A long-standing problem in development is understanding how progenitor cells transiently expressing genes contribute to complex anatomical and functional structures. In the developing nervous system an additional level of complexity arises when considering how cells of distinct lineages relate to newly established neural circuits. To address these problems, we used both cumulative marking with Cre/loxP and Genetic Inducible Fate Mapping (GIFM), which permanently and heritably marks small populations of progenitors and their descendants with fine temporal control using CreER/loxP. A key component used in both approaches is a conditional phenotyping allele that has the potential to be expressed in all cell types, but is quiescent because of a loxP flanked Stop sequence, which precedes a reporter allele. Upon recombination, the resulting phenotyping allele is 'turned on' and then constitutively expressed. Thus, the reporter functions as a high fidelity genetic lineage tracer in vivo. Currently there is an array of reporter alleles that can be used in marking strategies, but their recombination efficiency and applicability to a wide array of tissues has not been thoroughly described. To assess the recombination/marking potential of the reporters, we utilized CreER(T) under the control of a Wnt1 transgene (Wnt1-CreER(T)) as well as a cumulative, non-inducible En1(Cre) knock-in line in combination with three different reporters: R26R (LacZ reporter), Z/EG (EGFP reporter), and Tau-Lox-STOP-Lox-mGFP-IRES-NLS-LacZ (membrane-targeted GFP/nuclear LacZ reporter). We marked the Wnt1 lineage using each of the three reporters at embryonic day (E) 8.5 followed by analysis at E10.0, E12.5, and in the adult. We also compared cumulative marking of cells with a history of En1 expression at the same stages. We evaluated the reporters by whole-mount and section analysis and ascertained the strengths and weaknesses of each of the reporters. Comparative analysis with the reporters elucidated complexities of how the Wnt1 and En1 lineages contribute to developing embryos and to axonal projection patterns of neurons derived from these lineages.

Un probl&#xE8;me de longue date dans le d&#xE9;veloppement est de comprendre comment les cellules prog&#xE9;nitrices de mani&#xE8;re transitoire des g&#xE8;nes exprimant contribuer &#xE0; des structures anatomiques complexes et fonctionnel. Dans le syst&#xE8;me nerveux en d&#xE9;veloppement un niveau suppl&#xE9;mentaire de complexit&#xE9; se pose lorsque l&#x26;#39;on consid&#xE8;re comment les cellules de lign&#xE9;es distinctes se rapportent &#xE0; nouvellement cr&#xE9;&#xE9;es circuits neuronaux. Pour r&#xE9;soudre ces probl&#xE8;mes, nous avons utilis&#xE9; &#xE0; la fois cumulatif marquage avec Cre / loxP et cartographie g&#xE9;n&#xE9;tique destin inductible (GIFM), qui marque de fa&#xE7;on permanente et heritably de petites populations de prog&#xE9;niteurs et de leurs descendants avec un contr&#xF4;le temporel pr&#xE9;cis avec CREER / loxP. Nous avons marqu&#xE9; la lign&#xE9;e Wnt1 en utilisant chacun des trois journalistes &#xE0; jour embryonnaire (E) 8,5 suivie d&#x26;#39;une analyse &#xE0; E10.0, E12.5, et chez l&#x26;#39;adulte. Analyse comparative des all&#xE8;les Reporter conditionnelles dans le d&#xE9;veloppement de l&#x26;#39;embryon et embryonnaire du syst&#xE8;me nerveux

