BALB/cJ &#x5C0F;&#x9F20;&#x7684;&#x5148;&#x8FDB;&#x5149;&#x5468;&#x671F;&#x7684;&#x52A0;&#x901F;&#x7684; re-entrainment&#x3002;

Circadian rhythms in mammals are coordinated by the suprachiasmatic nuclei (SCN) of the hypothalamus, which are most potently synchronized to environmental light-dark cycles. Large advances in the light-dark cycle typically yield gradual advances in activity rhythms on the order of 1-2h per day until re-entrainment is complete due to limitations on the circadian system which are not yet understood. In humans, this delay until re-entrainment is accomplished is experienced as jetlag, with accompanying symptoms of malaise, decreased cognitive performance, sleep problems and gastrointestinal distress. In these experiments, locomotor rhythms of BALB/cJ mice monitored by running wheels were shown to re-entrain to large 6- or 8-hour shifts of the light-dark cycle within 1-2 days, as opposed to the 5-7 days required for C57BL/6J mice. A single-day 6-hour advance of the LD cycle followed by release to constant darkness yielded similar phase shifts, demonstrating that exaggerated re-entrainment is not explained by masking of activity by the light-dark cycle. Responses in BALB/cJ mice were similar when monitored instead by motion detectors, indicating that wheel-running exercise does not influence the magnitude of responses. Neither brief (15 min) light exposure late during subjective nighttime nor 6-hour delays of the light-dark cycle produced exaggerated locomotor phase shifts, indicating that BALB/cJ mice do not merely experience enhanced sensitivity to light. Fos protein was expressed in cells of the SCN following acute light exposure at ZT10 of their previous light-dark cycle, a normally non-responsive time in the circadian cycle, but only in BALB/cJ (and not C57BL/6J) mice that had been subjected two days earlier to a single-day 6-hour advance of the light-dark cycle, indicating that their SCN had been advanced by that treatment. BALB/cJ mice may thus serve as a useful comparative model for studying molecular and physiological processes that limit responsiveness of circadian clocks to photic input.

Accelerated re-entrainment to advanced light cycles in BALB/cJ mice.

Los ritmos circadianos en mam&iacute;feros son coordinados por los n&uacute;cleos supraquiasm&aacute;ticos (SCN) del hipot&aacute;lamo, que sincronizan m&aacute;s potentemente a ciclos de luz-oscuridad ambientales. Grandes avances en el ciclo de luz-oscuridad t&iacute;picamente producen avances graduales en ritmos de actividad del orden de 1-2 horas diarias hasta re-arrastre debido a limitaciones en el sistema circadiano que todav&iacute;a no se entienden. En los seres humanos, este retraso hasta que se logra re-arrastre se vive como jet-lag, con el acompa&ntilde;amiento de s&iacute;ntomas de malestar, disminuy&oacute; el rendimiento cognitivo, problemas para dormir y dolor gastrointestinal. En estos experimentos, ritmos locomotores de ratones BALB/cJ monitoreados ejecutando las ruedas fueron demostradas para re-entrain a los cambios de hora grande de 6 o 8 del ciclo luz-oscuridad dentro de 1-2 d&iacute;as, en comparaci&oacute;n con los 5-7 d&iacute;as para ratones C57BL/6J. Un avance de 6 horas de un d&iacute;a del ciclo LD seguido por a la oscuridad constante rindi&oacute; defasajes similares, demostrando re-arrastre exagerada no se explica por el enmascaramiento de la actividad por el ciclo de luz / oscuridad. Respuestas en ratones BALB/cJ fueron similares cuando supervisado en su lugar por los detectores de movimiento, indicando que rueda duraci&oacute;n ejercicio no influye en la magnitud de las respuestas. Ni breve (15 min) exposici&oacute;n a la luz tarde durante la noche y subjetiva ni 6 horas retrasos del ciclo luz-oscuridad producci&oacute;n cambios de fase locomotor exagerada, indicando que ratones BALB/cJ no s&oacute;lo experimentan mayor sensibilidad a la luz. La prote&iacute;na Fos fue expresada en las c&eacute;lulas de la escena despu&eacute;s de la exposici&oacute;n aguda a la luz en ZT10 de su anterior ciclo luz-oscuridad, uno normalmente no responden en el ciclo circadiano, pero s&oacute;lo en BALB/cJ (y no C57BL/6J) ratones que hab&iacute;an sido sometidos dos d&iacute;as antes a un avance de 6 horas de un d&iacute;a del ciclo luz-oscuridad, indicando que su escena hab&iacute;a sido avanzada por que el tratamiento. Ratones BALB/cJ as&iacute; pueden servir como un modelo comparativo &uacute;til para estudiar los procesos fisiol&oacute;gicos y moleculares que limitan la capacidad de respuesta de los relojes circadianos a entrada luminosa.
