The clinical use of stem cells for regenerative medicine is critically dependent on preclinical studies in animal models. In this review we examine some of the key issues and challenges in the use of animal models to study human stem cell biology-experimental standardization, body size, immunological barriers, cell survival factors, fusion of host and donor cells, and in vivo imaging and tracking. We focus particular attention on the various imaging modalities that can be used to track cells in living animals, comparing their strengths and weaknesses and describing technical developments that are likely to lead to new opportunities for the dynamic assessment of stem cell behavior in vivo. We then provide an overview of some of the most commonly used animal models, their advantages and disadvantages, and examples of their use for xenotypic transplantation of human stem cells, with separate reviews of models involving rodents, ungulates, nonhuman primates, and the chicken embryo. As the use of human somatic, embryonic, and induced pluripotent stem cells increases, so too will the range of applications for these animal models. It is likely that increasingly sophisticated uses of human/animal chimeric models will be developed through advances in genetic manipulation, cell delivery, and in vivo imaging.

Dans cette revue, nous examinons certains des principaux enjeux et d&#xE9;fis dans l&#x26;#39;utilisation de mod&#xE8;les animaux pour &#xE9;tudier les cellules souches humaines biologie exp&#xE9;rimentale normalisation, la taille du corps, les barri&#xE8;res immunologiques, des facteurs de survie cellulaire, la fusion de l&#x26;#39;h&#xF4;te et les cellules du donneur, et l&#x26;#39;imagerie in vivo et suivi. L&#39;uso clinico di cellule staminali per la medicina rigenerativa dipende criticamente studi preclinici in modelli animali. In questa recensione noi esaminare alcune questioni chiave e sfide nell&#39;uso di modelli animali per studiare biologia sperimentale standardizzazione sulle cellule staminali umane, dimensione corporea, le barriere immunologiche, fattori di sopravvivenza delle cellule, fusione delle cellule ospite e donatore e imaging in vivo e di rilevamento. Ci concentriamo particolare attenzione sui vari di imaging che pu&ograve; essere utilizzato per tenere traccia delle cellule in animali viventi, confrontando i loro punti di forza e di debolezza e descrivendo gli sviluppi tecnici che potrebbero portare a nuove opportunit&agrave; per la valutazione dinamica del comportamento delle cellule staminali in vivo. El uso cl&#xED;nico de c&#xE9;lulas madre en medicina regenerativa es cr&#xED;ticamente dependiente de los estudios precl&#xED;nicos en modelos animales. En esta revisi&#xF3;n se examinan algunos de los principales problemas y desaf&#xED;os en el uso de modelos animales para estudiar la biolog&#xED;a de c&#xE9;lulas madre humana-experimental de normalizaci&#xF3;n, el tama&#xF1;o del cuerpo, barreras inmunol&#xF3;gicas, factores de supervivencia celular, la fusi&#xF3;n de acogida y las c&#xE9;lulas del donante, y en vivo de im&#xE1;genes y seguimiento. Nos centramos la atenci&#xF3;n en particular sobre las diversas modalidades de imagen que pueden ser utilizados para rastrear las c&#xE9;lulas en animales vivos, la comparaci&#xF3;n de sus puntos fuertes y d&#xE9;biles y la descripci&#xF3;n de los avances t&#xE9;cnicos que puedan dar lugar a nuevas oportunidades para la evaluaci&#xF3;n din&#xE1;mica del comportamiento de c&#xE9;lulas madre in vivo. Proper control of movement and posture occurs partly via descending projections from the vestibular nuclei to spinal motor circuits. Days before birth in rodents, vestibulospinal neurons develop axonal projections that extend to the spinal cord. How functional these projections are just after birth is unknown. Our goal was to assess the overall functional organization of vestibulospinal inputs to spinal motoneurons in a brainstem-spinal cord preparation of the neonatal mouse (postnatal day (P) 0-5). Using calcium imaging, we recorded responses evoked by electrical stimulation of the VIIIth nerve, in many motoneurons simultaneously throughout the spinal cord (C2, C6, T7, L2 and L5 segments), in the medial and lateral motor columns. Selective lesions in the brainstem and/or spinal cord distinguished which tracts contributed to the responses: those in the cervical cord originated primarily from the medial vestibulospinal tracts but with a substantial contribution from the lateral vestibulospinal tract; those in the thoracolumbar cord originated exclusively from the lateral vestibulospinal tract. In the thoracolumbar but not the cervical cord, excitatory commissural connections mediated vestibular responses in contralateral motoneurons. Pharmacological blockade of GABA(A) receptors showed that responses involved a convergence of excitatory and inhibitory inputs which in combination produced temporal response patterns specific for different segmental levels. Our results show that by birth vestibulospinal projections in rodents have already established functional synapses and are organized to differentially regulate activity in neck and limb motoneurons in a tract- and segment-specific pattern similar to that in adult mammals. Thus, this particular set of descending projections develops several key features of connectivity appropriately at prenatal stages. We also present novel information about vestibulospinal inputs to axial motoneurons in mammals, providing a more comprehensive platform for future studies into the overall organization of vestibulospinal inputs and their role in regulating postural stability.

