Caenorhabditis elegans TOM-1 is orthologous to vertebrate tomosyn, a cytosolic syntaxin-binding protein implicated in the modulation of both constitutive and regulated exocytosis. To investigate how TOM-1 regulates exocytosis of synaptic vesicles in vivo, we analyzed C. elegans tom-1 mutants. Our electrophysiological analysis indicates that evoked postsynaptic responses at tom-1 mutant synapses are prolonged leading to a two-fold increase in total charge transfer. The enhanced response in tom-1 mutants is not associated with any detectable changes in postsynaptic response kinetics, neuronal outgrowth, or synaptogenesis. However, at the ultrastructural level, we observe a concomitant increase in the number of plasma membrane-contacting vesicles in tom-1 mutant synapses, a phenotype reversed by neuronal expression of TOM-1. Priming defective unc-13 mutants show a dramatic reduction in plasma membrane-contacting vesicles, suggesting these vesicles largely represent the primed vesicle pool at the C. elegans neuromuscular junction. Consistent with this conclusion, hyperosmotic responses in tom-1 mutants are enhanced, indicating the primed vesicle pool is enhanced. Furthermore, the synaptic defects of unc-13 mutants are partially suppressed in tom-1 unc-13 double mutants. These data indicate that in the intact nervous system, TOM-1 negatively regulates synaptic vesicle priming.

Caenorhabditis elegans TOM-1 est orthologue &#xE0; vert&#xE9;br&#xE9;s tomosyn, un cytosolique syntaxine-binding protein impliqu&#xE9; dans la modulation de l&#x26;#39;exocytose &#xE0; la fois constitutive et r&#xE9;gul&#xE9;e. Pour &#xE9;tudier comment TOM-1 r&#xE9;gule l&#x26;#39;exocytose des v&#xE9;sicules synaptiques in vivo, nous avons analys&#xE9; C. elegans tam-1 mutants. Notre analyse indique que &#xE9;lectrophysiologique &#xE9;voqu&#xE9;s r&#xE9;ponses postsynaptiques &#xE0; Tom-1 synapses mutantes sont prolong&#xE9;s conduisant &#xE0; une augmentation de deux fois dans le transfert de charge totale. Cependant, au niveau ultrastructural, on observe une augmentation concomitante du nombre de la membrane plasmique en contact avec des v&#xE9;sicules dans Tom-1 synapses mutants, un ph&#xE9;notype invers&#xE9; par l&#x26;#39;expression neuronale de TOM-1. Amor&#xE7;age d&#xE9;fectueux-13 UNC mutants montrent une r&#xE9;duction spectaculaire de la membrane plasmique en contact avec des v&#xE9;sicules, sugg&#xE9;rant que ces v&#xE9;sicules repr&#xE9;sentent surtout la piscine v&#xE9;sicule amorc&#xE9;e &#xE0; la jonction neuromusculaire C. elegans. Conform&#xE9;ment &#xE0; cette conclusion, les r&#xE9;ponses hyperosmotiques dans Tom-1 mutants sont am&#xE9;lior&#xE9;es, ce qui indique la piscine v&#xE9;sicule amorc&#xE9;e est renforc&#xE9;e. En outre, les d&#xE9;fauts synaptiques de l&#x26;#39;UNC-13 mutants sont partiellement supprim&#xE9;e en tom-1 unc-13 doubles mutants. Ces donn&#xE9;es indiquent que dans le syst&#xE8;me nerveux intact, TOM-1 r&#xE9;gule n&#xE9;gativement l&#x26;#39;amor&#xE7;age des v&#xE9;sicules synaptiques.

