A digital microfluidic (DMF) device was applied to a heterogeneous sandwich immunoassay. The digital approach to microfluidics manipulates samples and reagents in the form of discrete droplets, as opposed to the streams of fluid used in microchannels. Since droplets are manipulated on relatively generic 2-D arrays of electrodes, DMF devices are straightforward to use, and are reconfigurable for any desired combination of droplet operations. This flexibility makes them suitable for a wide range of applications, especially those requiring long, multistep protocols such as immunoassays. Here, we developed an immunoassay on a DMF device using Human IgG as a model analyte. To capture the analyte, an anti-IgG antibody was physisorbed on the hydrophobic surface of a DMF device, and DMF actuation was used for all washing and incubation steps. The bound analyte was detected using FITC-labeled anti-IgG, and fluorescence after the final wash was measured in a fluorescence plate reader. A non-ionic polymer surfactant, Pluronic F-127, was added to sample and detection antibody solutions to control non-specific binding and aid in movement via DMF. Sample and reagent volumes were reduced by nearly three orders of magnitude relative to conventional multiwell plate methods. Since droplets are in constant motion, the antibody-antigen binding kinetics is not limited by diffusion, and total analysis times were reduced to less than 2.5 h per assay. A multiplexed device comprising several DMF platforms wired in series further increased the throughput of the technique. A dynamic range of approximately one order of magnitude was achieved, with reproducibility similar to the assay when performed in a 96-well plate. In bovine serum samples spiked with human IgG, the target molecule was successfully detected in the presence of a 100-fold excess of bovine IgG. It was concluded that the digital microfluidic format is capable of carrying out qualitative and quantitative sandwich immunoassays with a dramatic reduction in reagent usage and analysis time compared to macroscale methods.

A digital microfluidic approach to heterogeneous immunoassays.

Depuis gouttelettes sont manipul&#xE9;s sur relativement g&#xE9;n&#xE9;riques en 2-D r&#xE9;seaux d&#x26;#39;&#xE9;lectrodes, dispositifs DMF sont simples &#xE0; utiliser, et sont reconfigurables pour toute combinaison souhait&#xE9;e des op&#xE9;rations de gouttelettes. Une approche num&#xE9;rique microfluidique pour immunoessais h&#xE9;t&#xE9;rog&#xE8;nes

Ein digitaler mikrofluidische (DMF) Ger&auml;t wurde auf eine heterogene Sandwich-Immunoassay angewendet. Der digitale Ansatz f&uuml;r Mikrofluidik manipuliert Proben und Reagenzien in Form von diskreten Tr&ouml;pfchen, im Gegensatz zu die Str&ouml;me der Fl&uuml;ssigkeit in Mikrokan&auml;len verwendet. Seit Tr&ouml;pfchen auf relativ generisch zweidimensionalen Arrays von Elektroden manipuliert werden, sind DMF-Ger&auml;te einfach zu bedienen, und sind f&uuml;r jede gew&uuml;nschte Kombination von Tr&ouml;pfchen Operationen rekonfigurierbare. Diese Flexibilit&auml;t eignen sie sich f&uuml;r eine Vielzahl von Anwendungen, insbesondere solche, die lange, multistep-Protokolle wie Immunoassays. Hier entwickelten wir ein Immunoassay auf einem DMF-Ger&auml;t mit Human IgG, als ein Modell-Analyt. Erfassen von Analyten, ein Anti-IgG Antik&ouml;rper wurde Physisorbed an der hydrophoben Oberfl&auml;che eines DMF-Ger&auml;ts, und DMF-Bet&auml;tigung f&uuml;r alle Wasch- und Inkubation Schritte verwendet wurde. Da Tr&ouml;pfchen in st&auml;ndiger Bewegung sind, die Antik&ouml;rper-Antigen-Bindung-Kinetik ist nicht begrenzt durch Diffusion und Gesamt-Analyse-Mal wurden auf weniger als 2,5 h pro Probe reduziert. Ein Multiplex-Ger&auml;t bestehend aus mehreren DMF-Plattformen, die in Serie weiter verdrahtet erh&ouml;ht den Durchsatz der Technik. Ein Dynamikumfang von ca. eine Gr&ouml;&szlig;enordnung wurde erreicht, mit Reproduzierbarkeit, die &auml;hnlich wie die Probe in einem 96-Well-Platte durchgef&uuml;hrt. In Rinderserum Beispielen gespickt mit menschlichen IgG wurde das Zielmolek&uuml;l erfolgreich in Anwesenheit einer hundertfachen &Uuml;berschuss an Rinder-IgG festgestellt. Es wurde beschlossen, dass das Format digital mikrofluidische Durchf&uuml;hrung von qualitativen und quantitativen Sandwich Immunoassays mit einem dramatischen R&uuml;ckgang der Reagenz-Nutzung und Analyse-Zeit im Vergleich zu makroskopischen Methoden kann.

