrane potential dynamics describe how the electrical voltage across a cell membrane changes over time, a process that enables neurons and other excitable cells to communicate. These changes arise from ion concentration gradients and selective membrane permeability, as sodium, potassium, calcium, and chloride move through leak or voltage-gated channels, altering the membrane’s charge and driving depolarization, repolarization, or hyperpolarization. In neuroscience, analyzing these dynamics explains resting potential, action potential generation, synaptic integration, and signal propagation along axons. It also supports research into neural coding, sensory processing, and disorders caused by disrupted ion-channel function.

Overview The relative difference in electrical charge, or voltage, between the inside and the outside of a cell membrane, is called the membrane potential. It is generated by differences in permeability of the membrane to various ions and the concentrations of these ions across the membrane. The Inside of a Neuron Is More Negative The membrane potential of a cell can be measured by inserting a microelectrode into a cell and comparing the charge to a reference electrode in the extracellular...

High-Resolution Fluorespirometry to Assess Dynamic Changes in Mitochondrial Membrane Potential in Human Immune Cells

2024

Methods for studying mitochondrial bioenergetics under physiologically relevant substrate concentrations in immune cells are limited. We provide a detailed protocol that uses high-resolution fluorespirometry to assess changes in the response of the mitochondrial membrane potential to energy demand in human T-cells, monocytes, and peripheral mononuclear cells.
