In order to prove that ectopic over-expression of Pim-2 could induce malignant transformation of human liver cell line L02, three groups of cells were set up including human liver cell line L02 (L02), L02 cells transfected with Pim-2 gene (L02/Pim-2) and L02 cells transfected with empty-vector (L02/Vector). Pim-2 expression levels were detected. The morphology, proliferation level, apoptosis rate and migration ability of the cells were detected respectively. Then the cells were subcutaneously inoculated into athymic mice and the microstructures of the neoplasm were observed. Compared with the controls, Pim-2 expression levels were significantly higher in L02/Pim-2 cells (P<0.05), and their morphology had obvious malignant changes. They also showed a significantly increased proliferation rate (P<0.05) and migration capacity (P<0.05), as well as a significantly decreased apoptosis rate (P<0.05). Only the athymic mice inoculated with L02/Pim-2 cells could generate neoplasm, and the morphology of the neoplasm coincided with that of the hepatoma. The results manifest that ectopic Pim-2 gene could be stably expressed in L02/Pim-2 cells. Both the morphological and biological changes of L02/Pim-2 cells demonstrate the trend of malignant transformation. L02/Pim-2 cells could generate hepatoma in athymic mice. In conclusion, Pim-2 could induce malignant transformation of human liver cell line L02.

Ectopic over-expression of oncogene Pim-2 induce malignant transformation of nontumorous human liver cell line L02.

Self-propelled nano/micromotors and pumps are considered to be next generation drug delivery systems since the carriers can either propel themselves ("motor"-based drug delivery) or be delivered ("pump"-based drug delivery) to the target in response to specific biomarkers. Recently, there has been significant advancement towards developing nano/microtransporters into proof-of-concept tools for biomedical applications. This review encompasses the progress made to date on the design of synthetic nano/micromotors and pumps with respect to transportation and delivery of cargo at specific locations. Looking ahead, it is possible to imagine a day when intelligent machines navigate through the human body and perform challenging tasks.

Intelligent, self-powered, drug delivery systems.

We report a Ag(3)PO(4) microparticle system showing collective behaviors in aqueous medium. Transition between two emergent patterns, namely "exclusion" and "schooling", can be triggered by shift in chemical equilibrium upon the addition or removal of NH(3) or in response to UV light. The transitions are consistent with a self-diffusiophoresis mechanism resulting from ion gradients. The reported system is among the few examples of nanomotors that are based on a reversible nonredox reaction and demonstrates new design principles for micro/nanomotors. Potential applications of the reported system in logic gates, microscale pumping, and hierarchical assembly have been demonstrated.

Transition between collective behaviors of micromotors in response to different stimuli.

We report the design of autonomous motors powered by the rapid depolymerization reaction of poly(2-ethyl cyanoacrylate) (PECA), an FDA-approved polymer. Motors were fabricated in two different length scales, 3 cm and 300 μm. The motion of the motors is induced by self-generated surface tension gradients along their bodies. The motors are capable of moving in various media, including salt solutions and artificial serum.

Depolymerization-powered autonomous motors using biocompatible fuel.

Electrophoretic motion at low potentials is facilitated by redox chemistry occurring in a supercapacitor-based electrochemical energy storage system during charge and discharge. We show that MnO2 -modified electrodes can effectively alleviate the electrode surface polarization, the main factor that leads to inefficient electrophoresis at low voltages. A self-powered electrophoretic system based on a discharging battery has been also fabricated.

Enhanced electrophoretic motion using supercapacitor-based energy storage system.