Un bon contr&#xF4;le de mouvement et la posture se produit en partie par ordre d&#xE9;croissant des projections &#xE0; partir des noyaux vestibulaires &#xE0; des circuits moteurs spinaux. Quelques jours avant la naissance chez les rongeurs, les neurones vestibulo d&#xE9;velopper projections axonales qui s&#x26;#39;&#xE9;tendent de la moelle &#xE9;pini&#xE8;re. Comment fonctionnelle de ces projections sont juste apr&#xE8;s la naissance est inconnue. Notre objectif &#xE9;tait d&#x26;#39;&#xE9;valuer l&#x26;#39;organisation fonctionnelle globale des intrants vestibulo &#xE0; motoneurones spinaux dans une pr&#xE9;paration cordon tronc c&#xE9;r&#xE9;bral-spinal de la souris n&#xE9;onatales (jour postnatal (P) 0-5). Dans la thoraco-lombaire, mais pas la moelle cervicale, excitateurs connexions commissurales m&#xE9;diation des r&#xE9;ponses vestibulaires dans les motoneurones controlat&#xE9;raux. Le blocage pharmacologique de GABA (A) a montr&#xE9; que les r&#xE9;cepteurs impliqu&#xE9;s r&#xE9;ponses &#xE0; une convergence des intrants excitateurs et inhibiteurs qui, en combinaison produites sch&#xE9;mas de r&#xE9;ponse temporels sp&#xE9;cifiques pour les diff&#xE9;rents niveaux segmentaires. Nos r&#xE9;sultats montrent que par la naissance des projections vestibulo chez les rongeurs ont d&#xE9;j&#xE0; mis en place des synapses fonctionnelles et sont organis&#xE9;es pour r&#xE9;guler l&#x26;#39;activit&#xE9; diff&#xE9;rentielle dans les motoneurones du cou et des membres dans un mod&#xE8;le des voies et des segments sp&#xE9;cifiques, similaire &#xE0; celle chez les mammif&#xE8;res adultes. Ainsi, cet ensemble particulier de projections descendantes d&#xE9;veloppe plusieurs caract&#xE9;ristiques cl&#xE9;s de la connectivit&#xE9; de fa&#xE7;on appropri&#xE9;e &#xE0; des stades pr&#xE9;natal. Ordnungsgem&auml;&szlig;e Kontrolle der Bewegung und K&ouml;rperhaltung erfolgt teilweise &uuml;ber absteigende Projektionen aus den vestibul&auml;ren Kernen zu spinalen motorischen Schaltungen. Tage vor der Geburt bei Nagern, Vestibulospinal Neuronen entwickeln axonale Projektionen, die auf das R&uuml;ckenmark zu erweitern. Es ist unbekannt, wie diese Projektionen nur nach der Geburt funktionieren. Unser Ziel war es die insgesamt funktionelle Organisation des Vestibulospinal Eingaben f&uuml;r spinal Motoneurons in einer Hirnstamm-R&uuml;ckenmark-Zubereitung der Neugeborenen Maus zu beurteilen (postnatale Tag (P) 0-5). Selektive L&auml;sionen im Hirnstamm und/oder R&uuml;ckenmark zu unterscheiden, welche Landstriche zu Antworten beigetragen: die im zervikalen R&uuml;ckenmark stammen haupts&auml;chlich aus den medialen Vestibulospinal Tracts aber mit wesentlich von seitlichen Vestibulospinal und Darm; diejenigen im thoracolumbar R&uuml;ckenmark stammt ausschlie&szlig;lich aus den seitlichen Vestibulospinal-Trakt. Die thoracolumbar aber nicht am Geb&auml;rmutterhals Kabel vermittelte exzitatorischen kommissurale Verbindungen vestibul&auml;re Reaktionen in den kontralateralen Motoneurons. Pharmakologische Blockade GABA(A) Rezeptoren zeigte, dass Antworten eine Konvergenz von exzitatorischen und inhibitorischen beteiligt Eing&auml;nge die in Kombination produziert zeitliche Antwort Muster speziell f&uuml;r segmentale Ebenen. Unsere Ergebnisse zeigen, dass durch Geburt Vestibulospinal Projektionen bei Nagern funktionale Synapsen bereits haben und angeordnet, differentiell T&auml;tigkeit in Hals und Gliedma&szlig;en Motoneurons in einem Trakt und Segment-spezifische Muster &auml;hnlich wie bei Erwachsenen S&auml;ugetieren zu Regeln. So, entsteht diese besondere Gruppe von absteigende Projektionen mehrere wichtige Features der Konnektivit&auml;t entsprechend in vorgeburtlichen Stadien. Wir pr&auml;sentieren auch neuartige Informationen &uuml;ber Vestibulospinal Eingaben f&uuml;r axiale Motoneurons bei S&auml;ugetieren, eine umfassendere Plattform f&uuml;r zuk&uuml;nftige Studien in der Gesamtorganisation der Vestibulospinal ein- und ihre Rolle bei der Regulierung posturale Stabilit&auml;t.

