&#x4EBA;&#x7C7B;&#x80DA;&#x80CE;&#x5E72;&#x7EC6;&#x80DE;&#x7E41;&#x6B96;&#x7684;&#x4EBA;&#x6E90;&#x6027;&#x78B1;&#x6027;&#x6210;&#x7EA4;&#x7EF4;&#x7EC6;&#x80DE;&#x7684;&#x7EC6;&#x80DE;&#x5916;&#x57FA;&#x8D28;&#x6210;&#x5206;&#x7684;&#x8868;&#x5F81;&#x3002;

Recent studies from our laboratory have shown that acellular substrates generated from human fibroblasts successfully maintained human pluripotent stem cells (hPSCs) in their undifferentiated state for extended periods. Aiming at better characterization, we conducted proteomic analyses to identify the extracellular matrix (ECM) proteins in mouse embryonic- and two human fibroblast-derived acellular substrates. Our studies identified heparan sulfate proteoglycan (HSPG) as a core component of these substrates and immunocytochemical analyses confirmed the presence of HSPG as well as other ECM proteins identified through proteomic analyses. In our attempt to develop surfaces that mimic fibroblast-deposited ECM and their self-renewal capabilities, substrates comprising HSPG and other core ECM proteins were formulated and assessed for the function of hPSC self-renewal. WA09 and BG01v hPSCs maintained on these substrates exhibit multiple characteristics of pluripotency, including (i) tight colony formation with typical stem cell morphology; (ii) positive expression of alkaline phosphatase, (iii) positive expression of SSEA3, SSEA4 and Oct4 based on immunocytochemical analyses; (iv) POU5F1, NANOG and SOX2 mRNA expression; and (v) in vitro differentiation and expression of germ-layer-specific markers. Our studies also reveal that although HSPG by itself-does not support hPSC self-renewal, a substrate that combines HSPG and fibronectin is sufficient for undifferentiated propagation of hPSCs. These studies form the basis for identification of appropriate ECM components in a substrate that synergistically promotes activation of adhesion and signaling pathways responsible for hPSC self-renewal.

Characterization of human fibroblast-derived extracellular matrix components for human pluripotent stem cell propagation.

Des &#xE9;tudes r&#xE9;centes de notre laboratoire ont montr&#xE9; que les substrats acellulaires g&#xE9;n&#xE9;r&#xE9;s &#xE0; partir de fibroblastes humains r&#xE9;ussi &#xE0; maintenir les cellules souches humaines pluripotentes (hPSCs) dans leur &#xE9;tat indiff&#xE9;renci&#xE9; pendant des p&#xE9;riodes prolong&#xE9;es. Visant &#xE0; mieux caract&#xE9;riser, nous avons effectu&#xE9; des analyses prot&#xE9;omiques pour identifier les matrice extracellulaire (ECM) des prot&#xE9;ines chez la souris embryonnaires humaines-et deux des fibroblastes d&#xE9;riv&#xE9;s substrats acellulaires. Nos &#xE9;tudes ont identifi&#xE9; l&#x26;#39;h&#xE9;parane sulfate prot&#xE9;oglycanes (HSPG) comme une composante essentielle de ces substrats et des analyses immunocytochimiques a confirm&#xE9; la pr&#xE9;sence de HSPG ainsi que d&#x26;#39;autres prot&#xE9;ines de la MEC identifi&#xE9;s par des analyses prot&#xE9;omiques. Dans notre tentative de d&#xE9;velopper des surfaces qui imitent des fibroblastes-d&#xE9;pos&#xE9; ECM et leurs capacit&#xE9;s d&#x26;#39;auto-renouvellement, substrats comprenant HSPG et d&#x26;#39;autres principales prot&#xE9;ines de la MEC ont &#xE9;t&#xE9; formul&#xE9;s et &#xE9;valu&#xE9;s pour la fonction de HPSC auto-renouvellement. HPSCs WA09 et BG01v maintenues sur ces substrats pr&#xE9;sentent des caract&#xE9;ristiques multiples de pluripotence, y compris (i) la formation de colonies &#xE9;troite avec la morphologie des cellules souches typiques; (ii) l&#x26;#39;expression positive de la phosphatase alcaline, (iii) l&#x26;#39;expression positive de SSEA3, SSEA4 et Oct4 bas&#xE9;e sur immunocytochimique analyses; (iv) POU5F1, NANOG et expression de l&#x26;#39;ARNm SOX2, et (v) la diff&#xE9;renciation in vitro et l&#x26;#39;expression de germes couche marqueurs sp&#xE9;cifiques. Nos &#xE9;tudes r&#xE9;v&#xE8;lent &#xE9;galement que, bien que HSPG par elle-m&#xEA;me ne prend pas en charge HPSC auto-renouvellement, un substrat qui combine HSPG et de la fibronectine est suffisante pour la propagation indiff&#xE9;renci&#xE9;e de hPSCs. Ces &#xE9;tudes constituent la base pour l&#x26;#39;identification des composants de l&#x26;#39;ECM appropri&#xE9;es dans un substrat qui favorise l&#x26;#39;activation synergique de l&#x26;#39;adh&#xE9;rence et les voies de signalisation responsables de HPSC auto-renouvellement.

