E2 &#x86CB;&#x767D;&#x7684;&#x9AD8;&#x548C;&#x4F4E;&#x98CE;&#x9669;&#x4EBA;&#x7C7B;&#x4E73;&#x5934;&#x7624;&#x75C5;&#x6BD2;&#x7C7B;&#x578B;&#x4E0D;&#x540C;&#x7684;&#x7ED1;&#x5B9A; p53 &#x548C;&#x8BF1;&#x5BFC;&#x7EC6;&#x80DE;&#x51CB;&#x4EA1;&#x7684;&#x80FD;&#x529B;&#x3002;

The E2 proteins from oncogenic (high-risk) human papillomaviruses (HPVs) can induce apoptotic cell death in both HPV-transformed and non-HPV-transformed cells. Here we show that the E2 proteins from HPV type 6 (HPV6) and HPV11, two nononcogenic (low-risk) HPV types, fail to induce apoptosis. Unlike the high-risk HPV16 E2 protein, these low-risk E2 proteins fail to bind p53 and fail to induce p53-dependent transcription activation. Interestingly, neither the ability of p53 to activate transcription nor the ability of p53 to bind DNA, are required for HPV16 E2-induced apoptosis in non-HPV-transformed cells. However, mutations that reduce the binding of the HPV16 E2 protein to p53 inhibit E2-induced apoptosis in non-HPV-transformed cells. In contrast, the interaction between HPV16 E2 and p53 is not required for this E2 protein to induce apoptosis in HPV-transformed cells. Thus, our data suggest that this high-risk HPV E2 protein induces apoptosis via two pathways. One pathway involves the binding of E2 to p53 and can operate in both HPV-transformed and non-HPV-transformed cells. The second pathway requires the binding of E2 to the viral genome and can only operate in HPV-transformed cells.

Les prot&#xE9;ines oncog&#xE9;niques (E2 de risque &#xE9;lev&#xE9;) papillomavirus humains (HPV) peut induire la mort cellulaire par apoptose dans les cellules HPV-transform&#xE9;es et non-HPV-transform&#xE9;. Ici, nous montrons que les prot&#xE9;ines E2 de HPV de type 6 (HPV6) et HPV11, deux non oncog&#xE8;ne (faible risque) HPV types, ne parviennent pas &#xE0; induire l&#x26;#39;apoptose. Contrairement &#xE0; la prot&#xE9;ine &#xE0; haut risque HPV16 E2, ces prot&#xE9;ines &#xE0; faible risque ne parviennent pas &#xE0; se lier E2 p53 et ne parviennent pas &#xE0; induire l&#x26;#39;activation de la transcription p53-d&#xE9;pendante. Int&#xE9;ressant, ni la capacit&#xE9; de p53 &#xE0; activer la transcription ou la capacit&#xE9; de lier l&#x26;#39;ADN de p53 &#xE0;, sont n&#xE9;cessaires pour HPV16 E2-apoptose induite dans les cellules non-HPV-transform&#xE9;es. Cependant, des mutations qui r&#xE9;duisent la liaison de la prot&#xE9;ine E2 HPV16 &#xE0; p53 inhiber l&#x26;#39;apoptose induite par E2 dans des cellules non-HPV-transform&#xE9;es. En revanche, l&#x26;#39;interaction entre HPV16 E2 et p53 n&#x26;#39;est pas n&#xE9;cessaire pour cette prot&#xE9;ine E2 pour induire l&#x26;#39;apoptose dans les cellules HPV-transform&#xE9;es. Ainsi, nos donn&#xE9;es sugg&#xE8;rent que cette VPH &#xE0; risque &#xE9;lev&#xE9; prot&#xE9;ine E2 induit l&#x26;#39;apoptose par deux voies. Une voie passe par la liaison de E2 &#xE0; p53 et peut fonctionner dans les deux cellules HPV-transform&#xE9;es et non-HPV-transform&#xE9;. La deuxi&#xE8;me voie n&#xE9;cessite la liaison de E2 dans le g&#xE9;nome viral et ne peut fonctionner que dans des cellules HPV-transform&#xE9;es.

Prot&#xE9;ines E2 de haute et &#xE0; faible risque papillomavirus humains de types se distinguent par leur capacit&#xE9; &#xE0; se lier P53 et induire la mort cellulaire par apoptose