In campioni di siero bovino a spillo con IgG umane, la molecola bersaglio &egrave; stato correttamente rilevata in presenza di un eccesso di 100 volte di IgG bovine. Si &egrave; concluso che il formato digitale microfluidica &egrave; in grado di effettuare immunodosaggi qualitativa e quantitativa il sandwich con una drastica riduzione nel tempo analisi e utilizzo di reagente rispetto ai metodi di macroscala.

Un digital de microfluidos (DMF) dispositivo se aplic&#xF3; a un inmunoensayo s&#xE1;ndwich heterog&#xE9;neo. El enfoque digital a microflu&#xED;dica manipula las muestras y reactivos en forma de gotitas discretas, a diferencia de las corrientes de fluido usados &#x200B;&#x200B;en microcanales. Dado que las gotitas son manipulados en relativamente gen&#xE9;ricos 2-D conjuntos de electrodos, los dispositivos de DMF son f&#xE1;ciles de usar, y son reconfigurables para cualquier combinaci&#xF3;n deseada de las operaciones de gotitas. Esta flexibilidad hace adecuadas para una amplia gama de aplicaciones, especialmente aquellas que requieren protocolos de largo, de varias etapas, tales como inmunoensayos. En este sentido, hemos desarrollado un inmunoensayo en un dispositivo de DMF con IgG humana como un analito modelo. Para capturar el analito, un anticuerpo anti-IgG se adsorbidas sobre la superficie hidr&#xF3;foba de un dispositivo de DMF, y DMF accionamiento se utiliz&#xF3; para todos los pasos de lavado y de incubaci&#xF3;n. El analito unido se detect&#xF3; utilizando marcado con FITC anti-IgG, y la fluorescencia despu&#xE9;s del lavado final se midi&#xF3; en un lector de placas de fluorescencia. Un tensioactivo no i&#xF3;nico pol&#xED;mero, Pluronic F-127, se a&#xF1;adi&#xF3; a las soluciones de muestra y la detecci&#xF3;n de anticuerpos para controlar la uni&#xF3;n no espec&#xED;fica y ayuda en movimiento a trav&#xE9;s de DMF. Vol&#xFA;menes de muestra y reactivo se redujeron en casi tres &#xF3;rdenes de magnitud con relaci&#xF3;n a los m&#xE9;todos convencionales de placa de pocillos m&#xFA;ltiples. Puesto que las gotas est&#xE1;n en constante movimiento, la cin&#xE9;tica de uni&#xF3;n ant&#xED;geno-anticuerpo no est&#xE1; limitada por difusi&#xF3;n, y los tiempos totales de an&#xE1;lisis se redujo a menos de 2,5 h por ensayo. Un dispositivo de multiplexado que comprende varias plataformas DMF conectados en serie aument&#xF3; a&#xFA;n m&#xE1;s el rendimiento de la t&#xE9;cnica. Un rango din&#xE1;mico de aproximadamente un orden de magnitud se logr&#xF3;, con una reproducibilidad similar al ensayo cuando se realiza en una placa de 96 pocillos. En las muestras de suero bovino enriquecidas con IgG humana, la mol&#xE9;cula diana se detect&#xF3; con &#xE9;xito en la presencia de un exceso de 100 veces de IgG bovino. Se concluy&#xF3; que el formato de microfluidos digital es capaz de llevar a cabo inmunoensayos s&#xE1;ndwich cualitativos y cuantitativos con una reducci&#xF3;n dram&#xE1;tica en el uso de reactivos y tiempo de an&#xE1;lisis en comparaci&#xF3;n con los m&#xE9;todos de macroescala.
