Melanopsin (Opn4) is a photopigment found in a subset of retinal ganglion cells (RGCs) that project to various brain areas. These neurons are intrinsically photosensitive (ipRGCs) and are implicated in nonimage-forming responses to environmental light such as the pupillary light reflex and circadian entrainment. Recent evidence indicates that ipRGCs respond to light at birth, but questions remain as to whether and when they undergo significant functional changes. We used bacterial artificial chromosome transgenesis to engineer a mouse line in which enhanced green fluorescent protein (EGFP) is expressed under the control of the melanopsin promoter. Double immunolabeling for EGFP and melanopsin demonstrates their colocalization in ganglion cells of mutant mouse retinas. Electrophysiological recordings of ipRGCs in neonatal mice (postnatal day 0 [P0] to P7) demonstrated that these cells responded to light with small and sluggish depolarization. However, starting at P11 we observed ipRGCs that responded to light with a larger and faster onset (<1 s) and offset (<1 s) depolarization. These faster, larger depolarizations were observed in most ipRGCs by early adult ages. However, on application of a cocktail of synaptic blockers, we found that all cells responded to light with slow onset (>2.5 s) and offset (>10 s) depolarization, revealing the intrinsic, melanopsin-mediated light responses. The extrinsic, cone/rod influence on ipRGCs correlates with their extensive dendritic stratification in the inner plexiform layer. Collectively, these results demonstrate that ipRGCs make use of melanopsin for phototransduction before eye opening and that these cells further integrate signals derived from the outer retina as the retina matures.

M&#xE9;lanopsine (Opn4) est un photopigment trouve dans un sous-ensemble de cellules ganglionnaires de la r&#xE9;tine (CGR) de ce projet pour diff&#xE9;rentes zones du cerveau. Ces neurones sont intrins&#xE8;quement photosensibles (ipRGCs) et sont impliqu&#xE9;s dans nonimage formant des r&#xE9;ponses &#xE0; la lumi&#xE8;re de l&#x26;#39;environnement tels que l&#x26;#39;entra&#xEE;nement du r&#xE9;flexe pupillaire et circadienne de la lumi&#xE8;re. Des donn&#xE9;es r&#xE9;centes indiquent que ipRGCs r&#xE9;agissent &#xE0; la lumi&#xE8;re &#xE0; la naissance, mais des questions demeurent quant &#xE0; savoir si et quand ils subissent d&#x26;#39;importantes modifications fonctionnelles. Nous avons utilis&#xE9; la transgen&#xE8;se bact&#xE9;rienne chromosome artificiel de concevoir une lign&#xE9;e de souris dans lequel la prot&#xE9;ine fluorescente verte am&#xE9;lior&#xE9;e (EGFP) est exprim&#xE9;e sous le contr&#xF4;le du promoteur m&#xE9;lanopsine. Immunomarquage double pour EGFP et m&#xE9;lanopsine d&#xE9;montre leur colocalisation dans les cellules ganglionnaires de la r&#xE9;tine de souris mutantes. Des enregistrements &#xE9;lectrophysiologiques de ipRGCs chez la souris nouveau-n&#xE9;s (jour postnatal 0 [P0] &#xE0; P7) ont d&#xE9;montr&#xE9; que ces cellules ont r&#xE9;pondu &#xE0; la lumi&#xE8;re avec la d&#xE9;polarisation petite et lente. Cependant, &#xE0; partir de P11, nous avons observ&#xE9; ipRGCs qui ont r&#xE9;pondu &#xE0; la lumi&#xE8;re avec un d&#xE9;but plus et plus vite (&#x26;lt;1 s) et le d&#xE9;calage (&#x26;lt;1 s) la d&#xE9;polarisation. Ces rapides, les plus grandes d&#xE9;polarisations ont &#xE9;t&#xE9; observ&#xE9;es dans la plupart des ipRGCs par adulte pr&#xE9;coce. Toutefois, sur demande d&#x26;#39;un cocktail d&#x26;#39;inhibiteurs synaptiques, nous avons constat&#xE9; que toutes les cellules ont r&#xE9;pondu &#xE0; la lumi&#xE8;re avec l&#x26;#39;apparition lente (&#x26;gt; 2,5 s) et le d&#xE9;calage (&#x26;gt; 10 s) de d&#xE9;polarisation, r&#xE9;v&#xE9;lant la valeur intrins&#xE8;que, les r&#xE9;ponses de lumi&#xE8;re m&#xE9;lanopsine m&#xE9;diation. Le extrins&#xE8;que, c&#xF4;ne / tige influence sur ipRGCs en corr&#xE9;lation avec leur stratification dendritique vaste dans la couche plexiforme interne. Collectivement, ces r&#xE9;sultats d&#xE9;montrent que ipRGCs faire usage de la m&#xE9;lanopsine de la phototransduction avant l&#x26;#39;ouverture des yeux et que ces cellules d&#x26;#39;int&#xE9;grer davantage les signaux provenant de la r&#xE9;tine externe comme la r&#xE9;tine m&#xFB;rit.

