Cholesterinester-Ester-Transferprotein (CETP) vermittelt die net &Uuml;bertragung von Cholesterinestern aus Atheroprotective High-Density-Lipoproteine atherogenic Low-Density-Lipoproteine oder sehr-Low-Density-Lipoproteine. Hemmung der CETP erh&ouml;ht High-density-Lipoprotein-Cholesterin (das gute Cholesterin) Ebenen und reduziert low density Lipoprotein Cholesterin (schlechtes Cholesterin) Ebenen, so dass es viel versprechende Medikament Ziel f&uuml;r die Pr&auml;vention und Behandlung der koronaren Herzkrankheit. Obwohl die Kristallstruktur von CETP bestimmt wurde, ist die molekulare Mechanismen, die Vermittlung Cholesterinester Ester Transfer noch unbekannt, sogar die strukturellen Merkmale des CETP in einer physiologischen Umgebung weiter ausbleiben. Wir spielten Molekulardynamik-Simulationen zum strukturellen Merkmale der CETP in einer w&auml;ssrigen L&ouml;sung zu erforschen. Ergebnisse zeigen, dass die Flexibilit&auml;t der distalen Teil der N-terminale &beta;-Fass-Dom&auml;ne in L&ouml;sung als in Kristall erheblich h&ouml;her ist; umgekehrt ist die Flexibilit&auml;t der Helix X etwas weniger. W&auml;hrend der Simulation erweitert das distale Ende der C-terminale &beta;-Fass-Dom&auml;ne w&auml;hrend der hydrophile Oberfl&auml;che, die mehr als die Hydrophobe Oberfl&auml;che erh&ouml;hen. Dar&uuml;ber hinaus wurde eine neue Oberfl&auml;che Pore in dieser Dom&auml;ne generiert. Diese Oberfl&auml;che Pore und alle Hohlr&auml;ume CETP sind stabil. Diese Ergebnisse legen nahe, dass die Bildung eines kontinuierlichen Tunnels innerhalb CETP durch die Verbindung von Hohlr&auml;umen in L&ouml;sung gestattet ist. Cholesteryl ester transfer protein (CETP) mediates the net transfer of cholesteryl esters from atheroprotective high-density lipoproteins to atherogenic low-density lipoproteins or very-low-density lipoproteins. Inhibition of CETP raises high-density lipoprotein cholesterol (good cholesterol) levels and reduces low-density lipoprotein cholesterol (bad cholesterol) levels, making it a promising drug target for the prevention and treatment of coronary heart disease. Although the crystal structure of CETP has been determined, the molecular mechanism mediating cholesteryl esters transfer is still unknown, even the structural features of CETP in a physiological environment remain elusive. We performed molecular dynamics simulations to explore the structural features of CETP in an aqueous solution. Results show that the distal portion flexibility of N-terminal Î²-barrel domain is considerably greater in solution than in crystal; conversely, the flexibility of helix X is slightly less. During the simulations the distal end of C-terminal Î²-barrel domain expanded while the hydrophilic surface increasing more than the hydrophobic surface. In addition a new surface pore was generated in this domain. This surface pore and all cavities in CETP are stable. Structural features of cholesteryl ester transfer protein: A molecular dynamics simulation study.