Tomosyn Inhibe amor&#xE7;age des v&#xE9;sicules synaptiques chez Caenorhabditis elegans

Caenorhabditis Elegans TOM-1 ist Orthologous zu vertebrate Tomosyn, ein cytosolisches Syntaxin-verbindliches Protein in der Modulation der konstitutiven und die regulierten Exozytose verwickelt. Um zu untersuchen, wie TOM-1 regelt die Exozytose von in-vivo synaptische Vesikel, analysierten wir C. Elegans-Tom-1-Mutanten. Unsere elektrophysiologische Analyse zeigt, dass evozierte postsynaptisches Antworten auf Tom-1 mutant Synapsen zu einem zweifachen Anstieg Gesamtrechnungsbetrages Transfer verl&auml;ngert werden. Die verst&auml;rkte Reaktion in Tom-1 Mutanten ist nicht nachweisbaren &Auml;nderungen in Myasthenische Reaktion Kinetik, neuronale Auswuchs oder Synaptogenese zugeordnet. Jedoch auf ultrastrukturelle Ebene beobachten wir eine damit einhergehende Zunahme der Zahl der Plasmamembran-Kontaktaufnahme mit Bl&auml;schen in Tom-1 mutant Synapsen, einem Ph&auml;notyp umgekehrt durch neuronale Ausdruck von TOM-1. Grundierung defekte Unc-13 Mutanten zeigen eine drastische Reduzierung der Plasmamembran-Kontaktaufnahme mit Bl&auml;schen, was darauf hindeutet, dass diese Bl&auml;schen weitgehend den pr&auml;parierten Vesikel-Pool an der neuromuskul&auml;ren Synapse von C. Elegans darstellen. In &Uuml;bereinstimmung mit dieser Schlussfolgerung, sind Hyperosmotic Antworten im Tom-1 Mutanten verbessert, dass der pr&auml;parierte Vesikel-Pool noch verst&auml;rkt wird. Dar&uuml;ber hinaus werden die synaptische defekte Unc-13-Mutanten in Tom-1 Unc-13 Doppelzimmer Mutanten teilweise unterdr&uuml;ckt. Diese Daten zeigen, dass im intakten Nervensystem, TOM-1 negativ synaptische Vesikel Grundierung reguliert.

Caenorhabditis elegans TOM-1 &egrave; orthologous a vertebrati tomosyn, una citosolica Sintaxina-proteina implicata nella modulazione dell&#39;esocitosi costitutiva e regolata. Per indagare come TOM-1 regola esocitosi delle vescicole sinaptiche in vivo, abbiamo analizzato mutanti tom-1 c. elegans. La nostra analisi elettrofisiologica indicano che evocate postsinaptiche risposte a tom-1 mutante sinapsi sono prorogate portando ad un aumento di due volte nel trasferimento della carica totale. La risposta avanzata in tom-1 mutanti non &egrave; associata con eventuali cambiamenti rilevabili nella cinetica di risposta postsinaptica, escrescenza neuronale o pervasivi. Tuttavia, a livello ultrastrutturale, osserviamo un concomitante aumento del numero delle vescicole di membrana plasmatica-contattare nelle sinapsi mutante tom-1, un fenotipo invertito dall&#39;espressione neuronale di TOM-1. Innesco difettosi unc-13 mutanti mostrano una drastica riduzione nella membrana plasmatica contattando le vescicole, suggerendo che queste vescicole in gran parte rappresentano il pool di vescicola innescato alla giunzione neuromuscolare c. elegans. Coerente con questa conclusione, risposte iperosmotico tom-1 mutanti sono esaltate, indicando che piscina adescata vescicola &egrave; migliorata. Inoltre, i difetti sinaptici di unc-13 mutanti sono parzialmente soppressa nel tom-1 unc-13 doppi mutanti. Questi dati indicano che nel sistema nervoso intatto, TOM-1 regola negativamente innesco della vescicola sinaptica.

