The discovery of endothelial progenitor cells in the 1990s challenged the paradigm of angiogenesis by showing that cells derived from hematopoietic stem cells are capable of forming new blood vessels even in the absence of a pre-existing vessel network, a process termed vasculogenesis. Since then, the majority of studies in the field have found a strong association between circulating endothelial progenitor cells and cardiovascular risk. Several studies have also reported that inflammation influences the mobilization and differentiation of endothelial progenitor cells. In this review, we discuss the emerging role of endothelial progenitor cells as biomarkers of cardiovascular disease as well as the interplay between inflammation and endothelial progenitor cell biology. We will also review the challenges in the field of endothelial progenitor cell-based therapy.

La d&#xE9;couverte de cellules prog&#xE9;nitrices endoth&#xE9;liales dans les ann&#xE9;es 1990 a contest&#xE9; le paradigme de l&#x26;#39;angiogen&#xE8;se en montrant que les cellules d&#xE9;riv&#xE9;es de cellules souches h&#xE9;matopo&#xEF;&#xE9;tiques sont capables de former de nouveaux vaisseaux sanguins, m&#xEA;me en l&#x26;#39;absence d&#x26;#39;un r&#xE9;seau de vaisseaux pr&#xE9;-existant, un processus appel&#xE9; vasculogen&#xE8;se. Depuis lors, la majorit&#xE9; des &#xE9;tudes sur le terrain ont constat&#xE9; une forte association entre les cellules prog&#xE9;nitrices endoth&#xE9;liales circulantes et le risque cardiovasculaire. Plusieurs &#xE9;tudes ont &#xE9;galement signal&#xE9; que l&#x26;#39;inflammation influe sur la mobilisation et la diff&#xE9;renciation des cellules prog&#xE9;nitrices endoth&#xE9;liales. Die Entdeckung der endothelialen Vorl&auml;uferzellen in den 1990er Jahren forderte das Paradigma von Angiogenesis zeigt, dass Zellen aus h&auml;matopoetischen Stammzellen der Bildung neuer Blutgef&auml;&szlig;e auch in Abwesenheit eines bereits vorhandenen Schiff-Netzwerks f&auml;hig sind, bezeichnet ein Prozess Vaskulogenese. Seitdem haben die meisten Studien im Bereich eine starke Assoziation zwischen zirkulierenden endothelialen Vorl&auml;uferzellen und kardiovaskul&auml;res Risiko gefunden. Mehrere Studien haben auch berichtet, dass Entz&uuml;ndungen die Mobilisierung und die Differenzierung von endothelialen Progenitorzellen beeinflusst. In diesem Bericht diskutieren wir die aufstrebende Rolle der endothelialen Vorl&auml;uferzellen als Biomarker der Herz-Kreislauf-Erkrankungen sowie das Zusammenspiel zwischen Entz&uuml;ndung und EPCs Zellbiologie. La scoperta di cellule progenitrici endoteliali negli anni novanta ha sfidato il paradigma dell&#39;angiogenesi mostrando che le cellule derivate da cellule staminali ematopoietiche sono in grado di formare nuovi vasi sanguigni anche in assenza di una rete di vasi preesistenti, un processo definito vasculogenesi. Da allora, la maggior parte degli studi in campo hanno trovato una forte associazione tra cellule progenitrici endoteliali e rischio cardiovascolare in circolazione. Numerosi studi hanno anche riportato che l&#39;infiammazione influenza la mobilitazione e la differenziazione delle cellule progenitrici endoteliali. In questa recensione, discutiamo il ruolo emergente delle cellule progenitrici endoteliali come biomarkers di malattia cardiovascolare, nonch&eacute; l&#39;interazione tra infiammazione e biologia delle cellule progenitrici endoteliali. Rivedremo anche le sfide nel campo della terapia basate sulle cellule progenitrici endoteliali.
