Usher syndrome 3A (USH3A) is an autosomal recessive disorder characterized by progressive loss of hearing and vision due to mutation in the clarin-1 (CLRN1) gene. Lack of an animal model has hindered our ability to understand the function of CLRN1 and the pathophysiology associated with USH3A. Here we report for the first time a mouse model for ear disease in USH3A. Detailed evaluation of inner ear phenotype in the Clrn1 knockout mouse (Clrn1(-/-)) coupled with expression pattern of Clrn1 in the inner ear are presented here. Clrn1 was expressed as early as embryonic day 16.5 in the auditory and vestibular hair cells and associated ganglionic neurons, with its expression being higher in outer hair cells (OHCs) than inner hair cells. Clrn1(-/-) mice showed early onset hearing loss that rapidly progressed to severe levels. Two to three weeks after birth (P14-P21), Clrn1(-/-) mice showed elevated auditory-evoked brainstem response (ABR) thresholds and prolonged peak and interpeak latencies. By P21, approximately 70% of Clrn1(-/-) mice had no detectable ABR and by P30 these mice were deaf. Distortion product otoacoustic emissions were not recordable from Clrn1(-/-) mice. Vestibular function in Clrn1(-/-) mice mirrored the cochlear phenotype, although it deteriorated more gradually than cochlear function. Disorganization of OHC stereocilia was seen as early as P2 and by P21 OHC loss was observed. In sum, hair cell dysfunction and prolonged peak latencies in vestibular and cochlear evoked potentials in Clrn1(-/-) mice strongly indicate that Clrn1 is necessary for hair cell function and associated neural activation.

L&#x26;#39;absence d&#x26;#39;un mod&#xE8;le animal a entrav&#xE9; notre capacit&#xE9; &#xE0; comprendre la fonction de CLRN1 et la physiopathologie associ&#xE9;e &#xE0; USH3A. Nous rapportons ici pour la premi&#xE8;re fois un mod&#xE8;le de souris pour la maladie de l&#x26;#39;oreille dans USH3A. Deux &#xE0; trois semaines apr&#xE8;s la naissance (P14-P21), les Clrn1 (- / -) des souris a montr&#xE9; &#xE9;lev&#xE9;es auditif &#xE9;voqu&#xE9; du tronc c&#xE9;r&#xE9;bral (ABR) et seuils de pointe prolong&#xE9;e et les latences Interpeak. Par P21, environ 70% de Clrn1 (- / -) chez la souris n&#x26;#39;a pas eu ABR d&#xE9;tectable et par P30 ces souris &#xE9;taient sourds. Les &#xE9;missions de produits de distorsion oto n&#x26;#39;&#xE9;taient pas enregistrable &#xE0; partir Clrn1 (- / -) chez la souris. La fonction vestibulaire dans Clrn1 (- / -) souris refl&#xE8;te le ph&#xE9;notype cochl&#xE9;aire, m&#xEA;me si elle s&#x26;#39;est d&#xE9;t&#xE9;rior&#xE9;e plus graduellement que la fonction cochl&#xE9;aire. D&#xE9;sorganisation des OHC st&#xE9;r&#xE9;ocils a &#xE9;t&#xE9; observ&#xE9;e d&#xE8;s que P2 et P21 par la perte OHC a &#xE9;t&#xE9; observ&#xE9;e. En somme, le dysfonctionnement des cellules cili&#xE9;es et prolong&#xE9;es latences des pics en vestibulaire et cochl&#xE9;aire des potentiels &#xE9;voqu&#xE9;s dans Clrn1 (- / -) chez la souris indiquent fortement que Clrn1 est n&#xE9;cessaire pour la fonction des cellules cili&#xE9;es et associ&#xE9; activation neuronale.

