Mitochondrial dysfunction and associated oxidant stress have been linked with numerous complex diseases and aging largely by in vitro determination of mitochondria oxidant production and scavenging. We applied targeted in vivo fluorescence analyses of mitochondria-dense pharyngeal tissue in Caenorhabditis elegans to better understand relative mitochondrial effects, particularly on matrix oxidant burden, of respiratory chain complex, MnSOD, and insulin receptor mutants displaying variable longevity. The data demonstrate significantly elevated in vivo matrix oxidant burden in the short-lived complex I mutant, gas-1(fc21), which was associated with limited superoxide scavenging capacity despite robust MnSOD induction, as well as decreased mitochondria content and membrane potential. Significantly increased MnSOD activity was associated with in vivo matrix oxidant levels similar to wild-type in the long-lived respiratory chain complex III mutant, isp-1(qm150). Yet, despite greater superoxide scavenging capacity in the complex III mutant than in the significantly longer-lived insulin receptor mutant, daf-2(e1368), only the former showed modest oxidative stress sensitivity. Furthermore, increased longevity was seen in MnSOD knockout mutants (sod-2(ok1030) and sod-2(gk257)) that had decreased MnSOD scavenging capacity and increased in vivo matrix oxidant burden. Thus, factors beside oxidant stress must underlie RC mutant longevity in C. elegans. This work highlights the utility of the C. elegans model as a tractable means to non-invasively monitor multi-dimensional in vivo consequences of primary mitochondrial dysfunction.

Mitochondrial respiratory chain dysfunction variably increases oxidant stress in Caenorhabditis elegans.

Dysfonctionnement mitochondrial et stress oxydant associ&eacute;es ont &eacute;t&eacute; associ&eacute;es &agrave; nombreuses maladies complexes et vieillissement en grande partie par la d&eacute;termination in vitro de la production de mitochondries oxydant et le nettoyage. Nous avons appliqu&eacute; des analyses cibl&eacute;es de fluorescence in vivo des mitochondries-dense tissu pharyng&eacute; chez Caenorhabditis elegans afin de mieux comprendre les effets mitochondriales relatifs, en particulier sur le fardeau oxydant matrice, de la cha&icirc;ne respiratoire complexe MnSOD et mutants de r&eacute;cepteur de l&#39;insuline affichant une long&eacute;vit&eacute; variable. Les donn&eacute;es d&eacute;montrent nettement fardeau oxydant &eacute;lev&eacute; matrice in vivo dans le complexe de courte dur&eacute;e j&#39;ai mutant, gas-1(fc21), qui a &eacute;t&eacute; associ&eacute;e de superoxyde limit&eacute;e nettoyage capacit&eacute; malgr&eacute; l&#39;induction MnSOD robuste, ainsi que diminue la teneur en mitochondries et potentiel de membrane. Activit&eacute; consid&eacute;rablement accrue de MnSOD a &eacute;t&eacute; associ&eacute;e &agrave; des niveaux d&#39;oxydant in vivo matrice semblables &agrave; la cha&icirc;ne sauvage dans la longue dur&eacute;e de vie respiratoire complexe III mutant, isp-1(qm150). Pourtant, malgr&eacute; le superoxyde grand nettoyage capacit&eacute; chez le mutant III complexe que dans l&#39;&eacute;tendrait consid&eacute;rablement l&#39;insuline r&eacute;cepteur mutant, daf-2(e1368), seul l&#39;ancien montr&eacute; sensibilit&eacute; modeste de stress oxydatif. Par ailleurs, long&eacute;vit&eacute; accrue a &eacute;t&eacute; observ&eacute;e dans les mutants de knockout MnSOD (sod-2(ok1030) et sod-2(gk257)) qui avait diminu&eacute; de MnSOD nettoyage capacit&eacute; et fardeau oxydant accru matrice in vivo. Ainsi, les facteurs &agrave; c&ocirc;t&eacute; de stress oxydant doivent sous-tendre RC long&eacute;vit&eacute; mutante chez c. elegans. Ce travail met en &eacute;vidence l&#39;utilit&eacute; du mod&egrave;le c. elegans comme moyen de surveillance non invasive des cons&eacute;quences multidimensionnelles in vivo du primaire dysfonctionnement mitochondrial tractable.

