The expression of RHAMM and other centrosome-associated genes are known to correlate with the extent of centrosome amplification in multiple myeloma, and with poor prognosis. RHAMM has a significant interaction with TPX2, a protein which regulates the localization and action of Aurora A kinase (AURKA) at the spindle poles. AURKA is known to be a central determinant of centrosome and spindle function and is a target for cancer therapy. Given these observations, we investigated the role of Aurora kinases as therapeutic targets in myeloma. Here we report that AURKA is expressed ubiquitously in myeloma, to varying degrees, in both cell lines and patients' bone marrow plasma cells. siRNA targeting AURKA induces apoptotic cell death in myeloma cell lines. The Aurora kinase inhibitor VE-465 also induces apoptosis and death in myeloma cell lines and primary myeloma plasma cells. The combination of VE-465 and dexamethasone improves cell killing compared with the use of either agent alone, even in cells resistant to the single agents. The phenotype of myeloma cells treated with VE-465 is consistent with published reports on the effects of Aurora kinase inhibition. Aurora kinase inhibitors should be pursued as potential treatments for myeloma.

Gene transcription is constrained by the nucleosomal nature of chromosomal DNA. This nucleosomal barrier is modulated by FACT, a conserved histone-binding heterodimer. FACT mediates transcription-linked nucleosome disassembly and also nucleosome reassembly in the wake of the RNA polymerase II transcription complex, and in this way maintains the repression of 'cryptic' promoters found within some genes. Here we focus on a novel mutant version of the yeast FACT subunit Spt16 that supplies essential Spt16 activities but impairs transcription-linked nucleosome reassembly in dominant fashion. This Spt16 mutant protein also has genetic effects that are recessive, which we used to show that certain Spt16 activities collaborate with histone acetylation and the activities of a Bur-kinase/Spt4-Spt5/Paf1C pathway that facilitate transcription elongation. These collaborating activities were opposed by the actions of Rpd3S, a histone deacetylase that restores a repressive chromatin environment in a transcription-linked manner. Spt16 activity paralleling that of HirC, a co-repressor of histone gene expression, was also found to be opposed by Rpd3S. Our findings suggest that Spt16, the Bur/Spt4-Spt5/Paf1C pathway, and normal histone abundance and/or stoichiometry, in mutually cooperative fashion, facilitate nucleosome disassembly during transcription elongation. The recessive nature of these effects of the mutant Spt16 protein on transcription-linked nucleosome disassembly, contrasted to its dominant negative effect on transcription-linked nucleosome reassembly, indicate that mutant FACT harbouring the mutant Spt16 protein competes poorly with normal FACT at the stage of transcription-linked nucleosome disassembly, but effectively with normal FACT for transcription-linked nucleosome reassembly. This functional difference is consistent with the idea that FACT association with the transcription elongation complex depends on nucleosome disassembly, and that the same FACT molecule that associates with an elongation complex through nucleosome disassembly is retained for reassembly of the same nucleosome.

ABSTRACT: BACKGROUND: Lung cancer is a common cause of cancer-related death. Staging typically includes positron emission tomography (PET) scanning, in which 18F-fluoro-2-dexoy-D-glucose (FDG) is taken up by cells proportional to metabolic activity, thus aiding in differentiating benign and malignant pulmonary nodules. Uptake of FDG can also occur in the abdomen. The clinical significance of incidental intraabdominal FDG uptake in the setting of pulmonary nodules is not well established. Our objective was to report on the clinical significance of incidental intra-abdominal FDG activity in the setting of lung cancer. METHODS: Fifteen hundred FDG-PET imaging studies performed for lung cancer were retrospectively reviewed for the presence of incidental FDG-positive intraabdominal findings. Patient charts with positive findings were then reviewed and information extracted. RESULTS: Twenty-five patients (25/1500) demonstrated incidental intraabdominal FDG uptake thought to be significant (1.7%) with a mean patient age of 71 years. Colonic uptake was most common (n = 17) with 9 (52%) being investigated further. Of these 9 cases, a diagnosis of malignancy was made in 3 patients, pre-malignant adenomas in 2 patients, a benign lipoma in 1 patient and no abnormal findings in the remaining patients. 8 patients were not investigated further (3 diagnosed with metastatic lung cancer and 2 were of advanced age) secondary to poor prognosis. CONCLUSION: Incidental abdominal findings in the colon on FDG-PET scan for work-up of pulmonary nodules need to be further investigated by colonoscopy.

