Towards the goal of developing a real-time monitoring device for microorganisms, we demonstrate the use of microcantilevers as resonant mass sensors for detection of Bacillus anthracis Sterne spores in air and liquid. The detection scheme was based on measuring resonant frequency decrease driven by thermally induced oscillations, as a result of the added mass of the spores with the use of a laser Doppler vibrometer (LDV). Viscous effects were investigated by comparing measurements in air and deionized (DI) water along with theoretical values. Moreover, biological experiments were performed which involved suspending spores onto the cantilevers and performing mass detection in air and water. For detection of spores in water, the cantilevers were functionalized with antibodies in order to fix the spores onto the surface. We demonstrate that as few as 50 spores on the cantilever can be detected in water using the thermal noise as excitation source. Measurement sensitivity of 9.23 Hz/fg for air and 0.1 Hz/fg for water were obtained. These measurements were compared with theoretical values and sources of improvement in cantilever sensitivity in a viscous medium were also discussed. It is expected that by driving the cantilevers and using higher order modes, detection of a single spore in liquids should be achievable.

Vers le but de d&#xE9;velopper un dispositif de surveillance en temps r&#xE9;el pour les micro-organismes, nous d&#xE9;montrons l&#x26;#39;utilisation de capteurs de masse comme microleviers r&#xE9;sonance pour la d&#xE9;tection des spores de Bacillus anthracis Sterne dans l&#x26;#39;air et de liquide. Le syst&#xE8;me de d&#xE9;tection bas&#xE9; sur la mesure a &#xE9;t&#xE9; diminution de fr&#xE9;quence de r&#xE9;sonance entra&#xEE;n&#xE9;s par des oscillations induites thermiquement, &#xE0; la suite de la masse ajout&#xE9;e des spores avec l&#x26;#39;utilisation d&#x26;#39;un vibrom&#xE8;tre laser &#xE0; effet Doppler (LDV). Effets visqueux ont &#xE9;t&#xE9; &#xE9;tudi&#xE9;s en comparant les mesures dans l&#x26;#39;air et d&#xE9;sionis&#xE9;e (DI) de l&#x26;#39;eau ainsi que les valeurs th&#xE9;oriques. En outre, les exp&#xE9;riences biologiques ont &#xE9;t&#xE9; r&#xE9;alis&#xE9;es qui consiste &#xE0; suspendre les spores sur les consoles et effectuer la d&#xE9;tection de masse dans l&#x26;#39;air et l&#x26;#39;eau. Pour la d&#xE9;tection des spores dans l&#x26;#39;eau, les consoles ont &#xE9;t&#xE9; fonctionnalis&#xE9;s avec des anticorps afin de fixer les spores sur la surface. Nous d&#xE9;montrons que aussi peu que 50 spores sur le cantilever peut &#xEA;tre d&#xE9;tect&#xE9;e dans l&#x26;#39;eau en utilisant le bruit thermique comme source d&#x26;#39;excitation. Sensibilit&#xE9; de la mesure de 9,23 Hz / fg pour l&#x26;#39;air et de 0,1 Hz / fg pour l&#x26;#39;eau ont &#xE9;t&#xE9; obtenus. Ces mesures ont &#xE9;t&#xE9; compar&#xE9;es avec les valeurs th&#xE9;oriques et les sources d&#x26;#39;am&#xE9;lioration de la sensibilit&#xE9; en porte &#xE0; faux dans un milieu visqueux ont &#xE9;galement &#xE9;t&#xE9; discut&#xE9;s. Das Ziel der Entwicklung einer Echtzeit-&Uuml;berwachungsger&auml;t f&uuml;r Mikroorganismen zeigen wir den Einsatz von Microcantilevers als resonanten Masse Sensoren zur Erkennung von Bacillus Anthracis, die Sterne in die Luft und Fl&uuml;ssigkeit Sporen. Die Nachweis-Regelung beruhte auf der Messung der Resonanzfrequenz Abnahme von thermisch induzierten Schwingungen, durch die Masse der Sporen mit dem Einsatz von einem Laser-Doppler-Vibrometer (LDV) angetrieben. Z&auml;hfl&uuml;ssige Effekte wurden untersucht, durch Messungen in Luft und deionisiertem Wasser absp&uuml;len (DI) zusammen mit theoretischen Werten vergleichen. Dar&uuml;ber hinaus wurden biologische Experimente durchgef&uuml;hrt, die Aussetzung der Sporen auf den Ausleger und Durchf&uuml;hren von mass-Erkennung in Luft und Wasser beteiligt. Zum Nachweis von Sporen in Wasser wurden die Ausleger mit Antik&ouml;rpern funktionalisierten, um die Sporen auf die Oberfl&auml;che zu befestigen. Wir zeigen, dass so wenig wie 50 Sporen auf der Ausladung im Wasser mit das thermisches Rauschen als Anregungsquelle erkannt werden k&ouml;nnen. Messempfindlichkeit von 9.23 Hz/fg f&uuml;r Luft und 0,1 Hz/fg f&uuml;r Wasser wurden erzielt. Diese Messungen wurden mit theoretischen Werten verglichen und Quellen der Freischwinger Empfindlichkeit in einem viskosen Medium wurden ebenfalls diskutiert. Es wird erwartet, dass durch Nutzung der Ausleger und die Verwendung h&ouml;herer Ordnung-Modi, Erkennung von einem einzigen Spore in Fl&uuml;ssigkeiten erreichbar sein sollte.