Nano- and microscale motors powered by catalytic reactions exhibit collective behavior such as swarming, predator-prey interactions, and chemotaxis that resemble those of biological microorganisms. A quantitative understanding of the catalytically generated forces between particles that lead to these behaviors has so far been lacking. Observations and numerical simulations of pairwise interactions between gold-platinum nanorods in hydrogen peroxide solutions show that attractive and repulsive interactions arise from the catalytically generated electric field. Electrokinetic effects drive the assembly of staggered doublets and triplets of nanorods that are moving in the same direction. None of these behaviors are observed with nanorods composed of a single metal. The motors also collect tracer microparticles at their head or tail, depending on the charge of the particles, actively assembling them into close-packed rafts and aggregates of rafts. These motor-tracer particle interactions can also be understood in terms of the catalytically generated electric field around the ends of the nanorod motors.

Catalytically powered dynamic assembly of rod-shaped nanomotors and passive tracer particles.

DNA polymerase is responsible for synthesizing DNA, a key component in the running of biological machinery. Using fluorescence correlation spectroscopy, we demonstrate that the diffusive movement of a molecular complex of DNA template and DNA polymerase enhances during nucleotide incorporation into the growing DNA template. The diffusion coefficient of the complex also shows a strong dependence on its inorganic cofactor, Mg2+ ions. When exposed to gradients of either nucleotide or cofactor concentrations, an ensemble of DNA polymerase complex molecules shows collective movement toward regions of higher concentrations. By immobilizing the molecular complex on a patterned gold surface, we demonstrate the fabrication of DNA polymerase-powered fluid pumps. These miniature pumps are capable of transporting fluid and tracer particles in a directional manner with the pumping speed increasing in the presence of the cofactor. The role of DNA polymerase as a micropump opens up avenues for designing miniature fluid pumps using enzymes as engines.

DNA polymerase as a molecular motor and pump.

A self-powered polymeric micropump based on boronate chemistry is described. The pump is triggered by the presence of glucose in ambient conditions and induces convective fluid flows, with pumping velocity proportional to the glucose concentration. The pumping is due to buoyancy convection that originates from reaction-associated heat flux, as verified from experiments and finite difference modeling. As predicted, the fluid flow increases with increasing height of the chamber. In addition, pumping velocity is enhanced on replacing glucose with mannitol because of the enhanced exothermicity associated with the reaction of the latter.

Self-powered glucose-responsive micropumps.

Bimetallic gold-ruthenium microrods are propelled in opposite directions in water by ultrasound and by catalytic decomposition of hydrogen peroxide. This property was used to effect reversible swarming, to stall and reverse autonomous axial propulsion, and to study the chemically powered movement of acoustically levitated microrods.

A tale of two forces: simultaneous chemical and acoustic propulsion of bimetallic micromotors.

Aspirin has been reported to regulate lipid metabolism. However, the mechanism underlying the regulation is not clear. We presently investigated aspirin's promotion of AMP-activated protein kinase (AMPK) pathway activation in human hepatoma HepG2 cells by examining AMPK expression, the promotion of AMPK activation. Then we investigated the influence of aspirin-promoted AMPK signaling on fatty acid oxidation in HepG2 cells. The results demonstrated that aspirin treatment did not regulate the expression of AMPK and its downstream target, Acetyl-Coenzyme A Carboxylase (ACC), but activated the AMPK signaling pathway by promoting the phosphorylation of AMPK and ACC. And, interestingly, the promotion by aspirin is dependent of cellular esterase, which catalyzes aspirin to salicylate. Moreover, the activated AMPK signaling promoted the fatty acid oxidation, by promoting expression of Carnitine palmitoyltransferase I (CPT1) and Medium-Chain Acyl-CoA Dehydrogenase (MCAD) in both mRNA and protein levels. Thus, we confirmed in this study that aspirin promoted lipid oxidation by upregulating the AMPK signaling pathway.

Aspirin regulates hepatocellular lipid metabolism by activating AMPK signaling pathway.