Usher Syndrom 3A (USH3A) ist eine autosomal rezessive Erkrankung mit progressiven Verlust der H&ouml;rf&auml;higkeit und Vision durch Genmutation Clarin-1 (CLRN1). Mangel an einem Tiermodell hat unsere F&auml;higkeit zu verstehen, die Funktion des CLRN1 und der Pathophysiologie zugeordnete USH3A behindert. Hier berichten wir zum ersten Mal einem Mausmodell f&uuml;r Ohr-Krankheit in USH3A. Detaillierte Auswertung der Innenohr-Ph&auml;notyp in der Clrn1-Knockout-Maus (Clrn1(-/-)) gekoppelt mit Muster f&uuml;r den Ausdruck des Clrn1 im Innenohr werden hier vorgestellt. Clrn1 war so fr&uuml;h wie embryonale Tag 16,5 im auditorischen und vestibul&auml;ren Haarzellen und damit verbundenen ganglionic Neuronen, mit seinen Ausdruck, die h&ouml;her sind als innere Haarzellen in &auml;u&szlig;eren Haarzellen (elektrische) ausgedr&uuml;ckt. Clrn1(-/-) M&auml;usen zeigte fr&uuml;h beginnende H&ouml;rverlust, die rasch zu schweren Ebenen Motiven. Zwei bis drei Wochen nach der Geburt (P14-P21), Clrn1(-/-) M&auml;use zeigte erh&ouml;hte auditive hervorgerufen Hirnstamm Response (ABR) Schwellen und l&auml;ngere Spitze und interpeak Wartezeiten. Von P21 ungef&auml;hr 70 % des Clrn1(-/-) M&auml;use hatte keine nachweisbaren ABR und P30 diese M&auml;use waren taub. Verzerrung Produkt Otoakustische Emissionen waren nicht beschreibbare Clrn1(-/-) M&auml;use. Vestibul&auml;ren Funktion bei Clrn1(-/-) M&auml;usen gespiegelt der Cochlea Ph&auml;notyp, obwohl es als Cochlea Funktion verschlechtert. Desorganisation der OHC Stereozilien wurde gesehen, wie fr&uuml;h als P2 und von P21 OHC Verlust beobachtet wurde. Zusammenfassend l&auml;sst sich sagen zufolge Haarzelle Dysfunktion und verl&auml;ngerte Spitze Latenzen im vestibul&auml;ren und cochlear evozierte Potentiale bei Clrn1(-/-) M&auml;usen stark Clrn1 f&uuml;r Haarzelle Funktion und damit verbundenen neuronalen Aktivierung notwendig ist.

3A la sindrome di Usher (USH3A) &egrave; una malattia autosomica recessiva caratterizzata dalla progressiva perdita di udito e visione dovuta a mutazione del gene clarin-1 (CLRN1). Mancanza di un modello animale ha ostacolato la nostra capacit&agrave; di comprendere la funzione di CLRN1 e fisiopatologia associato USH3A. Riportiamo per la prima volta un modello del mouse per la malattia dell&#39;orecchio in USH3A. Valutazione dettagliata del fenotipo dell&#39;orecchio interno nel topo knockout Clrn1 (Clrn1(-/-)) accoppiato con pattern di espressione di Clrn1 nell&#39;orecchio interno sono presentati qui. Clrn1 &egrave; stata espressa fin dal giorno embrionale 16,5 nelle cellule ciliate uditive e vestibolari e neuroni ganglionica associati, con la sua espressione in cellule ciliate esterne (OHC) superiore interne delle cellule dei capelli. Clrn1(-/-) topi ha mostrato precoce perdita di udito di esordio che ha progredito rapidamente a livelli gravi. Aqu&#xED; mostramos por primera vez un modelo de rat&#xF3;n para la enfermedad de la oreja en USH3A. Evaluaci&#xF3;n detallada del fenotipo del o&#xED;do interno en el rat&#xF3;n knock-out Clrn1 (Clrn1 (- / -)), junto con el patr&#xF3;n de expresi&#xF3;n de Clrn1 en el o&#xED;do interno se presentan aqu&#xED;.