Verso l&#39;obiettivo di sviluppare un dispositivo di monitoraggio in tempo reale per i microorganismi, dimostriamo l&#39;uso di microcantilevers come risonante massa sensori per il rilevamento di Bacillus anthracis che Sterne spore nell&#39;aria e liquido. Lo schema di rilevazione era basato sulla misurazione diminuzione frequenza risonante guidato da oscillazioni indotte termicamente, in seguito la massa aggiunta delle spore con l&#39;uso di un laser Doppler Vibrometro (LDV). Confrontando le misure in aria e acqua deionizzata (DI) insieme con i valori teorici sono stati indagati gli effetti viscosi. Inoltre, esperimenti biologici sono stati eseguiti che ha coinvolto la sospensione di spore sul cantilever e l&#39;esecuzione di massa rilevamento in aria e acqua. Per la rilevazione delle spore in acqua, il cantilever sono stati funzionalizzati con anticorpi per fissare le spore sulla superficie. Dimostriamo che come pochi come 50 spore sul cantilever possono essere rilevate in acqua usando il rumore termico come fonte di eccitazione. Sensibilit&agrave; di misura di 9.23 Hz/fg per aria e 0.1 Hz/fg per l&#39;acqua sono stati ottenuti. Queste misurazioni sono state confrontate con i valori teorici e fonti di miglioramento nella sensibilit&agrave; a sbalzo in un mezzo viscoso sono stati anche discussi. &Egrave; previsto che guida il cantilever e utilizzando la modalit&agrave; di ordine superiore, rilevazione di una singola spora in liquidi dovrebbe essere realizzabile.

Hacia el objetivo de desarrollar un dispositivo de vigilancia en tiempo real para los microorganismos, que demuestran el uso de microcantilevers como sensores de masa resonantes para la detecci&#xF3;n de esporas de Bacillus anthracis Sterne en el aire y l&#xED;quido. El esquema de detecci&#xF3;n se basa en la medici&#xF3;n de disminuci&#xF3;n de la frecuencia resonante impulsado por las oscilaciones inducidas t&#xE9;rmicamente, como resultado de la masa a&#xF1;adida de las esporas con el uso de un vibr&#xF3;metro l&#xE1;ser Doppler (LDV). Los efectos viscosos se investigaron mediante la comparaci&#xF3;n de las mediciones en el aire y agua desionizada (DI) de agua junto con los valores te&#xF3;ricos. Por otra parte, los experimentos biol&#xF3;gicos se realizaron suspensi&#xF3;n de esporas que participaron en los voladizos y la realizaci&#xF3;n de la detecci&#xF3;n de masas en el aire y el agua. Para la detecci&#xF3;n de las esporas en el agua, los voladizos se funcionalizado con anticuerpos con el fin de fijar las esporas sobre la superficie. Se demuestra que tan s&#xF3;lo 50 esporas en el voladizo se puede detectar en agua utilizando el ruido t&#xE9;rmico como fuente de excitaci&#xF3;n. La medici&#xF3;n de la sensibilidad 9,23 Hz / fg para el aire y 0,1 Hz / fg de agua se obtuvieron. Estas mediciones se compararon con los valores te&#xF3;ricos y las fuentes de mejora en la sensibilidad en voladizo en un medio viscoso se examinaron tambi&#xE9;n. The size of a cell is a fundamental physiological property and is closely regulated by various environmental and genetic factors. Optical or confocal microscopy can be used to measure the dimensions of adherent cells, and Coulter counter or flow cytometry (forward scattering light intensity) can be used to estimate the volume of single cells in a flow. Although these methods could be used to obtain the mass of single live cells, no method suitable for directly measuring the mass of single adherent cells without detaching them from the surface is currently available. We report the design, fabrication, and testing of 'living cantilever arrays', an approach to measure the mass of single adherent live cells in fluid using silicon cantilever mass sensor. HeLa cells were injected into microfluidic channels with a linear array of functionalized silicon cantilevers and the cells were subsequently captured on the cantilevers with positive dielectrophoresis. The captured cells were then cultured on the cantilevers in a microfluidic environment and the resonant frequencies of the cantilevers were measured. The mass of a single HeLa cell was extracted from the resonance frequency shift of the cantilever and was found to be close to the mass value calculated from the cell density from the literature and the cell volume obtained from confocal microscopy. This approach can provide a new method for mass measurement of a single adherent cell in its physiological condition in a non-invasive manner, as well as optical observations of the same cell. We believe this technology would be very valuable for single cell time-course studies of adherent live cells.