Caract&#xE9;risation de l&#x26;#39;homme des fibroblastes d&#xE9;riv&#xE9;s des composants de la matrice extracellulaire pour la propagation de cellules souches pluripotentes humaines

Neuere Studien aus unserem Labor zufolge azellul&auml;re Substrate aus menschlichen Fibroblasten erfolgreich generiert menschlichen pluripotenten Stammzellen (hPSCs) in ihrem undifferenzierte Zustand &uuml;ber einen l&auml;ngeren Zeitraum aufrechterhalten. Mit dem Ziel bessere Charakterisierung, f&uuml;hrten wir Proteomic Analysen der extrazellul&auml;ren Matrix (ECM)-Proteine in embryonalen Maus zu identifizieren- und zwei menschlichen Fibroblasten abgeleitete azellul&auml;re Substrate. Unsere Studien identifiziert Heparan Sulfat Proteoglycans (HSPG) als eine Kernkomponente dieser Substrate und Analysen des Prozesses Immunocytochemical HSPG sowie andere ECM-Proteine identifiziert durch Proteomic Analysen best&auml;tigt. In unserem Versuch, Oberfl&auml;chen zu entwickeln, dass Mimik Fibroblast-hinterlegt ECM und ihre Selbsterneuerung F&auml;higkeiten, Substrate, bestehend aus HSPG und andere Kern ECM-Proteine wurden formuliert und f&uuml;r die Funktion der hPSC Selbsterneuerung bewertet. WA09 und BG01v hPSCs auf diese Substrate gepflegt weisen mehrere Merkmale der Pluripotenz, einschlie&szlig;lich (i) enge Kolonie Formation mit typischen Stammzelle Morphologie; (Ii) positive Ausdruck der alkalischen Phosphatase, (Iii) positive Ausdruck von SSEA3, SSEA4 und Oct4, die auf der Grundlage des Prozesses Immunocytochemical Analysen; (iv) POU5F1, NANOG und SOX2 mRNA Ausdruck; und (V) in-vitro-Differenzierung und Ausdruck der Keim-Schicht-spezifische Marker. Unsere Studien zeigen auch, dass obwohl HSPG durch selbst-tut nicht unterst&uuml;tzen hPSC Selbsterneuerung, ein Substrat, das HSPG und Fibronektin verbindet undifferenzierte Verbreitung des hPSCs ausreicht. Diese Studien bilden die Grundlage f&uuml;r die Identifikation von geeigneten ECM-Komponenten in einem Substrat, die Aktivierung von Adh&auml;sion und Signalwege verantwortlich f&uuml;r hPSC Selbsterneuerung synergistisch f&ouml;rdert.