Caenorhabditis elegans TOM-1 es orthologous a la vertebraci&#xF3;n de tomosyn, una citos&#xF3;lica syntaxin vinculante prote&#xED;na implicada en la modulaci&#xF3;n de la exocitosis constitutiva y regulada. Para investigar c&#xF3;mo TOM-1 regula la exocitosis de las ves&#xED;culas sin&#xE1;pticas en vivo, se analizaron los C. elegans tom-1 mutantes. Nuestro an&#xE1;lisis indica que electrofisiol&#xF3;gico respuestas evocadas postsin&#xE1;pticos en el Tom-1 sinapsis mutantes se prolongan dando lugar a un aumento de dos veces en la transferencia de carga total. El aumento de la respuesta en el tom-1 mutantes no est&#xE1; asociado con cambios detectables en la cin&#xE9;tica de la respuesta postsin&#xE1;ptica neuronal, crecimiento, o la sinaptog&#xE9;nesis. Sin embargo, a nivel ultraestructural, se observa un incremento concomitante en el n&#xFA;mero de la membrana plasm&#xE1;tica de contacto con ves&#xED;culas en tom-1 sinapsis mutantes, un fenotipo neuronal revertido por expresi&#xF3;n de TOM-1. Cebado defectuosos unc-13 mutantes muestran una reducci&#xF3;n dram&#xE1;tica en la membrana plasm&#xE1;tica de contacto con las ves&#xED;culas, lo que sugiere estas ves&#xED;culas en gran medida representan la piscina ves&#xED;cula imprimado en la uni&#xF3;n neuromuscular C. elegans. The SNARE proteins, syntaxin, SNAP-25 and synaptobrevin form a tertiary complex essential for vesicle fusion. Proteins that influence SNARE complex assembly are therefore likely to be important regulators of fusion events. In this study we have focused on tomosyn, a highly conserved, neuronally enriched, syntaxin-binding protein that has been implicated in the regulation of vesicle exocytosis. To directly test the role of tomosyn in neurosecretion we analysed loss-of-function mutants in the single Caenorhabditis elegans tomosyn gene, tom-1. These mutants exhibit enhanced synaptic transmission based on electrophysiological analysis of neuromuscular junction activity. This phenotype is the result of increased synaptic vesicle priming. In addition, we present evidence that tom-1 mutants also exhibit enhanced peptide release from dense core vesicles. These results indicate that tomosyn negatively regulates secretion for both vesicle types, possibly through a common mechanism, interfering with SNARE complex formation, thereby inhibiting vesicle fusion.

Les prot&#xE9;ines SNARE, la syntaxine, SNAP-25 et synaptobr&#xE9;vine former un complexe tertiaire essentiel pour la fusion des v&#xE9;sicules. Les prot&#xE9;ines qui influencent assemblage complexe SNARE sont donc susceptibles d&#x26;#39;&#xEA;tre importants r&#xE9;gulateurs de la fusion des &#xE9;v&#xE9;nements. Dans cette &#xE9;tude nous nous sommes concentr&#xE9;s sur tomosyn, un tr&#xE8;s conserv&#xE9;e, enrichie neuronale, la syntaxine-binding protein qui a &#xE9;t&#xE9; impliqu&#xE9;e dans la r&#xE9;gulation de l&#x26;#39;exocytose des v&#xE9;sicules. Ce ph&#xE9;notype est le r&#xE9;sultat de l&#x26;#39;amor&#xE7;age des v&#xE9;sicules synaptiques accrue. En outre, nous pr&#xE9;sentons des preuves que Tom-1 mutants pr&#xE9;sentent &#xE9;galement la production de peptide renforc&#xE9;e &#xE0; partir de v&#xE9;sicules de base denses. Las prote&#xED;nas SNARE, syntaxin, SNAP-25 y sinaptobrevina formar un complejo terciario esencial para la fusi&#xF3;n de ves&#xED;culas. Las prote&#xED;nas que influyen en el conjunto complejo SNARE que por ello puedan ser reguladores importantes de los eventos de fusi&#xF3;n. En este estudio nos hemos centrado en tomosyn, altamente conservada, neuronalmente enriquecido, syntaxin prote&#xED;na de uni&#xF3;n que se ha implicado en la regulaci&#xF3;n de la exocitosis de ves&#xED;culas. Para probar directamente el papel de tomosyn en neurosecreci&#xF3;n se analiz&#xF3; la p&#xE9;rdida de la funci&#xF3;n de los mutantes en el Caenorhabditis elegans, un solo gen tomosyn, tom-1. Estos mutantes presentan una mayor transmisi&#xF3;n sin&#xE1;ptica basado en el an&#xE1;lisis electrofisiol&#xF3;gico de la actividad de la uni&#xF3;n neuromuscular. Este fenotipo es el resultado de un aumento de cebado ves&#xED;culas sin&#xE1;pticas. Adem&#xE1;s, se presentan pruebas de que Tom-1 mutantes tambi&#xE9;n muestran una mayor liberaci&#xF3;n de p&#xE9;ptidos de las ves&#xED;culas n&#xFA;cleo denso. Estos resultados indican que tomosyn regula negativamente la secreci&#xF3;n para ambos tipos de ves&#xED;culas, posiblemente a trav&#xE9;s de un mecanismo com&#xFA;n, interfiriendo con la formaci&#xF3;n de complejo SNARE, la fusi&#xF3;n de ves&#xED;culas inhibiendo con ello.

