Hashimoto thyroiditis can be partially reproduced in mice by immunization with thyroglobulin or, more recently, thyroperoxidase. This experimental autoimmune thyroiditis (EAT) model has been extensively characterized during early disease phases (up to d 35 after immunization). By extending the analysis of EAT to 100 d after immunization, we noted a remarkable regenerative capacity of the thyroid and the expression of Oct-4, suggesting in vivo the existence of adult thyroid stem cells. After an almost complete destruction of the follicular architecture, occurring between d 21 and 28, the thyroid was capable of restoring its follicles and reducing the mononuclear infiltration, so that by d 100 after immunization, it regained its normal morphology and function. During this regeneration process, thyrocytes expressed high levels of CD24. We therefore assessed the role of CD24 in thyroid regeneration by inducing EAT in mice lacking CD24. Regeneration was faster in the absence of CD24, likely a consequence of the effect of CD24 on the infiltrating lymphocytes. The study suggests that the EAT model can also be used as a tool to investigate adult thyroid stem cells.

Regenerative potentials of the murine thyroid in experimental autoimmune thyroiditis: role of CD24.

La thyro&#xEF;dite de Hashimoto peut &#xEA;tre reproduit partiellement chez la souris par immunisation avec la thyroglobuline ou, plus r&#xE9;cemment, la thyroperoxydase. Cette thyro&#xEF;dite auto-immune exp&#xE9;rimentale (EAE) mod&#xE8;le a &#xE9;t&#xE9; largement caract&#xE9;ris&#xE9; au cours des phases pr&#xE9;coces de la maladie (jusqu&#x26;#39;&#xE0; D 35 apr&#xE8;s la vaccination). En &#xE9;tendant l&#x26;#39;analyse de EAT &#xE0; 100 jours apr&#xE8;s la vaccination, nous avons constat&#xE9; une remarquable capacit&#xE9; de r&#xE9;g&#xE9;n&#xE9;ration de la thyro&#xEF;de et l&#x26;#39;expression de Oct-4, ce qui sugg&#xE8;re l&#x26;#39;existence in vivo de cellules souches adultes thyro&#xEF;de. Diese experimentellen Autoimmun-Thyreoiditis (EAT)-Modell war umfassend in fr&uuml;hen Phasen der Erkrankung (bis zu 35 d nach Immunisierung) charakterisiert. Erweitern die Analyse von EAT 100D nach Immunisierung, bekannt wir eine bemerkenswerte Regenerationsf&auml;higkeit der Schilddr&uuml;se und der Ausdruck der Oct-4, in-vivo schl&auml;gt das Vorhandensein von Stammzellen von Erwachsenen Schilddr&uuml;se. Nach einer fast vollst&auml;ndigen Zerst&ouml;rung der follikul&auml;ren Architektur, zwischen 21 und 28 d, konnte die Schilddr&uuml;se Wiederherstellen der Follikel und die Verringerung der mononukle&auml;ren Infiltration, so dass es von 100 d nach Immunisierung, ihre normale Morphologie und Funktion wiedererlangt. W&auml;hrend dieses Vorgangs Regenerierung ausgedr&uuml;ckt Thyrocytes hohe CD24. Wir beurteilt daher die Rolle der CD24 bei Schilddr&uuml;se Regeneration induzierende EAT in M&auml;usen CD24 fehlt. Regenerierung war schneller in Ermangelung CD24, wahrscheinlich eine Folge der Auswirkungen der CD24 auf die infiltrierende Lymphozyten. Die Studie legt nahe, dass das EAT-Modell auch als Werkzeug verwendet werden kann, um Stammzellen von Erwachsenen Schilddr&uuml;se zu untersuchen.

Lo studio suggerisce che il modello mangiare pu&ograve; essere utilizzato anche come strumento per indagare le cellule staminali adulte della tiroide.

Interferon (IFN)-gamma has been involved in the pathogenesis of Hashimoto thyroiditis. It is a cytokine released by infiltrating mononuclear cells that mediates its actions mainly through signal transducer and activator of transcription-1 (STAT1) but also through other transcription factors. To dissect the effect of IFN gamma on thyroid morphology and function, we crossed transgenic mice that express IFN gamma specifically in the thyroid gland to mice deficient in STAT1. Lack of STAT1 ameliorated the abnormal thyroid morphology and the primary hypothyroidism typical of IFN gamma transgenic mice but not the suppressed iodine accumulation. Interestingly, lack of STAT1 alone decreased iodine accumulation, seemingly through expression of TGFbeta. These results indicate that STAT1 is required to mediate some but not all of the phenotypic changes induced by IFN gamma and that it also regulates iodine accumulation via TGFbeta signaling.

