Cockayne syndrome (CS) is a human hereditary disease belonging to the group of segmental progerias, and the clinical phenotype is characterized by postnatal growth failure, neurological dysfunction, cachetic dwarfism, photosensitivity, sensorineural hearing loss, and retinal degradation. CS-B cells are defective in transcription-coupled DNA repair, base excision repair, transcription, and chromatin structural organization. Using array analysis, we have examined the expression profile in CS complementation group B (CS-B) fibroblasts after exposure to oxidative stress (H2O2) before and after complete complementation with the CSB gene. The following isogenic cell lines were compared: CS-B cells (CS-B null), CS-B cells complemented with wild-type CSB (CS-B wt), and a stably transformed cell line with a point mutation in the ATPase domain of CSB (CS-B ATPase mutant). In the wt rescued cells, we detected significant induction (two-fold) of 112 genes out of the 6912 analysed. The patterns suggested an induction or upregulation of genes involved in several DNA metabolic processes including DNA repair, transcription, and signal transduction. In both CS-B mutant cell lines, we found a general deficiency in transcription after oxidative stress, suggesting that the CSB protein influenced the regulation of transcription of certain genes. Of the 6912 genes, 122 were differentially regulated by more than two-fold. Evidently, the ATPase function of CSB is biologically important as the deficiencies seen in the ATPase mutant cells are very similar to those observed in the CS-B-null cells. Some major defects are in the transcription of genes involved in DNA repair, signal transduction, and ribosomal functions.

The transcriptional response after oxidative stress is defective in Cockayne syndrome group B cells.

Syndrome de Cockayne (CS) est une maladie humaine h&#xE9;r&#xE9;ditaire appartenant au groupe des progerias segmentaires, et le ph&#xE9;notype clinique est caract&#xE9;ris&#xE9;e par un retard de croissance postnatal, des troubles neurologiques, nanisme cachectique, la photosensibilit&#xE9;, la perte auditive neurosensorielle, et la d&#xE9;gradation de la r&#xE9;tine. CS-B cellules sont d&#xE9;fectueux r&#xE9;paration de l&#x26;#39;ADN de transcription coupl&#xE9;, la r&#xE9;paration par excision de base, la transcription et la chromatine organisation structurelle. Utilisation de l&#x26;#39;analyse r&#xE9;seau, nous avons examin&#xE9; le profil d&#x26;#39;expression dans le groupe de compl&#xE9;mentation B CS (CS-B) fibroblastes apr&#xE8;s exposition &#xE0; un stress oxydatif (H2O2) avant et apr&#xE8;s la compl&#xE9;mentation avec le g&#xE8;ne CSB. Dans les cellules en poids sauv&#xE9;s, nous avons d&#xE9;tect&#xE9; une induction significative (deux fois) de 112 g&#xE8;nes de la 6912 analys&#xE9;s. Les mod&#xE8;les ont sugg&#xE9;r&#xE9; une induction ou la r&#xE9;gulation positive des g&#xE8;nes impliqu&#xE9;s dans plusieurs processus m&#xE9;taboliques, y compris d&#x26;#39;ADN r&#xE9;paration de l&#x26;#39;ADN, la transcription et la transduction du signal. Dans les deux lignes CS-B des cellules mutantes, nous avons trouv&#xE9; un d&#xE9;ficit g&#xE9;n&#xE9;ral dans la transcription apr&#xE8;s un stress oxydatif, ce qui sugg&#xE8;re que la prot&#xE9;ine CSB influenc&#xE9; la r&#xE9;gulation de la transcription de certains g&#xE8;nes. Sindrome di Cockayne (CS) &egrave; una malattia ereditaria umana appartiene al gruppo di progerias segmentali e il fenotipo clinico &egrave; caratterizzato da insufficienza di crescita postnatale, disfunzione neurologica, nanismo cachetic, fotosensibilit&agrave;, ipoacusia neurosensoriale e degradazione della retina. CS-B cellule sono difettose in trascrizione-accoppiato di riparazione del DNA, riparazione di base di escissione, trascrizione e organizzazione strutturale della cromatina. Utilizzando l&#39;analisi array, abbiamo esaminato il profilo di espressione in fibroblasti di CS complementazione gruppo B (CS-B) dopo l&#39;esposizione a stress ossidativo (H2O2) prima e dopo il completo complementazione con il gene CSB. Le seguenti linee cellulari isogeni sono state confrontate: CS-B cellule (null-CS-B), CS-B completato con selvaggio-tipo CSB (wt-CS-B) e una cella stabile trasformata in linea con una mutazione puntiforme nel dominio dell&#39;atpasi del CSB (mutante ATPasi CS-B). Nelle cellule wt salvato, abbiamo rilevato significativi ad induzione (due volte) di 112 geni fuori 6912 analizzati. I modelli suggerito un&#39;induzione o upregulation di geni coinvolti in numerosi processi metabolici del DNA compreso la riparazione del DNA, trascrizione e trasduzione del segnale. In due linee di cellule mutanti CS-B, abbiamo trovato un deficit generale di trascrizione dopo lo stress ossidativo, suggerendo che la proteina CSB influenzato la regolazione della trascrizione di alcuni geni. Dei geni 6912, 122 differenzialmente erano regolati da pi&ugrave; di due volte. Evidentemente, la funzione dell&#39;atpasi del CSB &egrave; biologicamente importante come le carenze vista nelle cellule mutanti ATPasi sono molto simili a quelle osservate nelle cellule B da CS-null. S&#xED;ndrome de Cockayne (CS) es una enfermedad humana hereditaria que pertenece al grupo de progerias segmentarias, y el fenotipo cl&#xED;nico se caracteriza por retraso del crecimiento postnatal, disfunci&#xF3;n neurol&#xF3;gica, enanismo caqu&#xE9;ctico, fotosensibilidad, p&#xE9;rdida de audici&#xF3;n neurosensorial, y la degradaci&#xF3;n de la retina. CS-c&#xE9;lulas B son defectuosas en la reparaci&#xF3;n del ADN transcripci&#xF3;n de acoplamiento, la reparaci&#xF3;n por escisi&#xF3;n de base, la transcripci&#xF3;n, y la organizaci&#xF3;n estructural de la cromatina. Mediante un an&#xE1;lisis de conjunto, hemos examinado el perfil de expresi&#xF3;n en el CS de complementaci&#xF3;n del grupo B (CS-B), los fibroblastos despu&#xE9;s de la exposici&#xF3;n al estr&#xE9;s oxidativo (H2O2) antes y despu&#xE9;s de complementaci&#xF3;n completa con el gen CSB. Los patrones sugieren una inducci&#xF3;n o la regulaci&#xF3;n positiva de genes implicados en diversos procesos metab&#xF3;licos, incluyendo ADN de reparaci&#xF3;n del ADN, transcripci&#xF3;n y transducci&#xF3;n de se&#xF1;ales. En ambas l&#xED;neas celulares mutantes CS-B, se encontr&#xF3; una deficiencia general en la transcripci&#xF3;n despu&#xE9;s de un estr&#xE9;s oxidativo, lo que sugiere que la prote&#xED;na CSB influido en la regulaci&#xF3;n de la transcripci&#xF3;n de ciertos genes. Werner syndrome (WS) is a premature aging disorder, displaying defects in DNA replication, recombination, repair, and transcription. It has been hypothesized that several WS phenotypes are secondary consequences of aberrant gene expression and that a transcription defect may be crucial to the development of the syndrome. We used cDNA microarrays to characterize the expression of 6,912 genes and ESTs across a panel of 15 primary human fibroblast cell lines derived from young donors, old donors, and WS patients. Of the analyzed genes, 6.3% displayed significant differences in expression when either WS or old donor cells were compared with young donor cells. This result demonstrates that the WS transcription defect is specific to certain genes. Transcription alterations in WS were strikingly similar to those in normal aging: 91% of annotated genes displayed similar expression changes in WS and in normal aging, 3% were unique to WS, and 6% were unique to normal aging. We propose that a defect in the transcription of the genes as identified in this study could produce many of the complex clinical features of WS. The remarkable similarity between WS and normal aging suggests that WS causes the acceleration of a normal aging mechanism. This finding supports the use of WS as an aging model and implies that the transcription alterations common to WS and normal aging represent general events in the aging process.

