The Islets of Langerhans form a nutrient sensing network spread throughout the pancreas. They are tightly connected to the source organ, the intestine, and the target organs--liver, muscle, and fat cells. The expression of a unique set of proteins enables beta cells, the most frequent islet cell type, to detect elevated blood glucose levels and secrete insulin accordingly. Clustered beta-cells achieve tighter regulation of glucose-induced insulin secretion by coordination through cell surface proteins. They also adjust their secretory capacity and flow to avoid being damaged. The immediate reaction of the beta cell to nutrients is regulated by translational mechanisms, while longer term adaptations involve changes in transcription. Glucose increases overall protein synthesis in beta-cells but selectively boosts translation of some secretory proteins including insulin. This may be mediated through recognition of RNA motifs in the untranslated regions of those messengers. If essential molecular components of this nutrient sensing system are broken or fail due to repeated stress, beta cells malfunction, which on a larger scale manifest as diseases like diabetes mellitus.

Regulatory pathways intersect at but also originate from the insulin granule membrane. Examples include the small G-proteins Rab3a and Rab27a, which influence granule movement, and the transmembrane proteins (tyrosine phosphatase receptors type N) PTPRN and PTPRN2, which upregulate Î²-cell transcription and proliferation. In addition, many cosecreted compounds possess regulatory functions, often related to energy metabolism. For instance, ATP and Î³-amino butyric acid (GABA) modulate insulin and glucagon secretion, respectively; C-peptide protects Î²-cells and kidney cells; and amylin reduces gastric emptying and food intake via the brain. In this paper, we review the current knowledge of the insulin granule proteome and discuss its regulatory functions.

Polypyrimidine-tract binding protein 1 (PTBP1) is an important cellular regulator of messenger RNAs influencing the alternative splicing profile of a cell as well as its mRNA stability, location and translation. In addition, it is diverted by some viruses to facilitate their replication. Here, we used a novel PTBP1 knockout mouse to analyse the tissue expression pattern of PTBP1 as well as the effect of its complete removal during development. We found evidence of strong PTBP1 expression in embryonic stem cells and throughout embryonic development, especially in the developing brain and spinal cord, the olfactory and auditory systems, the heart, the liver, the kidney, the brown fat and cartilage primordia. This widespread distribution points towards a role of PTBP1 during embryonic development. Homozygous offspring, identified by PCR and immunofluorescence, were able to implant but were arrested or retarded in growth. At day 7.5 of embryonic development (E7.5) the null mutants were about 5x smaller than the control littermates and the gap in body size widened with time. At mid-gestation, all homozygous embryos were resorbed/degraded. No homozygous mice were genotyped at E12 and the age of weaning. Embryos lacking PTBP1 did not display differentiation into the 3 germ layers and cavitation of the epiblast, which are hallmarks of gastrulation. In addition, homozygous mutants displayed malformed ectoplacental cones and yolk sacs, both early supportive structure of the embryo proper. We conclude that PTBP1 is not required for the earliest isovolumetric divisions and differentiation steps of the zygote up to the formation of the blastocyst. However, further post-implantation development requires PTBP1 and stalls in homozygous null animals with a phenotype of dramatically reduced size and aberration in embryonic and extra-embryonic structures.

