Mycobacterium avium subsp. paratuberculosis (M. paratuberculosis) enters intestinal epithelial cells of cattle and other ruminants via a mechanism that remains to be fully elucidated. This study showed that a gene encoding the M. paratuberculosis 35 kDa major membrane protein (MMP) is expressed at a higher level in low-oxygen and high-osmolarity conditions that are similar to the environment of the intestine. In addition, cattle with Johne's disease produced antibodies against MMP, suggesting that the protein is present during infection. The gene encoding MMP was cloned and expressed as a fusion protein with the maltose-binding protein (MBP-MMP) in Escherichia coli. Rabbit antisera were raised against a M. paratuberculosis whole-cell sonicate and MMP-specific antibodies were purified from these sera by affinity chromatography. MMP was localized to the surface of M. paratuberculosis by immunoelectron microscopy and by immunoblot analysis of fractionated protein lysates. Both anti-MMP antibodies and MBP-MMP protein inhibited M. paratuberculosis invasion of cultured Madin-Darby bovine kidney cells by 30 %. In similar invasion experiments with M. paratuberculosis incubated in low oxygen tension, these antibodies and protein decreased invasion by 60 %. Collectively, these data show that the 35 kDa MMP is a surface exposed protein that plays a role in invasion of epithelial cells. The authors suggest that the MMP is a virulence factor of M. paratuberculosis that may be important in the initiation of infection in vivo.

Francisella tularensis is a gram-negative coccobacillus that is capable of causing severe, fatal disease in a number of mammalian species, including humans. Little is known about the proteins that are surface exposed on the outer membrane (OM) of F. tularensis, yet identification of such proteins is potentially fundamental to understanding the initial infection process, intracellular survival, virulence, immune evasion and, ultimately, vaccine development. To facilitate the identification of putative F. tularensis outer membrane proteins (OMPs), the genomes of both the type A strain (Schu S4) and type B strain (LVS) were subjected to six bioinformatic analyses for OMP signatures. Compilation of the bioinformatic predictions highlighted 16 putative OMPs, which were cloned and expressed for the generation of polyclonal antisera. Total membranes were extracted from both Schu S4 and LVS by spheroplasting and osmotic lysis, followed by sucrose density gradient centrifugation, which separated OMs from cytoplasmic (inner) membrane and other cellular compartments. Validation of OM separation and enrichment was confirmed by probing sucrose gradient fractions with antibodies to putative OMPs and inner membrane proteins. F. tularensis OMs typically migrated in sucrose gradients between densities of 1.17 and 1.20 g/ml, which differed from densities typically observed for other gram-negative bacteria (1.21 to 1.24 g/ml). Finally, the identities of immunogenic proteins were determined by separation on two-dimensional sodium dodecyl sulfate-polyacrylamide gel electrophoresis and mass spectrometric analysis. This is the first report of a direct method for F. tularensis OM isolation that, in combination with computational predictions, offers a more comprehensive approach for the characterization of F. tularensis OMPs.

Francisella Tularensis ist ein gramnegatives Coccobacillus, das schwere, t&ouml;dlich in eine Reihe von Arten von S&auml;ugetieren, einschlie&szlig;lich Menschen hervorrufen kann. Wenig ist &uuml;ber die Proteine, die Fl&auml;che auf die &auml;u&szlig;ere Membran (OM) F. Tularensis sind bekannt, doch Identifizierung von solchen Proteinen ist potenziell grundlegend f&uuml;r das Verst&auml;ndnis der Erstinfektion Prozess, intrazellul&auml;re &uuml;berleben, Virulenz, immun ausweichen und letztlich Impfstoffentwicklung. Zur Erleichterung der Identifizierung von vermeintlichen F. Tularensis &auml;u&szlig;ere Membranproteine (OMPs), die Genome der Typ A-Stamm (Schu S4) und Typ B-Stamm wurden sechs bioinformatische Analysen f&uuml;r OMP Signaturen (LVS) unterzogen. Zusammenstellung der bioinformatische Vorhersagen hervorgehoben 16 vermeintliche OMPs, die geklont und zur Erzeugung von polyklonale Antiseren ausgedr&uuml;ckt wurden. Gesamte Membranen wurden extrahiert aus Schu S4 und LVS durch Spheroplasting und osmotische Lyse, gefolgt von Saccharose Dichte Gradienten Zentrifugation, die OMs aus andere zellulare F&auml;cher und zytoplasmatischen (innere) Membran getrennt. Validierung von OM Trennung und Bereicherung wurde durch Sondierung Saccharose gradient Br&uuml;che mit Antik&ouml;rper gegen vermeintliche OMPs und inneren Membranproteinen best&auml;tigt. F. Tularensis OMs migriert in der Regel in Saccharose Steigungen zwischen dichten von 1.17 und 1,20 g/ml, differierten von dichten, die in der Regel f&uuml;r andere gramnegativen Bakterien (1.21 auf 1,24 g/ml) beobachtet. Tularensis OMPs bietet.