The syntaxin-interacting protein tomosyn is thought to be a key regulator of exocytosis, although its precise mechanism of action has yet to be elucidated. Here we examined the role of tomosyn in peptide secretion in Caenorhabditis elegans tomosyn (tom-1) mutants. Ultrastructural analysis of tom-1 mutants revealed a 50% reduction in presynaptic dense-core vesicles (DCVs) corresponding to enhanced neuropeptide release. Conversely, overexpression of TOM-1 led to an accumulation of DCVs. Together, these data provide the first in vivo evidence that TOM-1 negatively regulates DCV exocytosis. In C. elegans, neuropeptide release is promoted by the calcium-dependent activator protein for secretion (CAPS) homolog UNC-31. To test for a genetic interaction between tomosyn and CAPS, we generated tom-1;unc-31 double mutants. Loss of TOM-1 suppressed the behavioral, electrophysiological, and DCV ultrastructural phenotypes of unc-31 mutants, indicating that TOM-1 antagonizes UNC-31-dependent DCV release. Because unc-31 mutants exhibit synaptic transmission defects, we postulated that loss of DCV release in these mutants and the subsequent suppression by tom-1 mutants could simply reflect alterations in synaptic activity, rather than direct regulation of DCV release. To distinguish between these two possibilities, we analyzed C. elegans Rim mutants (unc-10), which have a comparable reduction in synaptic transmission to unc-31 mutants, specifically attributed to defects in synaptic vesicle (SV) exocytosis. Based on this analysis, we conclude that the changes in DCV release in tom-1 and unc-31 mutants reflect direct effects of TOM-1 and UNC-31 on DCV exocytosis, rather than altered SV release.

Cells normally grow to a certain size before they enter mitosis and divide. Entry into mitosis depends on the activity of Cdk1, which is inhibited by the Wee1 kinase and activated by the Cdc25 phosphatase. However, how cells sense their size for mitotic commitment remains unknown. Here we show that an intracellular gradient of the dual-specificity tyrosine-phosphorylation regulated kinase (DYRK) Pom1, which emanates from the ends of rod-shaped Schizosaccharomyces pombe cells, serves to measure cell length and control mitotic entry. Pom1 provides positional information both for polarized growth and to inhibit cell division at cell ends. We discovered that Pom1 is also a dose-dependent G2-M inhibitor. Genetic analyses indicate that Pom1 negatively regulates Cdr1 and Cdr2, two previously described Wee1 inhibitors of the SAD kinase family. This inhibition may be direct, because in vivo and in vitro evidence suggest that Pom1 phosphorylates Cdr2. Whereas Cdr1 and Cdr2 localize to a medial cortical region, Pom1 forms concentration gradients from cell tips that overlap with Cdr1 and Cdr2 in short cells, but not in long cells. Disturbing these Pom1 gradients leads to Cdr2 phosphorylation and imposes a G2 delay. In short cells, Pom1 prevents precocious M-phase entry, suggesting that the higher medial Pom1 levels inhibit Cdr2 and promote a G2 delay. Thus, gradients of Pom1 from cell ends provide a measure of cell length to regulate M-phase entry.

