Pole Test Assessment

Pole Test Assessment is a behavioral assay used to measure motor coordination, balance, and movement initiation in laboratory rodents, making it valuable for neuroscience research on motor-system function. During the test, an animal is placed head-up on a vertically oriented rough-surfaced pole, and researchers record the time required to turn downward and descend, along with related movement errors; impaired performance can indicate deficits in coordination or bradykinesia. The assay is widely applied in studies of Parkinson’s disease, basal ganglia circuitry, neurodegeneration, and motor effects of drugs or genetic alterations, providing a relatively simple quantitative outcome for evaluating disease models and treatments.

This video demonstrates the technique used for preparation of organizer and animal pole explants from Xenopus laevis embryos, including the use of the eyebrow knife - a specialized dissection tool made of one's eyebrow. The protocol for assembling an adhesion assay is also given, which probes for the presence of key adhesion molecules present on the surface organizer or animal pole cells that are critical for proper development.

The aim was to substantiate optimal use of data collection techniques for Nordic walking gait and posture analysis. Three-dimensional motion capture should be used during short duration analysis (i.e. single gait cycle), while accelerometry should be employed for longer duration analysis (i.e. repeated cycles) like a 6 Minute Walk Test.
