Complete lack of transcription factor PDX-1 leads to pancreatic agenesis, whereas heterozygosity for PDX-1 mutations has been recently noted in some individuals with maturity-onset diabetes of the young (MODY) and in some individuals with type 2 diabetes. To determine how alterations in PDX-1 affect islet function, we examined insulin secretion and islet physiology in mice with one PDX-1 allele inactivated. PDX-1(+/-) mice had a normal fasting blood glucose and pancreatic insulin content but had impaired glucose tolerance and secreted less insulin during glucose tolerance testing. The expression of PDX-1 and glucose transporter 2 in islets from PDX-1(+/-) mice was reduced to 68 and 55%, respectively, whereas glucokinase expression was not significantly altered. NAD(P)H generation in response to glucose was reduced by 30% in PDX-1(+/-) mice. The in situ perfused pancreas of PDX-1(+/-) mice secreted about 45% less insulin when stimulated with 16.7 mm glucose. The K(m) for insulin release was similar in wild type and PDX-1(+/-) mice. Insulin secretion in response to 20 mm arginine was unchanged; the response to 10 nm glucagon-like peptide-1 was slightly increased. However, insulin secretory responses to 10 mm 2-ketoisocaproate and 20 mm KCl were significantly reduced (by 61 and 66%, respectively). These results indicate that a modest reduction in PDX-1 impairs several events in glucose-stimulated insulin secretion (such as NAD(P)H generation, mitochondrial function, and/or mobilization of intracellular Ca(2+)) and that PDX-1 is important for normal function of adult pancreatic islets.

In muscle, insulin enhances influx of glucose and its conversion to glucose 6-phosphate (G6P) by hexokinase (HK). While effects of insulin on glucose transport have been demonstrated, its effect on the activity of HK of cells has not. In L6 myotubes treated for 24 h with insulin there was increased expression of the HK isoform, HKII, and increased glucose phosphorylation without a concomitant increase in glucose transport, indirectly suggesting that phosphorylation of glucose was a target of insulin action [Osawa, Printz, Whitesell and Granner (1995) Diabetes 44, 1426-1432]. In the present work the same treatment led to a 2-fold rise in G6P, suggesting that transport and/or HK were important targets of insulin action. We used a method to identify the site of rate control involving the specificity of phosphorylation towards 2-deoxy-[1-14C]glucose and D-[2-3H]glucose. Glucose transport does not greatly discriminate between these two tracers while HK shows increased specificity for glucose. Specificity of the glucose phosphorylation of the cells increased with addition of insulin and when extracellular glucose was raised. Specificity was reduced at low glucose concentrations or when the inhibitor of transport, cytochalasin B, was added. We conclude that transport and HK share nearly equal control over glucose phosphorylation in these cells. A computer program was used to test models for compatibility with the different types of experiments. The predicted intracellular glucose and transport rates associated with phosphorylation activity were lower than their measured values for the whole cell. In the most likely model, 15+/-4% of the glucose transporters serve a proportionate volume of the cytoplasm. Insulin activation of glucose phosphorylation might then result from stimulation of these transporters together with HK recruitment or relief from inhibition by G6P.