Melanopsin (Opn4) ist eine Photopigment in eine Teilmenge der retinalen Ganglienzellen (Routinggruppenconnectors) das Projekt auf verschiedene Bereiche des Gehirns gefunden. Diese Neuronen sind intrinsisch lichtempfindliche (IpRGCs) und bei der Bildung von eingebundener verwickelt sind Antworten zu vorhandenem Licht wie die Pupillen Licht-Reflex und circadiane Entrainment. Neuere Beweise gibt, dass IpRGCs auf Licht bei der Geburt reagieren, aber noch Fragen, ob und wann sie bedeutende funktionelle Ver&auml;nderungen durchmachen. Wir verwendeten bacterial artificial Chromosome Transgenese Ingenieur eine Maus-Linie in dem Gr&uuml;n verbessert, die fluoreszierenden Proteins (EGFP) unter der Kontrolle des Projekttr&auml;gers Melanopsin ausgedr&uuml;ckt wird. Jedoch beginnend am P11 wir beobachteten IpRGCs, die Licht mit einer gr&ouml;&szlig;eren und schnelleren Beginn beantwortet hat (&lt; 1 s) und offsetdruck (&lt; 1 s) depolarisierung. Diese schneller, gr&ouml;&szlig;ere Depolarizations in den meisten IpRGCs von fr&uuml;hen erwachsenen altersgruppen. Jedoch auf antrag von einem Cocktail von synaptischen blocker beobachtet wurden, fanden wir, dass alle zellen auf licht mit langsamen beginn reagiert (&gt; 2,5 s) und Offsetdruck (&gt; 10 s) Depolarisierung, offenbart die systeminternen, Melanopsin-vermittelte leichten Antworten. Die extrinsischen, Kegel/Rod Einfluss auf IpRGCs korreliert mit ihren umfangreichen dendritische Schichtung in die innere plexiforme Schicht. Gemeinsam diese Ergebnisse zeigen, dass IpRGCs machen Verwendung von Melanopsin f&uuml;r Signaltransduktion vor dem &Ouml;ffnen der Augen und dass diese Zellen weitere Signale, die von der &auml;u&szlig;eren Netzhaut abgeleitet werden, wie die Netzhaut reift zu integrieren.

