&#x6279;&#x91CF;&#x751F;&#x4EA7;&#x7684;&#x4EBA;-&#x9F20; heterohybridoma &#x7EC6;&#x80DE;&#x4F53;&#x5916;&#x7EC6;&#x80DE;&#x7535;&#x878D;&#x5408;&#x7684;&#x4F18;&#x5316;&#x3002;

Cell electrofusion is a phenomenon that occurs, when cells are in close contact and exposed to short high-voltage electric pulses. The consequence of exposure to pulses is transient and nonselective permeabilization of cell membranes. Cell electrofusion and permeabilization depend on the values of electric field parameters including amplitude, duration and number of electric pulses and direction of the electric field. In our study, we first investigated the influence of the direction of the electric field on cell fusion in two cell lines. In both cell lines, applications of pulses in two directions perpendicular to each other were the most successful. Cell electrofusion was finally used for production of human-mouse heterohybridoma cells with modified Koehler and Milstein hybridoma technology, which was not done previously. The results, obtained by cell electrofusion, are comparable to usually used polyethylene glycol mediated fusion on the same type of cells.

Optimization of bulk cell electrofusion in vitro for production of human-mouse heterohybridoma cells.

&#xC9;lectrofusion cellulaire est un ph&#xE9;nom&#xE8;ne qui se produit, lorsque les cellules sont en contact &#xE9;troit et expos&#xE9; &#xE0; de courtes impulsions &#xE0; haute tension &#xE9;lectriques. La cons&#xE9;quence de l&#x26;#39;exposition aux impulsions est perm&#xE9;abilisation transitoire et non s&#xE9;lectif des membranes cellulaires. &#xC9;lectrofusion cellule et perm&#xE9;abilisation d&#xE9;pendent des valeurs de param&#xE8;tres du champ &#xE9;lectrique dont l&#x26;#39;amplitude, dur&#xE9;e et le nombre d&#x26;#39;impulsions &#xE9;lectriques et &#xE0; l&#x26;#39;orientation du champ &#xE9;lectrique. Dans notre &#xE9;tude, nous avons d&#x26;#39;abord &#xE9;tudi&#xE9; l&#x26;#39;influence de la direction du champ &#xE9;lectrique sur la fusion cellulaire dans deux lign&#xE9;es cellulaires. Dans les deux lignes de cellules, les applications d&#x26;#39;impulsions dans deux directions perpendiculaires les unes aux autres sont les plus efficace. &#xC9;lectrofusion cellulaire a finalement &#xE9;t&#xE9; utilis&#xE9;e pour la production de cellules h&#xE9;t&#xE9;rohybridome humaine &#xE0; la souris avec modification Koehler et Milstein technique de l&#x26;#39;hybridome, qui n&#x26;#39;a pas &#xE9;t&#xE9; fait auparavant. Les r&#xE9;sultats obtenus par &#xE9;lectrofusion cellule, sont comparables &#xE0; la fusion du poly&#xE9;thyl&#xE8;ne glycol &#xE0; m&#xE9;diation habituellement utilis&#xE9; sur le m&#xEA;me type de cellules.

Zelle Elektroschweiss ist ein Ph&auml;nomen, das auftritt, wenn Zellen sich in engem Kontakt und kurze Hochspannungs-elektrische Impulse ausgesetzt. Die Folge der Exposition auf Impulse ist verg&auml;nglich und unselektiv Permeabilization der Zellmembranen. Zelle Elektroschweiss und Permeabilization h&auml;ngen die Werte der elektrischen Feldparameter wie Amplitude, Dauer und Anzahl der elektrischen Impulsen und Richtung des elektrischen Feldes. In unserer Studie untersuchten wir zun&auml;chst den Einfluss der Richtung des elektrischen Feldes auf Zellfusion in beiden Zelllinien. In beiden Zelllinien wurden Anwendungen von Impulsen in zwei senkrecht zueinander stehenden Richtungen die erfolgreichste. Zelle Elektroschweiss wurde schlie&szlig;lich zur Herstellung von Mensch-Maus-Heterohybridoma Zellen mit ver&auml;nderter K&ouml;hler und Milstein Hybridom-Technik, verwendet, die nicht zuvor getan wurde. Von Zelle Heizwendelschwei&szlig;en, erzielten Ergebnisse sind vergleichbar mit in der Regel verwendete Polyethylenglykol vermittelte Fusion auf die gleiche Art von Zellen.

Cella elettrofusione &egrave; un fenomeno che si verifica quando le cellule sono in stretto contatto ed esposti a brevi impulsi elettrici ad alta tensione. La conseguenza dell&#39;esposizione a impulsi &egrave; transitoria e non selettivi permeabilization delle membrane cellulari. Cella Elettrosaldabile e permeabilization dipendono i valori dei parametri di campo elettrico tra cui ampiezza, durata e numero di impulsi elettrici e la direzione del campo elettrico. Nel nostro studio, abbiamo studiato in primo luogo l&#39;influenza della direzione del campo elettrico sulla fusione cellulare in due linee cellulari. In entrambe le linee cellulari, applicazioni di impulsi in due direzioni perpendicolari tra loro erano il maggior successo. Cella elettrofusione infine &egrave; stato utilizzato per la produzione di cellule umane-mouse heterohybridoma con modificata Koehler e Milstein ibridoma tecnologia, che non &egrave; stato fatto in precedenza. I risultati, ottenuti da cellule elettrofusione, sono paragonabili al solito usato polietilenglicole mediata fusione sullo stesso tipo di cellule.

