Regulation of corticotropin-releasing hormone (CRH) activity is critical for the animal's adaptation to stressful challenges, and its dysregulation is associated with psychiatric disorders in humans. However, the molecular mechanism underlying this transcriptional response to stress is not well understood. Using various stress paradigms in mouse and zebrafish, we show that the hypothalamic transcription factor Orthopedia modulates the expression of CRH as well as the splicing factor Ataxin 2-Binding Protein-1 (A2BP1/Rbfox-1). We further show that the G protein coupled receptor PAC1, which is a known A2BP1/Rbfox-1 splicing target and an important mediator of CRH activity, is alternatively spliced in response to a stressful challenge. The generation of PAC1-hop messenger RNA isoform by alternative splicing is required for termination of CRH transcription, normal activation of the hypothalamic-pituitary-adrenal axis and adaptive anxiety-like behavior. Our study identifies an evolutionarily conserved biochemical pathway that modulates the neuronal adaptation to stress throughÂ transcriptional activation and alternative splicing.

R&#xE8;glement de la corticotropine-releasing hormone (CRH) l&#x26;#39;activit&#xE9; est critique pour l&#x26;#39;adaptation de l&#x26;#39;animal &#xE0; des d&#xE9;fis de stress, et sa d&#xE9;r&#xE9;gulation est associ&#xE9;e &#xE0; des troubles psychiatriques chez les humains. Cependant, le m&#xE9;canisme mol&#xE9;culaire qui sous-tend cette r&#xE9;ponse transcriptionnelle au stress n&#x26;#39;est pas bien comprise. Nous montrons en outre que le r&#xE9;cepteur coupl&#xE9; aux prot&#xE9;ines G PAC1, qui est une cible connue A2BP1/Rbfox-1 &#xE9;pissage et un m&#xE9;diateur important de l&#x26;#39;activit&#xE9; CRH, est un &#xE9;pissage alternatif en r&#xE9;ponse &#xE0; un d&#xE9;fi stressant. Regelung der Corticotropin-releasing Hormon (CRH) T&auml;tigkeit ist entscheidend f&uuml;r das Tier Anpassung an stressigen Herausforderungen, und seine Dysregulation psychiatrische Erkrankungen beim Menschen zugeordnet ist. Die molekulare Mechanismen, die hinter diesem transkriptionelle Reaktion auf stress ist jedoch nicht gut verstanden. Mit verschiedenen Druck-Paradigmen in Maus und Zebrafisch, zeigen wir, dass der Hypothalamus &Uuml;bertragungfaktor Orthopedia Ausdruck des CRH sowie der Splei&szlig;en Faktor Ataxin 2-Binding Protein-1 (A2BP1/Rbfox-1) moduliert. Wir zeigen weiter, dass der G-Protein gekoppelt-Rezeptor PAC1, der ein bekannter A2BP1/Rbfox-1-Splei&szlig;en-Ziel und ein wichtiger Vermittler der CRH-Aktivit&auml;t ist, als Reaktion auf eine stressige Herausforderung Alternativ gesplei&szlig;t wird. Die Generation der PAC1-Hop Boten-RNA-Isoform durch alternatives Splei&szlig;en ist f&uuml;r Beendigung des CRH-Transkription, normale Aktivierung der Hypothalamus-Hypophysen-Nebennieren-Achse und adaptive Angst-&auml;hnliches Verhalten erforderlich. Unsere Studie zeigt eine evolution&auml;r konservierte biochemischen Stoffwechselweg, der die Verz&ouml;gerung die neuronale Anpassung durch transkriptionelle Aktivierung und alternatives Splei&szlig;en betonen.

Sin embargo, el mecanismo molecular que subyace a esta respuesta transcripcional a estr&#xE9;s no se entiende bien. Utilizando diversos paradigmas de estr&#xE9;s en el rat&#xF3;n y el pez cebra, se muestra que el factor de transcripci&#xF3;n Ortopedia hipot&#xE1;lamo modula la expresi&#xF3;n de CRH, as&#xED; como el factor de empalme ataxina 2 prote&#xED;na de uni&#xF3;n-1 (A2BP1/Rbfox-1). Adem&#xE1;s, muestran que la prote&#xED;na G del receptor acoplado PAC1, que es un conocido A2BP1/Rbfox-1 objetivo de empalme y un mediador importante de la actividad CRH, es empalmados alternativamente en respuesta a un reto estresante. La generaci&#xF3;n del mensajero de la isoforma PAC1-hop ARN por splicing alternativo se requiere para la terminaci&#xF3;n de la transcripci&#xF3;n de la CRH, la activaci&#xF3;n normal del eje hipotal&#xE1;mico-pituitario-adrenal y la conducta adaptativa, como la ansiedad. Nuestro estudio identifica una v&#xED;a bioqu&#xED;mica conservadas evolutivamente que modula la adaptaci&#xF3;n neuronal al estr&#xE9;s mediante la activaci&#xF3;n de la transcripci&#xF3;n y splicing alternativo.
