Biological oxygen measurements by phosphorescence quenching make use of exogenous phosphorescent probes, which are introduced directly into the medium of interest (e.g. blood or interstitial fluid) where they serve as molecular sensors for oxygen. The byproduct of the quenching reaction is singlet oxygen, a highly reactive species capable of damaging biological tissue. Consequently, potential probe phototoxicity is a concern for biological applications. Herein, we compared the ability of polyethyleneglycol (PEG)-coated Pd tetrabenzoporphyrin (PdTBP)-based dendritic nanoprobes of three successive generations to sensitize singlet oxygen. It was found that the size of the dendrimer has practically no effect on the singlet oxygen sensitization efficiency in spite of the strong attenuation of the triplet quenching rate with an increase in the dendrimer generation. This unexpected result is due to the fact that the lifetime of the PdTBP triplet state in the absence of oxygen increases with dendritic generation, thus compensating for the concomitant decrease in the rate of quenching. Nevertheless, in spite of their ability to sensitize singlet oxygen, the phosphorescent probes were found to be non-phototoxic when compared with the commonly used photodynamic drug Photofrin in a standard cell-survival assay. The lack of phototoxicity is presumably due to the inability of PEGylated probes to associate with cell surfaces and/or penetrate cellular membranes. In contrast, conventional photosensitizers bind to cell components and act by generating singlet oxygen inside or in the immediate vicinity of cellular organelles. Therefore, PEGylated dendritic probes are safe to use for tissue oxygen measurements as long as the light doses are less than or equal to those commonly employed in photodynamic therapy.

Evaluation of phototoxicity of dendritic porphyrin-based phosphorescent oxygen probes: an in vitro study.

Ce r&#xE9;sultat inattendu est d&#xFB; au fait que la dur&#xE9;e de vie de l&#x26;#39;&#xE9;tat triplet PdTBP en l&#x26;#39;absence de l&#x26;#39;oxyg&#xE8;ne augmente avec la g&#xE9;n&#xE9;ration dendritique, compensant ainsi la diminution concomitante du taux de trempe. N&#xE9;anmoins, en d&#xE9;pit de leur capacit&#xE9; &#xE0; sensibiliser les l&#x26;#39;oxyg&#xE8;ne singulet, les sondes phosphorescentes ont &#xE9;t&#xE9; jug&#xE9;s non-phototoxique par rapport &#xE0; la Photofrin m&#xE9;dicament photodynamique couramment utilis&#xE9;s dans une norme cellulaire de survie dosage. Es wurde festgestellt, dass die Gr&ouml;&szlig;e der die Dendrimere hat praktisch keine Auswirkungen auf die Singulett-Sauerstoff-Sensibilisierung-Effizienz trotz der starken D&auml;mpfung des Triplets abschrecken Rate mit einer Zunahme der Dendrimere-Generation. Dennoch wurden trotz ihrer F&auml;higkeit, Singulett-Sauerstoff zu sensibilisieren, die phosphoreszierenden Sonden gefunden nicht phototoxisch im Vergleich mit der h&auml;ufig verwendeten photodynamische Droge Photofrin in einer Norm Zelle-&uuml;berleben-Assay. Der Mangel an Phototoxizit&auml;t ist vermutlich aufgrund der Unf&auml;higkeit zu ordnen Sie Zelle Oberfl&auml;chen bzw. Zellmembranen zu durchdringen pegyliertes Pr&uuml;fpunkte. We report the synthesis, calibration, and examples of application of two new phosphorescent probes, Oxyphor R4 and Oxyphor G4, optimized specifically for in vivo oxygen imaging by phosphorescence quenching. These "protected" dendritic probes can operate in either albumin-rich (blood plasma) or albumin-free (interstitial space) environments at all physiological oxygen concentrations, from normoxic to deep hypoxic conditions. Oxyphors R4 and G4 are derived from phosphorescent Pd-meso-tetra-(3,5-dicarboxyphenyl)-porphyrin (PdP) or Pd-meso-tetra-(3,5-dicarboxyphenyl)-tetrabenzoporphyrin (PdTBP), respectively, and possess features common for protected dendritic probes, i.e., hydrophobic dendritic encapsulation of phosphorescent metalloporphyrins and hydrophilic PEGylated periphery. The new Oxyphors are highly soluble in aqueous environments and do not permeate biological membranes. The probes were calibrated under physiological conditions (pH 6.4-7.8) and temperatures (22-38 Â°C), showing high stability, reproducibility of signals, and lack of interactions with biological solutes. Oxyphor G4 was used to dynamically image intravascular and interstitial oxygenation in murine tumors in vivo. The physiological relevance of the measurements was demonstrated by dynamically recording changes in tissue oxygenation during application of anesthesia (isofluorane). These experiments revealed that changes in isofluorane concentration significantly affect tissue oxygenation.

Se presenta la s&#xED;ntesis, la calibraci&#xF3;n, y ejemplos de aplicaci&#xF3;n de dos nuevas sondas fosforescentes, Oxyphor R4 y G4 Oxyphor, optimizados espec&#xED;ficamente para la formaci&#xF3;n de im&#xE1;genes in vivo de ox&#xED;geno por temple fosforescencia. Estos &#x26;quot;protegidos&#x26;quot; sondas dendr&#xED;ticas pueden operar en cualquiera de alb&#xFA;mina ricos (plasma sangu&#xED;neo) o (espacio intersticial) la alb&#xFA;mina de ambientes libres de ox&#xED;geno en todas las concentraciones fisiol&#xF3;gicas, de normoxic a las profundas condiciones de hipoxia. Oxyphors R4 y G4 se derivan de fosforescente Pd-meso-tetra-(3,5-dicarboxifenil)-porfirina (PDP) o Pd-meso-tetra-(3,5-dicarboxifenil)-tetrabenzoporphyrin (PdTBP), respectivamente, y poseen caracter&#xED;sticas comunes de las sondas protegidas dendr&#xED;ticas, es decir, la encapsulaci&#xF3;n dendr&#xED;ticas hidrof&#xF3;bica de metaloporfirinas fosforescentes y la periferia pegilado hidrof&#xED;lico. Los nuevos Oxyphors son altamente solubles en medios acuosos y no permeado membranas biol&#xF3;gicas. Las sondas fueron calibrados bajo condiciones fisiol&#xF3;gicas (pH 6.4 hasta 7.8) y temperaturas (22-38 &#xB0; C), que muestra una alta estabilidad, la reproducibilidad de las se&#xF1;ales, y la falta de interacciones con solutos biol&#xF3;gicos. Oxyphor G4 se utiliz&#xF3; para la imagen intravascular de forma din&#xE1;mica y la oxigenaci&#xF3;n intersticial en los tumores murinos in vivo. La importancia fisiol&#xF3;gica de las mediciones se ha demostrado de forma din&#xE1;mica el registro de cambios en la oxigenaci&#xF3;n de los tejidos durante la aplicaci&#xF3;n de la anestesia (isofluorano). Estos experimentos revelaron que los cambios en la concentraci&#xF3;n de isofluorano afectan significativamente oxigenaci&#xF3;n de los tejidos.