Il est une cytokine lib&#xE9;r&#xE9;e par les cellules mononucl&#xE9;aires infiltrant qui assure la m&#xE9;diation de ses actions principalement par de transduction de signal et l&#x26;#39;activateur de la transcription-1 (STAT1) mais aussi &#xE0; travers d&#x26;#39;autres facteurs de transcription. Afin de diss&#xE9;quer l&#x26;#39;effet de l&#x26;#39;interf&#xE9;ron gamma sur la morphologie de la thyro&#xEF;de et la fonction, nous avons travers&#xE9; des souris transg&#xE9;niques qui expriment l&#x26;#39;interf&#xE9;ron gamma en particulier dans la glande thyro&#xEF;de pour les souris d&#xE9;ficientes en STAT1. Manque de STAT1 am&#xE9;lior&#xE9; la morphologie de la thyro&#xEF;de anormale et l&#x26;#39;hypothyro&#xEF;die primaire typique de souris transg&#xE9;niques IFN gamma, mais pas l&#x26;#39;accumulation d&#x26;#39;iode supprim&#xE9;. Fait int&#xE9;ressant, l&#x26;#39;absence de STAT1 seule diminution de l&#x26;#39;accumulation d&#x26;#39;iode, en apparence &#xE0; travers l&#x26;#39;expression de TGFbeta. Ces r&#xE9;sultats indiquent que STAT1 est n&#xE9;cessaire pour servir de m&#xE9;diateur certains mais pas tous les changements ph&#xE9;notypiques induites par l&#x26;#39;interf&#xE9;ron gamma et qu&#x26;#39;il r&#xE9;glemente &#xE9;galement l&#x26;#39;accumulation de l&#x26;#39;iode par l&#x26;#39;interm&#xE9;diaire de signalisation TGFbeta.

Es ist ein Zytokin, ver&ouml;ffentlicht von MONONUKLE&Auml;RE Zellen einzuschleusen, die ihre Aktionen vor allem durch Signal-Wandler und Aktivator der Transkription-1 (STAT1), sondern auch durch andere Transkriptionsfaktoren vermittelt. Um die Wirkung von IFN-Gamma auf Schilddr&uuml;se Morphologie und Funktion zu zergliedern, &uuml;berquerten wir transgene M&auml;use, die IFN-Gamma, speziell in der Schilddr&uuml;se an M&auml;usen mangelhaft ist in STAT1 Ausdr&uuml;cken. Mangelnde STAT1 verbessert die abnormale Schilddr&uuml;sen-Morphologie und die prim&auml;re Hypothyreose, die typisch f&uuml;r IFN-Gamma transgene M&auml;use aber nicht die unterdr&uuml;ckte Jod-Ansammlung. Interessanterweise fehlen STAT1 allein verminderte Jod Akkumulation, scheinbar durch Ausdruck der TGFbeta. Diese Ergebnisse zeigen, dass STAT1 ben&ouml;tigt wird, um einige zu vermitteln, aber nicht alle die ph&auml;notypischen Ver&auml;nderungen von IFN-Gamma induziert und es auch Jod Akkumulation &uuml;ber TGFbeta zu signalisieren reguliert.

Interferone (IFN)-gamma &egrave; stato coinvolto nella patogenesi della tiroidite di Hashimoto. &Egrave; una citochina rilasciata da infiltrazione di cellule mononucleate che media le azioni principalmente attraverso il trasduttore di segnale e attivatore di trascrizione-1 (STAT1) ma anche attraverso altri fattori di trascrizione. Per sezionare l&#39;effetto dell&#39;IFN gamma sulla tiroide morfologia e funzione, abbiamo attraversato topi transgenici che esprimono IFN gamma specificamente nella ghiandola tiroide a topi carenti di STAT1. Mancanza di STAT1 migliorata la morfologia anomala della tiroide e l&#39;ipotiroidismo primario tipico di topi transgenici IFN gamma, ma non l&#39;accumulo di iodio soppressi. &Egrave; interessante notare, mancanza di STAT1 solo diminuito accumulo di iodio, apparentemente attraverso expression of TGFbeta. Questi risultati indicano che STAT1 &egrave; necessario mediare alcuni ma non tutti i cambiamenti fenotipici indotta da IFN gamma e che regola anche accumulo di iodio tramite TGFbeta signaling.