Francisella tularensis &egrave; un coccobacillo Gram-negativo che &egrave; in grado di causare malattia grave, fatale in un certo numero di specie di mammiferi, inclusi gli esseri umani. Poco si sa circa le proteine che sono superficie esposti sulla membrana esterna (OM) di F. tularensis, ancora potenzialmente fondamentali per comprendere il processo di infezione iniziale, la sopravvivenza intracellulare, la virulenza, l&#39;identificazione di tali proteine &egrave; evasione immune e, in ultima analisi, lo sviluppo del vaccino. Francisella tularensis is a gram-negative intracellular bacterium and the causative agent of the zoonotic disease tularemia. F. tularensis is a category A select agent and thus a potential agent of bioterrorism. Whereas an F. tularensis live, attenuated vaccine strain (LVS) is the basis of an investigational vaccine, this vaccine is not licensed for human use because of efficacy and safety concerns. In the present study, we immunized mice with isolated native outer membrane proteins (OMPs), ethanol-inactivated LVS (iLVS), or purified LVS lipopolysaccharide (LPS) and assessed the ability of each vaccine preparation to protect mice against pulmonary challenge with the virulent type A F. tularensis strain SchuS4. Antibody isotyping indicated that both Th1 and Th2 antibody responses were generated in mice after immunization with OMPs or iLVS, whereas LPS immunization resulted in only immunoglobulin A production. In survival studies, OMP immunization provided the greatest level of protection (50% survival at 20 days after infection with SchuS4), and there were associated 3-log reductions in the spleen and liver bacterial burdens (compared to nonvaccinated mice). Cytokine quantitation for the sera of SchuS4-challenged mice indicated that OMP and iLVS immunizations induced high levels of tumor necrosis factor alpha and interleukin-2 (IL-2) production, whereas only OMP immunization induced high levels of IL-10 production. By comparison, high levels of proinflammatory cytokines, including RANTES, granulocyte colony-stimulating factor, IL-6, IL-1alpha, IL-12p40, and KC, in nonvaccinated mice indicated that these cytokines may facilitate disease progression. Taken together, the results of this study demonstrate the potential utility of an OMP subunit (acellular) vaccine for protecting mammals against type A F. tularensis.

Francisella tularensis es una bacteria intracelular gram-negativa y el agente causante de la tularemia enfermedad zoon&#xF3;tica. F. tularensis es una categor&#xED;a de un agente de selecci&#xF3;n y por lo tanto un posible agente de bioterrorismo. Considerando que F. tularensis en vivo, vacuna contra la cepa atenuada (LVS) es la base de una vacuna en investigaci&#xF3;n, esta vacuna no est&#xE1; aprobada para uso humano debido a las preocupaciones de seguridad y eficacia. En el presente estudio, se inmunizaron ratones con las prote&#xED;nas nativas aisladas de membrana externa (OMP), las inactivadas etanol-LVS (ILVS), o lipopolisac&#xE1;rido purificado LVS (LPS) y se evalu&#xF3; la capacidad de cada preparaci&#xF3;n de la vacuna para proteger a los ratones contra el desaf&#xED;o pulmonar con la virulenta tipo A, cepa F. tularensis SchuS4. Isotipado anticuerpos indicaron que tanto las respuestas de anticuerpos Th1 y Th2 se generaron en ratones despu&#xE9;s de la inmunizaci&#xF3;n con OMPs o ILVS, mientras que la inmunizaci&#xF3;n LPS dio lugar a una producci&#xF3;n &#xFA;nica inmunoglobulina. En estudios de supervivencia, la inmunizaci&#xF3;n OMP siempre el mayor nivel de protecci&#xF3;n (50% de supervivencia a 20 d&#xED;as despu&#xE9;s de la infecci&#xF3;n con el SchuS4), y no se asociaron 3-registro de las reducciones en el bazo y las cargas bacterianas del h&#xED;gado (en comparaci&#xF3;n con los ratones no vacunados). Cuantificaci&#xF3;n de citoquinas para los sueros de los ratones SchuS4-impugnadas indic&#xF3; que OMP y vacunas ILVS inducida altos niveles de factor de necrosis tumoral y la interleucina 2 (IL-2) de producci&#xF3;n, mientras que s&#xF3;lo la inmunizaci&#xF3;n OMP inducida altos niveles de IL-10. Francisella species secrete a polycarboxylate siderophore that resembles rhizoferrin to acquire ferric iron. Several of the Francisella siderophore synthesis genes are contained in a Fur-regulated operon (designated fig or fsl) comprised of at least seven ORFs including fur. Reverse transcriptase-PCR showed transcriptional linkage between figD and figE and between figE and figF. Mutations were constructed in four of these ORFs (figB, figC, figD, and figE) in Francisella novicida U112. All four of these new mutants and a F. novicida figA mutant grew at rates comparable to that of wild type under iron-replete conditions but growth of all five mutants was stunted in iron-limiting media. When ferric rhizoferrin was added to the iron-limited media, growth of the figA, figB, figC, and figD mutants was restored to levels similar to those obtained in iron-replete media. However, this exogenously added siderophore could not rescue the figE mutant. When Chrome Azurol S assays were used to measure siderophore production, the figA, figB, and figC mutants were markedly deficient in their ability to synthesize siderophore whereas the figD and figE mutants produced siderophore at levels equivalent to the wild-type parent strain.

La transcriptasa inversa-PCR mostr&#xF3; una vinculaci&#xF3;n entre la transcripci&#xF3;n y la figD FIGE y entre FIGE y FIGF. Many bacterial pathogens rely on a conserved membrane histidine sensor kinase, QseC, to respond to host adrenergic signaling molecules and bacterial signals in order to promote the expression of virulence factors. Using a high-throughput screen, we identified a small molecule, LED209, that inhibits the binding of signals to QseC, preventing its autophosphorylation and consequently inhibiting QseC-mediated activation of virulence gene expression. LED209 is not toxic and does not inhibit pathogen growth; however, this compound markedly inhibits the virulence of several pathogens in vitro and in vivo in animals. Inhibition of signaling offers a strategy for the development of broad-spectrum antimicrobial drugs.