Dysfonctionnement de la cha&icirc;ne respiratoire mitochondriale augmente variablement stress oxydant chez Caenorhabditis elegans.

Mitochondriale Dysfunktion und damit verbundenen oxidativen Stress sind verbunden mit zahlreichen komplexen Erkrankungen und weitgehend durch in-vitro-Bestimmung der Mitochondrien Oxidationsmittel Produktion Alterung und des Aufr&auml;umvorgangs. Wir angewandt gezielte in-vivo Fluoreszenz Analyse der Mitochondrien-Dichte pharyngis Gewebe im Fadenwurm Caenorhabditis Elegans, relative mitochondriale Auswirkungen, insbesondere auf Matrix oxidativen Belastung besser zu verstehen, der Atmungskette, die komplexe, MnSOD und Insulin-Rezeptor-Mutanten, die angezeigte Variable Langlebigkeit. Die Daten zeigen deutlich erh&ouml;hten in-vivo-Matrix Oxidationsmittel belasten in der kurzlebigen Komplex ich Mutant, gas-1(fc21), die begrenzte Superoxid Aufr&auml;umvorgang Kapazit&auml;t trotz robuster MnSOD Induktion zugeordnet wurde, ebenso wie nahm der Mitochondrien Inhalt und Membrane m&ouml;glichen. Deutlich erh&ouml;hter MnSOD Aktivit&auml;t wurde in-vivo-Matrix Oxidationsmittel &auml;hnliche Niveaus zugeordnet Wildtyp in der langlebigen Atemwege Kette Komplex III mutant, isp-1(qm150). Doch zeigte trotz gr&ouml;&szlig;erer Superoxid Aufr&auml;umvorgang Kapazit&auml;t in den Komplex III-Mutanten als in der wesentlich langlebigeren Insulin Rezeptor mutant, daf-2(e1368), nur erstere bescheiden oxidativen Stress Empfindlichkeit. Dar&uuml;ber hinaus erh&ouml;hte Langlebigkeit galt im MnSOD Knockout-Mutanten (sod-2(ok1030) und sod-2(gk257)), die MnSOD Aufr&auml;umvorgang Kapazit&auml;t und h&ouml;here in-vivo-Matrix oxidativen Belastung abgenommen hatte. Daher m&uuml;ssen Faktoren neben oxidativen Stress RC mutant Langlebigkeit in C. Elegans zugrunde liegen. Diese Arbeit zeigt das Dienstprogramm des Modells C. Elegans als beeinflussende Mittel zur mehrdimensionale in-vivo Folgen der prim&auml;ren mitochondriale Dysfunktion nicht-invasiv zu &uuml;berwachen.

Mitochondrialen Dysfunktion erh&ouml;ht variabel oxidativen Stress bei Caenorhabditis Elegans.

