Extrapancreatic autoimmune manifestations in type 1 diabetes patients and their first-degree relatives: a multicenter study.

Two non-psychotropic cannabinoids, cannabidiol (CBD) and cannabidiol-dimethylheptyl (CBD-DMH), induced apoptosis in a human acute myeloid leukemia (AML) HL-60 cell line. Apoptosis was determined by staining with bisBenzimide and propidium iodide. A dose dependent increase of apoptosis was noted, reaching 61 and 43% with 8 microg/ml CBD and 15 microg/ml CBD-DMH, respectively, after a 24 h treatment. Prior exposure of the cells to gamma-irradiation (800 cGy) markedly enhanced apoptosis, reaching values of 93 and 95%, respectively. Human monocytes from normal individuals were resistant to either cannabinoids or gamma-irradiation. Caspase-3 activation was observed after the cannabinoid treatment, and may represent a mechanism for the apoptosis. Our data suggest a possible new approach to treatment of AML.

Although skin dendritic cells (DCs) have been shown to directly present Ag to CD8(+) T cells after intradermal immunization with lentivectors, the contribution of the different skin DC subsets to this process remains unclear. Using langerin-diphtheria toxin receptor transgenic mice we demonstrated that ablation of langerhans cells and langerin-expressing positive dermal DCs (Ln(+)dDCs) did not interfere with the generation of CD8(+) T cells by lentiviral vectors. Consistent with these findings, the absence of langerhans cells and Ln(+)dDCs did not hamper the presentation level of lentiviral-derived Ag by skin DCs in vitro. We further demonstrated that only dDCs and Ln(+)dDCs were capable of presenting Ag, however, the number of dDCs migrating to the draining lymph nodes was 6-fold higher than that of Ln(+)dDCs. To study how the duration of DC migration influences CD8(+) T cell responses, we analyzed the kinetics of Ag expression at the injection site and manipulated DC migration by excising the injected skin at various times after immunization. A low level of Ag expression was seen 1 wk after the immunization; peaked during week 2, and was considerably cleared by week 3 via a perforin-dependent fas-independent mechanism. Removing the injection site 3 or 5 d, but not 10 d, after the immunization, resulted in a reduced CD8(+) T cell response. These findings suggest that dDCs are the main APCs active after intradermal lentiviral-mediated immunization, and migration of dDCs in the initial 10-d period postimmunization is required for optimal CD8(+) T cell induction.

The role of skin-derived dendritic cells in CD8+ T cell priming following immunization with lentivectors.

Bien que les cellules dendritiques (DCs) de la peau montrent directement pr&eacute;senter Ag aux cellules T CD8 apr&egrave;s vaccination intradermique avec lentivecteurs, la contribution des sous-ensembles DC peau diff&eacute;rente &agrave; ce processus reste floue. &Agrave; l&#39;aide de souris transg&eacute;niques du r&eacute;cepteur toxine dipht&eacute;rique langerin nous avons d&eacute;montr&eacute; que l&#39;ablation des cellules de langerhans et exprimant langerin positives DCs par voie cutan&eacute;e (Ln(+)dDCs) n&#39;interf&eacute;raient pas avec la g&eacute;n&eacute;ration des cellules T CD8 de vecteurs LENTIVIRAUX. Conform&eacute;ment &agrave; ces conclusions, l&#39;absence de cellules de langerhans et Ln (+) DSAD n&#39;a pas g&ecirc;n&eacute; au niveau de la pr&eacute;sentation des Ag des g&egrave;nes d&eacute;riv&eacute;s de peau in vitro de DCs. De plus, nous avons d&eacute;montr&eacute; que seulement DSAD et DSAD Ln (+) &eacute;tait capables de pr&eacute;senter l&#39;Ag, le nombre de DSAD migrer vers les ganglions lymphatiques de drainage &eacute;tait cependant 6 fois sup&eacute;rieur &agrave; celui de DSAD Ln (+). Afin d&#39;&eacute;tudier comment la dur&eacute;e de migration DC influe sur les lymphocytes T CD8, nous avons analys&eacute; la cin&eacute;tique d&#39;expression Ag au point d&#39;injection et manipul&eacute; la migration DC en excisant la peau inject&eacute;e &agrave; des moments diff&eacute;rents apr&egrave;s la vaccination. Un faible niveau d&#39;expression de l&#39;Ag a &eacute;t&eacute; vu 1 sem apr&egrave;s la vaccination ; a atteint un sommet au cours de la semaine 2 et a &eacute;t&eacute; consid&eacute;rablement autoris&eacute; par semaine 3 via un m&eacute;canisme ind&eacute;pendant du SAF de perforine-d&eacute;pendante. Retirer l&#39;injection site 3 ou 5 d, mais pas 10 jours, apr&egrave;s la vaccination, a entra&icirc;n&eacute; une r&eacute;ponse r&eacute;duite des cellules T CD8. Ces r&eacute;sultats sugg&egrave;rent que DSAD est les principaux TTB actifs apr&egrave;s la vaccination intradermique par l&#39;interm&eacute;diaire des g&egrave;nes, et migration de DSAD dans les s&eacute;rologies p&eacute;riode initiale de 10-d est n&eacute;cessaire pour l&#39;induction de cellules T CD8 optimale.