Alors que les effets de l&#x26;#39;insuline sur le transport du glucose ont &#xE9;t&#xE9; d&#xE9;montr&#xE9;es, son effet sur l&#x26;#39;activit&#xE9; de HK de cellules n&#x26;#39;a pas. Dans L6 myotubes trait&#xE9;s pendant 24 h avec de l&#x26;#39;insuline il a &#xE9;t&#xE9; augmentation de l&#x26;#39;expression de l&#x26;#39;isoforme HK, HKII, et la phosphorylation accrue de glucose, sans une augmentation concomitante de transport du glucose, de mani&#xE8;re indirecte ce qui sugg&#xE8;re que la phosphorylation du glucose a &#xE9;t&#xE9; une cible de l&#x26;#39;insuline [Osawa, Printz, Whitesell et Granner (1995) du diab&#xE8;te 44, 1426-1432]. Dans le pr&#xE9;sent travail le m&#xEA;me traitement a conduit &#xE0; une augmentation de 2 fois en G6P, ce qui sugg&#xE8;re que le transport et / ou HK sont des cibles importantes de l&#x26;#39;insuline. Nous avons utilis&#xE9; une m&#xE9;thode pour identifier le site de contr&#xF4;le du taux de participation de la sp&#xE9;cificit&#xE9; de la phosphorylation vers 2-d&#xE9;soxy-[1-14C] glucose et du D-[2-3H] glucose. Le transport du glucose n&#x26;#39;a pas grandement la distinction entre ces deux traceurs tout HK pr&#xE9;sente une sp&#xE9;cificit&#xE9; accrue pour le glucose. Sp&#xE9;cificit&#xE9; de la phosphorylation du glucose des cellules a augment&#xE9; avec l&#x26;#39;ajout de l&#x26;#39;insuline et de glucose extracellulaire lorsque a &#xE9;t&#xE9; soulev&#xE9;e. La sp&#xE9;cificit&#xE9; a &#xE9;t&#xE9; r&#xE9;duite &#xE0; des concentrations de glucose faible ou lorsque l&#x26;#39;inhibiteur du transport, la cytochalasine B, a &#xE9;t&#xE9; ajout&#xE9;. Nous concluons que le transport et la part HK contr&#xF4;le &#xE0; peu pr&#xE8;s &#xE9;gale sur la phosphorylation du glucose dans ces cellules. Un programme d&#x26;#39;ordinateur a &#xE9;t&#xE9; utilis&#xE9; pour tester la compatibilit&#xE9; avec les mod&#xE8;les pour les diff&#xE9;rents types d&#x26;#39;exp&#xE9;riences. Le glucose intracellulaire et pr&#xE9;dit des taux de transport associ&#xE9;s &#xE0; l&#x26;#39;activit&#xE9; de phosphorylation ont &#xE9;t&#xE9; plus faibles que leurs valeurs mesur&#xE9;es pour la cellule enti&#xE8;re. Dans le mod&#xE8;le le plus probable, 15% + / -4 des transporteurs de glucose servir un volume proportionnel du cytoplasme. Nel muscolo, insulina aumenta afflusso di glucosio e la sua conversione a glucosio-6-fosfato (G6P) da esochinasi (HK). Mentre sono stati dimostrati effetti dell&#39;insulina sul trasporto di glucosio, suo effetto sull&#39;attivit&agrave; della HK di cellule non ha. In myotubes L6 trattati per 24 h con insulina c&#39;era aumentata espressione della isoforma HK, HKII e fosforilazione di glucosio aumentato senza un concomitante aumento del trasporto di glucosio, suggerendo indirettamente che la fosforilazione del glucosio &egrave; stato un bersaglio di azione di insulina [Osawa, Printz, Whitesell e Granner (1995) diabete 44, 1426-1432]. Nel presente lavoro lo stesso trattamento ha portato ad un aumento di 2 volte a G6P, suggerendo che il trasporto e/o HK erano obiettivi importanti di azione di insulina. Abbiamo usato un metodo per identificare il sito di controllo di tasso che coinvolgono la specificit&agrave; di fosforilazione verso 2 - deossi-[1-14 C] glucosio e glucosio D-[2-3 H]. Trasporto di glucosio notevolmente non discrimina tra questi due elementi traccianti mentre HK Mostra maggiore specificit&agrave; per il glucosio. Specificit&agrave; della fosforilazione di glucosio delle cellule &egrave; aumentato con l&#39;aggiunta di insulina e quando &egrave; stato generato il glucosio extracellulare. Specificit&agrave; &egrave; stata ridotta a concentrazioni di glucosio basso o quando l&#39;inibitore del trasporto, citocalasina B, &egrave; stato aggiunto. Possiamo concludere che il trasporto e HK condividono quasi uguale controllo sulla fosforilazione di glucosio in queste cellule. Un programma per computer &egrave; stato utilizzato per testare i modelli per la compatibilit&agrave; con i diversi tipi di esperimenti. Il predetto glucosio intracellulare e tassi di trasporto connessi con attivit&agrave; di fosforilazione sono stati inferiori a loro valori misurati per l&#39;intera cellula. Nel modello pi&ugrave; probabile, 15 + /-4% dei trasportatori di glucosio servono un volume proporzionato del citoplasma. Attivazione di insulina di fosforilazione di glucosio quindi potrebbe derivare dalla stimolazione di questi trasportatori insieme al reclutamento di HK o sollievo dall&#39;inibizione da G6P.

