Understanding the interplay of different cellular proteins and their substrates is of major interest in the postgenomic era. For this purpose, selective isolation and identification of proteins from complex biological samples is necessary and targeted isolation of enzyme families is a challenging task. Over the last years, methods like activity-based protein profiling (ABPP) and capture compound mass spectrometry (CCMS) have been developed to reduce the complexity of the proteome by means of protein function in contrast to standard approaches, which utilize differences in physical properties for protein separation. To isolate and identify the subproteome consisting of S-adenosyl-L-methionine (SAM or AdoMet)-dependent methyltransferases (methylome), we developed and synthesized trifunctional capture compounds containing the chemically stable cofactor product S-adenosyl-L-homocysteine (SAH or AdoHcy) as selectivity function. SAH analogues with amino linkers at the N6 or C8 positions were synthesized and attached to scaffolds containing different photocrosslinking groups for covalent protein modification and biotin for affinity isolation. The utility of these SAH capture compounds for selective photoinduced protein isolation is demonstrated for various methyltransferases (MTases) acting on DNA, RNA and proteins as well as with Escherichia coli cell lysate. In addition, they can be used to determine dissociation constants for MTase-cofactor complexes.

Comprendre l&#39;interaction entre les prot&eacute;ines cellulaires diff&eacute;rentes et leurs substrats est d&#39;un int&eacute;r&ecirc;t majeur dans l&#39;&egrave;re postg&eacute;nomique. &Agrave; cette fin, isolement s&eacute;lectif et l&#39;identification des prot&eacute;ines &agrave; partir d&#39;&eacute;chantillons biologiques complexes est n&eacute;cessaire et isolement cibl&eacute;e des familles d&#39;enzymes est une t&acirc;che difficile. SAH analogues avec les linkers amin&eacute;s aux positions N6 ou C8 ont &eacute;t&eacute; synth&eacute;tis&eacute;s et attach&eacute;s aux &eacute;chafaudages contenant psoralen diff&eacute;rents groupes pour modification covalente de prot&eacute;ine et la biotine pour l&#39;isolement de l&#39;affinit&eacute;. L&#39;utilit&eacute; de ces compos&eacute;s de capture SAH pour l&#39;isolement des prot&eacute;ines s&eacute;lective photoinduit est d&eacute;montr&eacute;e pour divers m&eacute;thyltransf&eacute;rases (MTases), agissant sur l&#39;ADN, l&#39;ARN et les prot&eacute;ines, ainsi qu&#39;avec la cellule de Escherichia coli lysat. Verstehen das Zusammenspiel der verschiedenen zellul&auml;re Proteine und ihre Substrate ist von besonderem Interesse in der postgenomic-&Auml;ra. Zu diesem Zweck selektive Isolierung und Identifizierung von Proteinen aus komplexen biologischen Proben ist notwendig und gezielte Isolation der Enzym-Familien ist eine anspruchsvolle Aufgabe. In den letzten Jahren Methoden wie Activity based Protein Profilerstellung (ABPP) und Erfassung zusammengesetzte Massenspektrometrie (CCMS) wurden entwickelt, um die Reduzierung der Komplexit&auml;t der Proteome mittels Proteinfunktion im Gegensatz zu standard-Ans&auml;tzen, die Unterschiede in den physikalischen Eigenschaften f&uuml;r Protein Trennung zu nutzen. Isoliert und identifiziert die Subproteome bestehend aus S-Adenosyl-L-Methionin (SAM oder AdoMet)-abh&auml;ngige Methyltransferasen (Methylome), haben wir entwickelt und synthetisiert trifunktionalen Erfassung Verbindungen, die chemisch stabil Cofaktor Produkt S-Adenosyl-L-Homocystein (SAH oder AdoHcy) als Funktion der Selektivit&auml;t. SAH-Analoga mit amino Verkn&uuml;pfungsprogramme auf die N6 oder C8 Positionen wurden synthetisiert und an Ger&uuml;sten mit verschiedenen Photocrosslinking Gruppen f&uuml;r kovalente Protein &Auml;nderung und Biotin f&uuml;r die Affinit&auml;t Isolierung. Das Dienstprogramm dieser SAH Erfassung Verbindungen f&uuml;r selektive photoinduzierten Proteinisolierung wird f&uuml;r verschiedene Methyltransferasen (MTases) handeln auf DNA, RNA und Proteine sowie mit Escherichia coli Zelle lysate demonstriert. Dar&uuml;ber hinaus k&ouml;nnen sie verwendet werden, Dissoziation-Konstanten f&uuml;r MTasen-Cofaktor-komplexe zu bestimmen.

Comprendere l&#39;interazione delle proteine cellulari diverse e i loro substrati &egrave; di maggiore interesse in epoca postgenomic. Per questo scopo, isolamento selettivo e l&#39;identificazione di proteine da campioni biologici complessi &egrave; necessario e mirato l&#39;isolamento delle famiglie di enzima &egrave; un compito impegnativo. Negli ultimi anni, metodi come proteina basata su attivit&agrave; di profilatura (ABPP) e spettrometria di massa composto cattura (CCMS) sono stati sviluppati per ridurre la complessit&agrave; del proteoma mediante la funzione della proteina in contrasto con gli approcci standard, che utilizzano le differenze nelle propriet&agrave; fisiche per la separazione di proteine. Analoghi SAH con amino linkers alle posizioni N6 o C8 sono stati sintetizzati e associate a ponteggi contenenti gruppi diversi photocrosslinking per modifica covalente proteine e biotina per isolamento di affinit&agrave;. L&#39;utilit&agrave; di questi composti di cattura SAH per isolamento della proteina fotoindotti selettiva &egrave; dimostrata per i methyltransferases vari (MTases) che agiscono sul DNA, RNA e proteine, nonch&eacute; con lysate delle Escherichia coli. Inoltre, pu&ograve; essere utilizzati per determinare le costanti di dissociazione per complessi MTase-cofattore.

There is a variety of approaches to reduce the complexity of the proteome on the basis of functional small molecule-protein interactions. We describe a generic approach based on trifunctional Capture Compounds, in which the initial equilibrium-driven interaction between a small molecule probe and target proteins is irreversibly fixed upon photo-crosslinking between an independent photo-activable reactivity function of the Capture Compound and the surface of the target protein(s). Subsequently, Capture Compound - protein conjugates are isolated from complex biological mixtures via the sorting function of the Capture Compound. Here, we describe the application of a trifunctional Capture Compound that carries the methyltransferase product inhibitor S-Adenosyl-L -homocysteine as the selectivity function for the isolation of methyltransferases from a complex lysate of Escherichia coli DH5Î± cells. Photo-activated crosslinking enhances yield and sensitivity of the experiment, and the specificity can be readily tested for in competition experiments using an excess of free S-Adenosyl-L -homocysteine.

Posteriormente, compuesto de captura - prote&iacute;na conjugados son aislados de mezclas biol&oacute;gicas complejas mediante la funci&oacute;n de clasificaci&oacute;n de los compuestos de captura.
