The use of mice to study models of human disease has resulted in a surge of interest in developing mouse MRI. The ability to take 3D, high-resolution images of live mice allows significant insight into anatomy and function. However, with imaging times on the order of hours, high throughput of specimens has been problematic. To facilitate high throughput, concurrent imaging of multiple mice has been developed; however, this poses further complexities regarding the ease and rapidity of loading several animals. In this study, custom-built equipment was developed to streamline the preparation process and to safely maintain seven mice during a multiple-mouse imaging session. Total preparation time for seven mice was approximately 24 min. ECG and temperature were monitored throughout the scan and maintained by regulating anesthetic and heating. Proof of principle was demonstrated in a 3-h imaging session of seven mice.

El uso de ratones para estudiar modelos de enfermedades humanas se ha traducido en un aumento del inter&#xE9;s en el desarrollo del rat&#xF3;n de resonancia magn&#xE9;tica. La capacidad de tomar en 3D, im&#xE1;genes de alta resoluci&#xF3;n de ratones vivos permite una informaci&#xF3;n valiosa sobre la anatom&#xED;a y funci&#xF3;n. Sin embargo, con tiempos de formaci&#xF3;n de im&#xE1;genes en el orden de horas, de alto rendimiento de las muestras ha sido problem&#xE1;tico. Cardiac MR imaging in small animals presents some difficulties due to shorter cardiac cycles and smaller dimensions than in human beings, but prospectively gated techniques have been successfully applied. As with human imaging, there may be certain applications in animal imaging for which retrospective gating is preferable to prospective gating. For example, cardiac imaging in multiple mice simultaneously is one such application. In this work we investigate the use of retrospective gating for cardiac imaging in a mouse. Using a three-dimensional imaging protocol, we show that image quality with retrospective gating is comparable to prospectively gated imaging. We conclude that retrospective gating is applicable for small animal cardiac MRI and show how it can be applied to the problem of cardiac MRI in multiple mice.

Kardiologische MR-Tomographie bei kleinen Tieren pr&auml;sentiert einige Schwierigkeiten aufgrund der k&uuml;rzeren kardiale Zyklen und kleinere Abmessungen als beim Menschen, aber prospektiv nicht&ouml;ffentliche Techniken erfolgreich angewendet wurden. Wie mit menschlichen Bildgebung, m&ouml;glicherweise gibt es bestimmte Anwendungen in der tierischen Bildgebung ist f&uuml;r welche retrospektiven gating prospektive Anspritzung vorzuziehen. La RM card&#xED;aca en animales peque&#xF1;os presenta algunas dificultades debido a la reducci&#xF3;n de los ciclos card&#xED;acos y dimensiones m&#xE1;s peque&#xF1;as que en los seres humanos, pero las t&#xE9;cnicas de prospectiva, barrios cerrados se han aplicado con &#xE9;xito. Como con la imagen humana, puede haber ciertas aplicaciones en las im&#xE1;genes de animales para los cuales sincronizaci&#xF3;n retrospectiva es preferible al de una puerta prospectivo. Por ejemplo, im&#xE1;genes card&#xED;acas en ratones de forma simult&#xE1;nea es una de esas aplicaciones. En este trabajo se investiga el uso de la sincronizaci&#xF3;n retrospectiva de im&#xE1;genes card&#xED;acas en un rat&#xF3;n. El uso de un protocolo de im&#xE1;genes en tres dimensiones, que muestran que la calidad de imagen con sincronizaci&#xF3;n retrospectiva es comparable a la imagen prospectiva cerrada. Se concluye que la sincronizaci&#xF3;n retrospectiva es aplicable para los peque&#xF1;os animales CRM y mostrar c&#xF3;mo se puede aplicar al problema de la resonancia magn&#xE9;tica cardiaca en ratones m&#xFA;ltiples.

With the introduction of mouse models for the study of cardiac morphogenesis, there arises a need for new imaging protocols that can capture both morphological and functional information. High-resolution 2D cardiac cine MRI has often been used to quantify left and right ventricular function. In this study we propose a 3D isotropic cardiac cine MRI protocol with a voxel size of 200 microm(3) as a means of studying cardiac multi-chamber morphology and function. A black blood sequence was used to enhance blood myocardium contrast. Manual segmentation of the ventricles was used to measure ventricular volumes at end diastole and end systole. This method is demonstrated on an Irx4-deficient mouse model. We have been able to identify the volumes of both ventricles dynamically and to show differences in ejection fraction in the mutant. We have also identified an abnormality of the papillary muscle in the mutant that had been missed in previous phenotyping with ultrasound and histology.

