Only one broadly neutralizing anti-HIV antibody, 2G12, recognises the envelope sugars of HIV. In the present study, we show that 2G12 also recognises Candida albicans and Candida tropicalis with high affinity (11 nmol/l) through a carbohydrate-dependent interaction (50% inhibitory concentration for D-fructose, 12 mmol/l). This is the first report of a neutralizing HIV antibody displaying cross-reactivity with another pathogen, revealing that the carbohydrate neutralization determinant of HIV, defined by 2G12, is more widespread amongst immunogenic, microbial surfaces than previously recognized.

We solved the X-ray structures of two Escherichia coli tRNA(Ser) acceptor stem microhelices. As both tRNAs are aminoacylated by the same seryl-tRNA-synthetase, we performed a comparative structure analysis of both duplexes to investigate the helical conformation, the hydration patterns and magnesium binding sites. It is well accepted, that the hydration of RNA plays an important role in RNA-protein interactions and that the extensive solvent content of the minor groove has a special function in RNA. The detailed comparison of both tRNA(Ser) microhelices provides insights into the structural arrangement of the isoacceptor tRNA aminoacyl stems with respect to the surrounding water molecules and may eventually help us to understand their biological function at atomic resolution.

Superposition of two tRNASer acceptor stem crystal structures: comparison of structure, ligands and hydration.

Nous avons r&eacute;solu les structures des deux microhelices de souches Escherichia coli tRNA(Ser) accepteur. Comme les deux ARNt est tRNALeu par le m&ecirc;me s&eacute;ryl-ARNt-synth&eacute;tase, nous avons effectu&eacute; une analyse de la structure comparative de deux duplex d&#39;enqu&ecirc;ter sur la conformation h&eacute;lico&iuml;dale, les patrons de l&#39;hydratation et les sites de fixation de magn&eacute;sium. Il est bien reconnu, que l&#39;hydratation de l&#39;ARN joue un r&ocirc;le important dans les interactions ARN-prot&eacute;ine et que le contenu exhaustif de solvant de la rainure mineure a une fonction particuli&egrave;re dans l&#39;ARN. La comparaison d&eacute;taill&eacute;e des deux microhelices de tRNA(Ser) permet de mieux comprendre l&#39;agencement structurel de l&#39;isoacceptor tiges aminoacyl ARNt en ce qui concerne les mol&eacute;cules d&#39;eau environnantes et peut &eacute;ventuellement nous aider &agrave; comprendre leur fonction biologique &agrave; r&eacute;solution atomique.

Wir l&ouml;sen die R&ouml;ntgen-Strukturen der beiden Escherichia coli tRNA(Ser) Akzeptor Stamm Microhelices. Wie beide tRNAs Aminoacylated von der gleichen Seryl-tRNA-Synthetase, f&uuml;hrten wir eine vergleichende Strukturanalyse der beide Duplices, die spiralf&ouml;rmige Konformation, die Hydratation Muster und Magnesium-Bindungsstellen zu untersuchen. Es wird auch akzeptiert, dass die Hydratation von RNA spielt eine wichtige Rolle in RNA-Protein-Wechselwirkungen und die umfangreiche L&ouml;semittelgehalt der kleinen Nut hat eine spezielle Funktion in RNA. Der detaillierte Vergleich der beiden tRNA(Ser)-Microhelices bietet Einblicke in die strukturelle Anordnung der Isoacceptor tRNA Aminoacyl Stielen in Bezug auf die umgebenden Wassermolek&uuml;le und kann schlie&szlig;lich dazu beitragen, uns zu verstehen, ihre biologische Funktion mit atomarer Aufl&ouml;sung.

Abbiamo risolto le strutture dei raggi x di due Escherichia coli tRNA(Ser) acceptor stem microhelices. Come entrambi tRNA &egrave; amminocilato dalla stessa seryl-tRNA-sintetasi, abbiamo eseguito un&#39;analisi comparativa della struttura di entrambi duplex per indagare la conformazione elicoidale, i modelli di idratazione e siti di legame di magnesio. &Egrave; ben accettato, che l&#39;idratazione del RNA gioca un ruolo importante nelle interazioni RNA-proteina e che il contenuto di solvente esteso del solco minore ha una funzione speciale in RNA. Il confronto dettagliato di entrambi microhelices tRNA(Ser) fornisce intuizioni la disposizione strutturale dell&#39;isoacceptor gambi amminoacil tRNA per quanto riguarda le molecole d&#39;acqua circostanti e alla fine possa aiutarci a capire la loro funzione biologica a risoluzione atomica.

RNA transcripts are subject to posttranscriptional gene regulation involving hundreds of RNA-binding proteins (RBPs) and microRNA-containing ribonucleoprotein complexes (miRNPs) expressed in a cell-type dependent fashion. We developed a cell-based crosslinking approach to determine at high resolution and transcriptome-wide the binding sites of cellular RBPs and miRNPs. The crosslinked sites are revealed by thymidine to cytidine transitions in the cDNAs prepared from immunopurified RNPs of 4-thiouridine-treated cells. We determined the binding sites and regulatory consequences for several intensely studied RBPs and miRNPs, including PUM2, QKI, IGF2BP1-3, AGO/EIF2C1-4 and TNRC6A-C. Our study revealed that these factors bind thousands of sites containing defined sequence motifs and have distinct preferences for exonic versus intronic or coding versus untranslated transcript regions. The precise mapping of binding sites across the transcriptome will be critical to the interpretation of the rapidly emerging data on genetic variation between individuals and how these variations contribute to complex genetic diseases.

Las transcripciones de ARN est&#xE1;n sujetos a la regulaci&#xF3;n de genes postranscripcional que participaron cientos de ARN-prote&#xED;nas de uni&#xF3;n a las pr&#xE1;cticas comerciales restrictivas () y que contienen complejos de microARN-ribonucleoprote&#xED;na (miRNPs) expresados &#x200B;&#x200B;en forma de c&#xE9;lulas de tipo dependiente. Hemos desarrollado un enfoque reticulaci&#xF3;n a base de c&#xE9;lulas para determinar en alta resoluci&#xF3;n y en todo el transcriptoma de los sitios de uni&#xF3;n de las pr&#xE1;cticas comerciales restrictivas y miRNPs celulares. Los sitios reticuladas son reveladas por timidina a las transiciones de citidina en los ADNc preparadas a partir de immunopurified RNPs de 4-tiouridina c&#xE9;lulas tratadas. Se determinaron los sitios de uni&#xF3;n y las consecuencias reglamentarias para varias pr&#xE1;cticas comerciales restrictivas intensamente estudiados y miRNPs, incluyendo PUM2, QKI, IGF2BP1 3, AGO/EIF2C1-4 y TNRC6A-C. Nuestro estudio revel&#xF3; que estos factores se unen miles de sitios que contengan la secuencia de motivos definidos y tienen distintas preferencias de exonic contra intr&#xF3;nica o codificaci&#xF3;n en comparaci&#xF3;n con las regiones no traducidas de transcripci&#xF3;n. La asignaci&#xF3;n exacta de los sitios de uni&#xF3;n de todo el transcriptoma ser&#xE1; fundamental para la interpretaci&#xF3;n de los datos que emergen r&#xE1;pidamente sobre la variaci&#xF3;n gen&#xE9;tica entre los individuos y c&#xF3;mo estas variaciones contribuyen a enfermedades gen&#xE9;ticas complejas.