Ein anhaltendes Problem in der Entwicklung ist verstehen, wie Vorl&auml;uferzellen, die vor&uuml;bergehend Ausdr&uuml;cken Gene zu komplexen anatomischen und funktionalen Strukturen beitragen. In das sich entwickelnde Nervensystem entsteht ein zus&auml;tzliches Ma&szlig; an Komplexit&auml;t, wenn unter Ber&uuml;cksichtigung der Zellen unterschiedliche Linien neu Beziehung zu neuronale Schaltkreise gegr&uuml;ndet. Um diese Probleme zu beheben, haben wir beide kumulative Kennzeichnung mit Cre/LoxP und genetische Inducible Schicksal Mapping (GIFM), die dauerhaft und heritably kleine Populationen der Vorfahren und ihre Nachkommen mit feinen zeitliche Steuerung mit CreER/LoxP Marken verwendet. Eine Schl&uuml;sselkomponente in beide Ans&auml;tze verwendet ist eine bedingte pathologies-Allel, die das Potenzial hat, alle Zelltypen ausgedr&uuml;ckt werden, aber ruhige aufgrund einer LoxP flankiert Stop-Sequenz, die ein Reporter-Allel vorausgeht. Bei der Rekombination ist die resultierende pathologies Allel &#39;eingeschaltet&#39; und dann konstitutiv ausgedr&uuml;ckt. Hier markieren wir die Wnt1-Linie, die mit den einzelnen der drei Reporter bei embryonalen Tag (E) 8.5 mit anschlie&szlig;ender Analyse auf E10.0, E12.5, und in den Erwachsenen. Wir verglichen auch kumulative Markierung von Zellen mit einer Geschichte von En1 Ausdruck in den gleichen Phasen. Wir die Reporter von ganz-Mount und Abschnitt Analyse ausgewertet und festgestellt, die St&auml;rken und Schw&auml;chen der einzelnen Reportern. Un annoso problema in via di sviluppo &egrave; capire come le cellule progenitrici transitoriamente espressione dei geni contribuiscono a complesse strutture anatomiche e funzionali. Abbiamo segnato il lignaggio di Wnt1 con ciascuno dei tre giornalisti a giorno embrionale (E) 8.5 seguita da analisi a E10.0, E12.5 e nell&#39;adulto. Abbiamo confrontato anche cumulativo marcatura delle cellule con una storia di En1 espressione presso le stesse fasi. Abbiamo valutato i reporter dall&#39;intero-monta e sezione analisi e accertata la forza e debolezza di ciascuno dei giornalisti. Analisi comparativa con la complessit&agrave; chiarito ai giornalisti di come i lignaggi Wnt1 ed En1 contribuiscono allo sviluppo di embrioni e di modelli di proiezione assonali dei neuroni derivati da questi lignaggi.

En el sistema nervioso en desarrollo de un nivel adicional de complejidad surge cuando se considera c&#xF3;mo las c&#xE9;lulas de diferentes linajes se refieren a los reci&#xE9;n establecidos los circuitos neuronales. Para solucionar estos problemas, hemos utilizado tanto acumulado marcado con el Cre / loxP y mapeo gen&#xE9;tico inducible Destino (GIFM), que marca de forma permanente y heritably peque&#xF1;as poblaciones de progenitores y sus descendientes, con un control preciso temporal usando CREER / loxP. Un componente clave utilizada en ambos enfoques es un alelo fenotipado condicional que tiene el potencial de ser expresado en todos los tipos celulares, pero est&#xE1; en reposo a causa de un loxP flanqueado secuencia de parada, que precede a un alelo reportero. Para evaluar la recombinaci&#xF3;n / marcado potencial de los reporteros, hemos utilizado CREER (T) bajo el control de un transg&#xE9;n Wnt1 (Wnt1-CREER (T)), as&#xED; como un acumulativo, no inducible En1 (Cre) en cadena en l&#xED;nea en combinaci&#xF3;n con tres reporteros diferentes: R26R (lacZ reportero), Z / EG (EGFP reportero), y Tau-Lox-STOP-Lox-mGFP-IRES-NLS-LacZ (membrana orientada GFP / nuclear reportero LacZ). Se marc&#xF3; el linaje Wnt1 utilizando cada uno de los tres reporteros en d&#xED;a embrionario (E) 8,5 seguido por an&#xE1;lisis en E10.0, E12.5, y en el adulto. Asimismo, en comparaci&#xF3;n acumulada marcado de las c&#xE9;lulas con una historia de En1 expresi&#xF3;n en las mismas etapas. Se evalu&#xF3; a los periodistas por todo el montaje y el an&#xE1;lisis de la secci&#xF3;n y comprobar las fortalezas y debilidades de cada uno de los periodistas. El an&#xE1;lisis comparativo con los periodistas dilucidado complejidades de c&#xF3;mo los Wnt1 y en1 linajes contribuir a los embriones en desarrollo y los patrones de proyecci&#xF3;n axonal de las neuronas derivadas de estos linajes.

Conditional marking and gene inactivation experiments are valuable approaches used to understand developmental and molecular mechanisms. CreER(T) is a fundamental component in recombinase-based conditional strategies and is used to gain temporal control subsequent to tamoxifen administration. We tested the hypothesis that tamoxifen dose linearly correlates with recombination efficiency in vivo. Wnt1-CreER(T) and tamoxifen administration were used to mark progenitors that contributed to the trigeminal ganglia. We executed a dose response study to determine the number of neurons that had undergone recombination in response to tamoxifen administered at doses ranging from 50 to 500 mg/kg. Our findings show a substantial variability in the amount of recombination within and between dose groups with no clear correlation between tamoxifen dose and the number of marked cells. This is the first study that we are aware of in which cell counts, robust quantitative data, and statistical analyses were performed on sections obtained from embryos marked in response to a wide range of tamoxifen dose in vivo. We provide an important quantitative and statistical framework for designing CreER(T)-based experiments and choosing tamoxifen dosing paradigms.