The assembly of SNARE complexes between syntaxin, SNAP-25 and synaptobrevin is required to prime synaptic vesicles for fusion. Since Munc18 and tomosyn compete for syntaxin interactions, the interplay between these proteins is predicted to be important in regulating synaptic transmission. We explored this possibility, by examining genetic interactions between C. elegans unc-18(Munc18), unc-64(syntaxin) and tom-1(tomosyn). We have previously demonstrated that unc-18 mutants have reduced synaptic transmission, whereas tom-1 mutants exhibit enhanced release. Here we show that the unc-18 mutant release defect is associated with loss of two morphologically distinct vesicle pools; those tethered within 25â€‰nm of the plasma membrane and those docked with the plasma membrane. In contrast, priming defective unc-13 mutants accumulate tethered vesicles, while docked vesicles are greatly reduced, indicating tethering is UNC-18-dependent and occurs in the absence of priming. C. elegans unc-64 mutants phenocopy unc-18 mutants, losing both tethered and docked vesicles, whereas overexpression of open syntaxin preferentially increases vesicle docking, suggesting UNC-18/closed syntaxin interactions are responsible for vesicle tethering. Given the competition between vertebrate tomosyn and Munc18, for syntaxin binding, we hypothesized that C. elegans TOM-1 may inhibit both UNC-18-dependent vesicle targeting steps. Consistent with this hypothesis, tom-1 mutants exhibit enhanced UNC-18 plasma membrane localization and a concomitant increase in both tethered and docked synaptic vesicles. Furthermore, in tom-1;unc-18 double mutants the docked, primed vesicle pool is preferentially rescued relative to unc-18 single mutants. Together these data provide evidence for the differential regulation of two vesicle targeting steps by UNC-18 and TOM-1 through competitive interactions with syntaxin.

In contrasto, difettosi mutanti unc-13 adescamento accumulano tethered vescicole, mentre ancorate le vescicole sono notevolmente ridotti, che indica il tethering &egrave; UNC-18-dipendente e si verifica in assenza di innesco. C. elegans unc-64 mutanti fenocopia unc-18 mutanti, perdendo sia legato e ancorato vescicole, mentre sovraespressione di Sintaxina aperto aumenta preferenzialmente vescicola docking, suggerendo Sintaxina UNC-18/chiuso interazioni sono responsabili della vescicola tethering. Data la concorrenza tra i vertebrati tomosyn e Munc18, per l&#39;associazione Sintaxina, abbiamo ipotizzato che il c. elegans TOM-1 possono inibire entrambi vescicola UNC-18-dipendente passi di targeting. Coerente con questa ipotesi, tom-1 mutanti exhibit enhanced UNC-18 membrana plasmatica localizzazione e un concomitante aumento di entrambi legato e ancorato le vescicole sinaptiche. El conjunto de los complejos SNARE entre syntaxin, SNAP-25 y sinaptobrevina est&#xE1; obligado a primeros ves&#xED;culas sin&#xE1;pticas para la fusi&#xF3;n. Desde Munc18 tomosyn y competir por las interacciones syntaxin, la interacci&#xF3;n entre estas prote&#xED;nas se prev&#xE9; que sea importante en la regulaci&#xF3;n de la transmisi&#xF3;n sin&#xE1;ptica. Hemos explorado esta posibilidad, mediante el examen de las interacciones gen&#xE9;ticas entre C. elegans unc-18 (Munc18), UNC-64 (sintaxina) y Tom-1 (tomosyn). Hemos demostrado anteriormente que la UNC-18 mutantes han reducido la transmisi&#xF3;n sin&#xE1;ptica, mientras que Tom-1 mutantes presentan una mayor liberaci&#xF3;n. Concentration gradients regulate many cell biological and developmental processes. In rod-shaped fission yeast cells, polar cortical gradients of the DYRK family kinase Pom1 couple cell length with mitotic commitment by inhibiting a mitotic inducer positioned at midcell. However, how Pom1 gradients are established is unknown. Here, we show that Tea4, which is normally deposited at cell tips by microtubules, is both necessary and, upon ectopic cortical localization, sufficient to recruit Pom1 to the cell cortex. Pom1 then moves laterally at the plasma membrane, which it binds through a basic region exhibiting direct lipid interaction. Pom1 autophosphorylates in this region to lower lipid affinity and promote membrane release. Tea4 triggers Pom1 plasma membrane association by promoting its dephosphorylation through the protein phosphatase 1 Dis2. We propose that local dephosphorylation induces Pom1 membrane association and nucleates a gradient shaped by the opposing actions of lateral diffusion and autophosphorylation-dependent membrane detachment.