Pancreatic beta cells secrete insulin in response to changes in the extracellular glucose. However, prolonged exposure to elevated glucose exerts toxic effects on beta cells and results in beta cell dysfunction and ultimately beta cell death (glucose toxicity). To investigate the mechanism of how increased extracellular glucose is toxic to beta cells, we used two model systems where glucose metabolism was increased in beta cell lines by enhancing glucokinase (GK) activity and exposing cells to physiologically relevant increases in extracellular glucose (3.3-20 mm). Exposure of cells with enhanced GK activity to 20 mm glucose accelerated glycolysis, but reduced cellular NAD(P)H and ATP, caused accumulation of intracellular reactive oxygen species (ROS) and oxidative damage to mitochondria and DNA, and promoted apoptotic cell death. These changes required both enhanced GK activity and exposure to elevated extracellular glucose. A ROS scavenger partially prevented the toxic effects of increased glucose metabolism. These results indicate that increased glucose metabolism in beta cells generates oxidative stress and impairs cell function and survival; this may be a mechanism of glucose toxicity in beta cells. The level of beta cell GK may also be critical in this process.

Las c&#xE9;lulas beta pancre&#xE1;ticas secretar insulina en respuesta a los cambios en la glucosa extracelular. Sin embargo, la exposici&#xF3;n prolongada a niveles elevados de glucosa tiene efectos t&#xF3;xicos sobre las c&#xE9;lulas beta y los resultados en la disfunci&#xF3;n de las c&#xE9;lulas beta y la muerte celular en &#xFA;ltima instancia, beta (toxicidad de la glucosa). Para investigar el mecanismo de c&#xF3;mo glucosa extracelular mayor es t&#xF3;xico para las c&#xE9;lulas beta, se utilizaron dos sistemas modelo donde metabolismo de la glucosa se increment&#xF3; en l&#xED;neas de c&#xE9;lulas beta mediante la mejora de la glucoquinasa (GK) actividad y exponer las c&#xE9;lulas a los aumentos fisiol&#xF3;gicamente relevantes en la glucosa extracelular (3.3-20 mm). La exposici&#xF3;n de las c&#xE9;lulas con mayor actividad de GK a 20 mM de glucosa gluc&#xF3;lisis acelerada, pero redujo celular de NAD (P) H y el ATP, provoc&#xF3; la acumulaci&#xF3;n intracelular de especies reactivas del ox&#xED;geno (ROS) y el da&#xF1;o oxidativo a la muerte celular y el ADN mitocondrial, y promovi&#xF3; la apoptosis. Estos cambios se requiere tanto la actividad de GK mejorada y la exposici&#xF3;n a la glucosa extracelular elevada. Un captador de ROS parcialmente impedido que los efectos t&#xF3;xicos del metabolismo de la glucosa incrementada. Estos resultados indican que el metabolismo de la glucosa aument&#xF3; en las c&#xE9;lulas beta genera estr&#xE9;s oxidativo y altera la funci&#xF3;n celular y la supervivencia, lo que puede ser un mecanismo de toxicidad de la glucosa en las c&#xE9;lulas beta. El nivel de GK c&#xE9;lulas beta tambi&#xE9;n puede ser cr&#xED;tico en este proceso.

Galectins are a family of proteins that share an affinity for beta-galactoside containing glycoconjugates. In prostate, ovarian and breast cancer, downregulation of galectin-3 is associated with malignancy and tumor progression. Kaposi's sarcoma (KS) is characterized as an angioproliferative tumor of vascular endothelial cells and produces rare B cell lymphoproliferative diseases in the form of primary effusion lymphomas and some forms of multicentric Castleman's disease. Kaposi's sarcoma-associated herpesvirus (KSHV) is the etiological agent of KS. We found reduced levels of galectin-3 expression in a significant fraction of latency-associated nuclear antigen (LANA)-positive spindle cell regions in human archival KS tissue and as measured in KS tissue microarrays. Here we demonstrate that galectin-3 protein expression is downregulated 10-fold in 10-day KSHV-infected dermal microvascular endothelial cells (DMVEC) accompanied by downregulation of message. There is loss of galectin-3 staining in KSHV-infected DMVEC by dual labeled immunohistochemistry in LANA-positive spindle cells. We observed a consistent downregulation of galectin-3 by time-course transcriptional analysis. Of the galectins assayed, only galectin-1 was also downregulated in KSHV-infected DMVEC. We examined 86 KS tumors; 19 were LANA positive (22%) and 67 LANA negative (78%). All 86 tumors were found to be galectin-3 positive; 11 of 19 showed reduced expression of galectin-3 in LANA-positive spindle cell regions. Our data suggest that KSHV vFLIP and LANA are the viral genes targeting galectin-3 downregulation. The contribution of host factors to the pathogenesis of KS is essential for early detection and development of innovative therapies for treatment.

