Since 1995, raw vegetable sprouts have been implicated as the vehicle of infection in 15 foodborne outbreaks involving Salmonella and 2 foodborne outbreaks involving Escherichia coli O157:H7. To reduce the numbers of sprout-related outbreaks, the U.S. Food and Drug Administration (FDA) published Guidance for Industry: Reducing Microbial Food Safety Hazards for Sprouting Seeds in 1999. Between October 2000 and April 2001, 61.5% (16 of 26) of the known commercial sprout firms in California were enrolled in a survey to evaluate the industry practices of California sprouting operations and to determine compliance with FDA guidelines. A standardized questionnaire was used to collect data on firm demographics and seed disinfection practices. Additionally, free chlorine levels in seed disinfection solutions were measured, and 48-h spent irrigation water samples were collected from each firm. The irrigation water was screened for Salmonella and E. coli O157:H7 with FDA-recommended test kits. Free chlorine levels in the treatment solutions ranged from 50 to 35,000 mg/liter (ppm), with a median of 14,000 mg/liter (ppm). Free chlorine levels were higher for firms producing alfalfa sprouts than for those producing only mung bean or soybean sprouts (P=0.03). Levels of free chlorine tended to be higher for firms using a calcium hypochlorite treatment solution than for firms using a sodium hypochlorite treatment solution (P=0.067). All 32 irrigation water samples screened for Salmonella tested negative. Of the irrigation water samples tested for E. coil O157:H7, 75% (24 of 32) tested negative, and 25% (8 of 32) tested presumptive positive. The eight presumptive positive samples were found to be negative after further testing. These results indicate that producers of alfalfa sprouts are generally achieving the FDA-recommended calcium hypochlorite level of 20,000 mg/liter (ppm), whereas mung bean sprout producers are not.

Industry practices and compliance with U.S. Food and Drug Administration guidelines among California sprout firms.

The purpose of this study was to analyze the effect of various degrees of axial midline angulation on the attractiveness of a smile. We explored the influence of age, race, sex, direction of midline deviation, education, occupation, and dominant hand on each evaluator's perception of dental esthetics. Photographs of smiling subjects--one man and one woman--were altered to produce both left and right axial midline angulations in 5 degree increments. Fifty orthodontists and 50 laypeople evaluated these altered photographs by assigning both a numerical attractiveness rating and an acceptable or unacceptable rating to each. The results showed that attractiveness scores and acceptability ratings declined consistently as axial midline angulation increased. Statistical analysis showed that both sex of the subject and occupation of the judge were significant variables (P < .05) in the evaluation of the subjects. Age, race, sex of the judge, education level, direction of midline deviation, and dominant hand were not statistically significant. The mean acceptable midline angulation for the male subject was 6.6 +/- 4.5 degrees for orthodontists and 10.7 +/- 6.2 degrees for laypeople. For the female subject, the mean acceptable threshold was 6.4 +/- 4.0 degrees for orthodontists and 10.0 +/- 6.1 degrees for laypeople (P < .001). Discrepancies of 10 degrees were unacceptable by 68% of orthodontists and 41% of laypeople.

The effect of axial midline angulation on dental esthetics.

Filter metrics are used as a quick assessment of sequence trace files in order to sort data into different categories (i.e. high quality, review, and low quality) without human intervention. The filter metrics consist of two numerical parameters for sequence quality assessment: trace score (TS) and contiguous read length (CRL). Primer-specific settings for the TS and CRL were established using a calibration dataset of 2817 traces and validated using a concordance dataset of 5617 traces. Prior to optimization, 57% of the traces required manual review before import into a sequence analysis program, whereas after optimization only 28% of the traces required manual review. After optimization of primer-specific filter metrics for mitochondrial DNA sequence data, an overall reduction of review of trace files translates into increased throughput of data analysis and decreased time required for manual review.