A phosphorylation cycle shapes gradients of the DYRK family kinase Pom1 at the plasma membrane.

Odors are key sensory signals for social communication and food search in animals including insects. Drosophila melanogaster, is a powerful neurogenetic model commonly used to reveal molecular and cellular mechanisms involved in odorant detection. Males use olfaction together with other sensory modalities to find their mates. Here, we review known olfactory signals, their related olfactory receptors, and the corresponding neuronal architecture impacting courtship. OR67d receptor detects 11-cis-Vaccenyl Acetate (cVA), a male specific pheromone transferred to the female during copulation. Transferred cVA is able to reduce female attractiveness for other males after mating, and is also suspected to decrease male-male courtship. cVA can also serve as an aggregation signal, maybe through another OR. OR47b was shown to be activated by fly odors, and to enhance courtship depending on taste pheromones. IR84a detects phenylacetic acid (PAA) and phenylacetaldehyde (PA). These two odors are not pheromones produced by flies, but are present in various fly food sources. PAA enhances male courtship, acting as a food aphrodisiac. Drosophila males have thus developed complementary olfactory strategies to help them to select their mates.

Where and when cells divide are fundamental questions. In rod-shaped fission yeast cells, the DYRK-family kinase Pom1 is organized in concentration gradients from cell poles and controls cell division timing and positioning. Pom1 gradients restrict to mid-cell the SAD-like kinase Cdr2, which recruits Mid1/Anillin for medial division. Pom1 also delays mitotic commitment through Cdr2, which inhibits Wee1. Here, we describe quantitatively the distributions of cortical Pom1 and Cdr2. These reveal low profile overlap contrasting with previous whole-cell measurements and Cdr2 levels increase with cell elongation, raising the possibility that Pom1 regulates mitotic commitment by controlling Cdr2 medial levels. However, we show that distinct thresholds of Pom1 activity define the timing and positioning of division. Three conditions-a separation-of-function Pom1 allele, partial downregulation of Pom1 activity, and haploinsufficiency in diploid cells-yield cells that divide early, similar to pom1 deletion, but medially, like wild-type cells. In these cells, Cdr2 is localized correctly at mid-cell. Further, Cdr2 overexpression promotes precocious mitosis only in absence of Pom1. Thus, Pom1 inhibits Cdr2 for mitotic commitment independently of regulating its localization or cortical levels. Indeed, we show Pom1 restricts Cdr2 activity through phosphorylation of a C-terminal self-inhibitory tail. In summary, our results demonstrate that distinct levels in Pom1 gradients delineate a medial Cdr2 domain, for cell division placement, and control its activity, for mitotic commitment.