R&eacute;sum&eacute; : Contexte : cancer du poumon est une cause fr&eacute;quente de d&eacute;c&egrave;s li&eacute;s au cancer. Mise en sc&egrave;ne g&eacute;n&eacute;ralement comprend des &eacute;missions positons (TEP) num&eacute;risation, dans laquelle 18F-fluoro-2-dexoy-D-glucose (FDG) est absorb&eacute; par les cellules proportionnelle &agrave; l&#39;activit&eacute; m&eacute;tabolique, facilitant ainsi pour diff&eacute;rencier les nodules pulmonaires b&eacute;nignes et malignes. L&#39;absorption du FDG peut &eacute;galement se produire dans l&#39;abdomen. La signification clinique de l&#39;absorption FDG intraabdominale accessoire dans le cadre de nodules pulmonaires n&#39;est pas bien &eacute;tablie. CONCLUSION : Des d&eacute;couvertes fortuites abdominales dans le c&ocirc;lon, le FDG-PET scan pour un travail de nodules pulmonaires il fallait &ecirc;tre davantage &eacute;tudi&eacute;e par coloscopie.

ABSTRACT: BACKGROUND: Lungenkrebs ist eine h&auml;ufige Ursache des Krebs-in Verbindung stehenden Todes. Inszenierung in der Regel umfasst die Positronen Emissions Tomographie (PET) Scannen, in denen 18F-fluoro-2-dexoy-D-glucose (FDG) von Zellen proportional zur metabolischen Aktivit&auml;t, so hilft in der Unterscheidung benigner und maligner pulmonaler Kn&ouml;tchen eingenommen wird. Aufnahme von FDG kann auch auftreten, in den Bauch. Die klinische Bedeutung der beil&auml;ufigen intraabdominal FDG-Aufnahme in der Einstellung der pulmonalen Kn&ouml;tchen ist nicht weit verbreitet. Unser Ziel war es, einen Bericht &uuml;ber die klinische Bedeutung der beil&auml;ufigen intraabdominellen FDG-Aktivit&auml;ten in der Umgebung von Lungenkrebs. Methoden: F&uuml;nfzehn hundert FDG-PET Untersuchungen f&uuml;r Lungenkrebs durchgef&uuml;hrt wurden nachtr&auml;glich auf das Vorhandensein von beil&auml;ufig FDG-positiven intraabdominal Erkenntnisse &uuml;berpr&uuml;ft. Geduldige Diagramme mit positiven Ergebnissen wurden dann &uuml;berpr&uuml;ft und Informationen extrahiert. Ergebnisse: 25 Patienten (25/1500) demonstrierten beil&auml;ufigen intraabdominal FDG-Aufnahme vermutlich erhebliche (1,7 %) mit einem Patienten Durchschnittsalter von 71 Jahren. Kolon Aufnahme war am h&auml;ufigsten (n = 17) mit 9 (52 %) weiter untersucht. Dieser 9 F&auml;lle wurde eine Diagnose Hyperkalz&auml;mie 3 Patienten, benigne Adenome bei 2 Patienten, ein gutartiges Lipom bei 1 Patienten und keine anormale Ergebnisse in die &uuml;brigen Patienten. 8 Patienten wurden nicht weiter untersucht (3 metastasierendem Lungenkrebs und 2 diagnostiziert wurden von fortgeschrittenen Alters) sekund&auml;r zu schlechte Prognose. Fazit: Zuf&auml;llige Abdominal-Feststellungen im Doppelpunkt auf FDG-PET scan nach Aufarbeitung pulmonale Kn&ouml;tchen Notwendigkeit von Koloskopie weiter untersucht werden.