'Living cantilever arrays' for characterization of mass of single live cells in fluids.

Riportiamo la progettazione, fabbricazione e la sperimentazione di &#39;living matrici a sbalzo&#39;, un approccio per misurare la massa di singole cellule vive aderente nel fluido mediante sensore massa di silicio a sbalzo. Le cellule HeLa sono state iniettate nei canali microfluidici con un array lineare di silicio funzionalizzate cantilever e le cellule vennero successivamente catturate su cantilever con dielettroforesi positiva. Le cellule catturate poi sono state coltivate su cantilever in un ambiente di microfluidica e sono state misurate le frequenze risonanti del cantilever. La massa di una singola cellula HeLa &egrave; stato estratto dallo spostamento di frequenza di risonanza del cantilever ed &egrave; stata trovata per essere vicino al valore di massa calcolato la densit&agrave; delle cellule dalla letteratura e il volume delle cellule ottenute da microscopia confocal. Questo approccio pu&ograve; fornire un nuovo metodo per la misurazione di massa di una singola cella aderente nella sua condizione fisiologica in maniera non invasiva, come pure le osservazioni ottiche della cella stessa. Wir pr&auml;sentieren die Echtzeit&uuml;berwachung von luftgest&uuml;tzten Vaccinia-Viren mit Quarz Kristall Mikrowaage (QCM) in einer integrierten Weise. Vi presentiamo il rilevamento in tempo reale di virus di Vaccinia airborne utilizzando microbilancia a cristalli di quarzo (QCM) in modo integrato. Vaccinia virus erano aerosolizzati e neutralizzati utilizzando un generatore di aerosol electrospray, trasportati nella camera di QCM e catturato da un cristallo QCM. La cattura dei virus sul cristallo QCM ha provocato cambiamenti di frequenza proporzionale al numero di virus. Il tasso di cattura varia linearmente con la concentrazione delle sospensioni iniziali virus (8.5x10(8)-8.5x10(10) particlesml) alle portate di lmin 2.0 e 1.1. Questo lavoro dimostra il potenziale generale di rilevazione massa sensibile di entit&agrave; biologiche su scala nanometrica in aria.

We present real-time detection of airborne Vaccinia viruses using quartz crystal microbalance (QCM) in an integrated manner. Vaccinia viruses were aerosolized and neutralized using an electrospray aerosol generator, transported into the QCM chamber, and captured by a QCM crystal. The capture of the viruses on the QCM crystal resulted in frequency shifts proportional to the number of viruses. The capture rate varied linearly with the concentration of initial virus suspensions (8.5x10(8)-8.5x10(10) particlesml) at flow rates of 2.0 and 1.1 lmin. This work demonstrates the general potential of mass sensitive detection of nanoscale biological entities in air.

Detecci&#xF3;n en tiempo real de virus en el aire en un dispositivo sensible a la misa

This study evaluated predation with Bdellovibrio bacteriovorous and CO(2) aerosol spraying to remove fluorescent Escherichia coli biofilms from silicon chips. Initial tests found that 7.5×10(5) viable E. coli cells were dispersed into the surrounding environment during aerosol treatment. The total number dispersed per test decreased to only 16 for predated biofilms. This is nearly 50,000-fold lower compared to untreated chips and 1000-fold lower compared to chips soaked in HEPES buffer only. Both scanning electron microscopy (SEM) and fluorescent microscopy analyses confirmed that predation alone did not completely eradicate the biofilm population. When used in conjunction with CO(2) aerosols, however, no fluorescent signals remained and the SEM pictures showed a pristine surface devoid of bacteria. Consequently, this study demonstrates these two methods can be used with each other to significantly remove biofilms from surfaces while also significantly reducing the likelihood of human exposure to potential pathogens during their removal.

