Neurofibrillary tangles are composed of insoluble aggregates of the microtubule-associated protein tau. In Alzheimer's disease the accumulation of neurofibrillary tangles occurs in the absence of tau mutations. Here we present mice that develop pathology from non-mutant human tau, in the absence of other exogenous factors, including beta-amyloid. The pathology in these mice is Alzheimer-like, with hyperphosphorylated tau accumulating as aggregated paired helical filaments. This pathologic tau accumulates in the cell bodies and dendrites of neurons in a spatiotemporally relevant distribution.

The generation of neurons and glia from pluripotent embryonic stem (ES) cells represents a promising strategy for the study of CNS development and repair. ES cell-derived neural precursors have been shown to develop into morphologically mature neurons and glia when grafted into brain and spinal cord. However, there is a surprising shortage of data concerning the functional integration of ES cell-derived neurons (ESNs) into the host CNS tissue. Here, we use ES cells engineered to express enhanced green fluorescent protein (EGFP) only in neuronal progeny to study the functional properties of ESNs during integration into long-term hippocampal slice cultures. After incorporation into the dentate gyrus, EGFP+ donor neurons display a gradual maturation of their intrinsic discharge behavior and a concomitant increase in the density of voltage-gated Na+ and K+ channels. Integrated ESNs express AMPA and GABA(A) receptor subunits. Most importantly, neurons derived from ES cells receive functional glutamatergic and GABAergic synapses from host neurons. Specifically, we demonstrate that host perforant path axons form synapses onto integrated ESNs. These synapses between host and ES cell-derived neurons display pronounced paired-pulse facilitation indicative of intact presynaptic short-term plasticity. Thus, ES cell-derived neural precursors generate functionally active neurons capable of integrating into the brain circuitry.

Pluripotency and the potential for continuous self-renewal make embryonic stem (ES) cells an attractive donor source for neuronal cell replacement. Despite recent encouraging results in this field, little is known about the functional integration of transplanted ES cell-derived neurons on the single-cell level. To address this issue, ES cell-derived neural precursors exhibiting neuron-specific enhanced green fluorescent protein (EGFP) expression were introduced into the developing brain. Donor cells implanted into the cerebral ventricles of embryonic rats migrated as single cells into a variety of brain regions, where they acquired complex morphologies and adopted excitatory and inhibitory neurotransmitter phenotypes. Synaptic integration was suggested by the expression of PSD-95 (postsynaptic density-95) on donor cell dendrites, which in turn were approached by multiple synaptophysin-positive host axon terminals. Ultrastructural and electrophysiological data confirmed the formation of synapses between host and donor cells. Ten to 21 d after birth, all EGFP-positive donor cells examined displayed active membrane properties and received glutamatergic and GABAergic synaptic input from host neurons. These data demonstrate that, at the single-cell level, grafted ES cell-derived neurons undergo morphological and functional integration into the host brain circuitry. Antibodies to the region-specific transcription factors Bf1, Dlx, En1, and Pax6 were used to explore whether functional donor cell integration depends on the acquisition of a regional phenotype. Our data show that incorporated neurons frequently exhibit a lacking or ectopic expression of these transcription factors. Thus, the lack of an appropriate regional "code" does not preclude morphological and synaptic integration of ES cell-derived neurons.

Furthermore, gel shift assays indicate that transcriptional regulators SP-1 and AP-2 are important for basal expression but not necessary for neuron-specific expression of the tau transcript.

Proteolytic processing of the transcription factor Gli3, for example, occurs in primary cilia, and defects in intraflagellar transport (IFT), which is crucial for the maintenance of primary cilia, can lead to severe developmental defects and diseases. Here we report an essential role of primary cilia in forebrain development. Uncovered by N-ethyl-N-nitrosourea-mutagenesis, cobblestone is a hypomorphic allele of the IFT gene Ift88, in which Ift88 mRNA and protein levels are reduced by 70-80%. cobblestone mutants are distinguished by subpial heterotopias in the forebrain. Apoptosis of neurons in the maturing neocortex has been recorded in a wide variety of mammals, but very little is known about its effects on cortical differentiation. Recent research has implicated the RhoA GTPase subfamily in the control of apoptosis in the developing nervous system and in other tissue types. Rho GTPases are important components of the signaling pathways linking extracellular signals to the cytoskeleton. To investigate the role of the RhoA GTPase subfamily in neocortical apoptosis and differentiation, we have engineered a mouse line in which a dominant-negative RhoA mutant (N19-RhoA) is expressed from the Mapt locus, such that all neurons of the developing nervous system are expressing the N19-RhoA inhibitor. Postnatal expression of N19-RhoA led to no major changes in neocortical anatomy. Six layers of the neocortex developed and barrels (whisker-related neural modules) formed in layer IV. However, the density and absolute number of neurons in the somatosensory cortex increased by 12-26% compared with wild-type littermates. This was not explained by a change in the migration of neurons during the formation of cortical layers but rather by a large decrease in the amount of neuronal apoptosis at postnatal day 5, the developmental maximum of cortical apoptosis. In addition, overexpression of RhoA in cortical neurons was seen to cause high levels of apoptosis. These results demonstrate that RhoA-subfamily members play a major role in developmental apoptosis in postnatal neocortex of the mouse but that decreased apoptosis does not alter cortical cytoarchitecture and patterning.

