Termites are known to have an extraordinary reproductive plasticity and capacity, but the underlying genetic patterns of termite reproductive biology are relatively understudied. The goal of this study was to identify genes for which expression levels differ between dealated precopulatory females (virgins) and egg-laying queens of the Formosan subterranean termite, Coptotermes formosanus Shiraki. We constructed a normalized polyphenic expressed sequence tag (EST) library that represents genomic material from most of the castes and life stages of the Formosan subterranean termite. Microarrays were designed using probes from this EST library and public genomic resources. Virgin females and queens were competitively hybridized to these microarrays and differentially expressed candidate genes were identified. Differential expression of eight genes was subsequently confirmed via reverse transcriptase quantitative PCR (RT-QPCR). When compared to virgins, queens had higher expression of genes coding for proteins related to immunity (gram negative binding protein), nutrition (e.g., termite-derived endo-beta-1,4-glucanase), protein storage, regulation of caste differentiation and reproduction (hexamerin, juvenile hormone binding protein). Queens also had higher transcript levels for genes involved in metabolism of xenobiotics, fat, and juvenile hormone (glutathione-S-transferase-like proteins, and cytochrome P450), among others. In particular, hexamerin, juvenile hormone binding protein, and a cytochrome P450 from the 4C subfamily are likely to be involved in initiating the inactive period during the reproductive cycle of the queen. Vice versa, virgins had higher expression than queens of genes related to respiration, probably due to recent flight activity, and several genes of unknown function.

Transcriptome profiling of female alates and egg-laying queens of the Formosan subterranean termite.

Profilage du transcriptome de alates femmes et de ponte de la Queens termites de Formose souterrain

Termiten sind daf&uuml;r bekannt, einen au&szlig;erordentlichen reproduktiven Plastizit&auml;t und Kapazit&auml;t verf&uuml;gen, aber die zugrunde liegenden genetischen Muster der Termite Reproduktionsbiologie sind relativ Sarjetschnaja. Ziel dieser Studie war es, Gene zu identifizieren, f&uuml;r die Stufen zwischen dealated precopulatory Frauen (Jungfrauen) und eierlegenden K&ouml;niginnen die formosische unterirdische Termite, Coptotermes Formosanus Shiraki unterscheiden. Konstruierten wir eine normalisierten Polyphenic ausgedr&uuml;ckt-Sequenz-Tag (EST)-Bibliothek, die meisten der Kasten und Lebensphasen einer die formosische unterirdische Termite genomische Material darstellt. Queens hatte auch h&ouml;here Transkript f&uuml;r Genen, die im Stoffwechsel von Xenobiotica, Fett und Juvenilhormon (Glutathion-S-Transferase-wie Proteine und Cytochrom P450), unter anderem. Insbesondere d&uuml;rften Hexamerin, Juvenilhormon verbindliches Protein und ein Zellfarbstoff P450 aus der Unterfamilie der 4C initiieren den inaktiven Zeitraum w&auml;hrend der reproduktiven Zyklus der K&ouml;nigin beteiligt sein. Umgekehrt hatte die Jungfrauen h&ouml;heren Ausdruck als K&ouml;niginnen von Genen mit Bezug zu Atmung, wahrscheinlich wegen den letzten Flug-Aktivit&auml;t, und mehrere Gene unbekannter Funktion.

L&#39;obiettivo di questo studio era di identificare geni per cui i livelli di espressione differiscono tra dealated precopulatory femmine (vergini) e le regine di deposizione delle uova della termite sotterranea formosane, Coptotermes formosanus Shiraki. In particolare, hexamerin, proteina ormone giovanile e un citocromo P450 da sottofamiglia C 4 rischiano di essere coinvolti in occasione dell&#39;avvio del periodo inattivo durante il ciclo riproduttivo della regina. Las termitas se sabe que tienen una extraordinaria plasticidad y la capacidad reproductiva, pero los patrones gen&#xE9;ticos subyacentes de la biolog&#xED;a reproductiva de las termitas son relativamente poco estudiado. El objetivo de este estudio fue identificar los genes que difieren entre los niveles de expresi&#xF3;n dealated precopulatorio mujeres (v&#xED;rgenes) y la postura-reinas de la termita subterr&#xE1;nea de Formosa, Coptotermes formosanus Shiraki. Hemos construido un polifen&#xF3;licos normalizado etiquetas de secuencias expresadas (EST) biblioteca que representa el material gen&#xF3;mico de la mayor&#xED;a de las castas y etapas de la vida de la termita subterr&#xE1;nea de Formosa. Microarrays fueron dise&#xF1;ados utilizando las sondas de esta biblioteca p&#xFA;blica EST y recursos gen&#xF3;micos. Hembras v&#xED;rgenes y reinas se hibrid&#xF3; competitivamente a estos microarrays de genes expresados &#x200B;&#x200B;diferencialmente y candidatos fueron identificados. La expresi&#xF3;n diferencial de los ocho genes se confirm&#xF3; posteriormente a trav&#xE9;s de PCR transcriptasa inversa cuantitativa (RT-QPCR). En comparaci&#xF3;n con las v&#xED;rgenes, reinas ten&#xED;an una mayor expresi&#xF3;n de genes que codifican para prote&#xED;nas relacionadas con la inmunidad (prote&#xED;na de uni&#xF3;n a gram-negativas), nutrici&#xF3;n (por ejemplo, las termitas derivado de endo-beta-1 ,4-glucanasa), el almacenamiento de prote&#xED;nas, regulaci&#xF3;n de la diferenciaci&#xF3;n de castas y reproducci&#xF3;n (hexamerin, la prote&#xED;na de uni&#xF3;n a hormonas juveniles). Queens tambi&#xE9;n ten&#xED;an altos niveles de transcripci&#xF3;n de genes implicados en el metabolismo de xenobi&#xF3;ticos, la grasa y la hormona juvenil (glutati&#xF3;n-S-transferasa-como las prote&#xED;nas, y el citocromo P450), entre otros. En particular, hexamerin, la prote&#xED;na de uni&#xF3;n a la hormona juvenil, y un citocromo P450 de la subfamilia 4C son propensos a estar involucrados en la iniciaci&#xF3;n del per&#xED;odo de inactividad durante el ciclo reproductivo de la reina. A la inversa, las v&#xED;rgenes ten&#xED;an una mayor expresi&#xF3;n de las reinas de los genes relacionados con la respiraci&#xF3;n, probablemente debido a la actividad de vuelo reciente, y de varios genes de funci&#xF3;n desconocida.

The goal of this study was to test whether the breeding system and/or the degree of inbreeding of field colonies of the Formosan subterranean termite, Coptotermes formosanus, Shiraki (Isoptera: Rhinotermitidae) influences bacterial load on the cuticle of foraging workers. We enumerated bacterial load on the cuticle of groups of workers foraging in 20 inground monitoring stations surrounding the French Market in New Orleans, LA, and identified bacteria species using 16S rRNA gene sequencing. We used microsatellite genotyping to assign the 20 worker groups to seven simple family colonies (headed by a single pair of reproductives) and four extended family colonies (headed by multiple inbreeding reproductives) with a wide range of degrees of inbreeding. Workers from extended family colonies had a higher bacterial load than those from simple family colonies; however, bacterial load was not significantly correlated to the degree of inbreeding, possibly because of confounding factors in colony life history, such as age and/or size of colonies. Colonies with high bacterial load did not have a higher proportion of entomopathogens, and thus, bacterial load is not necessarily an indicator for disease risk. The majority of bacteria cultured from the cuticle of termites were soil bacteria with no known pathology against termites.
