A new polymer, poloxamine-polylysine acrylate (PPA), was synthesized for tissue-engineering applications. Polylysine was used to confer endothelial cell attachment properties to the poloxamine-based polymer, while the acrylate groups made it photo-cross-linkable. The PPA polymer was synthesized in three reaction steps. (1) Polylysine was acrylated using N-hydroxysuccinimide acrylic acid ester and the reaction product was characterised by (1)H-NMR. (2) The hydroxyl groups on poloxamine were activated by tresylation with tresyl chloride and the reaction product (tresylated poloxamine) was characterised by ICP-AES for sulphur content. (3) The acrylated polylysine was reacted with the activated poloxamine to obtain the final product, poloxamine-polylysine acrylate. The final product was characterised by CHN elemental analysis. Aqueous solutions containing a mixture of PPA and poloxamine methacrylate were photo-cross-linked by exposure to a 365 nm UV light source in the presence of a photoinitiator to obtain hydrogels. The synthesized PPA polymer enhanced endothelial cell adhesion on poloxamine-based, photo-cross-linked hydrogel scaffolds. The same synthesis methods can be used in the future to introduce other desired functions (through other peptides) into the scaffold biomaterial.

A Poloxamine-Polylysine Acrylate Scaffold for Modular Tissue Engineering.

Un Poloxamine-polylysine acrylate d&#x26;#39;&#xE9;chafaudage modulaire pour le g&#xE9;nie tissulaire

F&uuml;r Tissue-Engineering-Anwendungen wurde eine neue Polymer, Poloxamine-Polylysin Acrylat (PPA), synthetisiert. Polylysin wurde verwendet, um Eigenschaften f&uuml;r Dateianlage endothelial Zellen zum Poloxamine-basierte Polymer, zu &uuml;bertragen, w&auml;hrend die Acrylat-Gruppen es Foto-Kreuz-Linkbar gemacht. (3) Die acrylated Polylysin wurde mit der aktivierten Poloxamine erhalten das Endprodukt, Poloxamine-Polylysin Acrylat reagiert. Das Endprodukt war gepr&auml;gt von CHN Elementaranalyse. W&auml;ssrige L&ouml;sungen mit einer Mischung aus PPA und Poloxamine wurden Hydrogele zu erhalten Foto-Kreuz-verbunden durch die Exposition gegen&uuml;ber einer 365 nm UV-Lichtquelle in Anwesenheit eines Fotoinitiators. Das synthetisierte PPA-Polymer verbessert endothelial Zellenadh&auml;sion auf Poloxamine basierende, Foto-Kreuz-chromosomale Hydrogel-Ger&uuml;sten. Die gleichen Synthese-Methoden k&ouml;nnen in Zukunft andere gew&uuml;nschten Funktionen (durch andere Peptide) Einf&uuml;hrung in das Ger&uuml;st Biomaterial verwendet werden.

Un nuovo polimero, poloxamine-polilisina acrilato (PPA), &egrave; stato sintetizzato per applicazioni di ingegneria tissutale. Polilisina &egrave; stato utilizzato per conferire propriet&agrave; attaccamento delle cellule endoteliali al polimero poloxamine-base, mentre i gruppi di acrilato fatto foto-croce-collegabili. Il polimero di PPA &egrave; stato sintetizzato in tre fasi di reazione. (1) Polilisina era stata utilizzando N-Idrossisuccinimide acido estere acrilico e il prodotto di reazione &egrave; stato caratterizzato da (1) H-NMR. (3) La polilisina allofanato era ha reagito con il poloxamine attivato per ottenere il prodotto finale, acrilato poloxamine-polilisina. Il prodotto finale &egrave; stato caratterizzato dall&#39;analisi elementare CHN. Soluzioni acquose contenenti una miscela di metacrilato PPA e poloxamine erano foto-cross-linked da esposizione a 365 nm UV fonte di luce in presenza di un fotoiniziatore per ottenere idrogel. Il polimero sintetizzato PPA migliorata adesione delle cellule endoteliali su ponteggi hydrogel poloxamine-based, foto-cross-linked. Gli stessi metodi di sintesi possono essere utilizzati in futuro per introdurre altre funzioni desiderati (attraverso altri peptidi) in biomateriale il patibolo.

Un nuevo pol&#xED;mero, poloxamina-polilisina acrilato (PPA), se sintetiz&#xF3; para aplicaciones de ingenier&#xED;a de tejidos. Polilisina se utiliza para conferir propiedades de uni&#xF3;n de c&#xE9;lulas endoteliales con el pol&#xED;mero basado en poloxamina, mientras que los grupos acrilato hizo foto-reticulable. El pol&#xED;mero PPA fue sintetizado en tres etapas de reacci&#xF3;n. (1) Polilisina se acrilado utilizando N-hidroxisuccinimida &#xE9;ster de &#xE1;cido acr&#xED;lico y el producto de reacci&#xF3;n se caracteriza por (1) H-RMN. (2) Los grupos hidroxilo en poloxamina fueron activadas por tresilaci&#xF3;n con cloruro de tresilo y el producto de reacci&#xF3;n (tresilados poloxamina) se caracteriz&#xF3; por ICP-AES para contenido de azufre. (3) La polilisina acrilado se hizo reaccionar con la poloxamina activado para obtener el producto final, poloxamina-polilisina acrilato. El producto final se caracteriz&#xF3; por an&#xE1;lisis elemental CHN. Las soluciones acuosas que contienen una mezcla de metacrilato de PPA y poloxamina fueron foto-reticulado por la exposici&#xF3;n a una fuente de luz UV de 365 nm, en presencia de un fotoiniciador para obtener hidrogeles. El pol&#xED;mero sintetizado PPA mejorada adhesi&#xF3;n de c&#xE9;lulas endoteliales en poloxamina basados, andamios hidrogel foto-reticulados. Los m&#xE9;todos de s&#xED;ntesis mismos puede utilizarse en el futuro para introducir otras funciones deseadas (a trav&#xE9;s de otros p&#xE9;ptidos) en el biomaterial andamio.
