The neural selector gene cut, a homeobox transcription factor, is required for the specification of the correct identity of external (bristle-type) sensory organs in Drosophila. Targets of cut function, however, have not been described. Here, we study bereft (bft) mutants, which exhibit loss or malformation of a majority of the interommatidial bristles of the eye and cause defects in other external sensory organs. These mutants were generated by excising a P element located at chromosomal location 33AB, the enhancer trap line E8-2-46, indicating that a gene near the insertion site is responsible for this phenotype. Similar to the transcripts of the gene nearest to the insertion, reporter gene expression of E8-2-46 coincides with Cut in the support cells of external sensory organs, which secrete the bristle shaft and socket. Although bft transcripts do not obviously code for a protein product, its expression is abolished in bft deletion mutants, and the integrity of the bft locus is required for (interommatidial) bristle morphogenesis. This suggests that disruption of the bft gene is the cause of the observed bristle phenotype. We also sought to determine what factors regulate the expression of bft and the enhancer trap line. The correct specification of individual external sensory organ cells involves not only cut, but also the lineage genes numb and tramtrack. We demonstrate that mutations of these three genes affect the expression levels at the bft locus. Furthermore, cut overexpression is sufficient to induce ectopic bft expression in the PNS and in nonneuronal epidermis. On the basis of these results, we propose that bft acts downstream of cut and tramtrack to implement correct bristle morphogenesis.

Through the direct analysis of cell behaviors, we address the mechanisms underlying anterior neural tube morphogenesis in the zebrafish and the role of the cell adhesion molecule N-cadherin (N-cad) in this process. We demonstrate that although the mode of neurulation differs at the morphological level between amphibians and teleosts, the underlying cellular mechanisms are conserved. Contrary to previous reports, the zebrafish neural plate is a multi-layered structure, composed of deep and superficial cells that converge medially while undergoing radial intercalation, to form a single cell-layered neural tube. Time-lapse recording of individual cell behaviors reveals that cells are polarized along the mediolateral axis and exhibit protrusive activity. In N-cad mutants, both convergence and intercalation are blocked. Moreover, although N-cad-depleted cells are not defective in their ability to form protrusions, they are unable to maintain them stably. Taken together, these studies uncover key cellular mechanisms underlying neural tube morphogenesis in teleosts, and reveal a role for cadherins in promoting the polarized cell behaviors that underlie cellular rearrangements and shape the vertebrate embryo.

The zebrafish is a well established model system for studying neural development, yet neurulation remains poorly understood in this organism. In particular, the morphogenetic movements that shape the posterior neural tube (PNT) have not been described. Using tools for imaging neural tissue and tracking the behavior of cells in real time, we provide the first comprehensive analysis of the cellular events shaping the PNT. We observe that this tissue is formed in a stepwise manner, beginning with merging of presumptive neural domains in the tailbud (Stage 1); followed by neural convergence and infolding to shape the neural rod (Stage 2); and continued elongation of the PNT, in absence of further convergence (Stage 3). We further demonstrate that cell proliferation plays only a minimal role in PNT elongation. Overall, these mechanisms resemble those previously described in anterior regions, suggesting that, in contrast to amniotes, neurulation is a fairly uniform process in zebrafish.

Mit Tools f&uuml;r Bildbearbeitung Nervengewebe und das Verhalten von Zellen in Echtzeit verfolgen, bieten wir die erste umfassende Analyse der Gestaltung der PNT zellul&auml;re Ereignisse. Wir beobachten, dass dieses Gewebe gebildet wird, in gewissem Sinne schrittweise, beginnend mit der Verschmelzung der mutma&szlig;lichen neuronale Dom&auml;nen in der Tailbud (Phase 1); gefolgt von neuronale Konvergenz und infolding gestalten den neuronalen Stab (Stufe 2); und anhaltende Dehnung der PNT, in Abwesenheit der weiteren Ann&auml;herung (Stufe 3). Weiter zeigen wir, dass die Zellproliferation bei PNT Dehnung nur eine minimale Rolle spielt. Insgesamt &auml;hneln diese Mechanismen den zuvor beschriebenen in den vorderen Regionen darauf hindeutet, dass, im Gegensatz zu Amnioten, Neuralplatte einen relativ einheitlichen Prozess im Zebrafisch.