Disfunzione mitocondriale e lo stress ossidante associato sono stati collegati con numerose malattie complesse e in gran parte dalla determinazione in vitro della produzione di ossidanti mitocondri e scavenging. Abbiamo applicato analisi mirate fluorescenza in vivo di mitocondri-denso tessuto faringeo nei elegans di Caenorhabditis per capire meglio i relativi effetti mitocondriali, particolarmente sull&#39;onere di ossidante matrix, della catena respiratoria complesso, MnSOD e mutanti di recettore insulina visualizzazione di durata variabile. I dati dimostrano in modo significativo ossidante matrix in vivo elevato onere nel complesso breve ho mutante, gas-1(fc21), che &egrave; stato associato con superossido limitato lo scavenging capacit&agrave; nonostante robusto MnSOD induzione, cos&igrave; come diminuito contenuto di mitocondri e potenziale di membrana. Significativamente aumentata attivit&agrave; MnSOD &egrave; stata associata con livelli di ossidanti matrix in vivo simili al wild-type nella longeva respiratorio catena complesso III mutanti, isp-1(qm150). Eppure, nonostante maggiore superossido lo scavenging capacit&agrave; nel complesso mutante III che nella significativamente balagh insulina recettore mutante, daf-2(e1368), solo l&#39;ex mostrato sensibilit&agrave; modesta dello stress ossidativo. Inoltre, &egrave; stato visto maggiore longevit&agrave; MnSOD mutanti knockout (sod-2(ok1030) e sod-2(gk257)) che aveva diminuito MnSOD lo scavenging capacit&agrave; e onere ossidante maggiore matrix in vivo. Cos&igrave;, fattori al lato dello stress ossidante devono sottostanno RC longevit&agrave; mutante in c. elegans. Questo lavoro mette in evidenza l&#39;utilit&agrave; del modello c. elegans come mezzo per il monitoraggio non invasivo conseguenze multidimensionali in vivo della disfunzione mitocondriale primaria trattabili.

La disfunci&oacute;n mitocondrial y estr&eacute;s oxidante asociado se han relacionado con numerosas enfermedades complejas y caducidad en gran medida por la determinaci&oacute;n in vitro de la producci&oacute;n de mitocondrias oxidante y el borrado. Se aplican an&aacute;lisis de fluorescencia in vivo espec&iacute;ficas de mitocondrias-denso tejido far&iacute;ngeo en Caenorhabditis elegans para comprender mejor los efectos relativos de la mitocondriales, especialmente en la carga oxidante de matriz, de complejo de cadena respiratoria, MnSOD y mutantes del receptor de insulina mostrando longevidad variable. Los datos demuestran significativamente oxidante de la matriz en vivo elevada carga en el complejo de breve duraci&oacute;n mutante, gas-1(fc21), que se asoci&oacute; con super&oacute;xido limitada capacidad a pesar de la inducci&oacute;n de MnSOD robusta de barrido, como tambi&eacute;n disminuy&oacute; el contenido de las mitocondrias y el potencial de membrana. Significativamente mayor actividad de la MnSOD se asoci&oacute; con niveles de oxidante de la matriz en vivo similares a cadena de tipo salvaje en la longeva respiratorio complejo III mutante, isp-1(qm150). Sin embargo, a pesar de super&oacute;xido mayor compactaci&oacute;n de capacidad en el complejo mutante III que en el significativamente m&aacute;s insulina receptor mutante, daf-2(e1368), s&oacute;lo el primero demostr&oacute; estr&eacute;s oxidativo modesta sensibilidad. Adem&aacute;s, se vio mayor longevidad en mutantes de nocaut de MnSOD (sod-2(ok1030) y sod-2(gk257)) que hab&iacute;a disminuido la MnSOD borrado capacidad y carga de oxidante mayor matriz in vivo. As&iacute;, factores al lado de estr&eacute;s oxidante deben subyacer RC longevidad mutante en C. elegans. Este trabajo pone de relieve la utilidad del modelo C. elegans como medio manejable monitorizar no invasiva consecuencias multidimensionales en vivo de la disfunci&oacute;n mitocondrial primaria.

Tail-anchored (TA) transmembrane proteins are targeted to membranes post- translationally. The proteins Get4 and Get5 form an obligate complex that catalyzes the transfer of TA proteins destined to the endoplasmic reticulum (ER) from Sgt2 to the cytosolic targeting factor Get3. Get5 forms a homodimer mediated by its carboxyl domain. We show here that a conserved motif exists within the carboxyl domain. A high-resolution crystal structure and solution NMR structures of this motif reveal a novel and stable helical dimerization domain. We additionally determined a solution NMR structure of a divergent fungal homolog and comparison of these structures allows annotation of specific stabilizing interactions. Using solution X-ray scattering and the structures of all folded domains, we present a model of the full length Get4/Get5 complex.

