Norton grape is one of the most important wine grapes in Southern and Midwestern states and generates massive pomace byproducts. The objective of this study is to characterize the antioxidant compounds and activity in Norton grape pomace extract (GPE) and further assess the potential health promoting properties of Norton GPE using an animal disease model. The total phenolic content and anthocyanins in Norton GPE were 475.4 mg of gallic acid equiv/g and 156.9 mg of cyanidin 3-glucoside equiv/g, respectively. Catechin and epicatechin in GPE were 28.6 and 24.5 mg/g, respectively. Other major antioxidants in GPE included quercetin (1.6 mg/g), trans-resveratrol (60 μg/g), gallic acid (867.2 μg/g), coutaric acid (511.8 μg/g), p-hydroxybenzoic acid (408.3 μg/g), and protocatechuic acid (371.5 μg/g). The antioxidant activity of GPE was evaluated by oxygen radical absorbance capacity (ORAC) and was 4133 μmol of Trolox equiv/g. Male diet-induced obese (DIO) mice were randomly divided to three treatment groups (n = 12): a normal diet (ND group), a high fat diet (HF group), and the high fat diet supplemented with GPE (HFGPE group). After 12-week treatment, mice in the high fat diet groups gained 29% more weight than the ND group. The GPE supplementation (estimated 250 mg/kg bw/d) lowered plasma C-reactive protein levels by 15.5% in the high fat diet fed mice (P < 0.05), suggesting a potential anti-inflammatory effect by dietary GPE. However, dietary GPE did not improve oxidative stress in DIO mice as determined by plasma ORAC, glutathione peroxidase, and liver lipid peroxidation. The results showed that GPE contained significant antioxidants and dietary GPE exerted an anti-inflammatory effect in diet induced obesity.

Effects of grape pomace antioxidant extract on oxidative stress and inflammation in diet induced obese mice.

Dietary antioxidants may provide a cost-effective strategy to promote health in obesity by targeting oxidative stress and inflammation. We recently found that the antioxidant-rich grape skin extract (GSE) also exerts a novel anti-hyperglycemic activity. This study investigated whether 3-month GSE supplementation can improve oxidative stress, inflammation, and hyperglycemia associated with a Western diet-induced obesity. Young diet-induced obese (DIO) mice were randomly divided to three treatment groups (n = 12): a standard diet (S group), a Western high fat diet (W group), and the Western diet plus GSE (2.4 g GSE/kg diet, WGSE group). By week 12, DIO mice in the WGSE group gained significantly more weight (24.6 g) than the W (20.2 g) and S groups (11.2 g); the high fat diet groups gained 80% more weight than the standard diet group. Eight of 12 mice in the W group, compared to only 1 of 12 mice in the WGSE group, had fasting blood glucose levels above 140 mg/dL. Mice in the WGSE group also had 21% lower fasting blood glucose and 17.1% lower C-reactive protein levels than mice in the W group (P < 0.05). However, the GSE supplementation did not affect oxidative stress in diet-induced obesity as determined by plasma oxygen radical absorbance capacity, glutathione peroxidase, and liver lipid peroxidation. Collectively, the results indicated a beneficial role of GSE supplementation for improving glycemic control and inflammation in diet-induced obesity.

Dietary supplementation of grape skin extract improves glycemia and inflammation in diet-induced obese mice fed a Western high fat diet.

Chemokines are small, secreted cytokine peptides known principally for their ability to induce migration and activation of leukocyte populations and regulate the immune response mechanisms. The cobia (Rachycentron canadum), a marine finfish species, has a great potential for net cage aquaculture in the South China Sea. We isolated and characterized a CC chemokine cDNA from cobia-designated RcCC2. Its cDNA is 783 bp in length and encodes a putative protein of 110 amino acids. Homology and phylogenetic analysis revealed that the RcCC2 gene, which contains four conserved cysteine residues, shares a high degree of similarity with other known CC chemokine sequences and is closest to the CCL19/21 clade. The mRNA of RcCC2 is expressed constitutively in all tested tissues, including gill, liver, muscle, spleen, kidney, head kidney, skin, brain, stomach, intestine and heart, but not blood, with the highest level of expression in gill and liver. The reverse transcription quantitative polymerase chain reaction was used to examine the expression of the RcCC2 gene in immune-related tissues, including head kidney, spleen and liver, following intraperitoneal injection of the viral mimic polyriboinosinic polyribocytidylic acid, formalin-killed Vibrio carchariae (bacterial vaccine) and phosphate-buffered saline as a control. RcCC2 gene expression was up-regulated differentially in head kidney, spleen and liver during 12 h after challenge. These results indicate that the RcCC2 gene is inducible and is involved in immune responses, suggesting RcCC2 has an important role in the early stage of viral and bacterial infections.