La apoptosis de las neuronas en el neoc&#xF3;rtex de maduraci&#xF3;n se ha registrado en una amplia variedad de mam&#xED;feros, pero se sabe muy poco acerca de sus efectos sobre la diferenciaci&#xF3;n cortical. La investigaci&#xF3;n reciente ha implicado a la subfamilia de GTPasa RhoA en el control de la apoptosis en el desarrollo del sistema nervioso y en otros tipos de tejidos. Rho GTPasas son componentes importantes de las v&#xED;as de se&#xF1;alizaci&#xF3;n que unen las se&#xF1;ales extracelulares con el citoesqueleto. Para investigar el papel de la subfamilia de GTPasa RhoA en la apoptosis y la diferenciaci&#xF3;n neocortical, hemos dise&#xF1;ado una l&#xED;nea de ratones en el que un dominante negativo de RhoA mutante (N19-RhoA) se expresa a partir del locus MAPT, de tal manera que todas las neuronas del sistema nervioso en desarrollo est&#xE1;n expresando el inhibidor de la N19-RhoA. Expresi&#xF3;n postnatal de N19-RhoA no condujo a cambios importantes en la anatom&#xED;a neocortical. Seis capas de la neocorteza y el desarrollo relacionados con el bigote de barriles (m&#xF3;dulos neuronales) se forman en la capa IV. Sin embargo, la densidad y el n&#xFA;mero absoluto de neuronas en la corteza somatosensorial aument&#xF3; en 26.12% en comparaci&#xF3;n con animales tipo salvaje. Esto no fue explicado por un cambio en la migraci&#xF3;n de las neuronas durante la formaci&#xF3;n de capas corticales sino m&#xE1;s bien por una gran disminuci&#xF3;n en la cantidad de la apoptosis neuronal en el d&#xED;a postnatal 5, el m&#xE1;ximo desarrollo de la apoptosis cortical. Adem&#xE1;s, la sobreexpresi&#xF3;n de RhoA en las neuronas corticales fue visto a causa de los altos niveles de apoptosis. Estos resultados demuestran que la subfamilia de RhoA miembros desempe&#xF1;an un papel importante en la apoptosis en el desarrollo post-natal en el neoc&#xF3;rtex del rat&#xF3;n, pero que disminuci&#xF3;n de la apoptosis no altera citoarquitectura cortical y patrones.

The primary cilium is a small microtubule-based organelle projecting from the plasma membrane of practically all cells in the mammalian body. In the past 8Â years, a flurry of papers has indicated a crucial role of this long-neglected organelle in the development of a wide variety of organs, including derivatives of all three germ layers. A common theme of these studies is the critical dependency of signal transduction of the Hedgehog pathway upon functionally intact cilia to regulate organogenesis. Another common theme is the role that the cilium plays, not necessarily in the determination of the embryonic anlagen of these organs, although this too occurs but rather in the proliferation and morphogenesis of the previously determined organ. We outline the various organ systems that are dependent upon primary cilia for their proper development and we discuss the cilia-dependent roles that Sonic and Indian Hedgehog play in these processes. In addition and most importantly for the field, we discuss the controversial involvement of another major developmental pathway, Wnt signaling, in cilia-dependent organogenesis.

Ein weiteres gemeinsames Thema ist die Rolle der Zilie, nicht unbedingt bei der Ermittlung des embryonalen Anlagen dieser Organe, obwohl dies auch geschieht, sondern in die Proliferation und Morphogenese der vorher festgelegte Orgel. Wir skizzieren die verschiedenen Organsysteme, die prim&auml;re Zilien f&uuml;r ihre ordnungsgem&auml;&szlig;e Entwicklung abh&auml;ngig sind, und wir besprechen die Zilien-abh&auml;ngige Rolle von Sonic und Indian Hedgehog in diesen Prozessen. Au&szlig;erdem und vor allem f&uuml;r das Feld diskutieren wir die umstrittene Beteiligung von einem anderen gro&szlig;en Entwicklungsst&ouml;rungen Weg, Wnt signalisieren, in Zilien-abh&auml;ngige Organogenese.

Prim&auml;re Zilien und Organogenese: ist Igel der einzige Bildhauer?

Il cilio primario &egrave; un piccolo organello microtubule-base sporgente dalla membrana del plasma di praticamente tutte le cellule nel corpo dei mammiferi. Negli ultimi 8 anni, una raffica di carte ha indicato un ruolo cruciale di questo organello lungo trascurato lo sviluppo di un&#39;ampia variet&agrave; di organi, compresi i derivati di tutti i tre strati germinali. Un tema comune di questi studi &egrave; la dipendenza critica della trasduzione del segnale di via del riccio su ciglia funzionalmente intatto per regolamentare l&#39;organogenesi. Un altro tema comune &egrave; il ruolo che il cilio riproduce, non necessariamente nella determinazione della anlagen embrionale di questi organi, anche se questo si verifica anche, ma piuttosto nella proliferazione e nella morfogenesi dell&#39;organo determinata in precedenza. Si delineano i vari sistemi di organi che dipendono dal ciliari per il loro corretto sviluppo e discutiamo i ruoli di cilia-dipendente che Sonic e Indian Hedgehog giocare in questi processi. In pi&ugrave; e la cosa pi&ugrave; importante per il campo, discutiamo il controverso coinvolgimento di un&#39;altra principale via dello sviluppo, Wnt segnalazione, nella organogenesi cilia-dipendente.