Purpose: To establish the zebrafish platinum mutant as a model for studying vision defects caused by syndromic albinism diseases such as Chediak-Higashi syndrome, Griscelli syndrome, and Hermansky-Pudlak syndrome (HPS). Methods: Bulked segregant analysis and candidate gene sequencing revealed that the zebrafish platinum mutation is a single nucleotide insertion in the vps11 (vacuolar protein sorting 11) gene. Expression of vps11 was determined by RT-PCR and in situ hybridization. Mutants were analyzed for pigmentation defects and retinal pathology by histology, immunohistochemistry, and transmission electron microscopy. Results: Phenocopy and rescue experiments determined that a loss of Vps11 results in the platinum phenotype. Expression of vps11 appeared ubiquitous during zebrafish development, with stronger expression in the developing retina and retinal pigmented epithelium (RPE). Zebrafish platinum mutants exhibited reduced pigmentation in the body and retinal pigmented epithelium (RPE), however, melanophore development, migration, and dispersion occurred normally. RPE, photoreceptors, and inner retinal neurons formed normally in zebrafish platinum mutants. However, a gradual loss of RPE, an absence of mature melanosomes and the subsequent degradation of RPE/photoreceptor interdigitation was observed. Conclusions: These data show that Vps11 is not required for normal retinal development or initiation of melanin biosynthesis, but is required for melanosome maturation and healthy maintenance of the RPE and photoreceptors.

Objetivo: Establecer el platino pez cebra mutante como un modelo para el estudio de los defectos de la visi&#xF3;n causados &#x200B;&#x200B;por enfermedades tales como albinismo s&#xED;ndromes de Chediak-Higashi, s&#xED;ndrome de Griscelli, y el s&#xED;ndrome de Hermansky-Pudlak (HPS). M&#xE9;todos: An&#xE1;lisis abultado segregante y la secuenciaci&#xF3;n de genes candidatos revel&#xF3; que la mutaci&#xF3;n de platino pez cebra es la inserci&#xF3;n de un solo nucle&#xF3;tido en el vps11 (prote&#xED;na vacuolar clasificaci&#xF3;n de 11) de genes. Expresi&#xF3;n de vps11 se determin&#xF3; por RT-PCR e hibridaci&#xF3;n in situ. Fgf signaling is required for many biological processes involving the regulation of cell proliferation and maintenance, including embryonic patterning, tissue homeostasis, wound healing, and cancer progression. Although the function of Fgf signaling is suggested in several different regeneration models, including appendage regeneration in amphibians and fin and heart regeneration in zebrafish, it has not yet been studied during zebrafish photoreceptor cell regeneration. Here we demonstrate that intravitreal injections of FGF-2 induced rod precursor cell proliferation and photoreceptor cell neuroprotection during intense light damage. Using the dominant-negative Tg(hsp70:dn-fgfr1) transgenic line, we found that Fgf signaling was required for homeostasis of rod, but not cone, photoreceptors. Even though fgfr1 is expressed in both rod and cone photoreceptors, we found that Fgf signaling differentially affected the regeneration of cone and rod photoreceptors in the light-damaged retina, with the dominant-negative hsp70:dn-fgfr1 transgene significantly repressing rod photoreceptor regeneration without affecting cone photoreceptors. These data suggest that rod photoreceptor homeostasis and regeneration is Fgf-dependent and that rod and cone photoreceptors in adult zebrafish are regulated by different signaling pathways.