The Get5 carboxyl terminal domain is a novel dimerization motif that tethers an extended Get4/Get5 complex.

Le complexe de prot&eacute;ines Get4 et Get5 forment un obligatoires qui catalyse le transfert des prot&eacute;ines de TA destin&eacute; vers le r&eacute;ticulum endoplasmique (ER) de Sgt2 &agrave; la cytosolique ciblant le facteur Get3. Get5 forme un homodim&egrave;re v&eacute;hicul&eacute;e par son domaine du carboxyle. Nous montrons ici qu&#39;un motif conserv&eacute; il existe au sein du domaine du carboxyle. Une structure cristalline &agrave; haute r&eacute;solution et structures NMR solution de ce motif r&eacute;v&egrave;lent un roman et le domaine de dim&eacute;risation h&eacute;lico&iuml;dale stable. Nous avons d&eacute;termin&eacute; par ailleurs une solution structure RMN d&#39;un homologue fongique divergent et permet de comparer ces structures annotation des interactions stabilisantes sp&eacute;cifiques. &Agrave; l&#39;aide de diffusion des rayons x de solution et les structures de tous les domaines pli&eacute;es, nous pr&eacute;sentons un mod&egrave;le de la longueur totale de Get4/Get5 complexes.

Eine hochaufl&ouml;sende Kristallstruktur und L&ouml;sung NMR Strukturen dieses Motivs zeigen eine neuartige und stabilen spiralf&ouml;rmigen Dimerisierung Dom&auml;ne. Wir entschlossen zus&auml;tzlich eine L&ouml;sung NMR Struktur eine abweichende pilzartige Homolog und Vergleich dieser Strukturen erm&ouml;glicht Annotation von spezifischen stabilisierende Wechselwirkungen. Mit R&ouml;ntgenstreuung L&ouml;sung und die Strukturen aller gefalteten Domains, pr&auml;sentieren wir ein Modell der vollen L&auml;nge Get4/Get5 Komplex.

Coda ancorata proteine transmembrana (TA) sono mirate a membrane post-translationally. Get5 carboxyl &#xD130;&#xBBF8;&#xB110; &#xB3C4;&#xBA54;&#xC778;&#xC740; &#xBCF5;&#xC7A1; &#xD55C; &#xD655;&#xC7A5;&#xB41C; Get4/Get5 tethers &#xC18C;&#xC124; dimerization &#xBAA8;&#xD2F0;&#xBE0C;.

Las prote&iacute;nas transmembrana (TA) anclada cola est&aacute;n dirigidas a membranas post-forma. El complejo de prote&iacute;nas Get4 y Get5 forman un obliga que cataliza a la transferencia de prote&iacute;nas TA destinado al ret&iacute;culo endopl&aacute;smico (ER) de Sgt2 a las citos&oacute;licas dirigidos a factor Get3. Get5 forma un homod&iacute;mero mediado por su dominio carboxilo. Aqu&iacute; mostramos que un motivo conservado existe dentro del dominio carboxilo. Una estructura cristalina de alta resoluci&oacute;n y soluci&oacute;n NMR estructuras de este motivo revelan una novela y dominio de dimerizaci&oacute;n helicoidal estable. Adem&aacute;s determinamos una soluci&oacute;n estructura NMR de un hom&oacute;logo del fungicida divergente y comparaci&oacute;n de estas estructuras permite anotaci&oacute;n de interacciones espec&iacute;ficas de estabilizaci&oacute;n. Mediante dispersi&oacute;n de rayos-x de soluci&oacute;n y las estructuras de todos los dominios doblados, presentamos un modelo de toda la duraci&oacute;n de Get4/Get5 complejo.
