Conserved ATP-dependent proteases ensure the quality control of mitochondrial proteins and control essential steps in mitochondrial biogenesis. Recent studies demonstrated that non-assembled mitochondrially encoded proteins are degraded to peptides and amino acids that are released from mitochondria. Here, we have characterized peptides extruded from mitochondria by mass spectrometry and identified 270 peptides that are exported in an ATP- and temperature-dependent manner. The peptides originate from 51 mitochondrially and nuclearly encoded proteins localized mainly in the matrix and inner membrane, indicating that peptides generated by the activity of all known mitochondrial ATP-dependent proteases can be released from the organelle. Pulse-labeling experiments in logarithmically growing yeast cells revealed that approximately 6-12% of preexisting and newly imported proteins is degraded and contribute to this peptide pool. Under respiring conditions, we observed an increased proteolysis of newly imported proteins that suggests a higher turnover rate of respiratory chain components and thereby rationalizes the predominant appearance of representatives of this functional class in the detected peptide pool. These results demonstrated a constant efflux of peptides from mitochondria and provided new insight into the stability of the mitochondrial proteome and the efficiency of mitochondrial biogenesis.

Gamma-aminobutyric acid (GABA)ergic neurons are a diverse group of inhibitory neurons playing crucial roles in information processing. We analyzed the gene expression of regionally defined GABAergic neurons from the cortex, olfactory bulb, striatum, and cerebellum of glutamate decarboxylase 67-green fluorescence protein (GAD67-GFP) knock-in mice. We introduce a generally applicable method for singularization of brain cells, flow cytometric enrichment, and global mRNA amplification for sensitive gene expression profiling. Systematic quantification elicited a high dynamic range of GABAergic cell numbers in different brain regions. Clustering of our gene expression results revealed major differences between hind and forebrain GABAergic neurons indicating that the development of GABAergic neurons depends on their regional location. While GABAergic neurons of the forebrain are characterized by three main groups of transcription factors of the Distal-less-family, the POU-family, and ETS/FOX family, specific members of the ZIC- and LHX-family of transcription factors appear to define hindbrain inhibitory neurons.

Nous avons analys&#xE9; l&#x26;#39;expression du g&#xE8;ne de la r&#xE9;gion d&#xE9;finis neurones GABAergiques du cortex, bulbe olfactif, le striatum et le cervelet de la d&#xE9;carboxylase 67-prot&#xE9;ine verte glutamate de fluorescence (GAD67-GFP) souris knock-in. After their generation and specification in periventricular regions, neuronal precursors maintain an immature and migratory state until their arrival in the respective target structures. Only here are terminal differentiation and synaptic integration induced. Although the molecular control of neuronal specification has started to be elucidated, little is known about the factors that control the latest maturation steps. We aimed at identifying factors that induce terminal differentiation during postnatal and adult neurogenesis, thereby focusing on the generation of periglomerular interneurons in the olfactory bulb. We isolated neuronal precursors and mature neurons from the periglomerular neuron lineage and analyzed their gene expression by microarray. We found that expression of the bHLH transcription factor NeuroD1 strikingly coincides with terminal differentiation. Using brain electroporation, we show that overexpression of NeuroD1 in the periventricular region in vivo leads to the rapid appearance of cells with morphological and molecular characteristics of mature neurons in the subventricular zone and rostral migratory stream. Conversely, shRNA-induced knockdown of NeuroD1 inhibits terminal neuronal differentiation. Thus, expression of a single transcription factor is sufficient to induce neuronal differentiation of neural progenitors in regions that normally do not show addition of new neurons. These results suggest a considerable potential of NeuroD1 for use in cell-therapeutic approaches in the nervous system.