Melanopsina (Opn4) es un fotopigmento se encuentra en un subconjunto de las c&#xE9;lulas ganglionares de la retina (CGR) que se proyectan hacia varias zonas del cerebro. Estas neuronas son intr&#xED;nsecamente fotosensibles (ipRGCs) y est&#xE1;n implicados en sin im&#xE1;genes que forman las respuestas a la luz del ambiente, como el arrastre de la luz pupilar reflejo y circadiano. La evidencia reciente indica que las ipRGCs responden a la luz al nacer, pero queda la pregunta de si, y cuando se someten a cambios funcionales significativos. Se utiliz&#xF3; la transg&#xE9;nesis cromosoma artificial bacteriano para dise&#xF1;ar una l&#xED;nea de ratones en los cuales se expresa el aumento de prote&#xED;na verde fluorescente (EGFP) bajo el control del promotor de melanopsina. Inmunomarcaci&#xF3;n doble para EGFP y melanopsina demuestra su colocalizaci&#xF3;n en las c&#xE9;lulas ganglionares de la retina de ratones mutantes. Registros electrofisiol&#xF3;gicos de las ipRGCs en ratones reci&#xE9;n nacidos (despu&#xE9;s del d&#xED;a 0 [P0] a P7) demostraron que estas c&#xE9;lulas respondieron a la luz con la despolarizaci&#xF3;n de las peque&#xF1;as y lento. Sin embargo, a partir de P11 se observ&#xF3; ipRGCs que respondieron a la luz con un inicio m&#xE1;s grande y m&#xE1;s r&#xE1;pida (&#x26;lt;1 s) y desplazamiento (&#x26;lt;1 s) de despolarizaci&#xF3;n. Estos r&#xE1;pidos y despolarizaciones mayores se observaron en la mayor&#xED;a de las ipRGCs por la edad adulta temprana. Sin embargo, sobre la aplicaci&#xF3;n de un c&#xF3;ctel de bloqueadores sin&#xE1;pticos, se encontr&#xF3; que todas las c&#xE9;lulas respond&#xED;an a la luz con inicio lento (&#x26;gt; 2,5 s) y desplazamiento (&#x26;gt; 10 s) de despolarizaci&#xF3;n, revelando los intr&#xED;nsecos, melanopsina las respuestas mediadas por la luz. La extr&#xED;nseca, cono / varilla de influencia en las ipRGCs se correlaciona con su estratificaci&#xF3;n dendr&#xED;ticas extensa en la capa plexiforme interna. En conjunto, estos resultados demuestran que las ipRGCs hacer uso de melanopsina para fototransducci&#xF3;n antes de la abertura del ojo y que estas c&#xE9;lulas una mayor integraci&#xF3;n de las se&#xF1;ales derivadas de la retina externa como la retina madura.

Morphological studies have confirmed the existence of at least three distinct subpopulations of ipRGCs, but studies of the physiology of ipRGCs at the single cell level have focused mainly on M1 cells, the dendrites of which stratify solely in sublamina a (OFF sublamina) of the retinal inner plexiform layer (IPL). Little work has been done to compare the functional properties of M1 cells to those of M2 cells, the dendrites of which stratify solely in sublamina b (ON sublamina) of the IPL. The goal of the current study was to compare the morphology, intrinsic light response, and intrinsic membrane properties of M1 and M2 cells in the mouse retina. Here we demonstrate additional morphological differences between M1 and M2 cells as well as distinct physiological characteristics of both the intrinsic light responses and intrinsic membrane properties. M2 cells displayed a more complex dendritic arborization and higher input resistance, yet showed lower light sensitivity and lower maximal light responses than M1 cells. These data indicate morphological and functional heterogeneity among ipRGCs.

Functional and morphological differences among intrinsically photosensitive retinal ganglion cells.

Un sous-ensemble de cellules ganglionnaires de la r&#xE9;tine des mammif&#xE8;res exprimer la m&#xE9;lanopsine photopigment et sont intrins&#xE8;quement photosensibles (ipRGCs). Diese Zellen sind in Bild-bildenden visuelle Antworten zu vorhandenem Licht, wie die Pupillen Licht-Reflex, saisonale Anpassungen in Physiologie, photic Hemmung der n&auml;chtlichen Melatonin-Freisetzung und Modulation der Schlaf, Wachheit und Aktivit&auml;t beteiligt. A small subset of retinal ganglion cells projecting to the suprachiasmatic nucleus and other brain areas, is implicated in non-image forming visual responses to environmental light such as the pupillary light reflex, seasonal adaptations in physiology, photic inhibition of nocturnal melatonin release, and modulation of sleep, alertness and activity. These cells are intrinsically photosensitive (ipRGCs) and express an opsin-like photopigment called melanopsin. Two recent studies utilizing selective genetic ablation of ipRGCs demonstrate the key role of these inner retinal cells in conveying luminance signals to the brain for non-image forming visual processing. These findings advance our understanding of functional organization of a novel photosensory system in the mammalian retina, demonstrating well-defined roles for ipRGCs in circadian timing and other homeostatic functions related to ambient illumination.