Haute r&#xE9;solution 2D cardiaque cin&#xE9; IRM a souvent &#xE9;t&#xE9; utilis&#xE9;e pour quantifier la fonction ventriculaire gauche et &#xE0; droite. Dans cette &#xE9;tude nous proposons une isotrope 3D cardiaque cin&#xE9; IRM protocole avec une taille de voxel de 200 microm (3) comme un moyen d&#x26;#39;&#xE9;tudier cardiaque multi-chambre de la morphologie et la fonction. Une s&#xE9;quence du sang noir a &#xE9;t&#xE9; utilis&#xE9; pour am&#xE9;liorer le sang myocarde contraste. Segmentation manuelle des ventricules a &#xE9;t&#xE9; utilis&#xE9; pour mesurer les volumes ventriculaires &#xE0; fin de la diastole et la systole fin. Cette m&#xE9;thode est d&#xE9;montr&#xE9;e sur un mod&#xE8;le de souris d&#xE9;ficientes en Irx4. Nous avons &#xE9;t&#xE9; en mesure d&#x26;#39;identifier les volumes des deux ventricules de mani&#xE8;re dynamique et de montrer les diff&#xE9;rences dans la fraction d&#x26;#39;&#xE9;jection dans le mutant. Nous avons &#xE9;galement identifi&#xE9; une anomalie du muscle papillaire chez le mutant qui avait &#xE9;t&#xE9; oubli&#xE9; dans le ph&#xE9;notypage pr&#xE9;c&#xE9;dente avec des ultrasons et de l&#x26;#39;histologie.

Mit der Einf&uuml;hrung der Mausmodelle f&uuml;r das Studium der kardialen Morphogenese entsteht es ein Bedarf f&uuml;r neue bildgebende Protokolle, die sowohl morphologische und funktionelle Informationen erfassen k&ouml;nnen. Hochaufl&ouml;sende 2D kardiale Cine MRI wurde h&auml;ufig verwendet, Links- und rechtsventrikul&auml;ren Funktion zu quantifizieren. In dieser Studie schlagen wir ein 3D isotrope kardiale Cine MRI-Protokoll mit einer Voxel-Gr&ouml;&szlig;e von 200 microm(3) als Mittel des Studiums kardiale Mehrkammer-Morphologie und Funktion. Eine Schwarzes Blut-Sequenz wurde verwendet, um Blut-Myokard-Kontrast verbessern. Manueller Segmentierung der Kammern wurde verwendet, um ventrikul&auml;re Volumen am Ende Diastole und Systole Ende messen. Diese Methode ist eine Irx4-defizienten Mausmodells unter Beweis gestellt. Ad alta risoluzione 2D cardiaca cine MRI &egrave; stato spesso usato per quantificare la funzione ventricolare destra e sinistra. In questo studio proponiamo un protocollo 3D isotropo cardiaca cine MRI con una dimensione del voxel di 200 microm(3) come mezzo per studiare la funzione e morfologia cardiaca multicamera. Una sequenza di sangue nero &egrave; stata utilizzata per migliorare il contrasto del miocardio sangue. Segmentazione manuale dei ventricoli &egrave; stata utilizzata per misurare i volumi ventricolari a fine diastole e sistole di fine. Questo metodo &egrave; dimostrato su un modello murino di Irx4-deficient. Siamo stati in grado di identificare in modo dinamico i volumi di entrambi i ventricoli e per mostrare le differenze nella frazione di eiezione in mutante. De alta resoluci&#xF3;n en 2D cine-RM card&#xED;aca a menudo se ha utilizado para cuantificar la funci&#xF3;n ventricular izquierda y la derecha. En este estudio se propone un isotr&#xF3;pica 3D cardiaca cine protocolo de estudio con un tama&#xF1;o de voxel de 200 micras (3) como un medio de estudiar cardiaca m&#xFA;ltiples c&#xE1;maras morfolog&#xED;a y funci&#xF3;n. Una secuencia de la sangre negro se utiliza para mejorar la sangre de contraste miocardio. Manual de segmentaci&#xF3;n de los ventr&#xED;culos se utiliz&#xF3; para medir los vol&#xFA;menes ventriculares en di&#xE1;stole y s&#xED;stole final. Este m&#xE9;todo se ha demostrado en un modelo de rat&#xF3;n deficiente en Irx4. Hemos podido identificar los vol&#xFA;menes de los dos ventr&#xED;culos y din&#xE1;micamente para mostrar diferencias en la fracci&#xF3;n de eyecci&#xF3;n en el mutante. We perform combined magnetic resonance and bioluminescence imaging of live mice for the purpose of improving the accuracy of bioluminescence tomography. The imaging is performed on three live nude mice in which tritium-powered light sources are surgically implanted. High-resolution magnetic resonance images and multispectral, multiview bioluminescence images are acquired in the same session. An anatomical model is constructed by segmenting the magnetic resonance images for all major tissues. The model is subsequently registered with nonlinear transformations to the 3-D light exittance (exiting intensity) surface map generated from the luminescence images. A Monte Carlo algorithm, along with a set of tissue optical properties obtained from in vivo measurements, is used to solve the forward problem. The measured and simulated light exittance images are found to differ by a factor of up to 2. The greatest cause of this moderate discrepancy is traced to the small errors in source positioning, and to a lesser extent to the optical properties used for the tissues. Discarding the anatomy and using a homogeneous model leads to a marginally worse agreement between the simulated and measured data.