Galectinas son una familia de prote&#xED;nas que comparten una afinidad para el beta-galact&#xF3;sido glicoconjugados que contienen. En la pr&#xF3;stata, ovario y c&#xE1;ncer de mama, regulaci&#xF3;n a la baja de la galectina-3 se asocia con enfermedad maligna y la progresi&#xF3;n tumoral. El sarcoma de Kaposi (SK) se caracteriza por ser un tumor angioproliferativa de c&#xE9;lulas endoteliales vasculares y produce enfermedades raras linfoproliferativos de c&#xE9;lulas B en la forma de los linfomas de efusi&#xF3;n primaria y algunas formas de enfermedad de Castleman multic&#xE9;ntrica. El sarcoma de Kaposi asociado a virus del herpes (HVSK) es el agente etiol&#xF3;gico del SK. Hemos encontrado niveles reducidos de la galectina-3 en una fracci&#xF3;n significativa de la latencia asociada a ant&#xED;geno nuclear (LANA)-positivos regiones de c&#xE9;lulas fusiformes en el tejido humano de archivo KS y como se mide en microarrays de tejido de los sarcomas. Kaposi's sarcoma is an angioproliferative tumor caused by Kaposi's sarcoma-associated herpesvirus (KSHV) infection of vascular endothelial cells. Fibulins, proteins that associate with extracellular matrix (ECM) proteins, may have both tumor-suppressive and oncogenic activities. We found that the expression of fibulin-2 protein and mRNA were decreased 50-fold and 26-fold, respectively, in 10-day KSHV-infected dermal microvascular endothelial cells (DMVEC). Using quantitative RT-PCR, we found a fivefold and 25-fold decrease of fibulin-2 extracellular matrix binding partners, fibronectin and tropoelastin, respectively. Time-course transcriptional analyses over 10 days showed that in addition to that of fibulin-2, expression of fibulins 3 and 5 was decreased in KSHV-infected DMVEC, fibulins 1C/1D were increased, and fibulins 4, 6, and 7 were unchanged. KSHV latency-associated nuclear antigen (LANA) transcription levels rose consistently over the same period. Addition of recombinant fibulin-3 or -5 for 48 hours to 10-day KSHV-infected cells caused a suppression of KSHV-induced vascular endothelial growth factor (VEGF) protein and mRNA levels. Recombinant fibulin-3 also significantly reduced VEGF receptor 3 expression. In pleural effusion lymphoma cell lines that express variable levels of KSHV lytic replication, we observed no detectable fibulin-2 or -5 expression. Finally, fibulin-2 expression was decreased in tissue microarrays from KSHV-infected, LANA-positive patient cells as compared to that in patient nontumor controls. Understanding the interactions between KSHV and the fibulins may lead to the development of novel therapies for treatment of Kaposi's sarcoma.

KSHV regulation of fibulin-2 in Kaposi's sarcoma: implications for tumorigenesis.

Le sarcome de Kaposi est une tumeur angioproliferative provoqu&#xE9;e par le sarcome de Kaposi associ&#xE9; au virus de l&#x26;#39;herp&#xE8;s (KSHV) l&#x26;#39;infection des cellules endoth&#xE9;liales vasculaires. Bacterial vaginosis (BV), a common condition seen in premenopausal women, is associated with preterm labor, pelvic inflammatory disease, and delivery of low birth weight infants. Gardnerella vaginalis is the predominant bacterial species associated with BV, although its exact role in the pathology of BV is unknown. Using immunofluorescence, confocal and transmission electron microscopy, we found that VK2 vaginal epithelial cells take up G. vaginalis after exposure to the bacteria. Confocal microscopy also indicated the presence of internalized G. vaginalis within vaginal epithelial cells obtained from a subject with BV. Using VK2 cells and (35)S labeled bacteria in an invasion assay, we found that a 1Â h uptake of G. vaginalis was 21.8-fold higher than heat-killed G. vaginalis, 84-fold compared to Lactobacillus acidophilus and 6.6-fold compared to Lactobacillus crispatus. Internalization was inhibited by pre-exposure of cells to cytochalasin-D. In addition, the cytoskeletal protein vimentin was upregulated in VK2 cells exposed to G. vaginalis, but there was no change in actin cytoskeletal polymerization/rearrangements or vimentin subcellular relocalization post exposure. Cytoskeletal protein modifications could represent a potential mechanism for G. vaginalis mediated internalization by vaginal epithelial cells. Finally, understanding vaginal bacteria/host interactions will allow us to better understand the underlying mechanisms of BV pathogenesis.