In mammals, intrinsically photosensitive retinal ganglion cells (ipRGCs) mediate various non-image-forming photic responses, such as circadian photoentrainment, pupillary light reflex and pineal melatonin suppression. ipRGCs directly respond to environmental light by activation of the photopigment melanopsin followed by the opening of an unidentified cation-selective channel. Studies in heterologous expression systems and in the native retina have strongly implicated diacylglycerol-sensitive transient receptor potential channels containing TRPC3, TRPC6 and TRPC7 subunits in melanopsin-evoked depolarization. Here we show that melanopsin-evoked electrical responses largely persist in ipRGCs recorded from early postnatal (P6-P8) and adult (P22-P50) mice lacking expression of functional TRPC3, TRPC6 or TRPC7 subunits. Multielectrode array (MEA) recordings performed at P6-P8 stages under conditions that prevent influences from rod/cone photoreceptors show comparable light sensitivity for the melanopsin-evoked responses in these mutant mouse lines in comparison to wild-type (WT) mice. Patch-clamp recordings from adult mouse ipRGCs lacking TRPC3 or TRPC7 subunits show intrinsic light-evoked responses equivalent to those recorded in WT mice. Persistence of intrinsic light-evoked responses was also noted in ipRGCs lacking TRPC6 subunits, although with significantly smaller magnitudes. These results demonstrate that the melanopsin-evoked depolarization in ipRGCs is not mediated by either TRPC3, TRPC6 or TRPC7 channel subunits alone. They also suggest that the melanopsin signaling pathway includes TRPC6-containing heteromeric channels in mature retinas.

A subpopulation of retinal ganglion cells (RGCs) expresses the photopigment melanopsin, rendering these cells intrinsically photosensitive (ipRGCs). These cells are critical for competent circadian entrainment, pupillary light reflex, and other non-imaging-forming photic responses. Research has now demonstrated the presence of multiple subpopulations of ipRGC based on the dendritic stratification in the inner plexiform layer (IPL), those monostratified in the Off sublamina (M1), those monostratified in the On sublamina (M2,4,5), and those bistratified in both the On and the Off sublaminae (M3). Despite evidence that M1 and M2 cells are distinct subpopulations of ipRGC based on distinct morphological and physiological properties, the inclusion of M3 cells as a distinct subtype has remained controversial. Aside from the identification of M3 cells as a morphological subpopulation of ipRGC, to date there have been no functional descriptions of M3 cell physiology or synaptic inputs. Our data provide the first in-depth description of M3 cell structural and functional properties. We report that M3 cells form a morphologically heterogeneous population but one that is physiologically homogeneous with properties similar to those of M2 cells.

