Kaposi sarcoma (KS) is the most common AIDS-associated malignancy and is characterized by angiogenesis and the presence of spindle cells. Kaposi sarcoma-associated herpesvirus (KSHV) is consistently associated with all clinical forms of KS, and in vitro infection of dermal microvascular endothelial cells (DMVECs) with KSHV recapitulates many of the features of KS, including transformation, spindle cell proliferation, and angiogenesis. To study the molecular mechanisms of KSHV pathogenesis, we compared the protein expression profiles of KSHV-infected and uninfected DMVECs. This comparison revealed that heme oxygenase-1 (HO-1), the inducible enzyme responsible for the rate-limiting step in heme catabolism, was up-regulated in infected endothelial cells. Recent evidence suggests that the products of heme catabolism have important roles in endothelial cell biology, including apoptosis and angiogenesis. Here we show that HO-1 mRNA and protein are up-regulated in KSHV-infected cultures. Comparison of oral and cutaneous AIDS-KS tissues with normal tissues revealed that HO-1 mRNA and protein were also up-regulated in vivo. Increased HO-1 enzymatic activity in vitro enhanced proliferation of KSHV-infected DMVECs in the presence of free heme. Treatment with the HO-1 inhibitor chromium mesoporphyrin IX abolished heme-induced proliferation. These data suggest that HO-1 is a potential therapeutic target for KS that warrants further study.

Sarcome de Kaposi (SK) est la plus courante malignit&eacute; associ&eacute;es au sida et se caract&eacute;rise par l&#39;angiogen&egrave;se et la pr&eacute;sence de cellules de tige. Herp&egrave;svirus de sarcome-associ&eacute; de Kaposi (KSHV) est constamment associ&eacute; &agrave; toutes les formes cliniques de KS et infection in vitro de cellules endoth&eacute;liales microvasculaires (DMVECs) par voie cutan&eacute;e avec KSHV reprend bon nombre des caract&eacute;ristiques de KS, y compris la transformation, la prolif&eacute;ration de cellules fusiformes et l&#39;angiogen&egrave;se. Sarcoma de Kaposi (KS) es la neoplasia m&aacute;s frecuente asociado al SIDA y se caracteriza por la angiog&eacute;nesis y la presencia de c&eacute;lulas fusiformes. Herpesvirus asociado sarcoma de Kaposi (KSHV) se asocia sistem&aacute;ticamente a todas las formas cl&iacute;nicas de KS, y la infecci&oacute;n in vitro de c&eacute;lulas endoteliales microvasculares cut&aacute;neas (DMVECs) con KSHV recoge muchas de las caracter&iacute;sticas de KS, incluyendo la transformaci&oacute;n, la proliferaci&oacute;n de c&eacute;lulas fusiformes y angiog&eacute;nesis. Para estudiar los mecanismos moleculares de la patogenesia KSHV, comparamos los perfiles de expresi&oacute;n de la prote&iacute;na de DMVECs KSHV infectados y no infectados. Esta comparaci&oacute;n revel&oacute; que hemo oxigenasa-1 (HO-1), la enzima inducible responsable para el paso de limitaci&oacute;n de velocidad en el catabolismo del grupo heme, era regulado hasta en c&eacute;lulas endoteliales infectadas. La evidencia reciente sugiere que los productos del catabolismo del grupo heme tienen un papel importante en la biolog&iacute;a de la c&eacute;lula endotelial, incluyendo la apoptosis y la angiog&eacute;nesis. Aqu&iacute; demostramos que HO-1 mRNA y prote&iacute;na son regulados por arriba en culturas KSHV infectados. We report here increased expression of KSHV lytic genes and increased release of progeny virus when synchronized cultures of body cavity-based lymphoma-1 cells are treated with a phorbol ester during S phase of the cell cycle.

Kaposi's sarcoma (KS) is a multifocal angioproliferative disorder affecting the skin, mucosa and viscera of individuals infected with human herpesvirus-8 (HHV-8; also Kaposi's sarcoma-associated herpesvirus [KSHV]). KS is the most common neoplasm in AIDS patients; the clinical outcome of AIDS-KS is significantly improved by highly active antiretroviral therapy (HAART). However, in Africa, where the severest manifestations of KS occur, there is limited access to these and other effective but expensive drugs. Here we present a review of current efforts to identify novel therapeutic targets for the treatment of KS using functional genomics, with recommendations regarding the development of economically feasible treatments for use in Africa.

KS est la tumeur plus courante chez les patients du sida ; le r&eacute;sultat clinique du sida-KS est nettement am&eacute;lior&eacute; par un traitement antir&eacute;troviral hautement actif (HAART). Cependant, en Afrique, o&ugrave; les manifestations les plus s&eacute;v&egrave;res de KS se produisent, il y a un acc&egrave;s limit&eacute; &agrave; ces et autre en vigueur mais les m&eacute;dicaments co&ucirc;teux. Nous pr&eacute;sentons ici une revue des efforts en cours pour identifier de nouvelles cibles th&eacute;rapeutiques pour le traitement de KS utilisant la g&eacute;nomique fonctionnelle, accompagn&eacute;s de recommandations concernant l&#39;&eacute;laboration de traitements &eacute;conomiquement faisables pour une utilisation en Afrique.

