Retinoblastoma protein (pRB) mediates cell-cycle withdrawal and differentiation by interacting with a variety of proteins. RB-Binding Protein 2 (RBP2) has been shown to be a key effector. We sought to determine transcriptional regulation by RBP2 genome-wide by using location analysis and gene expression profiling experiments. We describe that RBP2 shows high correlation with the presence of H3K4me3 and its target genes are separated into two functionally distinct classes: differentiation-independent and differentiation-dependent genes. The former class is enriched by genes that encode mitochondrial proteins, while the latter is represented by cell-cycle genes. We demonstrate the role of RBP2 in mitochondrial biogenesis, which involves regulation of H3K4me3-modified nucleosomes. Analysis of expression changes upon RBP2 depletion depicted genes with a signature of differentiation control, analogous to the changes seen upon reintroduction of pRB. We conclude that, during differentiation, RBP2 exerts inhibitory effects on multiple genes through direct interaction with their promoters.

Genome-wide analysis of the H3K4 histone demethylase RBP2 reveals a transcriptional program controlling differentiation.

Prot&#xE9;ine du r&#xE9;tinoblastome (pRB) sert d&#x26;#39;interm&#xE9;diaire du cycle cellulaire de retrait et de la diff&#xE9;renciation en interagissant avec une vari&#xE9;t&#xE9; de prot&#xE9;ines. RB-Binding Protein 2 (RBP2) a &#xE9;t&#xE9; montr&#xE9; pour &#xEA;tre un effecteur cl&#xE9;. Nous avons cherch&#xE9; &#xE0; d&#xE9;terminer la r&#xE9;gulation transcriptionnelle par RBP2 l&#x26;#39;&#xE9;chelle du g&#xE9;nome en utilisant une analyse emplacement et des exp&#xE9;riences de profilage d&#x26;#39;expression g&#xE9;nique. Analyse der Ausdruck &Auml;nderungen auf RBP2 Ersch&ouml;pfung dargestellt Gene mit einer Signatur der Differenzierung Kontrolle, analog zu den &Auml;nderungen, die nach Wiedereinf&uuml;hrung der pRB gesehen. Wir schlie&szlig;en, dass w&auml;hrend der Differenzierung, RBP2 hemmende Wirkung auf mehrere Gene durch direkte Interaktion mit ihren F&ouml;rderern aus&uuml;bt.

Prote&#xED;na retinoblastoma (pRB) es un mediador del ciclo celular retirada y la diferenciaci&#xF3;n mediante la interacci&#xF3;n con una variedad de prote&#xED;nas. RB-Binding Protein 2 (RBP2) ha demostrado ser una tecla efector. Hemos tratado de determinar la regulaci&#xF3;n transcripcional por RBP2 todo el genoma mediante el uso de an&#xE1;lisis de la ubicaci&#xF3;n y los experimentos de expresi&#xF3;n g&#xE9;nica de perfiles. Se describe que RBP2 muestra alta correlaci&#xF3;n con la presencia de H3K4me3 y sus genes diana se separan en dos clases distintas: funcionalmente independientes genes diferenciaci&#xF3;n-dependiente y diferenciaci&#xF3;n. La clase anterior se enriquece con los genes que codifican prote&#xED;nas mitocondriales, mientras que el segundo est&#xE1; representado por los genes del ciclo celular. Se demuestra el papel de RBP2 en la biog&#xE9;nesis mitocondrial, lo que implica la regulaci&#xF3;n de H3K4me3 modificados nucleosomas. An&#xE1;lisis de la expresi&#xF3;n de los cambios sobre el agotamiento de los genes RBP2 representados con una firma de control de la diferenciaci&#xF3;n, de forma an&#xE1;loga a los cambios observados en la reintroducci&#xF3;n del PRB. Llegamos a la conclusi&#xF3;n de que, durante la diferenciaci&#xF3;n, RBP2 ejerce efectos inhibitorios sobre varios genes a trav&#xE9;s de la interacci&#xF3;n directa con sus promotores.