Despu&#xE9;s de su generaci&#xF3;n y las especificaciones en las regiones periventriculares, precursores neuronales mantener un estado inmaduro y migratorias hasta su llegada a las estructuras de destino respectivos. S&#xF3;lo que aqu&#xED; son la diferenciaci&#xF3;n terminal y la integraci&#xF3;n sin&#xE1;ptica inducida. Aunque el control molecular de la especificaci&#xF3;n neuronal ha comenzado a ser dilucidado, se sabe poco sobre los factores que controlan los pasos m&#xE1;s recientes de maduraci&#xF3;n. El objetivo fue identificar los factores que inducen la diferenciaci&#xF3;n terminal durante la neurog&#xE9;nesis postnatal y adulta, con lo que se centra en la generaci&#xF3;n de las interneuronas periglomerulares en el bulbo olfatorio. Se aislaron los precursores neuronales y neuronas maduras a partir del linaje neuronal periglomerular y se analiza su expresi&#xF3;n g&#xE9;nica mediante microarrays. Hemos encontrado que la expresi&#xF3;n del factor de transcripci&#xF3;n bHLH NeuroD1 sorprendentemente coincide con la diferenciaci&#xF3;n terminal. Utilizando la electroporaci&#xF3;n del cerebro, nos muestran que la sobreexpresi&#xF3;n de NeuroD1 en la regi&#xF3;n periventricular en las derivaciones in vivo para la r&#xE1;pida aparici&#xF3;n de c&#xE9;lulas con caracter&#xED;sticas morfol&#xF3;gicas y moleculares de las neuronas maduras en la zona subventricular y la corriente migratoria rostral. A la inversa, ARNhc inducida desmontables de NeuroD1 inhibe la diferenciaci&#xF3;n neuronal de terminales. As&#xED;, la expresi&#xF3;n de un solo factor de transcripci&#xF3;n es suficiente para inducir la diferenciaci&#xF3;n neuronal de c&#xE9;lulas progenitoras neuronales en regiones que normalmente no muestran la adici&#xF3;n de nuevas neuronas. Estos resultados sugieren un potencial considerable de NeuroD1 para su uso en c&#xE9;lulas terap&#xE9;uticas enfoques en el sistema nervioso.

NeuroD1 induce la diferenciaci&#xF3;n neuronal de terminales en la neurog&#xE9;nesis olfatoria

We performed next generation sequencing- and microarray-based gene expression profiling of CD44(+)/CD24(-)/CD45(-) breast CSCs (cancer stem cells) isolated from primary ERα-positive breast cancer. By combining semi-automated dissociation of human tumor tissue, magnetic cell sorting and cDNA amplification less than 500 CSCs were required for transcriptome analyses. Besides overexpressing genes involved in maintenance of stemness, the CSCs showed higher levels of genes that drive the PI3K pathway, including EGFR, HB-EGF, PDGFRA/B, PDGF, MET, PIK3CA, PIK3R1 and PIK3R2. This suggests that, in CSCs of ERα-positive breast cancer, the PI3K pathway which is involved in endocrine resistance is hyperactive.

Highly sensitive profiling of CD44+/CD24- breast cancer stem cells by combining global mRNA amplification and next generation sequencing: evidence for a hyperactive PI3K pathway.

The study was undertaken to compare antitumor efficacy of electrochemotherapy (ECT) with cold plasma therapy (CP) in a melanoma mouse model. After melanoma implantation into the flank of C57BL/6N mice, CP by two different plasma sources (APPJ and DBD) was applied directly to the tumor surface. ECT was performed with bleomycin intravenously at a field strength of 1000 V/cm without or combined with CP. Primary endpoints were tumor growth acceleration (TGA), daily volume progression (DVP) and survival after treatment. Both plasma sources as single treatment showed a significant TGA delay, which proved less effective than ECT. CP (APPJ) combined with ECT (ECJ) significantly improved per cent mouse survival, with significant superiority compared with ECT. Plasma therapy alone albeit less effective seems a potential alternative to ECT in patients with melanoma and can be applied manifold in a session without general anaesthesia. Accordingly, CP alone and combined with ECT may serve as new option in palliative skin melanoma therapy.

De novo formation of beige/brite adipocytes from progenitor cells contributes to the thermogenic adaptation of adipose tissue and holds great potential for the therapeutic remodeling of fat as a treatment for obesity. Despite the recent identification of several factors regulating browning of white fat, there is a lack of physiological cell models for the mechanistic investigation of progenitor-mediated beige/brite differentiation. We have previously revealed prostacyclin (PGI2) as one of the few known endogenous extracellular mediators promoting de novo beige/brite formation by relaying β-adrenergic stimulation to the progenitor level. Here, we present a cell model based on murine primary progenitor cells defined by markers previously shown to be relevant for in vivo browning, including a simplified isolation procedure. We demonstrate the specific and broad induction of thermogenic gene expression by PGI2 signaling in the absence of lineage conversion, and reveal the previously unidentified nuclear relocalization of the Ucp1 gene locus in association with transcriptional activation. By profiling the time course of the progenitor response, we show that PGI2 signaling promoted progenitor cell activation through cell cycle and adhesion pathways prior to metabolic maturation toward an oxidative cell phenotype. Our results highlight the importance of core progenitor activation pathways for the recruitment of thermogenic cells and provide a resource for further mechanistic investigation.