Identification and expression analysis of a CC chemokine from cobia (Rachycentron canadum).

Chitosan coating plus different essential oils was developed and applied to fresh blueberries in order to find more natural treatments to preserve fresh fruit quality and safety during postharvest storage. Studies were 1st performed in vitro where wild-type Escherichia coli and Penicillium digitatum were grown in suitable media, and then subjected to 6 essential oils. Three compounds, carvacrol (CAR), cinnamaldehyde (CIN), and trans-cinnamaldehyde (ECIN) had high antimicrobial capacity and were selected for an in vivo study for postharvest storage of blueberries. The selected essential oils, 0.5% each, were added into a chitosan solution and coated on fresh blueberries. After storage at 5, 10, and 20 °C for various days, fruit firmness and microbial populations were evaluated. The chitosan coating substantially decreased bacteria and yeasts/molds on the fruit, and all 3 essential oils added to the antimicrobial activities. Further dosage experiments showed that the antimicrobial activity remained even when lowering CAR concentration to 0.1% and ECIN to 0.2%. Chitosan, CAR, and ECIN also maintained fruit firmness. Our results suggest that chitosan coatings containing essential oils are effective in extending the shelf life of fresh blueberries.

Effects of chitosan-essential oil coatings on safety and quality of fresh blueberries.

Bioassay-guided fractionation of the fruit powder of graviola (Annona muricata) yielded three novel compounds: muricins J, K, and L. The compounds are all C35 Annonaceous acetogenins with a mono-tetrahydrofuran ring and four hydroxyls. Their structures were elucidated by spectral methods and chemical modification after isolation via chromatographic techniques and HPLC purification. These three acetogenins demonstrated an antiproliferative against human prostate cancer PC-3 cells.

The effect of chlorine dioxide (ClO2) gas on the safety and quality of blueberries was studied. In vitro studies revealed that both ClO2 gas fumigation and ClO2 direct contact in water killed food pathogen bacterium Escherichia coli and fruit decay pathogen fungus Colletotrichum acutatum. In vivo studies were conducted using noninoculated berries and berries inoculated with postharvest decay and foodborne pathogens. Berries were inoculated with either E. coli (5.2 log CFU/g) or C. acutatum (3.9 log CFU/g). Inoculated fruit were dried for 2 h at room temperature in a climate-controlled laboratory and packed in perforated commercial clamshells, with or without ClO2 pads, and stored at 10°C for up to 9 days. The effects of ClO2 on microbial populations and fruit firmness were monitored during storage. In the inoculation experiment, treatment with ClO2 reduced populations of E. coli and C. acutatum by 2.2 to 3.3 and 1.3 to 2.0 log CFU/g, respectively. For the noninoculated blueberries, the initial total aerobic bacteria count and the yeast and mold count were 4.2 and 4.1 log CFU/g, respectively. ClO2 treatment reduced total aerobic bacteria count and yeast and mold count by 1.5 to 1.8 and 1.3 to 1.7 log CFU/g, respectively. The firmness of both inoculated and noninoculated blueberries was maintained by ClO2 treatment. Thus, controlled-release ClO2 gas fumigation technology shows promise as an effective and practical antimicrobial agent in commercial clamshell packaging of blueberry and other fruits.

Antimicrobial activity of controlled-release chlorine dioxide gas on fresh blueberries.

Chitosan films incorporated with various concentrations of the complex of β-cyclodextrin and essential oils (β-CD/EO) were prepared and investigated for antimicrobial, mechanical, and physical properties. Four bacterial strains that commonly contaminate food products were chosen as target bacteria to evaluate the antimicrobial activity of the prepared films. The incorporation of β-CD/EO significantly increased the antimicrobial activities of the chitosan films against Escherichia coli, Salmonella typhimurium, Staphylococcus aureus, and Listeria monocytogenes. It was also found that tensile strength (TS) of chitosan film was significantly increased with the incorporation of the β-cyclodextrin and 0.75% essential oils complex. The elongation at break (EB) decreased with the increasing concentrations of essential oils. Inclusion of the complex of β-cyclodextrin and 0.25% essential oils also significantly decreased water vapor permeability (WVP) of chitosan films. Our results suggest that chitosan films containing β-CD/EO could be used as active food-packaging material.

Antimicrobial and mechanical properties of β-cyclodextrin inclusion with essential oils in chitosan films.