Die E2-Proteine aus Onkogene (Hochrisiko) menschliche Papillomaviren (HPVs) k&ouml;nnen Apoptotic Zellentod in HPV-transformierten sowohl nicht-HPV-transformierten Zellen ausl&ouml;sen. Hier zeigen wir, dass die E2-Proteine aus HPV 6 (HPV6) eingeben und HPV11, zwei nononcogenic (risikoarmen)-HPV-Typen, Apoptose induzieren. Im Gegensatz zu den risikoreichen HPV16-E2-Protein nicht diese risikoarme E2 Proteine binden p53 und Scheitern p53-abh&auml;ngige Transkription Aktivierung veranlassen. Interessanterweise sind weder die F&auml;higkeit von p53, Transkription aktivieren als auch die F&auml;higkeit von p53, DNA, binden f&uuml;r HPV16-E2-induzierten Apoptose in HPV-transformierten Zellen erforderlich. Allerdings hemmen Mutationen, die die Bindung des Proteins HPV16-E2, p53 reduzieren E2-induzierten Apoptose in HPV-transformierten Zellen. Im Gegensatz dazu ist die Interaktion zwischen HPV16-E2 und p53 nicht erforderlich f&uuml;r dieses Protein E2 Apoptosis in HPV-transformierten Zellen veranlassen. So, unsere Daten deuten darauf hin, dass dieses Hochrisiko-HPV-E2-Protein Apoptose &uuml;ber zwei Wege induziert. Ein Weg beinhaltet die Bindung von E2 p53 und HPV-transformierten sowohl nicht-HPV-transformierten Zellen arbeiten kann. Der zweite Weg ist die Bindung der E2 des viralen Genoms erforderlich und kann nur in HPV-transformierten Zellen betrieben.

E2 Proteine aus - und Low-Risk humanen Papillomvirus-Typen unterscheiden sich in ihrer F&auml;higkeit, binden p53 und apoptotischen Zelltod zu induzieren.

Le proteine E2 da oncogenica (alto rischio) papillomavirus umano (HPV) possono indurre la morte cellulare apoptotica in cellule sia HPV-trasformati e non trasformati da HPV. Qui vi mostriamo che le proteine E2 da HPV di tipo 6 (HPV6) e HPV11, due tipi di HPV (basso rischio) HPV, non riescono a indurre l&#39;apoptosi. A differenza della proteina E2 HPV16 ad alto rischio, queste proteine E2 basso rischio non riescono a legare p53 e non riescono a indurre attivazione trascrizione p53-dipendente. Curiosamente, n&eacute; la possibilit&agrave; di attivare la trascrizione p53 n&eacute; la capacit&agrave; di legare il DNA, p53 sono necessari per HPV16 E2-induced apoptosis in cellule non trasformato da HPV. Tuttavia, mutazioni che riducono l&#39;associazione della proteina E2 HPV16 per p53 inibiscono E2-induced apoptosis in cellule non trasformato da HPV. Al contrario, l&#39;interazione tra HPV16 E2 e p53 non &egrave; richiesto per questa proteina E2 di indurre apoptosi nelle cellule trasformate di HPV. Cos&igrave;, i nostri dati suggeriscono che questa proteina E2 di HPV ad alto rischio induce l&#39;apoptosi tramite due percorsi. Un percorso comporta l&#39;associazione di E2 per p53 e pu&ograve; operare nelle cellule sia HPV-trasformati e non trasformati da HPV. Il secondo percorso richiede l&#39;associazione di E2 del genoma virale e pu&ograve; operare solo in cellule trasformate di HPV.

Proteine E2 da tipi di papillomavirus umano ad alto e basso rischio differiscono nella loro capacit&agrave; di legare p53 e indurre la morte delle cellule apoptotic.

Las prote&#xED;nas E2 de oncog&#xE9;nicos (de alto riesgo) del virus del papiloma humano (VPH) puede inducir la muerte celular por apoptosis en c&#xE9;lulas transformadas por el VPH y no el VPH transformado. Aqu&#xED; nos muestran que las prote&#xED;nas E2 del VPH tipo 6 (HPV6) y HPV11, dos no oncog&#xE9;nico (bajo riesgo), los tipos de VPH no inducir la apoptosis. A diferencia de la prote&#xED;na de alto riesgo HPV16 E2, estas prote&#xED;nas de bajo riesgo E2 no obligar a p53 y no para inducir p53 dependiente de la activaci&#xF3;n de la transcripci&#xF3;n. Curiosamente, ni la capacidad de p53 para activar la transcripci&#xF3;n ni la capacidad de p53 de unirse al ADN, son necesarios para HPV16 E2 inducida por la apoptosis en las c&#xE9;lulas no transformadas por VPH. Sin embargo, las mutaciones que reducen la uni&#xF3;n de la prote&#xED;na E2 de HPV 16 p53 inhibe la apoptosis inducida por E2 en c&#xE9;lulas no transformadas por VPH. Por el contrario, la interacci&#xF3;n entre el HPV16 E2 y p53 no es necesario para esta prote&#xED;na E2 para inducir la apoptosis en las c&#xE9;lulas transformadas por VPH. Por lo tanto, nuestros datos sugieren que esta VPH de alto riesgo prote&#xED;na E2 induce la apoptosis a trav&#xE9;s de dos v&#xED;as. Una v&#xED;a consiste en la uni&#xF3;n de E2 a p53 y puede operar tanto en las c&#xE9;lulas transformadas y HPV-no-HPV-transformado. La segunda v&#xED;a requiere la uni&#xF3;n de E2 con el genoma viral y s&#xF3;lo puede funcionar en las c&#xE9;lulas transformadas por VPH.