In this study, we used a newly-created transgenic zebrafish, Tg(nrd:egfp)/albino, to further characterize the expression of neurod in the developing and adult retina and to determine neurod expression during adult photoreceptor regeneration. We also provide observations regarding the expression of neurod in a variety of other tissues. In this line, EGFP is found in cells of the developing and adult retina, pineal gland, cerebellum, olfactory bulbs, midbrain, hindbrain, neural tube, lateral line, inner ear, pancreas, gut, and fin. Using immunohistochemistry and in situ hybridization, we compare the expression of the nrd:egfp transgene to that of endogenous neurod and to known retinal cell types. Consistent with previous data based on in situ hybridizations, we show that during retinal development, the nrd:egfp transgene is not expressed in proliferating retinal neuroepithelium, and is expressed in a subset of retinal neurons. In contrast to previous studies, nrd:egfp is gradually re-expressed in all rod photoreceptors. During photoreceptor regeneration in adult zebrafish, in situ hybridization reveals that neurod is not expressed in MÃ¼ller glial-derived neuronal progenitors, but is expressed in photoreceptor progenitors as they migrate to the outer nuclear layer and differentiate into new rod photoreceptors. During photoreceptor regeneration, expression of the nrd:egfp matches that of neurod. We conclude that Tg(nrd:egfp)/albino is a good representation of endogenous neurod expression, is a useful tool to visualize neurod expression in a variety of tissues and will aid investigating the fundamental processes that govern photoreceptor regeneration in adults.

Using the Tg(nrd:egfp)/albino zebrafish line to characterize in vivo expression of neurod.

Dans cette &#xE9;tude, nous avons utilis&#xE9; un nouvellement cr&#xE9;&#xE9; transg&#xE9;nique poisson-z&#xE8;bre, Tg (BDNI: egfp) / albinos, afin de mieux caract&#xE9;riser l&#x26;#39;expression de NeuroD dans la r&#xE9;tine en d&#xE9;veloppement et des adultes et pour d&#xE9;terminer l&#x26;#39;expression NeuroD lors de la r&#xE9;g&#xE9;n&#xE9;ration photor&#xE9;cepteur adulte. Nous fournissons &#xE9;galement des observations concernant l&#x26;#39;expression de NeuroD dans une vari&#xE9;t&#xE9; d&#x26;#39;autres tissus. Dans cette ligne, EGFP se trouve dans les cellules de la r&#xE9;tine en d&#xE9;veloppement et les adultes, la glande pin&#xE9;ale, le cervelet, le bulbe olfactif, le m&#xE9;senc&#xE9;phale, le rhombenc&#xE9;phale, le tube neural, la ligne lat&#xE9;rale, l&#x26;#39;oreille interne, le pancr&#xE9;as, l&#x26;#39;intestin, et la nageoire. Utilisation de l&#x26;#39;immunohistochimie et hybridation in situ, nous comparons l&#x26;#39;expression de la BDNI: transg&#xE8;ne EGFP &#xE0; celle de NeuroD endog&#xE8;ne et &#xE0; conna&#xEE;tre les types de cellules r&#xE9;tiniennes. Conform&#xE9;ment aux donn&#xE9;es ant&#xE9;rieures bas&#xE9;es sur des hybridations in situ, nous montrons que pendant le d&#xE9;veloppement de la r&#xE9;tine, la BDNI: transg&#xE8;ne EGFP n&#x26;#39;est pas exprim&#xE9;e dans la prolif&#xE9;ration neuro&#xE9;pith&#xE9;lium r&#xE9;tinien, et est exprim&#xE9; dans un sous-ensemble de neurones de la r&#xE9;tine. Contrairement aux &#xE9;tudes pr&#xE9;c&#xE9;dentes, la BDNI: EGFP est progressivement re-exprim&#xE9;e dans tous les photor&#xE9;cepteurs b&#xE2;tonnets. Lors de la r&#xE9;g&#xE9;n&#xE9;ration chez le poisson z&#xE8;bre adulte photor&#xE9;cepteur, hybridation in situ r&#xE9;v&#xE8;le que NeuroD n&#x26;#39;est pas exprim&#xE9;e dans l&#x26;#39;arr&#xEA;t M&#xFC;ller d&#xE9;riv&#xE9;s des cellules gliales prog&#xE9;nitrices neuronales, mais elle est exprim&#xE9;e dans les prog&#xE9;niteurs photor&#xE9;cepteurs comme ils migrent vers la couche nucl&#xE9;aire externe et se diff&#xE9;rencient en photor&#xE9;cepteurs nouvelle tige. Lors de la r&#xE9;g&#xE9;n&#xE9;ration photor&#xE9;cepteur, l&#x26;#39;expression de la BDNI: matchs eGFP que de NeuroD. Nous concluons que Tg (BDNI: egfp) / albinos est une bonne repr&#xE9;sentation de l&#x26;#39;expression NeuroD endog&#xE8;ne, est un outil utile pour visualiser l&#x26;#39;expression NeuroD dans une vari&#xE9;t&#xE9; de tissus et aidera enqu&#xEA;ter sur les processus fondamentaux qui r&#xE9;gissent la r&#xE9;g&#xE9;n&#xE9;ration photor&#xE9;cepteur chez les adultes.