Recenti studi dal nostro laboratorio hanno dimostrato che substrati acellulare generati da fibroblasti umani correttamente mantenuta cellule staminali pluripotenti umane (hPSCs) nel loro stato indifferenziato per periodi prolungati. Mirando alla migliore caratterizzazione, abbiamo condotto un&#39;analisi proteomic per identificare le proteine della matrice extracellulare (ECM) in embrionale del mouse- e due substrati umani derivato del fibroblasto acellulare. I nostri studi identificati eparan solfato proteoglicano (HSPG) come una componente fondamentale di questi substrati e analisi immunocitochimiche ha confermato la presenza di HSPG cos&igrave; come altre proteine ECM identificati attraverso analisi proteomica. Nel nostro tentativo di sviluppare superfici che mimica del fibroblasto-depositati ECM e le capacit&agrave; di auto-rinnovamento, substrati comprendente HSPG e altre proteine core ECM sono state formulate e valutate per la funzione di auto-rinnovamento di hPSC. HPSCs WA09 e BG01v mantenuto su questi substrati esibiscono caratteristiche multiple di pluripotenza, tra cui (i) formazione della Colonia stretto con morfologia tipica delle cellule staminali; (ii) positiva espressione della fosfatasi alcalina, (iii) espressione positiva di SSEA3, SSEA4 e Oct4 basato su analisi immunocitochimiche; (iv) POU5F1, NANOG e SOX2 espressione del mRNA; e (v) differenziazione in vitro e l&#39;espressione di marcatori di germe-strato-specifiche. I nostri studi rivelano anche che, sebbene HSPG da s&eacute;-non non supporta hPSC auto-rinnovamento, un substrato che unisce HSPG e fibronectina &egrave; sufficiente per la propagazione di indifferenziato di hPSCs. Questi studi costituiscono la base per l&#39;identificazione dei componenti appropriati di ECM in un substrato che sinergicamente promuove attivazione di adesione e signaling pathways responsabile hPSC self-renewal.

Estudios recientes de nuestro laboratorio han demostrado que los sustratos acelulares generados a partir de fibroblastos humanos mantenido con &#xE9;xito c&#xE9;lulas madre pluripotentes (hPSCs) en su estado indiferenciado durante largos per&#xED;odos. Con el objetivo de una mejor caracterizaci&#xF3;n, se realizaron an&#xE1;lisis prote&#xF3;micos para identificar a los de la matriz extracelular (ECM) de prote&#xED;nas en embriones de rat&#xF3;n de dos y humanos derivados de fibroblastos sustratos acelulares. Nuestros estudios identificaron proteoglicanos hepar&#xE1;n sulfato (HSPG) como un componente central de estos sustratos y an&#xE1;lisis inmunocitoqu&#xED;micos confirm&#xF3; la presencia de HSPG, as&#xED; como otras prote&#xED;nas ECM identificados a trav&#xE9;s de los an&#xE1;lisis prote&#xF3;micos. En nuestro intento de desarrollar superficies que imiten a fibroblastos depositan ECM y sus capacidades de auto-renovaci&#xF3;n, los sustratos que comprenden HSPG y otros principales prote&#xED;nas de la MEC se formularon y evaluaron la funci&#xF3;n de hPSC auto-renovaci&#xF3;n. HPSCs WA09 y BG01v mantienen en estos sustratos presentan m&#xFA;ltiples caracter&#xED;sticas de pluripotencialidad, incluyendo (i) la formaci&#xF3;n de colonias fuerte con una morfolog&#xED;a t&#xED;pica de c&#xE9;lulas madre, (ii) la expresi&#xF3;n positiva de la fosfatasa alcalina, (iii) la expresi&#xF3;n positiva de SSEA3, SSEA4 Oct4 y sobre la base de inmuno an&#xE1;lisis, (iv) POU5F1, NANOG y expresi&#xF3;n SOX2 ARNm, y (v) en la diferenciaci&#xF3;n in vitro y la expresi&#xF3;n de g&#xE9;rmenes capa de marcadores espec&#xED;ficos. Nuestros estudios tambi&#xE9;n revelan que a pesar de HSPG por s&#xED; mismo-no es compatible con hPSC auto-renovaci&#xF3;n, un sustrato que combina HSPG y fibronectina es suficiente para la propagaci&#xF3;n indiferenciada de hPSCs. Estos estudios constituyen la base para la identificaci&#xF3;n de los componentes de ECM apropiados en un sustrato que forma sin&#xE9;rgica promueve la activaci&#xF3;n de la adhesi&#xF3;n y las v&#xED;as de se&#xF1;alizaci&#xF3;n responsables de hPSC auto-renovaci&#xF3;n.