Sarcoma di Kaposi (KS) &egrave; una malattia multifocale angioproliferative che colpisce la pelle, mucosa e visceri delle persone infettate con herpesvirus-8 umano (HHV-8; anche associato al sarcoma di Kaposi herpesvirus [KSHV]). KS &egrave; la neoplasia pi&ugrave; comune nei pazienti con AIDS; l&#39;esito clinico del AIDS-KS &egrave; migliorato significativamente dalla terapia antiretrovirale altamente attiva (HAART). Tuttavia, in Africa, dove si verificano le manifestazioni pi&ugrave; severe della KS, c&#39;&egrave; un accesso limitato a queste e ad altre efficaci ma costosi farmaci. Sarcoma de Kaposi (KS) es un desorden de angioproliferative multifocal que afecta la piel, mucosa y v&iacute;sceras de los individuos infectados con herpesvirus-8 humano (HHV-8; tambi&eacute;n de sarcoma de Kaposi asociado herpesvirus [KSHV]). KS es la neoplasia m&aacute;s com&uacute;n en pacientes con SIDA; el resultado cl&iacute;nico de SIDA-KS se mejor&oacute; significativamente por la terapia antiretroviral altamente activa (HAART). Sin embargo, en &Aacute;frica, donde se producen las manifestaciones m&aacute;s severas de KS, hay acceso limitado a estos y otros efectivos pero costosos medicamentos. Aqu&iacute; presentamos una revisi&oacute;n de los actuales esfuerzos para identificar nuevas dianas terap&eacute;uticas para el tratamiento de KS utilizando la gen&oacute;mica funcional, con recomendaciones sobre el desarrollo de tratamientos econ&oacute;micamente factibles para uso en Africa.

Interferon (IFN)-Î³ is present in lesions of patients with Lyme disease and positively correlates with the severity of manifestations. To investigate the role of IFNÎ³ in the development of Lyme carditis, wild-type and IFNÎ³-deficient C57BL/6 mice were infected with the causative bacterium, Borrelia burgdorferi. Histological analysis revealed no change in the severity of carditis between wild-type and IFNÎ³-deficient mice at 14, 21, 25, and 28 days after infection. However, a distinct shift in the types of leukocytes within the hearts of IFNÎ³-deficient mice was observed at 25 days. In the absence of IFNÎ³, the number of neutrophils in the heart was increased, whereas the number of T lymphocytes was decreased. Bacterial loads within hearts were the same as in wild-type mice. Macrophages secrete chemokines that recruit immune cells, which could contribute to the accumulation of leukocytes in murine Lyme carditis. The ability of IFNÎ³ and B. burgdorferi to activate murine macrophages was examined, and the two stimuli synergistically induced chemoattractants for mononuclear cells (ie, CXCL9, CXCL10, CXCL11, CXCL16, and CCL12) and decreased those for neutrophils (ie, CXCL1, CXCL2, and CXCL3). IFNÎ³ and B. burgdorferi also synergistically enhanced secretion of CXCL9 and CXCL10 by murine cardiac endothelial cells. These results indicate that IFNÎ³ influences the composition of inflammatory infiltrates in Lyme carditis by promoting the accumulation of leukocytes associated with chronic inflammation and suppressing that of cells that typify acute inflammation.