Germinomas arising in the sella turcica are difficult to differentiate from autoimmune hypophysitis because of similar clinical and pathological features. This differentiation, nevertheless, is critical for patient care due to different treatments of the two diseases. We report the case of an 11-year-old girl who presented with diabetes insipidus and growth retardation, and was found to have an intra- and supra-sellar mass. Initial examination of the pituitary biopsy showed diffuse lymphocytic infiltration of the adenohypophysis and absent placental alkaline phosphatase expression, leading to a diagnosis of hypophysitis and glucocorticoid treatment. Because of the lack of clinical and radiological response, the pituitary specimen was re-examined, revealing this time the presence of scattered c-kit and Oct4 positive germinoma cells. The revised diagnosis prompted the initiation of radiotherapy, which induced disappearance of the pituitary mass. Immunological studies showed that the patient's serum recognized antigens expressed by the patient's own germinoma cells, as well as pituitary antigens like growth hormone and systemic antigens like the SjÃ¶gren syndrome antigen B and alpha-enolase. The study first reports the presence of pituitary and systemic antibodies in a patient with intrasellar germinoma, and reminds us that diffuse lymphocytic infiltration of the pituitary gland and pituitary antibodies does not always indicate a diagnosis of autoimmune hypophysitis.

Germinomas que se plantean en la silla turca son dif&#xED;ciles de diferenciar de hipofisitis autoinmune, debido a similares caracter&#xED;sticas cl&#xED;nicas y patol&#xF3;gicas. Esta diferenciaci&#xF3;n, sin embargo, es fundamental para el cuidado del paciente, debido a diferentes tratamientos de las dos enfermedades. Presentamos el caso de una ni&#xF1;a de 11 a&#xF1;os de edad que se present&#xF3; con diabetes ins&#xED;pida y retardo del crecimiento, y se encontr&#xF3; que ten&#xED;a una masa intra y supra selar. El examen inicial de la biopsia de gl&#xE1;ndula pituitaria mostr&#xF3; infiltraci&#xF3;n linfoc&#xED;tica difusa de la adenohip&#xF3;fisis y ausente la expresi&#xF3;n de fosfatasa alcalina placentaria, lo que lleva a un diagn&#xF3;stico de hipofisitis y el tratamiento con glucocorticoides. El diagn&#xF3;stico revisado llev&#xF3; a la iniciaci&#xF3;n de la radioterapia, que indujo la desaparici&#xF3;n de la masa pituitaria. Interleukin-12 (IL-12) is a pleiotropic cytokine that is elevated in the affected organs of patients with SjÃ¶gren's syndrome (SS). We have previously reported that overexpression of IL-12 in CBA mice leads to mononuclear infiltration of salivary and lacrimal glands, as well as to expansion of bronchial lymphoid tissue and decreased mucociliary clearance. Because xerostomia is one of the most important clinical features in SS patients, our main objective in the current study was to evaluate salivary gland function in IL-12-transgenic mice. Our secondary objective was to further characterize this animal model and to determine if the changes observed in these mice are representative of those observed in patients with SS overall.