Electrofusi&#xF3;n de c&#xE9;lulas es un fen&#xF3;meno que se produce, cuando las c&#xE9;lulas est&#xE1;n en contacto estrecho y expuesto a cortos pulsos de alta tensi&#xF3;n el&#xE9;ctrica. La consecuencia de la exposici&#xF3;n a pulsos es permeabilizaci&#xF3;n transitoria y no selectiva de las membranas celulares. Electrofusi&#xF3;n de la c&#xE9;lula y la permeabilizaci&#xF3;n depender&#xE1; de los valores de los par&#xE1;metros del campo el&#xE9;ctrico, incluyendo amplitud, duraci&#xF3;n y n&#xFA;mero de impulsos el&#xE9;ctricos y la direcci&#xF3;n del campo el&#xE9;ctrico. En nuestro estudio, primero investig&#xF3; la influencia de la direcci&#xF3;n del campo el&#xE9;ctrico en la fusi&#xF3;n celular en dos l&#xED;neas celulares. En ambas l&#xED;neas celulares, las aplicaciones de pulsos en dos direcciones perpendiculares entre s&#xED; fueron los m&#xE1;s exitosos. Electrofusi&#xF3;n de la c&#xE9;lula se utiliz&#xF3; finalmente para la producci&#xF3;n de c&#xE9;lulas humanas y de rat&#xF3;n heterohibridoma con modificaci&#xF3;n de Koehler y Milstein la tecnolog&#xED;a del hibridoma, que no se hizo anteriormente. Los resultados, obtenidos por electrofusi&#xF3;n celular, son comparables a la fusi&#xF3;n utiliza generalmente de polietileno glicol mediada en el mismo tipo de c&#xE9;lulas.

Efficient electroporation of cells in physical contact induces cell fusion, and this process is known as electrofusion. It has been shown that appropriate hypotonic treatment of cells before the application of electric pulses can cause a significant increase in electrofusion efficiency. First, the amplitudes of the electric field were determined spectrofluorometrically, where sufficient permeabilization in hypotonic buffer occurred for B16-F1 and CHO cells. In further electrofusion experiments 14 +/- 4% of fused cells for B16-F1 and 6 +/- 1% for CHO was achieved. These electrofusion efficiencies, determined by double staining and fluorescence microcopy, are comparable to those of other published studies. It was also confirmed that successful electroporation does not necessarily guarantee high electrofusion efficiency due to biological factors involved in the electrofusion process. Furthermore, not only the extension of electrofusion but also cell survival depends on the cell line used. Further studies are needed to improve overall cell survival after electroporation in hypotonic buffer, which was significantly reduced, especially for B16-F1 cells. Another contribution of this report is the description of a simple modification of the adherence method for formation of spontaneous cell contact, while cells preserve their spherical shape.

Electroporaci&#xF3;n eficiente de las c&#xE9;lulas en contacto f&#xED;sico induce la fusi&#xF3;n de c&#xE9;lulas, y este proceso se conoce como electrofusi&#xF3;n. Se ha demostrado que el tratamiento apropiado hipot&#xF3;nico de las c&#xE9;lulas antes de la aplicaci&#xF3;n de impulsos el&#xE9;ctricos pueden causar un aumento significativo de la eficiencia de electrofusi&#xF3;n. En primer lugar, las amplitudes del campo el&#xE9;ctrico se determinaron spectrofluorometrically, donde la permeabilizaci&#xF3;n suficiente en tamp&#xF3;n hipot&#xF3;nico producido por las c&#xE9;lulas B16-F1 y CHO. En experimentos adicionales de electrofusi&#xF3;n 14 + / - 4% de c&#xE9;lulas fusionadas para B16-F1 y 6 + / - 1% para CHO se logr&#xF3;. Estas eficiencias de electrofusi&#xF3;n, determinados por la doble tinci&#xF3;n y microscopia de fluorescencia, son comparables a los de otros estudios publicados. Tambi&#xE9;n se confirm&#xF3; que la electroporaci&#xF3;n &#xE9;xito no garantiza necesariamente la eficiencia de electrofusi&#xF3;n de alta debido a factores biol&#xF3;gicos que intervienen en el proceso de electrofusi&#xF3;n. Adem&#xE1;s, no s&#xF3;lo la extensi&#xF3;n de electrofusi&#xF3;n, sino tambi&#xE9;n la supervivencia celular depende de la l&#xED;nea celular utilizada. Se necesitan m&#xE1;s estudios para mejorar la supervivencia general de la c&#xE9;lula despu&#xE9;s de la electroporaci&#xF3;n en tamp&#xF3;n hipot&#xF3;nico, que se redujo significativamente, en especial para B16-F1 c&#xE9;lulas. Otra contribuci&#xF3;n de este informe es la descripci&#xF3;n de una simple modificaci&#xF3;n del m&#xE9;todo de adhesi&#xF3;n para la formaci&#xF3;n de contactos c&#xE9;lula espont&#xE1;nea, mientras que las c&#xE9;lulas preservar su forma esf&#xE9;rica.