L&#39;interf&eacute;ron (IFN)-&gamma; est pr&eacute;sent dans les l&eacute;sions des personnes atteintes de la maladie de Lyme et positivement est corr&eacute;l&eacute;e avec la s&eacute;v&eacute;rit&eacute; des manifestations. Afin d&#39;&eacute;tudier le r&ocirc;le de l&#39;IFN&gamma; dans le d&eacute;veloppement de Lyme cardite, sauvage et IFN&gamma; d&eacute;ficient des souris C57BL/6 &eacute;taient infect&eacute;s par la bact&eacute;rie Borrelia burgdorferi causale. L&#39;analyse histologique a r&eacute;v&eacute;l&eacute; aucun changement dans la s&eacute;v&eacute;rit&eacute; de cardite entre sauvage et les souris d&eacute;ficientes en IFN&gamma; &agrave; 14, 21, 25 et 28 jours apr&egrave;s l&#39;infection. Um die Rolle der IFN&gamma; in der Entwicklung der Lyme-Karditis zu untersuchen, wurden Wildtyp und IFN&gamma;-defizienten C57BL/6 M&auml;usen mit dem verursachenden Bakterium Borrelia Burgdorferi infiziert. Histologische Analyse ergab keine Ver&auml;nderung der Schweregrad der Karditis zwischen Wildtyp und IFN&gamma;-defizienten M&auml;usen bei 14, 21, 25 und 28 Tage nach der Infektion. Allerdings war eine deutliche Verschiebung in der Art der Leukozyten in die Herzen der IFN&gamma;-defizienten M&auml;usen bei 25 Tagen beobachtet. Interferone (IFN)-&gamma; &egrave; presente nelle lesioni dei pazienti con la malattia di Lyme e positivamente correla con la gravit&agrave; delle manifestazioni. Per indagare il ruolo di IFN&gamma; nello sviluppo di Lyme cardite, wild-type e IFN&gamma;-carenti topi C57BL/6 sono stati infettati con il batterio causativo, Borrelia burgdorferi. L&#39;analisi istologica ha rivelato alcun cambiamento nella gravit&agrave; della cardite tra wild-type e topi IFN&gamma;-carenti in 14, 21, 25 e 28 giorni dopo l&#39;infezione. Hodgkin's lymphoma is associated with immune dysregulation. Immune impairment often results in aberrant immune responses and lytic reactivation of ubiquitous Herpesviruses, such as Epstein-Barr virus (EBV) in mucosal tissues. Accordingly, the specificity of IgA to EBV early lytic antigens, which are important for reactivation, was evaluated to determine Hodgkin's lymphoma-specific sero-reactive patterns. Sera from 42 patients with Hodgkin's lymphoma were compared to sera from 17 patients with infectious mononucleosis (IM), another EBV-related condition that often presents in a similar manner; and to sera from 15 healthy EBV-seropositive subjects. Flow cytometry analysis demonstrated that like IM sera, most Hodgkin's lymphoma sera contained IgA that labeled cells expressing EBV early lytic antigens whereas healthy EBV-seropositive sera did not. Further evaluation to distinguish Hodgkin's lymphoma from IM showed that IgA in most Hodgkin's lymphoma, irrespective of the presence of EBV in primary tumors, detected only modified forms of EBV lytic Early Antigen-Diffuse (EA-D) while IM sera detected the un-modified form as well, further supporting the presence of immune dysregulation in Hodgkin's lymphoma patients. This IgA pattern distinguished Hodgkin's lymphoma from IM sera with a sensitivity of 92.9%, specificity 100%, positive predictive value 100%, and negative predictive value 85%. Our findings lay the groundwork for additional scientific and clinical investigation, particularly into the potential for developing Hodgkin's lymphoma-associated diagnostic and prognostic biomarkers.

Herpes simplex viruses 1 and 2 are human pathogens that lead to significant morbidity and mortality in certain clinical settings. The development of effective antiviral medications, however, has had little discernible impact on the epidemiology of these pathogens, largely because the majority of infections are clinically silent. Decades of work have gone into various candidate HSV vaccines, but to date none has demonstrated sufficient efficacy to warrant licensure. This review examines developments in HSV immunology and vaccine development published since 2010, and assesses the prospects for improved immunization strategies that may result in an effective, licensed vaccine in the near future.

Vaccine Responses and Immunologic Characteristics of Pediatric Patients With DiGeorge Syndrome.

Enterovirus D68 (EV-D68) is an emerging virus known to cause sporadic disease and occasional epidemics of severe lower respiratory tract infection. However, the true prevalence of infection with EV-D68 is unknown, due in part to the lack of a rapid and specific nucleic acid amplification test as well as the infrequency with which respiratory samples are analyzed by enterovirus surveillance programs. During the 2014 EV-D68 epidemic in the United States, we noted an increased frequency of "low-positive" results for human rhinovirus (HRV) detected in respiratory tract samples using the GenMark Diagnostics eSensor respiratory viral panel, a multiplex PCR assay able to detect 14 known respiratory viruses but not enteroviruses. We simultaneously noted markedly increased admissions to our Pediatric Intensive Care Unit for severe lower respiratory tract infections in patients both with and without a history of reactive airway disease. Accordingly, we hypothesized that these "low-positive" RVP results were due to EV-D68 rather than rhinovirus infection. Sequencing of the picornavirus 5' untranslated region (5'-UTR) of 49 samples positive for HRV by the GenMark RVP revealed that 33 (67.3%) were in fact EV-D68. Notably, the mean intensity of the HRV RVP result was significantly lower in the sequence-identified EV-D68 samples (20.3 nA) compared to HRV (129.7 nA). Using a cut-off of 40 nA for the differentiation of EV-D68 from HRV resulted in 94% sensitivity and 88% specificity. The robust diagnostic characteristics of our data suggest that the cross-reactivity of EV-D68 and HRV on the GenMark Diagnostics eSensor RVP platform may be an important factor to consider in making accurate molecular diagnosis of EV-D68 at institutions utilizing this system or other molecular respiratory platforms that may also cross-react.

Epidemic 2014 enterovirus D68 cross-reacts with human rhinovirus on a respiratory molecular diagnostic platform.

Kaposi's sarcoma (KS) is common in Africa, but economic constraints hinder successful treatment in most patients. Propranolol, a generic β-adrenergic antagonist, decreased proliferation of KS-associated herpesvirus (KSHV)-infected cells. Downregulation of cyclin A2 and cyclin-dependent kinase 1 (CDK1) recapitulated this phenotype. Additionally, propranolol induced lytic gene expression in association with downregulation of CDK6. Thus, propranolol has diverse effects on KSHV-infected cells, and this generic drug has potential as a therapeutic agent for KS.