Eine Subpopulation der retinalen Ganglienzellen (Routinggruppenconnectors) dr&uuml;ckt das Photopigment Melanopsin, rendern diese Zellen an sich lichtempfindliche (IpRGCs). Diese Zellen sind entscheidend f&uuml;r die zust&auml;ndigen circadian Mitnahme, pupillary Licht-Reflex und andere nicht-Bildgebung-bildenden photic Antworten. Forschung hat nun das Vorhandensein von mehreren Subpopulationen von IpRGC, die auf der Grundlage der dendritischen Schichtung in die innere plexiforme Schicht (IPL), diese Monostratified in die Off-Sublamina (M1), die Monostratified in der On-Sublamina (M2, 4, 5) und die Bistratified in der On und die Off-Sublaminae (M3) gezeigt. Queste cellule sono critiche per competente circadiano trascinamento, riflesso pupillare alla luce e altre non-imaging-forming fotica risposte. La ricerca ha ora dimostrato la presenza di molteplici sottopopolazioni di ipRGC basato sulla stratificazione dendritica nello strato plessiforme interno (IPL), quelli monostratified in Off sublamina (M1), quelli monostratified nella sublamina On (M2, 4, 5) e quelli bistratified nell&#39;On e l&#39;Off sublaminae (M3). For decades, rods and cones were thought to be the only photoreceptors in the mammalian retina. However, a population of atypical photoreceptive retinal ganglion cells (RGCs) expresses the photopigment melanopsin and is intrinsically photosensitive (ipRGCs). These ipRGCs are crucial for relaying light information from the retina to the brain to control circadian photoentrainment, pupillary light reflex, and sleep. ipRGCs were initially described as a uniform population involved solely in signaling irradiance for non-image forming functions. Recent work, however, has uncovered that ipRGCs are unexpectedly diverse at the molecular, cellular and functional levels, and could even be involved in image formation. This review summarizes our current understanding of the diversity of ipRGCs and their various roles in modulating behavior.

Melanopsin imparts an intrinsic photosensitivity to a subclass of retinal ganglion cells (ipRGCs). Generally thought of as irradiance detectors, ipRGCs target numerous brain regions involved in non-image-forming vision. ipRGCs integrate their intrinsic, melanopsin-mediated light information with rod/cone signals relayed via synaptic connections to influence light-dependent behaviors. Early observations indicated diversity among these cells and recently several specific subtypes have been identified. These subtypes differ in morphological and physiological form, controlling separate functions that range from biological rhythm via circadian photoentrainment, to protective behavioral responses including pupil constriction and light avoidance, and even image-forming vision. In this Mini-Symposium review, we will discuss some recent findings that highlight the diversity in both form and function of these recently discovered atypical photoreceptors.

Melanopsin vermittelt eine systeminterne Lichtempfindlichkeit zu einer Subklasse von retinalen Ganglienzellen (IpRGCs). Diese Subtypen unterscheiden sich in morphologischen und physiologischen Form, separate Kontrollfunktionen, die von biologischen Rhythmus &uuml;ber circadian Photoentrainment zu sch&uuml;tzenden Verhaltens Antworten Verengung der Pupille Licht vermeiden und sogar Bild-bildenden Vision. In diesem Mini-Symposium-Bericht wir diskutieren einige neuere Befunde, die die Vielfalt in beiden Form hervorheben und Funktion dieser vor kurzem entdeckte atypische Photorezeptoren.

Melanopsina impartisce una fotosensibilit&agrave; intrinseco di una sottoclasse di cellule gangliari della retina (ipRGCs). Generalmente considerati come rivelatori di irradianza, ipRGCs destinazione numerose regioni del cervello coinvolte nella visione non-immagine-formante. ipRGCs integrare le loro informazioni luce intrinseche, melanopsina-mediata con asta/cono segnali inoltrati tramite connessioni sinaptiche per influenzare i comportamenti dipendenti dalla luce. Considerado como detectores de radiaci&oacute;n, ipRGCs objetivo de numerosas regiones del cerebro implicadas en la visi&oacute;n no forman imagen. ipRGCs integrar su informaci&oacute;n luz intr&iacute;nseca, mediada por la melanopsina con barra/cono se&ntilde;ales transmitidas v&iacute;a conexiones sin&aacute;pticas influir en comportamientos dependientes de la luz. Primeras observaciones indicaron diversidad entre estas c&eacute;lulas y recientemente se han identificado varios subtipos espec&iacute;ficos. Estos subtipos difieren en forma morfol&oacute;gica y fisiol&oacute;gica, controlar funciones separadas que van desde el ritmo biol&oacute;gico mediante photoentrainment circadiano, a las respuestas del comportamiento protectoras incluyendo constricci&oacute;n de la pupila y evitar la luz y visi&oacute;n incluso formaci&oacute;n de imagen.