ABSTRACT: BACKGROUND: il cancro del polmone &egrave; una comune causa di morte per cancro. Stadiazione in genere include positron emission tomography (PET) scansione, in cui 18F-fluoro-2-dexoy-D-glucose (FDG) &egrave; preso dalle cellule proporzionale all&#39;attivit&agrave; metabolica, aiutando cos&igrave; a differenziare noduli polmonari benigni e maligni. Assorbimento di FDG pu&ograve; verificarsi anche nell&#39;addome. Il significato clinico dell&#39;assorbimento FDG intraaddominali incidentale nell&#39;impostazione dei noduli polmonari non &egrave; ben definito. Il nostro obiettivo era di segnalare il significato clinico di incidentali attivit&agrave; intra-addominale di FDG nella cornice del cancro del polmone. Metodi: Millecinquecento FDG-PET studi di imaging eseguiti per il cancro del polmone a posteriori sono stati esaminati per la presenza di fortuiti ritrovamenti intra-addominale FDG-positivo. Tabelle pazienti con risultati positivi sono stati quindi rivisti e informazioni estratte. RISULTATI: Venti-cinque pazienti (25/1500) dimostrato incidentali uptake FDG intraaddominali pensato per essere significativo (1,7%) con un&#39;et&agrave; media di paziente di 71 anni. Assorbimento del colon era pi&ugrave; comune (n = 17) con 9 (52%) indagato ulteriormente. Di questi 9 casi, una diagnosi di malignit&agrave; &egrave; stata effettuata in 3 pazienti, pre-maligne adenomi in 2 pazienti, un lipoma benigno in 1 paziente e senza risultati anormali nei restanti pazienti. 8 pazienti non sono stati studiati ulteriormente (3 con diagnosticata di cancro al polmone metastatico e 2 erano di et&agrave; avanzata) secondaria a prognosi infausta. CONCLUSIONE: Reperti incidentali addominale nel colon su FDG-PET scan per work-up di noduli polmonari devono essere ulteriormente indagato da colonscopia.

Resumen: Antecedentes: c&aacute;ncer de pulm&oacute;n es una causa com&uacute;n de muerte relacionada con el c&aacute;ncer. Escenificando t&iacute;picamente incluye la emisi&oacute;n de positrones (PET) exploraci&oacute;n, en el que 18F-fluoro-2-dexoy-D-glucose (FDG) es tomado por las c&eacute;lulas proporcional a la actividad metab&oacute;lica, ayudando as&iacute; a distinguir n&oacute;dulos pulmonares benignos y malignos. Captaci&oacute;n de FDG puede ocurrir tambi&eacute;n en el abdomen. La significaci&oacute;n cl&iacute;nica de la absorci&oacute;n FDG intrabdominal incidental en la configuraci&oacute;n de los n&oacute;dulos pulmonares no est&aacute; bien establecida. Nuestro objetivo era divulgar sobre la significaci&oacute;n cl&iacute;nica de la actividad FDG intrabdominal incidental en el ajuste del c&aacute;ncer de pulm&oacute;n. M&Eacute;TODOS: Estudios por im&aacute;genes de mil quinientos FDG-PET para c&aacute;ncer de pulm&oacute;n se revisaron retrospectivamente para detectar la presencia de resultados intrabdominales fortuitos de FDG positivo. Luego revisaron listas de pacientes con resultados positivos y la informaci&oacute;n extra&iacute;da. Resultados: Veinticinco pacientes (25/1500) demostraron incidental captaci&oacute;n FDG intrabdominal cree que significativa (1,7%) con una edad media de 71 a&ntilde;os. Absorci&oacute;n col&oacute;nica era m&aacute;s com&uacute;n (n = 17) con 9 (52%) est&aacute;n investigando m&aacute;s. De estos 9 casos, una diagnosis de la malignidad fue hecha en 3 pacientes, adenomas premalignos en 2 pacientes, un lipoma benigno en 1 paciente y ningunos resultados anormales en los pacientes. 8 pacientes no fueron investigados m&aacute;s (3 diagnosticados con c&aacute;ncer metast&aacute;sico de pulm&oacute;n y 2 eran de edad avanzada) secundaria a mal pron&oacute;stico. CONCLUSI&Oacute;N: Resultados fortuitos abdominales en el colon en FDG-PET scan para trabajo de n&oacute;dulos pulmonares deban ser investigada m&aacute;s a fondo por colonoscopia.

Background Positron emission tomography (PET) with 18-fluorodeoxyglucose (FDG) is routinely performed for investigating suspected lung cancers. Despite the known potential for false-positive FDG uptake in the head and neck, most suspicious lesions on FDG-PET are investigated. Methods Between October 2002 and January 2010, FDG-PET/PET-computed tomography (CT) reports showing significant incidental uptake were retrospectively analyzed using controls to compare time to treatment for lung cancer and the rate of secondary malignancy in those with and without incidental head and neck FDG uptake. Results In this study, 48/1,846 PET/PET-CT scan reports reviewed, and demonstrated nonthyroidal head and neck abnormalities. A total of 30 patients had proven non-small cell lung cancer (1.6%); 3/30 (10%) went on to have biopsy proven malignancy. Furthermore, there was a significant increase in time to treatment in those patients with incidental head and neck findings (p = 0.002). Conclusion These unexpected findings are mostly of no clinical significance and the patient should continue treatment without delay; however, aggressive work-up should be pursued if concerns for head and neck pathology are raised following clinical assessment.