En outre, il est d&eacute;tourn&eacute; par des virus pour faciliter leur reproduction. Ici, nous avons utilis&eacute; un roman PTBP1 la souris knock-out pour analyser le mod&egrave;le d&#39;expression tissulaire de PTBP1 ainsi que l&#39;effet de son retrait complet au cours du d&eacute;veloppement. Nous avons trouv&eacute; la preuve de la forte expression de PTBP1 dans les cellules souches embryonnaires et tout au long du d&eacute;veloppement embryonnaire, notamment dans le d&eacute;veloppement du cerveau et la moelle &eacute;pini&egrave;re, les syst&egrave;mes olfactifs et auditifs, coeur, le foie, le rein, la graisse brune et les primordiums cartilage. Cette vaste r&eacute;partition pointe vers un r&ocirc;le de PTBP1 durant le d&eacute;veloppement embryonnaire. Descendants homozygotes, identifi&eacute;es par PCR et immunofluorescence, ont &eacute;t&eacute; en mesure d&#39;implanter mais ont &eacute;t&eacute; arr&ecirc;t&eacute;s ou retard&eacute;s la croissance. Les mutants null jour 7.5 du d&eacute;veloppement embryonnaire (E7.5) &eacute;taient environ 5 x plus petit que la m&ecirc;me port&eacute;e de contr&ocirc;le et de l&#39;&eacute;cart de taille s&#39;est creus&eacute;e avec le temps. Au milieu, tous les embryons homozygotes ont &eacute;t&eacute; r&eacute;sorb&eacute;s/d&eacute;grad&eacute;s. Aucune souris homozygotes ont &eacute;t&eacute; g&eacute;notyp&eacute;s &agrave; E12 et l&#39;&acirc;ge du sevrage. Manque de PTBP1 des embryons n&#39;affichent pas de diff&eacute;renciation dans les 3 couches de germe et cavitation de l&#39;&eacute;piblaste, qui font la renomm&eacute;e de la gastrulation. De plus, les mutants homozygotes affichent ectoplacental malform&eacute; c&ocirc;nes et sacs vitellins, tant structure de soutien pr&eacute;coce de l&#39;embryon proprement dit. Nous concluons que la PTBP1 n&#39;est pas n&eacute;cessaire pour les premiers isovolum&eacute;triques divisions et les &eacute;tapes de la diff&eacute;renciation du zygote jusqu&#39;&agrave; la formation du blastocyste. Cependant, post-implantation d&eacute;veloppement n&eacute;cessite des PTBP1 et les &eacute;tals chez les animaux homozygotes null avec un ph&eacute;notype de taille consid&eacute;rablement r&eacute;duite et les aberrations dans les structures embryonnaires et extra-embryonnaires.

Am Tag 7,5 der embryonalen Entwicklung (E7.5) waren die null Mutanten ca. 5 X kleiner als die Steuerelement-Wurfgeschwister und die L&uuml;cke in der K&ouml;rpergr&ouml;&szlig;e, die mit der Zeit erweitert. Mid-gestation waren alle homozygoten Embryos resorbiert/abgebaut. Keinen homozygote M&auml;use waren genotypisierten bei E12 und dem Alter der Entw&ouml;hnung. Fehlt PTBP1 Embryonen wurden nicht angezeigt, Unterscheidung in 3 Keim Schichten und Kavitation von der Epiblast, die Stempel Gastrulation sind. Dar&uuml;ber hinaus angezeigt homozygote Mutanten aufgrund eines fehlerhaften Ectoplacental Zapfen und Eigelb Sacs, sowohl fr&uuml;he unterst&uuml;tzende Struktur des richtigen Embryos. Wir schlie&szlig;en, dass PTBP1 nicht f&uuml;r den fr&uuml;hesten Isovolumetric Divisionen und Differenzierung Schritte von der Zygote bis zu der Bildung der Blastozyste erforderlich ist. Jedoch post-implantation Weiterentwicklung erfordert PTBP1 und St&auml;nden in homozygot null Tiere mit einem Ph&auml;notyp deutlich geringere Gr&ouml;&szlig;e und Verirrung in embryonalen und extra-embryonic Strukturen.