Utilisation de la Tg (BDNI: egfp) / Ligne poisson z&#xE8;bre albinos pour caract&#xE9;riser l&#x26;#39;expression in vivo de NeuroD

In dieser Studie verwendeten wir eine neu erstellte transgene Zebrafisch, Tg(nrd:egfp)/Albino, Ausdruck der Neurod in der Retina entwickeln und Erwachsenen weiter zu charakterisieren und Neurod Ausdruck beim Erwachsenen Photorezeptor regenerieren zu bestimmen. Wir bieten auch Beobachtungen &uuml;ber den Ausdruck der Neurod in einer Vielzahl von anderen Geweben. In dieser Zeile findet sich in den Zellen der Entwicklungs- und Erwachsenen Netzhaut, Zirbeldr&uuml;se, Kleinhirn, Bulbus olfactorius, Mittelhirn, Hinterhirn, Neuralrohr, Seitenlinienorgan, Innenohr, Bauchspeicheldr&uuml;se, Darm und Fin EGFP. Mit Immunhistochemie und in-situ-Hybridisierung, vergleichen wir den Ausdruck der Nrd:egfp-Transgene der endogenen Neurod und bekannten Netzhaut Zelltypen. In &Uuml;bereinstimmung mit fr&uuml;heren Daten basierend auf in-situ-Hybridisierung, wir zeigen, dass w&auml;hrend der retinalen Entwicklung der Nrd:egfp-Transgene dr&uuml;ckt sich nicht in proliferierenden retinalen Neuroepithelium und dr&uuml;ckt sich in einer Teilmenge der Netzhaut Neuronen. Im Gegensatz zu fr&uuml;heren Untersuchungen ist Nrd:egfp in allen Stab Photorezeptoren allm&auml;hlich re-expressed. W&auml;hrend der Photorezeptoren Regenerierung in Erwachsenen Zebrafisch offenbart in situ Hybridisierung, Neurod in M&uuml;ller Gliazellen abgeleitete neuronalen Vorl&auml;uferzellen nicht angegeben, jedoch wird in Photorezeptoren Vorl&auml;uferzellen ausgedr&uuml;ckt, wie sie auf die nukleare Au&szlig;enschicht migrieren und differenzieren sich in neuen Stab Photorezeptoren. W&auml;hrend der Photorezeptoren Regenerierung entspricht Ausdruck der Nrd:egfp dem von Neurod. Wir schlie&szlig;en, dass Tg(nrd:egfp)/Albino eine gute Darstellung der endogenen Neurod Ausdruck ist, ein n&uuml;tzliches Werkzeug ist, Neurod Ausdruck in einer Vielzahl von Geweben zu visualisieren und wird helfen, die grundlegenden Prozesse &uuml;ber den Photorezeptoren Regenerierung bei Erwachsenen untersucht.

Benutzung der Tg(nrd:egfp)/Albino-Zebrafisch-Line in-vivo Ausdruck der Neurod charakterisieren.