Combined application of bacterial predation and carbon dioxide aerosols to effectively remove biofilms.

The present study evaluated the removal of Escherichia coli XL1-blue biofilms using periodic jets of carbon dioxide aerosols (a mixture of solid and gaseous CO(2)) with nitrogen gas. The aerosols were generated by the adiabatic expansion of high-pressure CO(2) gas through a nozzle and used to remove air-dried biofilms. The areas of the biofilms were measured from scanning electron micrographs before and after applying the aerosols. The removal efficiency of the aerosol treatment was measured with various air-drying times of the biofilms before the treatment, surface materials, and durations of CO(2) aerosols in each 8-s aerosol-nitrogen cleaning cycle. Nearly 100% of the fresh biofilms were removed from the various surfaces very reliably within 90 s. This technique can be useful for removing unsaturated biofilms on solid surfaces and has potential applications for cleaning bio-contaminated surfaces.

Gas-phase removal of biofilms from various surfaces using carbon dioxide aerosols.

The influenza virus has received extensive attention due to the recent H1N1 pandemics originating from swine. This study reports a label-free, highly sensitive, and selective electrical immunosensor for the detection of influenza virus H1N1 based on dielectrophoretically deposited single-walled carbon nanotubes (SWCNTs). COOH-functionalized SWCNTs were deposited on a self-assembled monolayer of polyelectrolyte polydiallyldimethyl-ammonium chloride (PDDA) between two gold electrodes by dielectrophoretic and electrostatic forces, which resulted in reproducible, uniform, aligned, and aggregation-free SWCNT channels (2-10 μm in length). Avidin was immobilized onto the PDDA-SWCNT channels, and viral antibodies were immobilized using biotin-avidin coupling. The resistance of the channels increased with the binding of the influenza viruses to the antibodies. These immunosensors showed linear behavior as the virus concentration was varied from 1 to 10(4) PFU ml(-1) along with a detection time of 30 min. The immunosensors with a 2 μm channel length detected 1 PFU ml(-1) of the influenza virus accurately (R(2) = 0.99) and selectively from MS2 bacteriophages. These immunosensors have the potential to become an important component of a point-of-care test kit that will enable a rapid clinical diagnosis.

Electrical immunosensor based on dielectrophoretically-deposited carbon nanotubes for detection of influenza virus H1N1.

Reusability of a biosensor has recently received considerable attention, and it is closely related with the effective desorption of probe molecules. We present a novel mechanical desorption technique to reuse biosensors by using periodic jets of carbon dioxide (CO2) aerosols (a mixture of solid and gaseous CO2), and demonstrate its feasibility by removing physically adsorbed and covalently bonded fluorescent proteins i.e., Escherichia coli fluorescein isothiocyanate antibody and bovine serum albumin (E. coli FITC-Ab and FITC-BSA) from silicon chips. The proteins on the chip surfaces were measured by fluorescent images before and after applying the aerosols. The removal efficiency of the aerosol treatment was measured for various concentrations (1-20 μg mL(-1)) of E. coli FITC-Ab and FITC-BSA with two different removal cycles (5 and 11 cycles; each cycle: 8s). We observed high removal efficiencies (>93.5% for physically adsorbed Ab and >84.6% for covalently bonded Ab) at 11 cycle aerosol treatment. This CO2 aerosol treatment did not undermine re-functionalization, which was confirmed by the fluorescent images of FITC-Abs for fresh and reused chips. Desorption of the immobilized layers was validated by Fourier transform infrared and X-ray photoelectron spectroscopic analyses. We also conducted an experiment on the regeneration of E. coli sensing chips using this aerosol treatment, and the chips were re-used 5 times successfully. This mechanical desorption technique is a highly effective and novel strategy for reusable biosensors.

Mechanical desorption of immobilized proteins using carbon dioxide aerosols for reusable biosensors.

Osteopontin (OPN) is involved in almost all steps of cancer development, and it is being investigated as a potential biomarker for a diagnosis and prognosis of prostate cancer. Here, we report a label-free, highly sensitive and transparent immunosensor based on single-walled carbon nanotubes (SWCNTs) for detection of OPN. A high density of COOH functionalized SWCNTs was deposited between two gold/indium tin oxide electrodes on a glass substrate by dielectrophoresis. Monoclonal antibodies specific to OPN were covalently immobilized on the SWCNTs. Relative resistance change of the immunosensors was measured as the concentration of OPN in phosphate buffer saline (PBS) and human serum was varied from 1 pg mL(-1) to 1 μg mL(-1) for different channel lengths of 2, 5, and 10 μm, showing a highly linear and reproducible behavior (R(2)>97%). These immunosensors were also specific to OPN against another test protein, bovine serum albumin, PBS and human serum, showing that a limit of detection for OPN was 0.3 pg mL(-1). This highly sensitive and transparent immunosensor has a great potential as a simple point-of-care test kit for various protein biomarkers.

