The application of nanotechnology in biological research is beginning to have a major impact leading to the development of new types of tools for human health. One focus of nanobiotechnology is the development of nanoparticle-based formulations for use in drug or gene delivery systems. However most of the nano probes currently in use have varying levels of toxicity in cells or whole organisms and therefore are not suitable for in vivo application or long-term use. Here we test the potential of a novel silica based nanoparticle (organically modified silica, ORMOSIL) in living neurons within a whole organism. We show that feeding ORMOSIL nanoparticles to Drosophila has no effect on viability. ORMOSIL nanoparticles penetrate into living brains, neuronal cell bodies and axonal projections. In the neuronal cell body, nanoparticles are present in the cytoplasm, but not in the nucleus. Strikingly, incorporation of ORMOSIL nanoparticles into the brain did not induce aberrant neuronal death or interfered with normal neuronal processes. Our results in Drosophila indicate that these novel silica based nanoparticles are biocompatible and not toxic to whole organisms, and has potential for the development of long-term applications.

L&#x26;#39;application de la nanotechnologie dans la recherche biologique commence &#xE0; avoir un impact majeur conduisant &#xE0; la mise au point de nouveaux types d&#x26;#39;outils pour la sant&#xE9; humaine. Un des objectifs de la nanobiotechnologie est le d&#xE9;veloppement de nanoparticules &#xE0; base de formulations pour utilisation dans les syst&#xE8;mes de d&#xE9;livrance de m&#xE9;dicaments ou d&#x26;#39;un g&#xE8;ne. Cependant, la plupart des sondes nano actuellement en usage ont diff&#xE9;rents niveaux de toxicit&#xE9; dans les cellules ou des organismes entiers et ne sont donc pas appropri&#xE9; pour une application in vivo ou utilisation &#xE0; long terme. Ici, nous testons le potentiel d&#x26;#39;une nanoparticule de silice roman bas&#xE9; sur (silice modifi&#xE9;e organiquement, ormosil) &#xE0; vivre au sein de neurones un organisme entier. Nous montrons que l&#x26;#39;alimentation des nanoparticules ormosil drosophile n&#x26;#39;a pas d&#x26;#39;effet sur la viabilit&#xE9;. Ormosil nanoparticules p&#xE9;n&#xE8;trent dans le cerveau de vie, les corps cellulaires neuronaux et des projections axonales. Dans le corps cellulaire neuronale, des nanoparticules sont pr&#xE9;sentes dans le cytoplasme, mais pas dans le noyau. &#xC9;tonnamment, l&#x26;#39;incorporation de nanoparticules dans le cerveau ormosil n&#x26;#39;a pas induit la mort neuronale aberrante ou interf&#xE9;r&#xE9; avec le processus normal de neurones. Nos r&#xE9;sultats chez la drosophile indiquent que ces nanoparticules de silice &#xE0; base de nouvelles sont biocompatibles et non toxique pour les organismes entiers, et poss&#xE8;de un potentiel pour le d&#xE9;veloppement &#xE0; long terme des applications.

Die Anwendung der Nanotechnologie in der biologischen Forschung beginnt stark zur Entwicklung neuer Arten von Werkzeugen f&uuml;r die menschliche Gesundheit auswirken. Ein Schwerpunkt der Nanobiotechnologie ist die Entwicklung von Nanopartikel-basierte Formulierungen f&uuml;r den Einsatz in Drogen- oder gen-Delivery-Systeme. Jedoch die meisten die Nano-Sonden zur Zeit im Einsatz haben unterschiedliche Ebenen der Toxizit&auml;t in Zellen oder ganze Organismen und sind daher nicht f&uuml;r in-vivo-Anwendung oder langfristigen Einsatz geeignet. Hier testen wir das Potenzial der eine neuartige Kiesels&auml;ure basierte Nanopartikel (organisch modifizierte Silika, ORMOSIL) in lebenden Neuronen innerhalb eines Organismus. Wir zeigen, dass die Verf&uuml;tterung von ORMOSIL Nanopartikel an Drosophila hat keinen Einfluss auf die Lebensf&auml;higkeit. ORMOSIL Nanopartikel dringen in die lebenden Gehirn, neuronalen Zelle K&ouml;rper und axonale Projektionen. In neuronale Zellk&ouml;rper, Nanopartikel sind vorhanden im Zytoplasma, aber nicht in den Zellkern. Auff&auml;llig ist, Einbeziehung der ORMOSIL Nanopartikel in das Gehirn diesen Versuchsbedingungen keine aberrante neuronalen Tod oder mischte sich mit normalen neuronalen Prozessen. Unsere Ergebnisse in Drosophila zeigen, dass diese neuartige Silica Basis-Nanopartikel biokompatibel und nicht f&uuml;r ganze Organismen giftig sind und hat das Potential f&uuml;r die Entwicklung von langfristigen Anwendungen.