Des images haute r&#xE9;solution par r&#xE9;sonance magn&#xE9;tique et multispectrales, des images de bioluminescence multivues sont acquises dans la m&#xEA;me session. Un mod&#xE8;le anatomique est construite en segmentant les images par r&#xE9;sonance magn&#xE9;tique pour tous les tissus majeurs. Le mod&#xE8;le est ensuite enregistr&#xE9; avec des transformations non lin&#xE9;aires &#xE0; la lumi&#xE8;re exittance 3-D (sortie intensit&#xE9;) carte de la surface g&#xE9;n&#xE9;r&#xE9;e &#xE0; partir des images de luminescence. Un algorithme de Monte Carlo, avec un ensemble de propri&#xE9;t&#xE9;s optiques du tissu obtenues &#xE0; partir de mesures in vivo, est utilis&#xE9;e pour r&#xE9;soudre le probl&#xE8;me de l&#x26;#39;avant. Les images mesur&#xE9;es et simul&#xE9;es exittance l&#xE9;gers se trouvent &#xE0; diff&#xE9;rer par un facteur allant jusqu&#x26;#39;&#xE0; 2. La principale cause de cet &#xE9;cart est imputable &#xE0; mod&#xE9;r&#xE9;e des petites erreurs dans le positionnement de source, et dans une moindre mesure dans les propri&#xE9;t&#xE9;s optiques utilis&#xE9;s pour les tissus. Wir durchf&uuml;hren kombinierten Magnetresonanz und Biolumineszenz Bildgebung Leben M&auml;use zum Zwecke der Verbesserung der Genauigkeit der Biolumineszenz-Tomographie. Das Modell ist mit nicht-linearen Transformationen zur 3-d leichte Exittance (Beenden Intensit&auml;t) Oberfl&auml;che Karte aus die Lumineszenz-Bilder generiert anschlie&szlig;end registriert. Eine Monte-Carlo-Algorithmus, sowie eine Reihe von Gewebe, die optische Eigenschaften von in-vivo Messungen bezogen wird verwendet, um das forward Problem zu l&ouml;sen. Realizamos im&#xE1;genes combinan la resonancia magn&#xE9;tica y la bioluminiscencia de ratones vivos con el fin de mejorar la exactitud de la tomograf&#xED;a por bioluminiscencia. La imagen se realiza en tres ratones vivos desnudos en la que las fuentes de tritio potencia de luz se implantan quir&#xFA;rgicamente. Im&#xE1;genes de alta resoluci&#xF3;n y resonancia magn&#xE9;tica de im&#xE1;genes multiespectrales, bioluminiscencia multivisi&#xF3;n se adquieren en la misma sesi&#xF3;n. Un modelo anat&#xF3;mico se construye mediante la segmentaci&#xF3;n de las im&#xE1;genes de resonancia magn&#xE9;tica para todos los tejidos principales. El modelo est&#xE1; inscrita con posterioridad, con transformaciones no lineales a la exittance la luz en 3-D (salida de intensidad) mapa de la superficie generada a partir de las im&#xE1;genes de luminiscencia. Un algoritmo de Monte Carlo, junto con un conjunto de propiedades &#xF3;pticas de tejidos obtenidos a partir de mediciones in vivo, se utiliza para resolver el problema hacia adelante. Las im&#xE1;genes de luz medidos y simulados exittance se encontr&#xF3; que difieren por un factor de hasta 2. La principal causa de esta discrepancia moderada se remonta a los peque&#xF1;os errores en el posicionamiento de origen y, en menor medida a las propiedades &#xF3;pticas utilizadas para los tejidos. El descarte de la anatom&#xED;a y el uso de un modelo homog&#xE9;neo conduce a un acuerdo ligeramente peor entre los datos simulados y medidos.
