Laser Sheet Imaging

Laser sheet imaging is an optical technique that illuminates a thin, planar section of a sample with a laser sheet while a camera records light from the illuminated plane. By directing excitation and detection along separate, typically perpendicular paths, the method restricts fluorescence or scattering signals to a defined slice and enables optical sectioning without scanning the entire volume point by point. Rapidly moving the sheet or sample produces three-dimensional image stacks with reduced out-of-focus background and photobleaching compared with wide-field illumination. In physics and related research, laser sheet imaging supports quantitative studies of fluid flow, particle motion, biological structure, and dynamic processes.

Light sheet fluorescence microscopy is an excellent tool for imaging embryonic development. It allows recording of long time-lapse movies of live embryos in near physiological conditions. We demonstrate its application for imaging zebrafish eye development across wide spatio-temporal scales and present a pipeline for fusion and deconvolution of multiview datasets.