Gene expression profiling in Werner syndrome closely resembles that of normal aging.

The accumulation of DNA damage and mutations is considered a major cause of cancer and aging. While it is known that DNA damage can affect changes in gene expression, transcriptional regulation after DNA damage is poorly understood. We characterized the expression of 6912 genes in human primary fibroblasts after exposure to three different kinds of cellular stress that introduces DNA damage: 4-nitroquinoline-1-oxide (4NQO), gamma-irradiation, or UV-irradiation. Each type of stress elicited damage specific gene expression changes of up to 10-fold. A total of 85 genes had similar changes in expression of 3-40-fold after all three kinds of stress. We examined transcription in cells from young and old individuals and from patients with Werner syndrome (WS), a segmental progeroid condition with a high incidence of cancer, and found various age-associated transcriptional changes depending upon the type of cellular stress. Compared to young individuals, both WS and old individuals had similarly aberrant transcriptional responses to gamma- and UV-irradiation, suggesting a role for Werner protein in stress-induced gene expression. Our results suggest that aberrant DNA damage-induced gene regulation may contribute to the aging process and the premature aging in WS.

Gene expression responses to DNA damage are altered in human aging and in Werner Syndrome.

Human progeroid syndromes are caused by mutations in single genes accelerating some but not all features of normal aging. Most progeroid disorders are linked to defects in genome maintenance, and while it remains unknown if similar processes underlie normal and premature aging, they provide useful models for the study of aging. Altered transcription is speculated to play a causative role in aging, and is involved in the pathology of most if not all progeroid syndromes. Previous studies demonstrate that there is a similar pattern of gene expression changes in primary cells from old and Werner syndrome compared to young suggesting a presence of common cellular aging mechanisms in old and progeria. Here we review the role of transcription in progeroid syndromes and discuss the implications of similar transcription aberrations in normal and premature aging.