Evidence for Gardnerella vaginalis uptake and internalization by squamous vaginal epithelial cells: implications for the pathogenesis of bacterial vaginosis.

La vaginose bact&#xE9;rienne (BV), une condition commune vu chez les femmes pr&#xE9;m&#xE9;nopaus&#xE9;es, est associ&#xE9;e &#xE0; un accouchement pr&#xE9;matur&#xE9;, la maladie inflammatoire pelvienne, et la livraison de faible poids de naissance. Vaginosi batterica (BV), una condizione comune vista in donne in premenopausa, sono associato con la consegna dei neonati di peso basso di nascita pretermine lavoro e malattia infiammatoria pelvica. Gardnerella vaginalis &egrave; la specie predominante batterica associata BV, anche se il suo esatto ruolo nella patologia di BV &egrave; sconosciuto. Gardnerella vaginalis &#xAE00;&#xADC0;&#xC640; &#xC9C8; &#xC0C1;&#xD53C; &#xC138;&#xD3EC;&#xB97C; &#xD3B8;&#xD3C9; &#xD558; &#xC5EC; &#xAD6D;&#xC81C;&#xD654;&#xC5D0; &#xB300; &#xD55C; &#xC99D;&#xAC70;: &#xC138;&#xADE0;&#xC131; vaginosis&#xC758; &#xBCD1; &#xC778;&#xC5D0; &#xB300; &#xD55C; &#xC758;&#xBBF8;.

La vaginosis bacteriana (VB), una condici&#xF3;n com&#xFA;n en las mujeres premenop&#xE1;usicas, se asocia con el parto prematuro, la enfermedad p&#xE9;lvica inflamatoria, y la entrega de reci&#xE9;n nacidos de bajo peso al nacer. Gardnerella vaginalis es la especie predominante de bacterias asociadas con la VB, aunque su papel exacto en la patolog&#xED;a de la vaginosis bacteriana es desconocida. Usando un microscopio de inmunofluorescencia, y la transmisi&#xF3;n electr&#xF3;nica confocal, se encontr&#xF3; que las c&#xE9;lulas epiteliales vaginales VK2 tomar hasta G. vaginalis despu&#xE9;s de la exposici&#xF3;n a la bacteria. La microscop&#xED;a confocal tambi&#xE9;n indic&#xF3; la presencia de G. vaginalis internalizada dentro de las c&#xE9;lulas epiteliales vaginales obtenidas a partir de un sujeto con BV. Usando VK2 c&#xE9;lulas y (35) bacterias S etiquetados en un ensayo de invasi&#xF3;n, hemos encontrado que una absorci&#xF3;n de 1 h de G. vaginalis fue 21,8 veces mayor que muertos por calor G. vaginalis, 84-veces en comparaci&#xF3;n con Lactobacillus acidophilus y 6,6 veces- en comparaci&#xF3;n con Crispatus Lactobacillus. La internalizaci&#xF3;n fue inhibida por la pre-exposici&#xF3;n de las c&#xE9;lulas a citocalasina-D. Adem&#xE1;s, el citoesqueleto de prote&#xED;nas vimentina se regul&#xF3; en VK2 las c&#xE9;lulas expuestas a G. vaginalis, pero no hubo ning&#xFA;n cambio en la polimerizaci&#xF3;n de la actina del citoesqueleto / o reordenamientos vimentina subcelular de la exposici&#xF3;n despu&#xE9;s de la relocalizaci&#xF3;n. Modificaciones del citoesqueleto prote&#xED;na podr&#xED;a representar un mecanismo potencial para la internalizaci&#xF3;n de G. vaginalis, mediada por las c&#xE9;lulas epiteliales vaginales. Por &#xFA;ltimo, la comprensi&#xF3;n de las bacterias vaginales e interacciones de acogida nos permitir&#xE1; comprender mejor los mecanismos subyacentes de la patog&#xE9;nesis de la VB.