Prot&eacute;ine de transfert CHOLESTERYL ester (CETP) intervient dans le transfert net des esters de cholest&eacute;rol des lipoprot&eacute;ines de haute densit&eacute; atheroprotective ath&eacute;rog&egrave;nes des lipoprot&eacute;ines de basse densit&eacute; ou lipoprot&eacute;ines de tr&egrave;s faible densit&eacute;. L&#39;inhibition de la CETP soul&egrave;ve HDL cholest&eacute;rol (bon cholest&eacute;rol) et r&eacute;duit le LDL cholest&eacute;rol (mauvais cholest&eacute;rol), ce qui en fait une cible de m&eacute;dicaments prometteurs pour la pr&eacute;vention et le traitement des maladies coronariennes. Les r&eacute;sultats montrent que la souplesse de la partie distale du domaine N-terminal de &beta;-baril est consid&eacute;rablement plus &eacute;lev&eacute;e dans la solution que dans le cristal ; &agrave; l&#39;inverse, la flexibilit&eacute; d&#39;h&eacute;lice X est l&eacute;g&egrave;rement inf&eacute;rieure. Proteina di trasferimento dell&#39;estere del colesterolo (CETP) media il trasferimento netto di esteri del colesterolo lipoproteine ad alta densit&agrave; atheroprotective aterogeno delle lipoproteine a bassa densit&agrave; o lipoproteine a densit&agrave; molto bassa. Inibizione di CETP genera livelli di lipoproteine ad alta densit&agrave; colesterolo (colesterolo buono) e riduce le lipoproteine a bassa densit&agrave; colesterolo (colesterolo cattivo), che lo rende un bersaglio promettente farmaco per la prevenzione e il trattamento della malattia coronarica. Anche se la struttura di cristallo del CETP &egrave; stata determinata, il meccanismo molecolare mediando il trasferimento di esteri del colesterolo &egrave; ancora sconosciuta, che anche le caratteristiche strutturali di CETP in un ambiente fisiologico rimangano inafferrabile. Abbiamo eseguito simulazioni di dinamica molecolare per esplorare le caratteristiche strutturali di CETP in soluzione acquosa. Risultati mostrano che la flessibilit&agrave; della porzione distale del &beta;-barilotto dominio N-terminale &egrave; notevolmente maggiore in soluzione che in cristallo; al contrario, la flessibilit&agrave; dell&#39;elica X &egrave; leggermente inferiore. Durante le simulazioni all&#39;estremit&agrave; distale del &beta;-barilotto dominio C-terminale espansa mentre la superficie idrofila aumentare di pi&ugrave; la superficie idrofoba. Prote&iacute;na de transferencia de &eacute;ster de colesterilo (CETP) interviene en la transferencia neta de &eacute;steres de colesterilo de lipoprote&iacute;nas de alta densidad ateroprotectores aterog&eacute;nicas de las lipoprote&iacute;nas de baja densidad o lipoprote&iacute;nas de muy baja densidad. Inhibici&oacute;n de la CETP eleva los niveles de lipoprote&iacute;na de alta densidad colesterol (colesterol bueno) y reduce los niveles de colesterol de lipoprote&iacute;na de baja densidad de (colesterol malo), lo que es un objetivo de drogas prometedoras para la prevenci&oacute;n y el tratamiento de la cardiopat&iacute;a coronaria. Aunque se ha determinado la estructura cristalina de CETP, el mecanismo molecular mediando transferencia de &eacute;steres de colesterilo sigue siendo desconocido, que incluso las caracter&iacute;sticas estructurales de CETP en un entorno fisiol&oacute;gico siendo esquivos. Realizamos simulaciones de din&aacute;mica molecular para explorar las caracter&iacute;sticas estructurales de CETP en una soluci&oacute;n acuosa. Los resultados muestran que la flexibilidad de la porci&oacute;n distal del dominio del &beta;-barril de N-terminal es considerablemente mayor en la soluci&oacute;n que en cristal; por el contrario, la flexibilidad de h&eacute;lice X es un poco menos. Durante las simulaciones del extremo distal del dominio del &beta;-barril c-terminal ampliado mientras la superficie hidrof&iacute;lica aumentando m&aacute;s la superficie. Adem&aacute;s se gener&oacute; un nuevo poro superficial en este dominio. Este poro superficial y todas las cavidades en CETP son estables. Caracter&iacute;sticas estructurales del &eacute;ster de colesterilo transferencia prote&iacute;na: un estudio de simulaci&oacute;n de din&aacute;mica molecular.

Peptides show much promise as potent and selective drug candidates. Fusing peptides to a scaffold monoclonal antibody produces a conjugated antibody which has the advantages of peptide activity yet also has the pharmacokinetics determined by the scaffold antibody. However, the conjugated antibody often has poor binding affinity to antigens that may be related to unknown structural changes. The study of the conformational change is difficult by conventional techniques because structural fluctuation under equilibrium results in multiple structures co-existing. Here, we employed our two recently developed electron microscopy (EM) techniques: optimized negative-staining (OpNS) EM and individual-particle electron tomography (IPET). Two-dimensional (2D) image analyses and three-dimensional (3D) maps have shown that the domains of antibodies present an elongated peptide-conjugated conformational change, suggesting that our EM techniques may be novel tools to monitor the structural conformation changes in heterogeneous and dynamic macromolecules, such as drug delivery vehicles after pharmacological synthesis and development.