In questo studio, abbiamo usato zebrafish transgenici appena creato, Tg(nrd:egfp)/albino, per caratterizzare ulteriormente l&#39;espressione di neurod nella retina in via di sviluppo e adulti e determinare neurod espressione durante la rigenerazione del fotorecettore adulto. Forniamo anche le osservazioni per quanto riguarda l&#39;espressione di neurod in una variet&agrave; di altri tessuti. In questa linea, EGFP si trova nelle cellule della retina in via di sviluppo e dell&#39;adulto, ghiandola pineale, cervelletto, bulbi olfattivi, mesencefalo, hindbrain, tubo neurale, linea laterale, orecchio interno, pancreas, intestino e pinna. Mediante immunoistochimica ed ibridazione in situ, confrontiamo l&#39;espressione del transgene nrd:egfp a quella di neurod endogeni e di tipi di cellule retiniche conosciuto. Coerente con i dati precedenti basati su ibridazioni in situ, ci mostrano che durante lo sviluppo della retina, il transgene nrd:egfp non &egrave; espressa nella proliferazione retinica neuroepithelium e si esprime in un subset di neuroni della retina. A differenza dei precedenti studi, &egrave; gradualmente riespresse in tutti i fotorecettori asta nrd:egfp. Durante la rigenerazione di fotorecettori in zebrafish adulto, ibridazione in situ rivela che neurod non &egrave; espressa in M&uuml;ller glial derivato progenitori neuronali, ma &egrave; espresso in progenitori fotorecettore come migrare verso lo strato nucleare esterno e differenziarsi in nuovi fotorecettori rod. Durante la rigenerazione del fotorecettore, espressione della nrd:egfp corrisponde a quello di neurod. Concludiamo che Tg(nrd:egfp)/albino &egrave; una buona rappresentazione dell&#39;espressione endogena neurod, &egrave; uno strumento utile per visualizzare neurod espressione in una variet&agrave; di tessuti e aiuteranno a indagare i processi fondamentali che regolano la rigenerazione fotorecettore negli adulti.

En este estudio, se utiliz&#xF3; una nueva creaci&#xF3;n de pez cebra transg&#xE9;nico, Tg (NRD: EGFP) / albino, para caracterizar mejor la expresi&#xF3;n de NeuroD en la retina en desarrollo y adultos y para determinar la expresi&#xF3;n NeuroD durante la regeneraci&#xF3;n de los fotorreceptores de adultos. Tambi&#xE9;n proporcionar observaciones relativas a la expresi&#xF3;n de NeuroD en una variedad de otros tejidos. En esta l&#xED;nea, EGFP se encuentra en las c&#xE9;lulas de la retina en desarrollo y el adulto, la gl&#xE1;ndula pineal, el cerebelo, bulbo olfatorio, mesenc&#xE9;falo, rombenc&#xE9;falo, el tubo neural, la l&#xED;nea lateral, el o&#xED;do interno, el p&#xE1;ncreas, el intestino, y la aleta. Durante la regeneraci&#xF3;n de los fotorreceptores en adultos de pez cebra, la hibridaci&#xF3;n in situ revela que NeuroD no se expresa en la gl&#xED;a de M&#xFC;ller derivados de progenitores neuronales, sino que se expresa en progenitores de fotorreceptores a medida que migran a la capa nuclear externa y se diferencian en nuevos fotorreceptores de los bastones. Zebrafish provide an attractive model to study the retinal response to photoreceptor apoptosis due to its remarkable ability to spontaneously regenerate retinal neurons following damage. There are currently two widely-used light-induced retinal degeneration models to damage photoreceptors in the adult zebrafish. One model uses constant bright light, whereas the other uses a short exposure to extremely intense ultraviolet light. Although both models are currently used, it is unclear whether they differ in regard to the extent of photoreceptor damage or the subsequent regeneration response. Here we report a thorough analysis of the photoreceptor damage and subsequent proliferation response elicited by each individual treatment, as well as by the concomitant use of both treatments. We show a differential loss of rod and cone photoreceptors with each treatment. Additionally, we show that the extent of proliferation observed in the retina directly correlates with the severity of photoreceptor loss. We also demonstrate that both the ventral and posterior regions of the retina are partially protected from light damage. Finally, we show that combining a short ultraviolet exposure followed by a constant bright light treatment largely eliminates the neuroprotected regions, resulting in widespread loss of rod and cone photoreceptors and a robust regenerative response throughout the retina.