Al giorno 7,5 dello sviluppo embrionale (E7.5) i mutanti null erano circa 5 x pi&ugrave; piccolo littermates il controllo e il divario nella dimensione corporea, ampliato con il tempo. A mid-gestation, tutti gli embrioni omozigoti erano riassorbiti e/o degradati. No topi omozigoti sono stati genotipizzati a E12 e l&#39;et&agrave; dello svezzamento. Embrioni privi di PTBP1 non hanno visualizzato la differenziazione in 3 strati germinali e cavitazione del epiblast, quali sono le caratteristiche distintive della gastrulazione. Inoltre, omozigoti mutanti visualizzato malformati ectoplacental coni e tuorlo sacs, entrambi primi struttura sostegno dell&#39;embrione corretta. Concludiamo che PTBP1 non &egrave; richiesto per le prime divisioni isovolumetric e passi di differenziazione di zigote fino alla formazione della blastocisti. Tuttavia, ulteriormente i sviluppo richiede PTBP1 e bancarelle in animali omozigoti null con un fenotipo di dimensioni notevolmente ridotte e aberrazione in strutture embrionale e fetale.

Prote&iacute;na de uni&oacute;n a Polypyrimidine-zona 1 (PTBP1) es un importante regulador celular de ARN mensajero que influyen en la alternativa que empalma el perfil de una celda, as&iacute; como su estabilidad del mRNA, localizaci&oacute;n y traducci&oacute;n. Adem&aacute;s, es desviado por algunos virus para facilitar su replicaci&oacute;n. Aqu&iacute;, utilizamos una novela rat&oacute;n knockout de PTBP1 para analizar el patr&oacute;n de expresi&oacute;n de tejido de PTBP1, as&iacute; como el efecto de su extirpaci&oacute;n completa durante el desarrollo. Se encontraron pruebas de fuerte expresi&oacute;n de PTBP1 en las c&eacute;lulas madre embrionarias y durante el desarrollo embrionario, especialmente en el cerebro en desarrollo y de la m&eacute;dula espinal, los sistemas olfativos y auditivos, el coraz&oacute;n, el h&iacute;gado, el ri&ntilde;&oacute;n, la grasa parda y primordios de cart&iacute;lago. Esta distribuci&oacute;n generalizada apunta hacia un papel de PTBP1 durante el desarrollo embrionario. Descendencia homocig&oacute;tica, identificado por PCR y la inmunofluorescencia, pudieron implantar pero fueron arrestado o retraso en el crecimiento. En d&iacute;a 7.5 del desarrollo embrionario (E7.5) los mutantes nulos eran unos 5 x menor de los hermanos de camada de control y la diferencia de tama&ntilde;o de cuerpo se ha ampliado con el tiempo. En mid-gestation, todos los embriones homocig&oacute;ticos fueron reabsorbidos/degradados. No ratones homocig&oacute;ticos fueron genotipados en E12 y la edad de destete. Embriones que carecen de PTBP1 no se mostraban diferenciaci&oacute;n en las 3 capas germinales y cavitaci&oacute;n del epiblasto, que son caracter&iacute;sticas distintivas de gastrulaci&oacute;n. Adem&aacute;s, mutantes homocig&oacute;ticos exhibi&oacute; ectoplacental malformados conos y sacos vitelinos, ambas primitiva estructura apoyo del embri&oacute;n adecuado. Concluimos que la PTBP1 no es necesario para las primeras divisiones de isovolum&eacute;trica y pasos de la diferenciaci&oacute;n del cigoto hasta la formaci&oacute;n del blastocisto. Sin embargo, tras la implantaci&oacute;n perfeccionamiento requiere PTBP1 y puestos en animales nulos homocig&oacute;ticos con un fenotipo de tama&ntilde;o reducido dram&aacute;ticamente y aberraci&oacute;n en estructuras embrionarias y ovulares.

PTBP1 se requiere para el desarrollo embrionario antes de gastrulaci&oacute;n.

Innate immunity is the oldest form of defense and is found to some degree in all species. It predates the adaptive immune system, consisting of antibodies, B cells, T cells, and the major histocompatibility antigens. These are found only in higher vertebrates and have been the focus of the majority of immunological research, particularly in mice and humans, over the years. Knowledge of immunity in lower vertebrate and invertebrate species is now increasing rapidly, shedding light on the evolution of immunity and in many cases adding to our understanding of the mammalian system. Several recurring structural, genetic, and developmental mechanisms are common features in these processes in both the cellular and the molecular aspects of innate immunity.