A new microsporidian species is described from farmed red sea bream Pagrus major (Temminck & Schlegel) (Teleostei: Sparidae). Large numbers of spherical whitish xenomas were observed throughout the visceral organs of the host. Histological examination showed that the microsporidia caused several xenomas that were embedded in the intestinal muscularis externa or submucosa. Light and transmission electron microscopy examination of the spores also revealed morphological features typical of species of Glugea Thélohan, 1891. This microsporidian parasite has two different types of mature spores: microspores and macrospores. The spores are elongate-ovoid, with a large posterior vacuole. The polaroplast is bi-partite, with anterior and posterior parts comprising densely packed lamellae and loose membranes, respectively, and occupies approximately the anterior half of the spore. The polar filament is anisofilar, with 12-13 coils in a single layer almost touching the posterior spore wall. Comparison of the small subunit rDNA sequences revealed 92.7-98.1% identity with the sequences available from other Glugea spp. from piscine hosts. Phylogenetic analysis demonstrated that the microsporidian species studied clustered within the Glugea clade with strong support. Based on the differences in the morphological characteristics and molecular data, the microsporidian infecting P. major is considered to represent a species new to science, Glugea pagri n. sp.

A new species of Glugea Thélohan, 1891 in the red sea bream Pagrus major (Temminck & Schlegel) (Teleostei: Sparidae) from China.

The sponge of Clathria cervicornis is commonly used in traditional medicine. This study aims to identify the active compound in C. cervicornis and to evaluate its antimicrobial activity. The purified active compound was determined to be crambescidin 800 and was found to be highly active against Acinetobacter baumannii (minimal inhibitory concentration, MIC=2 μg/ml), Klebsiella pneumoniae (MIC=1 μg/ml) and Pseudomonas aeruginosa (MIC=1 μg/ml). A potent antimicrobial compound, crambescidin 800, was isolated in Clathria cervicornis. It is extremely active against three common pathogenic bacteria.

Three polybrominated diphenyl ethers, 2-(2',4'-dibromophenoxy)-3,5-dibromophenol (1) and 2-(2',4'-dibromophenoxy)-3,4,5-tribromophenol (2) were isolated from the marine sponge Dysidea granulosa; and 2-(2',4'-dibromophenoxy)-4,6-dibromophenol (3) from Dysidea spp. They exhibited potent and broad spectrum in vitro antibacterial activity, especially against methicillin resistant Staphylococcus aureus (MRSA), methicillin sensitive Staphylococcus aureus (MSSA), Escherichia coli O157:H7, and Salmonella. Minimal inhibitory concentration (MIC) was evaluated against 12 clinical and standard strains of Gram positive and negative bacteria. The observed MIC range was 0.1-4.0mg/L against all the Gram positive bacteria and 0.1-16.0mg/L against Gram negative bacteria. 2-(2',4″-Dibromophenoxy)-3,5-dibromophenol showed stronger broad spectrum antibacterial activity than other two compounds. 2-(2',4″-Dibromophenoxy)-3,5-dibromophenol and 2-(2',4'-dibromophenoxy)-4,6-dibromophenol are thermo-stable. The results suggest that 2-(2',4'-dibromophenoxy)-3,5-dibromophenol could be used as a potential lead molecule for anti-MRSA, anti-E. coli O157:H7, and anti-Salmonella for drug development.

Polybrominated diphenyl ethers with potent and broad spectrum antimicrobial activity from the marine sponge Dysidea.

The effects of ultrasound treatment (UT) on the properties of methylcellulose (MC)/stearic acid (SA) blending films were studied. Film-forming emulsions were prepared with different UT conditions and characterized with respect to viscosity. The lipid aggregation and distribution in the blending dispersions were studied by the micrographs of Transmission Electron Microscopy (TEM). The micrographs of both surface and cross-section of the films were observed by scanning electron microscope (SEM) and the tensile strength (TS), elongation at break (E), water vapor permeability (WVP) and contact angles of the resulting films were determined as well. The intensification of the UT condition led to a decrease of viscosity of the MC-SA blending emulsions, a more homogeneous lipid distribution and a denser internal microstructure of the resulting films. UT exposure affected the mechanical, moisture barrier and surface hydrophobic properties. The optimal values of both TS and E was obtained from the sample treated for 10min and 180W power, while the sample treated for 10min and 270W presented the lowest value of WVP. However, an excessive exposure of UT led to a decrease of the mechanical and moisture barrier performance. By observing and analyzing the SEM graphs and the contact angles of the film surfaces, it was found that UT within the appropriate bounds had a notably positive effect on improving the surface hydrophobic property of the MC-SA blending films.

Winter melon (Benincasa hispida) is a widely consumed crop in Asia, and believed to impart special benefits to human health. The nutritional composition and sensory properties of four juice types, resulting from a combination of pulp levels (low/high pulp, LP/HP) and thermal processing (with/without boiling, B/NB), LPNB, HPNB, LPB and HPB, were compared.