The assembly of complex structures from simpler, individual units is a hallmark of biology. Examples include the pairing of DNA strands, the assembly of protein chains into quaternary structures, the formation of tissues and organs from cells, and the self-organization of bacterial colonies, flocks of birds, and human beings in cities. While the individual behaviors of biomolecules, bacteria, birds, and humans are governed by relatively simple rules, groups assembled from many individuals exhibit complex collective behaviors and functions that do not exist in the absence of the hierarchically organized structure. Self-assembly is a familiar concept to chemists who study the formation and properties of monolayers, crystals, and supramolecular structures. In chemical self-assembly, disorder evolves to order as the system approaches equilibrium. In contrast, living assemblies are typically characterized by two additional features: (1) the system constantly dissipates energy and is not at thermodynamic equilibrium; (2) the structure is dynamic and can transform or disassemble in response to stimuli or changing conditions. To distinguish them from equilibrium self-assembled structures, living (or nonliving) assemblies of objects with these characteristics are referred to as active matter. In this Account, we focus on the powered assembly and collective behavior of self-propelled colloids. These nano- and microparticles, also called nano- and micromotors or microswimmers, autonomously convert energy available in the environment (in the form of chemical, electromagnetic, acoustic, or thermal energy) into mechanical motion. Collections of these colloids are a form of synthetic active matter. Because of the analogy to living swimmers of similar size such as bacteria, the dynamic interactions and collective behavior of self-propelled colloids are interesting in the context of understanding biological active matter and in the development of new applications. The progression from individual particle motion to pairwise interactions, and then to multiparticle behavior, can be studied systematically with colloidal particles. Colloidal particles are also amenable to designs (in terms of materials, shapes, and sizes) that are not readily available in, for example, microbial systems. We review here our efforts and those of other groups in studying these fundamental interactions and the collective behavior that emerges from them. Although this field is still very new, there are already unique and interesting applications in analysis, diagnostics, separations, and materials science that derive from our understanding of how powered colloids interact and assemble.

From one to many: dynamic assembly and collective behavior of self-propelled colloidal motors.

Nature supports multifaceted forms of life. Despite the variety and complexity of these forms, motility remains the epicenter of life. The applicable laws of physics change upon going from macroscales to microscales and nanoscales, which are characterized by low Reynolds number (Re). We discuss motion at low Re in natural and synthetic systems, along with various propulsion mechanisms, including electrophoresis, electrolyte diffusiophoresis, and nonelectrolyte diffusiophoresis. We also describe the newly uncovered phenomena of motility in non-ATP-driven self-powered enzymes and the directional movement of these enzymes in response to substrate gradients. These enzymes can also be immobilized to function as fluid pumps in response to the presence of their substrates. Finally, we review emergent collective behavior arising from interacting motile species, and we discuss the possible biomedical applications of the synthetic nanobots and microbots.

Anatomy of Nanoscale Propulsion.

Synthetic nano- and microscale machines move autonomously in solution or drive fluid flows by converting sources of energy into mechanical work. Their sizes are comparable to analytes (sub-nano- to microscale), and they respond to signals from each other and their surroundings, leading to emergent collective behavior. These machines can potentially enable hitherto difficult analytical applications. In this article, we review the development of different classes of synthetic nano- and micromotors and pumps and indicate their possible applications in real-time in situ chemical sensing, on-demand directional transport, cargo capture and delivery, as well as analyte isolation and separation.

Synthetic Nano- and Micromachines in Analytical Chemistry: Sensing, Migration, Capture, Delivery, and Separation.

In advanced electrical grids of the future, electrochemically rechargeable fluids of high energy density will capture the power generated from intermittent sources like solar and wind. To meet this outstanding technological demand there is a need to understand the fundamental limits and interplay of electrochemical potential, stability, and solubility in low-weight redox-active molecules. By generating a combinatorial set of 1,4-dimethoxybenzene derivatives with different arrangements of substituents, we discovered a minimalistic structure that combines exceptional long-term stability in its oxidized form and a record-breaking intrinsic capacity of 161 mAh/g. The nonaqueous redox flow battery has been demonstrated that uses this molecule as a catholyte material and operated stably for 100 charge/discharge cycles. The observed stability trends are rationalized by mechanistic considerations of the reaction pathways.
