Brucella spp. are intracellular bacteria that cause the most frequent zoonosis in the world. Although recent work has advanced the field of Brucella vaccine development, there remains no safe human vaccine. In order to produce a safe and effective human vaccine, the immune response to Brucella spp. requires greater understanding. Induction of Brucella-specific CD8+ T cells is considered an important aspect of the host response; however, the CD8+ T-cell response is not clearly defined. Discovering the epitope containing antigens recognized by Brucella-specific CD8+ T cells and correlating them with microarray data will aid in determining proteins critical for vaccine development that cover a kinetic continuum during infection. Developing tools to take advantage of the BALB/c mouse model of Brucella melitensis infection will help to clarify the correlates of immunity and improve the efficacy of this model. Two H-2(d) CD8+ T-cell epitopes have been characterized, and a group of immunogenic proteins have provoked gamma interferon production by CD8+ T cells. RYCINSASL and NGSSSMATV induced cognate CD8+ T cells after peptide immunization that showed specific killing in vivo. Importantly, we found by microarray analysis that the genes encoding these epitopes are differentially expressed following macrophage infection, further emphasizing that these discordant genes may play an important role in the pathogenesis of B. melitensis infection.

Brucella spp. son bacterias intracelulares que causan la zoonosis m&#xE1;s frecuente en el mundo. A pesar de los trabajos recientes ha avanzado en el campo de desarrollo de la vacuna Brucella, no queda ninguna vacuna segura. Con el fin de producir una vacuna humana segura y efectiva, la respuesta inmune a la Brucella spp. requiere una mayor comprensi&#xF3;n. La inducci&#xF3;n de la Brucella espec&#xED;ficos de linfocitos T CD8 + se considera un aspecto importante de la respuesta del hu&#xE9;sped, sin embargo, los CD8 + respuesta de c&#xE9;lulas T no est&#xE1; claramente definido. Descubriendo el epitopo que contiene ant&#xED;genos reconocidos por Brucella espec&#xED;fica linfocitos T CD8 + y su correlaci&#xF3;n con los datos de microarrays ayudar&#xE1; en la determinaci&#xF3;n de prote&#xED;nas cr&#xED;ticas para el desarrollo de vacunas que cubren un continuo cin&#xE9;tica durante la infecci&#xF3;n. Desarrollo de herramientas para aprovechar el modelo de rat&#xF3;n BALB / c de la infecci&#xF3;n por Brucella melitensis, ayudar&#xE1;n a clarificar los correlatos de inmunidad y mejorar la eficacia de este modelo. Dos H-2 (d) CD8 + ep&#xED;topos de c&#xE9;lulas T se han caracterizado, y un grupo de prote&#xED;nas inmunog&#xE9;nicas han provocado la producci&#xF3;n de interfer&#xF3;n gamma por los linfocitos T CD8 +. RYCINSASL y NGSSSMATV afines CD8 + inducida por las c&#xE9;lulas T despu&#xE9;s de la inmunizaci&#xF3;n p&#xE9;ptido que mostraba destrucci&#xF3;n espec&#xED;fica in vivo. Es importante destacar que hemos encontrado por an&#xE1;lisis de microarrays que los genes que codifican estos ep&#xED;topes son expresados &#x200B;&#x200B;diferencialmente despu&#xE9;s de la infecci&#xF3;n de macr&#xF3;fagos, adem&#xE1;s haciendo hincapi&#xE9; en que estos genes discordantes pueden jugar un papel importante en la patog&#xE9;nesis de la infecci&#xF3;n por B. melitensis.