Obwohl direkt Ag CD8(+) T-Zellen pr&auml;sentieren nach Intradermaler Immunisierung mit Lentivectors Haut dendritische Zellen (DCs) nachgewiesen wurde, bleibt der Beitrag der verschiedenen Haut DC Teilmengen zu diesem Prozess unklar. Langerin-Diphtherie-Toxin Rezeptor transgene M&auml;use verwenden wir bewiesen die Ablation der Langerhans-Zellen und Langerin-Ausdruck positive dermal DCs (Ln(+)dDCs) nicht st&ouml;ren die Generation der CD8(+) T-Zellen durch Lentiviralen Vektoren. Mit diesen Ergebnissen das Fehlen von Langerhans-Zellen und Ln (+) DCS nicht der Pr&auml;sentation-Spiegel des Lentiviralen abgeleitete Ag von Haut DCs in-vitro-behindern. Wir gezeigt weiter, dass nur DCS und DCS Ln (+) pr&auml;sentieren Ag in der Lage waren, jedoch die Anzahl der DCS Migration zu Drainierende Lymphknoten 6-fold h&ouml;her als die der Ln (+) Transceiver. Um zu untersuchen, wie die Dauer der DC-Migration CD8(+) T Zelle Antworten beeinflusst, wir analysiert die Kinetik der Ag Ausdruck an der Injektionsstelle und manipuliert DC Migration gelenkten injizierte Haut zu verschiedenen Zeitpunkten nach der Immunisierung. Die Rolle des Haut-abgeleitete dendritische Zellen in CD8 + T-Zelle-Grundierung nach Immunisierung mit Lentivectors.

Anche se le cellule dendritiche (DCs) hanno dimostrate di presentare direttamente Ag a cellule cellule T dopo immunizzazione intradermica con lentivectors, il contributo di sottoinsiemi di DC il pelle diversa per questo processo rimane poco chiaro. CD8 + T &#xC138;&#xD3EC; &#xBABB;&#xC4F0;&#xAC8C; lentivectors&#xC640; &#xC608;&#xBC29; &#xC811;&#xC885;&#xC744; &#xB2E4;&#xC74C;&#xC5D0;&#xC11C; &#xD53C;&#xBD80; &#xC720;&#xB798; &#xC218;&#xC9C0;&#xC0C1; &#xC138;&#xD3EC;&#xC758; &#xC5ED;&#xD560;.

Aunque la piel de c&eacute;lulas dendr&iacute;ticas (DCs) han demostrado presentar directamente Ag c&eacute;lulas CD8 T despu&eacute;s de la vacunaci&oacute;n intrad&eacute;rmica con lentivectors, la contribuci&oacute;n de los subconjuntos de DC de piel diferentes a este proceso sigue siendo confusa. Utilizando ratones transg&eacute;nicos de receptor de la toxina de la difteria langerin demostramos que la ablaci&oacute;n de las c&eacute;lulas de langerhans y DCs cut&aacute;neos positivos langerin-expresando (Ln(+)dDCs) no interfirieron con la generaci&oacute;n de c&eacute;lulas T CD8 por vectores lentivirales. En concordancia con estos resultados, la ausencia de c&eacute;lulas de langerhans y Ln (+) dDCs no obstaculizar el nivel de presentaci&oacute;n de Ag lentivirales derivados por piel DCs in vitro. Adem&aacute;s, demostramos que s&oacute;lo dDCs y dDCs Ln (+) eran capaces de presentar Ag, sin embargo, el n&uacute;mero de dDCs migran a los ganglios linf&aacute;ticos drenantes fue doblez mayor que la de dDCs Ln (+). Para estudiar c&oacute;mo influye en la duraci&oacute;n de la migraci&oacute;n de DC respuestas de c&eacute;lulas T CD8, analizamos la cin&eacute;tica de expresi&oacute;n de la Ag en el sitio de inyecci&oacute;n y hab&iacute;a manipulado migraci&oacute;n DC suprimiendo la piel inyectada en diferentes momentos despu&eacute;s de la inmunizaci&oacute;n. Un bajo nivel de expresi&oacute;n de la Ag fue visto 1 semana despu&eacute;s de la inmunizaci&oacute;n; alcanz&oacute; su punto m&aacute;ximo durante dos semanas y considerablemente fue despejado por semana 3 a trav&eacute;s de un mecanismo independiente del fas de perforin-dependiente. Quitar la inyecci&oacute;n sitio 3 o 5 d, pero no de 10 d, despu&eacute;s de la inmunizaci&oacute;n, result&oacute; en una disminuci&oacute;n en la respuesta celular T CD8. Estos resultados sugieren que dDCs son el principal activo de APC despu&eacute;s de vacunaci&oacute;n intrad&eacute;rmica lentivirales-mediada, y migraci&oacute;n de dDCs en el posinmunizaci&oacute;n per&iacute;odo inicial de 10-d es necesaria para la inducci&oacute;n de c&eacute;lulas T CD8 &oacute;ptima.