Single-walled carbon nanotube based transparent immunosensor for detection of a prostate cancer biomarker osteopontin.

A periodic jet of carbon dioxide (CO2) aerosols is a very quick and effective mechanical technique to remove biofilms from various substrate surfaces. However, the impact of the aerosols on the viability of bacteria during treatment has never been evaluated. In this study, the effects of high-speed CO2 aerosols, a mixture of solid and gaseous CO2, on bacteria viability was studied. It was found that when CO2 aerosols were used to disperse biofilms of Escherichia coli, they led to a significant loss of viability, with approximately 50% of the dispersed bacteria killed in the process. By comparison, 75.6% of the biofilm-associated bacteria were viable when gently dispersed using Proteinase K and DNase I. Indirect proof that the aerosols are damaging the bacteria was found using a recombinant E. coli expressing the cyan fluorescent protein, as nearly half of the fluorescence was found in the supernatant after CO2 aerosol treatment, while the rest was associated with the bacterial pellet. In comparison, the supernatant fluorescence was only 9% when the enzymes were used to disperse the biofilm. As such, these CO2 aerosols not only remove biofilm-associated bacteria effectively but also significantly impact their viability by disrupting membrane integrity.

Effects of Carbon Dioxide Aerosols on the Viability of Escherichia coli during Biofilm Dispersal.

Rapidity and high sensitivity are critical factors for the diagnoses of heart attacks, and cardiac troponin I (cTnI) is at present a clinical standard for its diagnosis. Here we report a rapid, label-free, and highly sensitive single-walled carbon nanotube (SWCNT) electrical immunosensor, featuring two pairs of electrodes. Two concentration electrodes (gaps: 25 and 80µm) and two detection electrodes (source and drain; gap: 20µm; width: 50µm) were used for dielectrophoretic concentration of cTnI on the SWCNT channels and resistance measurements of the dielectrophoresis (DEP)-concentrated cTnI, respectively. The two concentration electrodes were imbedded between upper and lower dielectric layers, facing each other, underneath the -COOH-functionalized SWCNT channels deposited between the detection electrodes. Therefore, the gap between these imbedded concentration electrodes can be reduced to maximize the electric field intensity for DEP-mediated concentration of cTnI, thereby greatly reducing the detection time (1min) and enhancing the limit of detection (0.7-0.8pgmL(-)(1)). Relative resistance changes of the SWCNTs were measured as cTnI concentration in Tris-Borate-EDTA (TBE; 0.0025×) and human serum diluted 500-fold with 0.0025× TBE decreased from 100ngmL(-)(1) to 1pgmL(-1), and they were shown to be linear with the logarithm of cTnI concentration (R(2)=0.99 and 0.97, respectively). These immunosensors also showed high specificity over another cardiac biomarker, myoglobin, TBE medium (0.0025×), and 500-fold diluted human serum. The DEP-capture of cTnI depended on the frequency of the applied electric field, demonstrating the qualitative nature of the real part of the Clausius-Mossotti factor for cTnI.

Rapid electrical immunoassay of the cardiac biomarker troponin I through dielectrophoretic concentration using imbedded electrodes.

Measurements of airborne viruses via sampling have been critical issues. Most electrostatic samplers have been assessed for bacterial aerosols or micrometer-sized viral particles; however, sampling of submicrometer-sized airborne viruses is necessary, especially because of the high probability of their staying airborne and their deposition in the lower respiratory tract. Here, we present a novel personal electrostatic particle concentrator (EPC) for gentle sampling of submicrometer airborne virus particles. Owing to the enhanced electric field designed in this EPC, the collection efficiencies reached values as high as 99.3-99.8% for 0.05-2 μm diameter polystyrene particles at a flow rate of 1.2 L/min. Submicrometer-sized MS2 and T3 virus particles were also collected in the EPC, and the concentrations relative to their respective initial suspensions were more than 10 times higher than those in the SKC BioSampler. Moreover, the recovery rate of T3 was 982 times higher in the EPC (-2 kV) than in the BioSampler at 12.5 L/min because of the gentle sampling of the EPC. Gentle sampling is desirable because many bioaerosols suffer from significant viability losses during sampling. The influence of ozone generated, applied electrostatic field, and the flow rate on the viability of the viruses will also be discussed.