In particolare, non sono stati descritti i movimenti morfogenetici che forma il tubo neurale posteriore (PNT). Usando gli strumenti per imaging tessuto neurale e monitoraggio del comportamento delle cellule in tempo reale, forniamo la prima analisi completa degli eventi cellulari che modella il PNT. Osserviamo che questo tessuto &egrave; formato in modo graduale, cominciando con la fusione di domini neurale presuntiva in tailbud (fase 1); seguito dalla convergenza neurale e infolding per modellare l&#39;asta neurale (fase 2); e costante allungamento del PNT, in assenza di ulteriore convergenza (fase 3). Noi dimostrare ulteriormente che la proliferazione cellulare svolge solo un ruolo marginale in allungamento PNT. Nel complesso, questi meccanismi ricordano quelle precedentemente descritte nelle regioni anteriori, suggerendo che, a differenza di amnioti, neurulazione &egrave; un processo abbastanza uniforme nello zebrafish.

Microtubules are essential regulators of cell polarity, architecture and motility. The organization of the microtubule network is context-specific. In non-polarized cells, microtubules are anchored to the centrosome and form radial arrays. In most epithelial cells, microtubules are noncentrosomal, align along the apico-basal axis and the centrosome templates a cilium. It follows that cells undergoing mesenchyme-to-epithelium transitions must reorganize their microtubule network extensively, yet little is understood about how this process is orchestrated. In particular, the pathways regulating the apical positioning of the centrosome are unknown, a central question given the role of cilia in fluid propulsion, sensation and signaling. In zebrafish, neural progenitors undergo progressive epithelialization during neurulation, and thus provide a convenient in vivo cellular context in which to address this question. We demonstrate here that the microtubule cytoskeleton gradually transitions from a radial to linear organization during neurulation and that microtubules function in conjunction with the polarity protein Pard3 to mediate centrosome positioning. Pard3 depletion results in hydrocephalus, a defect often associated with abnormal cerebrospinal fluid flow that has been linked to cilia defects. These findings thus bring to focus cellular events occurring during neurulation and reveal novel molecular mechanisms implicated in centrosome positioning.

