Bidirectional trafficking of macromolecules between the cytoplasm and the nucleus is mediated by the nuclear pore complexes (NPCs) embedded in the nuclear envelope (NE) of eukaryotic cell. The NPC functions as the sole pathway to allow for the passive diffusion of small molecules and the facilitated translocation of larger molecules. Evidence shows that these two transport modes and the conformation of NPC can be regulated by calcium stored in the lumen of nuclear envelope and endoplasmic reticulum. However, the mechanism of calcium regulation remains poorly understood. In this review, we integrate data on the observations of calciumregulated structure and function of the NPC over the past years. Furthermore, we highlight challenges in the measurements of dynamic conformational changes and transient transport kinetics in the NPC. Finally, an innovative imaging approach, single-molecule superresolution fluorescence microscopy, is introduced and expected to provide more insights into the mechanism of calcium-regulated nucleocytoplasmic transport.

Le trafic bidirectionnel des macromol&#xE9;cules entre le cytoplasme et le noyau est m&#xE9;di&#xE9;e par les complexes des pores nucl&#xE9;aires (NPCs) embarqu&#xE9;s dans l&#x26;#39;enveloppe nucl&#xE9;aire (NE) de la cellule eucaryote. Les fonctions NPC que la seule voie pour permettre la diffusion passive de petites mol&#xE9;cules et de la translocation de mol&#xE9;cules plus grandes facilit&#xE9;. L&#x26;#39;exp&#xE9;rience montre que ces deux modes de transport et de la conformation de l&#x26;#39;APN peut &#xEA;tre r&#xE9;gul&#xE9;e par le calcium stock&#xE9; dans la lumi&#xE8;re de l&#x26;#39;enveloppe nucl&#xE9;aire et le r&#xE9;ticulum endoplasmique. Cependant, le m&#xE9;canisme de la r&#xE9;gulation du calcium reste mal comprise. Dans cette revue, nous int&#xE9;grons des donn&#xE9;es sur les observations de la structure et la fonction calciumregulated de l&#x26;#39;APN au cours des derni&#xE8;res ann&#xE9;es. En outre, nous mettons en &#xE9;vidence les d&#xE9;fis dans les mesures de dynamiques des changements conformationnels et de la cin&#xE9;tique de transport passagers dans le NPC. Bidirektionale Menschenhandel Makromolek&uuml;le zwischen Zytoplasma und Zellkern ist durch die nukleare Pore-komplexe (NPCs) eingebettet in die Kernh&uuml;lle (NE) der eukaryotischen Zelle vermittelt. Die NPC fungiert als der einzige Weg, um die passive Diffusion kleiner Molek&uuml;le und der erleichterten Translokation von gr&ouml;&szlig;eren Molek&uuml;len erm&ouml;glichen. Belege daf&uuml;r, dass diese zwei Verkehrstr&auml;ger und die Gestaltung des NPC von Kalzium in das Lumen der Kernh&uuml;lle und Endoplasmatisches Retikulum gespeicherten geregelt werden k&ouml;nnen. Der Mechanismus der Kalzium-Verordnung bleibt jedoch weitgehend unverstanden. In diesem Bericht integrieren wir Daten &uuml;ber die Beobachtungen der Calciumregulated-Struktur und Funktion des NPC in den letzten Jahren. Dar&uuml;ber hinaus beleuchten wir Herausforderungen in den Messungen des dynamischen Konformationsver&auml;nderungen und Transienten Transport Kinetik in der NPC. Schlie&szlig;lich einen innovativen bildgebenden Ansatz, Einzelmolek&uuml;l-Superresolution Fluoreszenzmikroskopie, eingef&uuml;hrt und an weitere Einblicke in den Mechanismus der Kalzium-regulierten Nucleocytoplasmic Transport.

El tr&#xE1;fico bidireccional de macromol&#xE9;culas entre el citoplasma y el n&#xFA;cleo est&#xE1; mediada por los complejos de poro nuclear (CPN) incrustados en la envoltura nuclear (NE) de la c&#xE9;lula eucariota. Las funciones NPC como la v&#xED;a &#xFA;nica para permitir la difusi&#xF3;n pasiva de mol&#xE9;culas peque&#xF1;as y la translocaci&#xF3;n facilitado de mol&#xE9;culas m&#xE1;s grandes. La evidencia indica que estos dos modos de transporte y la conformaci&#xF3;n de NPC puede ser regulada por calcio almacenado en el lumen de envoltura nuclear y el ret&#xED;culo endopl&#xE1;smico. Sin embargo, el mecanismo de regulaci&#xF3;n del calcio sigue siendo poco conocida. En esta revisi&#xF3;n, se integran los datos de las observaciones de la estructura y la funci&#xF3;n calciumregulated de la APN en los &#xFA;ltimos a&#xF1;os. Por otra parte, destacamos los desaf&#xED;os en la medici&#xF3;n de los cambios din&#xE1;micos en la conformaci&#xF3;n y la cin&#xE9;tica transitoria de transporte en la APN. Por &#xFA;ltimo, un enfoque innovador de la imagen, una sola mol&#xE9;cula de superresoluci&#xF3;n microscop&#xED;a de fluorescencia, se introduce y se espera que proporcione mayor informaci&#xF3;n sobre el mecanismo de transporte regulada por calcio nucleocytoplasmic.
