Tendon response to mechanical loading results in either homeostasis, improvement, or degeneration of tissue condition. In an effort to better understand the development of tendinopathies, this study investigated the mechanical and structural responses of tendons subjected to under- and over-stimulations (1.2% and 1.8% strain respectively, 1 Hz). The objective was to examine three sub-processes of tendon response: mechanical degradation, enzymatic degradation, and repair of the extracellular matrix. We subjected rat tail tendons to a 10-day stimulation protocol with four periods of 6 h each day: 30 min of stimulation and 5 h 30 min of rest. To investigate the contribution of the three sub-processes, we controlled the contribution of the cells through variations in the nutrient and protease inhibitor content in the in vitro solutions. Using nondestructive cyclic tests, we evaluated the daily changes in the peak stress. To assess structural changes, we carried out microscopic analyses at the end of the study period. We observed that the relative contributions of the sub-processes differed according to the stimulation amplitude. With over-stimulation of tendons immersed in DMEM, we succeeded in reducing enzymatic degradation and increasing peak stress. In under-stimulation, the addition of protease inhibitors was required to obtain the same result.

R&#xE9;ponse aux r&#xE9;sultats de chargement tendon m&#xE9;caniques dans les deux hom&#xE9;ostasie, l&#x26;#39;am&#xE9;lioration ou la d&#xE9;g&#xE9;n&#xE9;rescence de la condition du tissu. Dans un effort pour mieux comprendre le d&#xE9;veloppement des tendinopathies, cette &#xE9;tude a examin&#xE9; les r&#xE9;ponses m&#xE9;caniques et structurelles des tendons soumis &#xE0; la sous-et sur-stimulations (1,2% et 1,8% respectivement souche, 1 Hz). L&#x26;#39;objectif &#xE9;tait d&#x26;#39;examiner trois sous-processus de r&#xE9;ponse du tendon: la d&#xE9;gradation m&#xE9;canique, de la d&#xE9;gradation enzymatique, et la r&#xE9;paration de la matrice extracellulaire. Nous tendons de rats soumis &#xE0; un protocole de stimulation de 10 jours avec quatre p&#xE9;riodes de 6 h chaque jour: 30 min de stimulation et de 5 h 30 min de repos. Pour &#xE9;tudier la contribution des trois sous-processus, nous avons contr&#xF4;l&#xE9; la contribution des cellules &#xE0; travers les variations de la teneur en &#xE9;l&#xE9;ments nutritifs et inhibiteur de la prot&#xE9;ase dans le dans les solutions in vitro. L&#x26;#39;utilisation de tests non destructifs cycliques, nous avons &#xE9;valu&#xE9; les changements quotidiens dans le pic de contrainte. Pour &#xE9;valuer les changements structurels, nous avons effectu&#xE9; des analyses microscopiques &#xE0; la fin de la p&#xE9;riode d&#x26;#39;&#xE9;tude. Nous avons observ&#xE9; que les contributions relatives des sous-processus diff&#xE8;re selon l&#x26;#39;amplitude de stimulation. Avec la sur-stimulation des tendons immerg&#xE9;s dans du DMEM, nous avons r&#xE9;ussi &#xE0; r&#xE9;duire la d&#xE9;gradation enzymatique et en augmentant le stress de pointe. Sehne Reaktion auf mechanische Belastung f&uuml;hrt Hom&ouml;ostase, Verbesserung oder Degeneration der Gewebe Zustand. In dem Bem&uuml;hen, die Entwicklung der Tendinopathies besser zu verstehen untersuchte diese Studie die mechanischen und strukturellen Antworten der Sehnen unterworfen, unter - und over - stimulations (1,2 % und 1,8 % bzw. 1 Hz zu belasten). Ziel war es, drei Unterkategorien der Sehne Antwort pr&uuml;fen: mechanische Abbau, enzymatischen Abbau und Reparatur der extrazellul&auml;ren Matrix. Wir eine 10-t&auml;gige Stimulierung-Protokoll mit vier Perioden von 6 h pro Tag Ratte Schwanz sehnen ausgesetzt: 30 min der Stimulation und 5 h 30 min Rest. Um den Beitrag der drei Unterkategorien zu untersuchen, kontrollierten wir den Beitrag der Zellen durch Variationen in der N&auml;hrstoff- und Protease-Inhibitor-Inhalt in den in-vitro-L&ouml;sungen. Um strukturelle Ver&auml;nderungen zu beurteilen, durchgef&uuml;hrt wir mikroskopische Analysen am Ende des Untersuchungszeitraumes. Wir beobachteten, dass die relative Beitr&auml;ge von der Unterkategorien entsprechend der Anregung-Amplitude unterschieden. Mit Reiz&uuml;berflutung der Sehnen eingebettet in DMEM konnten wir verk&uuml;rzen enzymatischen Abbau und die Spitze-Spannung. In Under-stimulation war die Hinzuf&uuml;gung von Proteasehemmern erforderlich, um dasselbe Ergebnis zu erhalten.