La interleucina-12 (IL-12) es una citocina pleiotr&#xF3;pica que se eleva en los &#xF3;rganos afectados de pacientes con s&#xED;ndrome de Sj&#xF6;gren (SS). Hemos informado anteriormente que la sobreexpresi&#xF3;n de la IL-12 en ratones CBA conduce a la infiltraci&#xF3;n mononuclear de gl&#xE1;ndulas salivares y lagrimales, as&#xED; como a la expansi&#xF3;n del tejido linfoide bronquial y disminuci&#xF3;n del aclaramiento mucociliar. Debido a que la xerostom&#xED;a es una de las caracter&#xED;sticas cl&#xED;nicas m&#xE1;s importantes en los pacientes con SS, nuestro principal objetivo del presente estudio fue evaluar la funci&#xF3;n de la gl&#xE1;ndula salival en IL-12-ratones transg&#xE9;nicos. Pregnancy modulates autoimmune diseases through diverse and still incompletely defined mechanisms, in part operating at the decidua-placenta interface. To assess the immunological contribution of placenta, we administered mouse placental proteins to a mouse model of autoimmune hypophysitis, a disease known to be strongly associated with pregnancy. Emulsified placental proteins suppressed both the cellular and humoral aspects of hypophysitis. Suppression was specific to self antigens and not seen when two foreign antigens, tetanus toxoid or tuberculin purified protein derivative, were used. Proteomic analysis revealed high levels of soluble TNF receptor 1 in placenta, suggesting that blockade of the TNF-Î± pathway was a mechanism of disease suppression. Placentas derived from mice deficient in TNF receptor 1 lost the ability to suppress hypophysitis. Notably, hypophysitis suppression was seen only when the TNF-Î± pathway was blocked locally, at the site of immunization, and not systemically. These findings provide evidence that placenta contributes to the immune tolerance of pregnancy by locally inhibiting the TNF-Î± pathway.

Pour &#xE9;valuer la contribution immunologique du placenta, nous avons administr&#xE9; aux prot&#xE9;ines de souris placentaires &#xE0; un mod&#xE8;le murin de hypophysite auto-immune, une maladie connue pour &#xEA;tre fortement associ&#xE9;e &#xE0; la grossesse. &#xC9;mulsionn&#xE9;es prot&#xE9;ines placentaires supprim&#xE9; &#xE0; la fois les aspects cellulaires et humorales de hypophysite. R&#xE9;pression &#xE9;tait sp&#xE9;cifique aux antig&#xE8;nes du soi et de ne pas voir quand deux antig&#xE8;nes &#xE9;trangers, l&#x26;#39;anatoxine t&#xE9;tanique ou un d&#xE9;riv&#xE9; de prot&#xE9;ine purifi&#xE9;e, la tuberculine ont &#xE9;t&#xE9; utilis&#xE9;s. L&#x26;#39;analyse prot&#xE9;omique a r&#xE9;v&#xE9;l&#xE9; des niveaux &#xE9;lev&#xE9;s de r&#xE9;cepteur soluble du TNF 1 dans le placenta, ce qui sugg&#xE8;re que le blocage de la voie de TNF-&#x3B1; &#xE9;tait un m&#xE9;canisme de suppression de la maladie. Placentas issus de souris d&#xE9;ficientes en r&#xE9;cepteur 1 du TNF a perdu la capacit&#xE9; de supprimer hypophysite. Notamment, la suppression a &#xE9;t&#xE9; vu hypophysite que lorsque la voie de TNF-&#x3B1; a &#xE9;t&#xE9; bloqu&#xE9; &#xE0; proximit&#xE9;, au niveau du site de vaccination, et non syst&#xE9;mique. Ces r&#xE9;sultats fournissent la preuve que le placenta contribuent &#xE0; la tol&#xE9;rance immunitaire de la grossesse par des entreprises locales inhibition de la voie de TNF-&#x3B1;.

Proteomic Analyse ergab hohe Niveaus des l&ouml;slichen TNF-Rezeptors 1 in Plazenta, was darauf hindeutet, dass Blockade von TNF-&alpha;-Weg war ein Mechanismus der Krankheit Unterdr&uuml;ckung. Gravidanza modula malattie autoimmuni attraverso meccanismi diversi e ancora non completamente definiti, in parte operanti presso l&#39;interfaccia decidua-placenta. Per valutare il contributo immunologico della placenta, abbiamo somministrato proteine placentari del mouse in un modello murino di hypophysitis autoimmune, una malattia conosciuta per essere fortemente associato con la gravidanza. Proteine placentare emulsionate soppressi gli aspetti umorali e cellulari di hypophysitis. Soppressione era specifici per gli antigeni self e non si vede quando due antigeni estranei, Toxoide di tetano o tubercolina derivato proteico purificato sono stati utilizzati. Embarazo modula las enfermedades autoinmunes a trav&#xE9;s de diversos mecanismos y a&#xFA;n incompletamente determinada, en parte de operaci&#xF3;n en la interfaz decidua-placenta.