Characterization of multiple light damage paradigms reveals regional differences in photoreceptor loss.

Poisson-z&#xE8;bre constituent un mod&#xE8;le int&#xE9;ressant pour &#xE9;tudier la r&#xE9;ponse r&#xE9;tinienne &#xE0; l&#x26;#39;apoptose des photor&#xE9;cepteurs en raison de sa remarquable capacit&#xE9; &#xE0; r&#xE9;g&#xE9;n&#xE9;rer spontan&#xE9;ment neurones de la r&#xE9;tine apr&#xE8;s des dommages. En outre, nous montrons que la mesure de la prolif&#xE9;ration observ&#xE9;e dans la r&#xE9;tine est directement corr&#xE9;l&#xE9;e avec la s&#xE9;v&#xE9;rit&#xE9; de la perte de photor&#xE9;cepteurs. Pez cebra ofrecer un modelo atractivo para estudiar la respuesta de la retina a la apoptosis de fotorreceptores, debido a su notable capacidad de regeneraci&#xF3;n espont&#xE1;nea neuronas de la retina despu&#xE9;s del da&#xF1;o. Actualmente hay dos ampliamente utilizados inducidos por la luz los modelos de degeneraci&#xF3;n de la retina a los fotorreceptores de da&#xF1;o en el pez cebra adulto. Un modelo utiliza una luz brillante constante, mientras que el otro utiliza una corta exposici&#xF3;n a la luz ultravioleta muy intensa. Aqu&#xED; se presenta un an&#xE1;lisis detallado de los da&#xF1;os fotorreceptores y la respuesta posterior proliferaci&#xF3;n inducida por cada tratamiento individual, as&#xED; como por el uso concomitante de ambos tratamientos. Nos muestran una p&#xE9;rdida diferencial de fotorreceptores conos y bastones con cada tratamiento. Adem&#xE1;s, nos muestran que el grado de proliferaci&#xF3;n observado en la retina se correlaciona directamente con la severidad de la p&#xE9;rdida de fotorreceptores. Tambi&#xE9;n demuestran que tanto las regiones ventral y posterior de la retina est&#xE1;n parcialmente protegidos de da&#xF1;os luz. Por &#xFA;ltimo, nos muestran que la combinaci&#xF3;n de una exposici&#xF3;n ultravioleta corta seguida de un tratamiento con luz brillante constante elimina en gran medida las regiones neuroprotected, resultando en la p&#xE9;rdida generalizada de la barra y los conos y una s&#xF3;lida respuesta regenerativa a trav&#xE9;s de la retina.

Caracterizaci&#xF3;n de m&#xFA;ltiples paradigmas de da&#xF1;os causados &#x200B;&#x200B;por la luz revela las diferencias regionales en la p&#xE9;rdida de fotorreceptores

A reversed phase gradient ultra performance liquid chromatography tandem mass spectrometry (UPLC/MS/MS) method has been developed for the analysis of smokeless powders. A total of 20 different components were separated by UPLC and detected by MS/MS in multiple reaction monitoring (MRM) mode. These compounds included diphenylamines, centralites, nitrotoluenes, nitroglycerin, and various phthalates. Simultaneous positive and negative electrospray ionization (ESI) was used along with negative atmospheric pressure chemical ionization (APCI) to detect all compounds in a single analysis. Analysis times were under 8 min with a gradient of 10-73% organic at a flow rate of 0.500 mL/min. With this method, ultraviolet and MRM limits of detection ranging from 0.08 to 2.6 ng and 0.4-64 ng injected were achieved. Commercially available smokeless powders were also extracted with methylene chloride and characterized using the developed UPLC/MS/MS method. The procedure permits the determination of compositional differences between different brands as well as lot-to-lot variations.