Mikrotubuli sind wesentliche Regulatoren der Zellpolarit&auml;t, Architektur und Motilit&auml;t. Die Organisation des Microtubule-Netzes ist Kontext-sensitive. In nicht-polarisierten Zellen Mikrotubuli sind das Zentrosom verankert und bilden radial-Arrays. In den meisten Epithelzellen Mikrotubuli sind Noncentrosomal, entlang der Apico-basale Achse und die Zentrosom-Vorlagen eine Zilie ausrichten. Daraus folgt, dass Zellen, die &Uuml;bergangsphase zwischen Mesenchym-zu-Epithel befinden ihre Microtubule-Netzwerk ausgiebig, reorganisieren m&uuml;ssen noch wenig verstanden ist, &uuml;ber wie dieser Proze&szlig; inszeniert ist. Insbesondere sind die Wege, die Regulierung der apikalen Positionierung der Zentrosom unbekannt, eine zentrale Frage, die angesichts der Rolle der Zilien in fl&uuml;ssigen Antrieb, Empfindung und Signalisierung. Im Zebrafisch neuronale Vorl&auml;uferzellen progressive Heilungsabschluss w&auml;hrend der Neuralplatte zu unterziehen und somit bieten einen bequemen in-vivo zellul&auml;ren Kontext in dem mit dieser Frage besch&auml;ftigen. Hier zeigen wir, dass Microtubule Zytoskeletts aus einem Radial lineare Organisation w&auml;hrend der Neurulation und die Mikrotubuli-Funktion in Verbindung mit der Polarit&auml;t-Protein Pard3 Zentrosom Positionierung vermitteln allm&auml;hlich &uuml;bergeht. Neural progenitors are organized as a pseudostratified epithelium held together by adherens junctions (AJs), multiprotein complexes composed of cadherins and Î±- and Î²-catenin. Catenins are known to control neural progenitor division; however, it is not known whether they function in this capacity as cadherin binding partners, as there is little evidence that cadherins themselves regulate neural proliferation. We show here that zebrafish N-cadherin (N-cad) restricts cell proliferation in the dorsal region of the neural tube by regulating cell-cycle length. We further reveal that N-cad couples cell-cycle exit and differentiation, as a fraction of neurons are mitotic in N-cad mutants. Enhanced proliferation in N-cad mutants is mediated by ligand-independent activation of Hedgehog (Hh) signaling, possibly caused by defective ciliogenesis. Furthermore, depletion of Hh signaling results in the loss of junctional markers. We therefore propose that N-cad restricts the response of dorsal neural progenitors to Hh and that Hh signaling limits the range of its own activity by promoting AJ assembly. Taken together, these observations emphasize a key role for N-cad-mediated adhesion in controlling neural progenitor proliferation. In addition, these findings are the first to demonstrate a requirement for cadherins in synchronizing cell-cycle exit and differentiation and a reciprocal interaction between AJs and Hh signaling.

Wir zeigen hier, dass Zebrafisch N-Cadherin (N-Cad) Zellvermehrung in der dorsalen Region von dem Neuralrohr schr&auml;nkt durch die Regelung des Zellzyklus. Wir zeigen weiter, dass N-Cad Zellzyklus Ausfahrt und Differenzierung, Paare, wie ein Bruchteil der Neuronen sind mitotische in N-Cad-Mutanten. Verst&auml;rkte Verbreitung in N-Cad-Mutanten ist durch Ligand-unabh&auml;ngige Aktivierung Igel (Hh) signalisieren, m&ouml;glicherweise verursacht durch defekte Ciliogenesis vermittelt. Dar&uuml;ber hinaus f&uuml;hrt zum Verlust der junktionale Marker Ersch&ouml;pfung Hh zu signalisieren. Wir schlagen deshalb vor, dass N-Cad die Reaktion der dorsalen neuronalen Vorl&auml;uferzellen zu Hh schr&auml;nkt und Hh signalisieren den Bereich ihrer eigenen T&auml;tigkeit begrenzt durch die F&ouml;rderung von AJ-Versammlung. N cadherin &#xC911;&#xC7AC; &#xC138;&#xD3EC; &#xC811;&#xCC29; &#xB4F1; &#xCABD; &#xC2E0;&#xACBD; &#xD29C;&#xBE0C;&#xC5D0; &#xC138;&#xD3EC; &#xC99D;&#xC2DD;&#xC744; &#xC81C;&#xD55C;&#xD569;&#xB2C8;&#xB2E4;.

Progenitores neuronales se organizan como un epitelio pseudoestratificado mantienen unidos por uniones adherentes (AJS), compuestas de complejos multiproteicos cadherinas y &#x3B1;-catenina y la &#x3B2;-. Cateninas son conocidos por controlar la divisi&#xF3;n progenitoras neurales, sin embargo, no se sabe si funcionan en esta capacidad como socios cadherina vinculantes, ya que hay poca evidencia de que las cadherinas se regulan la proliferaci&#xF3;n neuronal. Mostramos aqu&#xED; que el pez cebra N-cadherina (N-cad) restringe la proliferaci&#xF3;n celular en la regi&#xF3;n dorsal del tubo neural mediante la regulaci&#xF3;n del ciclo celular longitud.