Abbiamo sottoposto i tendini di coda di ratto a un protocollo di stimolazione di 10 giorni con quattro periodi di 6 h al giorno: 30 min di stimolazione e 5 h 30 min di riposo. Per indagare il contributo di tre sottoprocessi, abbiamo controllato il contributo delle cellule attraverso le variazioni del contenuto di nutrienti e proteasi inibitore nelle soluzioni in vitro. Mediante prove non distruttive ciclici, abbiamo valutato le modifiche quotidiane nello sforzo di picco. Per valutare le modifiche strutturali, abbiamo effettuato analisi microscopiche alla fine del periodo di studio. Abbiamo osservato che i contributi relativi di sottoprocessi differivano secondo l&#39;ampiezza di stimolazione. Con eccesso di stimolazione dei tendini immersi in DMEM, siamo riusciti a ridurre la degradazione enzimatica e aumentare lo stress di picco. In under-stimulation, per ottenere lo stesso risultato era necessaria l&#39;aggiunta di inibitori della proteasi.

La respuesta a los resultados del tend&#xF3;n de carga mec&#xE1;nica, ya sea en la homeostasis, la mejora, o la degeneraci&#xF3;n de la condici&#xF3;n de los tejidos. En un esfuerzo por comprender mejor el desarrollo de las tendinopat&#xED;as, este estudio investig&#xF3; las respuestas mec&#xE1;nicas y estructurales de los tendones sometidos a bajo-y sobre-est&#xED;mulos (1,2% y 1,8% de deformaci&#xF3;n, respectivamente, de 1 Hz). El objetivo fue examinar las tres sub-procesos de respuesta del tend&#xF3;n: la degradaci&#xF3;n mec&#xE1;nica, la degradaci&#xF3;n enzim&#xE1;tica, y la reparaci&#xF3;n de la matriz extracelular. Estamos sometidos los tendones de cola de rata a un protocolo de estimulaci&#xF3;n de 10 d&#xED;as con cuatro per&#xED;odos de 6 horas cada d&#xED;a: 30 minutos de estimulaci&#xF3;n y 5 h 30 min de descanso. Para investigar la contribuci&#xF3;n de los tres sub-procesos, se control&#xF3; la contribuci&#xF3;n de las c&#xE9;lulas a trav&#xE9;s de variaciones en el valor nutritivo y contenido inhibidor de la proteasa en soluciones in vitro. El uso de pruebas no destructivas c&#xED;clicas, se evaluaron los cambios diarios en el pico de estr&#xE9;s. Para evaluar los cambios estructurales, se llev&#xF3; a cabo los an&#xE1;lisis microsc&#xF3;picos al final del per&#xED;odo de estudio. Hemos observado que las contribuciones relativas de los sub-procesos difieren de acuerdo a la amplitud de estimulaci&#xF3;n. Con la sobre-estimulaci&#xF3;n de los tendones inmersos en DMEM, tuvimos &#xE9;xito en la reducci&#xF3;n de la degradaci&#xF3;n enzim&#xE1;tica y el aumento de la tensi&#xF3;n m&#xE1;xima.