Corretto controllo della postura e del movimento avviene parzialmente attraverso decrescente proiezioni dai nuclei vestibolari ai circuiti motori spinali. Giorni prima della nascita nei roditori, i neuroni vestibulospinal sviluppano proiezioni assonali che si estendono dal midollo spinale. Come funzionale queste proiezioni sono appena dopo la nascita &egrave; sconosciuto. Il nostro obiettivo &egrave; stato quello di valutare l&#39;organizzazione nel complesso funzionale di vestibulospinal ingressi ai motoneuroni spinali in preparazione del tronco cerebrale-spinale del mouse neonatale (giorno postnatale (P) 0-5). Utilizzando immagini di calcio, abbiamo registrato risposte evocate dalla stimolazione elettrica del nervo VIII, in tanti motoneuroni simultaneamente in tutto il midollo spinale (segmenti C2, C6, T7, L2 e L5), nelle colonne motore mediale e laterale. Selettive lesioni del tronco encefalico e/o del midollo spinale distinguono che tratti contribuirono per le risposte: quelli del midollo cervicale ha originati principalmente da tratti vestibulospinal mediale, ma con un sostanza contributo nel tratto di vestibulospinal laterale; quelli del midollo toraco originato esclusivamente dal tratto vestibulospinal laterale. Il toraco ma non il midollo cervicale, connessioni eccitatorie commissural mediata risposte vestibolari nei motoneuroni controlaterale. Blocco farmacologico dei recettori GABA(A) ha mostrato che le risposte coinvolto una convergenza di inibitori ed eccitatori che ingressi in combinazione prodotto risposta temporale modelli specifici per i diversi livelli segmentali. I nostri risultati mostrano che dalla nascita vestibulospinal proiezioni nei roditori hanno gi&agrave; stabilito sinapsi funzionali e sono organizzate per differenzialmente regolare attivit&agrave; nel collo e arto motoneuroni in un modello del tratto e segmento specifico simile a quello nei mammiferi adulti. Cos&igrave;, questo particolare insieme di decrescente proiezioni si sviluppa diverse caratteristiche chiave di connettivit&agrave; in modo appropriato in fase prenatale. Siamo presenti anche romanzo informazioni su vestibulospinal ingressi ai motoneuroni assiali nei mammiferi, fornendo una piattaforma pi&ugrave; completa per gli studi futuri nell&#39;organizzazione complessiva della vestibulospinal ingressi e il loro ruolo nella regolazione della stabilit&agrave; posturale.

El control apropiado de movimiento y la postura se produce en parte a trav&#xE9;s descendente proyecciones de los n&#xFA;cleos vestibulares a los circuitos motoras espinales. D&#xED;as antes de su nacimiento en roedores, las neuronas vestibuloespinales desarrollar proyecciones axonales que se extienden a la m&#xE9;dula espinal. &#xBF;C&#xF3;mo funcional de estas proyecciones son s&#xF3;lo despu&#xE9;s de su nacimiento es desconocida. Nuestro objetivo fue evaluar la organizaci&#xF3;n funcional global de entradas a vestibuloespinales motoneuronas espinales en una preparaci&#xF3;n de la m&#xE9;dula del tronco cerebral-espinal del rat&#xF3;n reci&#xE9;n nacido (d&#xED;a postnatal (P) 0-5). Utilizando im&#xE1;genes de calcio, se registraron las respuestas evocadas por la estimulaci&#xF3;n el&#xE9;ctrica del nervio VIII, en las neuronas motoras de manera simult&#xE1;nea a lo largo de la m&#xE9;dula espinal (C2, C6, T7, L2 y L5 segmentos), en las columnas de motor medial y lateral. Las lesiones selectivas en el tronco cerebral y / o la m&#xE9;dula espinal que distinguen extensiones contribuido a las respuestas: las de la m&#xE9;dula cervical se origin&#xF3; principalmente de las v&#xED;as mediales vestibuloespinales pero con una contribuci&#xF3;n sustancial de la parte lateral del tracto vestibuloespinal, los de la m&#xE9;dula toracolumbar se origin&#xF3; exclusivamente de la lateral de las v&#xED;as vestibuloespinal. Por lo tanto, este conjunto de proyecciones descendentes desarrolla varias caracter&#xED;sticas clave de la conectividad adecuada en las etapas prenatales. Asimismo, la informaci&#xF3;n actual sobre la novela de entradas vestibuloespinales a las neuronas motoras axiales en los mam&#xED;feros, proporcionando una plataforma m&#xE1;s amplia para futuros estudios en la organizaci&#xF3;n general de los insumos vestibuloespinales y su papel en la regulaci&#xF3;n de la estabilidad postural.