Tuttavia la maggior parte delle sonde nano attualmente in uso hanno diversi livelli di tossicit&agrave; in cellule o interi organismi e pertanto non sono adatta per applicazioni in vivo o l&#39;uso a lungo termine. Qui ci prova il potenziale di una nanoparticella basata romanzo di silice (silice biologicamente modificata, ORMOSIL) nei neuroni viventi all&#39;interno di un intero organismo. Mostriamo che alimentazione ORMOSIL nanoparticelle di drosofila non ha alcun effetto sulla redditivit&agrave;. Le nanoparticelle ORMOSIL penetrano nel cervello vivente, corpi cellulari neuronali e proiezioni assonali. Nel corpo delle cellule neuronale, le nanoparticelle sono presenti nel citoplasma, ma non nel nucleo. Sorprendentemente, incorporazione di nanoparticelle ORMOSIL il cervello non indurre la morte neuronale aberrante o interferiva con i normali processi neuronali. I nostri risultati in drosofila indicano che queste nanoparticelle di silice romanzo basata sono biocompatibili e non tossico per gli organismi interi e ha il potenziale per lo sviluppo di applicazioni a lungo termine.

La aplicaci&#xF3;n de la nanotecnolog&#xED;a en la investigaci&#xF3;n biol&#xF3;gica est&#xE1; empezando a tener un impacto importante que lleva al desarrollo de nuevos tipos de herramientas para la salud humana. Uno de los objetivos de la nanobiotecnolog&#xED;a es el desarrollo de formulaciones a base de nanopart&#xED;culas para su uso en sistemas de administraci&#xF3;n de f&#xE1;rmacos o genes. Sin embargo la mayor&#xED;a de las sondas nano actualmente en uso tienen diferentes niveles de toxicidad en c&#xE9;lulas u organismos enteros y por lo tanto no son adecuados para su aplicaci&#xF3;n in vivo o el uso a largo plazo. Aqu&#xED; ponemos a prueba el potencial de las nanopart&#xED;culas de s&#xED;lice una novela basada en (s&#xED;lice modificada org&#xE1;nicamente, ormosil) en neuronas vivas dentro de un organismo completo. Se demuestra que la alimentaci&#xF3;n de nanopart&#xED;culas ormosil a Drosophila tiene ning&#xFA;n efecto sobre la viabilidad. Ormosil nanopart&#xED;culas penetran en los cerebros de vida, los &#xF3;rganos de c&#xE9;lulas neuronales y las proyecciones axonales. En el cuerpo de las c&#xE9;lulas neuronales, nanopart&#xED;culas est&#xE1;n presentes en el citoplasma, pero no en el n&#xFA;cleo. Sorprendentemente, la incorporaci&#xF3;n de nanopart&#xED;culas ormosil en el cerebro no induce muerte neuronal aberrante o interferido con los procesos normales de las neuronas. Nuestros resultados en Drosophila indican que estos nuevos basados &#x200B;&#x200B;en nanopart&#xED;culas de s&#xED;lice son biocompatibles y no t&#xF3;xico para los organismos enteros, y tiene un potencial para el desarrollo de aplicaciones a largo plazo.
