El crecimiento de la poblaci&#xF3;n de cero (ZPG) locus de Drosophila codifica una prote&#xED;na espec&#xED;fica de la l&#xED;nea germinal brecha de la uni&#xF3;n, Innexin 4, lo que se requiere para la supervivencia de la diferenciaci&#xF3;n de las c&#xE9;lulas germinales tempranos durante la gametog&#xE9;nesis en ambos sexos. Los animales con una mutaci&#xF3;n nula en ZPG son viables, pero est&#xE9;riles y tienen g&#xF3;nadas peque&#xF1;as. Proponemos que ZPG contienen hemicanales brecha ocasiones en la membrana plasm&#xE1;tica de la c&#xE9;lula germinal puede conectar con hemicanales hechos de otras isoformas innexin adyacentes en las c&#xE9;lulas som&#xE1;ticas. Brecha ocasiones la comunicaci&#xF3;n intercelular a trav&#xE9;s de estos canales puede mediar la aprobaci&#xF3;n de importantes mol&#xE9;culas peque&#xF1;as o se&#xF1;ales entre las c&#xE9;lulas som&#xE1;ticas y germinales de apoyo necesarios para la supervivencia y la diferenciaci&#xF3;n de las c&#xE9;lulas germinales tempranos en ambos sexos.

Germ cells require intimate associations and signals from the surrounding somatic cells throughout gametogenesis. The zero population growth (zpg) locus of Drosophila encodes a germline-specific gap junction protein, Innexin 4, that is required for survival of differentiating early germ cells during gametogenesis in both sexes. Animals with a null mutation in zpg are viable but sterile and have tiny gonads. Adult zpg-null gonads contain small numbers of early germ cells, resembling stem cells or early spermatogonia or oogonia, but lack later stages of germ cell differentiation. In the male, Zpg protein localizes to the surface of spermatogonia, primarily on the sides adjacent to the somatic cyst cells. In the female, Zpg protein localizes to germ cell surfaces, both those adjacent to surrounding somatic cells and those adjacent to other germ cells. We propose that Zpg-containing gap junctional hemichannels in the germ cell plasma membrane may connect with hemichannels made of other innexin isoforms on adjacent somatic cells. Gap junctional intercellular communication via these channels may mediate passage of crucial small molecules or signals between germline and somatic support cells required for survival and differentiation of early germ cells in both sexes.

In Drosophila, primordial germ cells (PGCs) are set aside from somatic cells and subsequently migrate through the embryo and associate with somatic gonadal cells to form the embryonic gonad. During larval stages, PGCs proliferate in the female gonad, and a subset of PGCs are selected at late larval stages to become germ line stem cells (GSCs), the source of continuous egg production throughout adulthood. However, the degree of similarity between PGCs and the self-renewing GSCs is unclear. Here we show that many of the genes that are required for GSC maintenance in adults are also required to prevent precocious differentiation of PGCs within the larval ovary. We show that following overexpression of the GSC-differentiation gene bag of marbles (bam), PGCs differentiate to form cysts without becoming GSCs. Furthermore, PGCs that are mutant for nanos (nos), pumilio (pum) or for signaling components of the decapentaplegic (dpp) pathway also differentiate. The similarity in the genes necessary for GSC maintenance and the repression of PGC differentiation suggest that PGCs and GSCs may be functionally equivalent and that the larval gonad functions as a "PGC niche".

In the fruit fly Drosophila melanogaster, both spermatogenesis and oogenesis rely on germ-line stem cells (GSCs). Intensive research has revealed many of the molecules and pathways that underlie GSC maintenance and differentiation in males and females. In this review, we discuss new studies that, some differences notwithstanding, highlight the similarities in the structural and molecular strategies used by the two sexes in GSC maintenance and differentiation. These include the tight control that somatic support cells exert on every aspect of GSC function and the similar molecular mechanisms for physical attachment, cell-cell signaling and gap-junction communication. Some common principles underlying GSC biology in the fly may be applied to stem cells in other organisms.

The ability of organs such as the liver or the lymphoid system to maintain their original size or regain it after injury is well documented. However, little is known about how these organs sense that equilibrium is breached, and how they cease changing when homeostasis is reached. Similarly, it remains unclear how, during normal development, different cell types within an organ coordinate their growth. Here we show that during gonad development in the fruitfly Drosophila melanogaster the proliferation of primordial germ cells (PGCs) and survival of the somatic intermingled cells (ICs) that contact them are coordinated by means of a feedback mechanism composed of a positive signal and a negative signal. PGCs express the EGF receptor (EGFR) ligand Spitz, which is required for IC survival. In turn, ICs inhibit PGC proliferation. Thus, homeostasis and coordination of growth between soma and germ line in the larval ovary is achieved by using a sensor of PGC numbers (EGFR-mediated survival of ICs) coupled to a correction mechanism inhibiting PGC proliferation. This feedback loop ensures that sufficient numbers of PGCs exist to fill all the stem-cell niches that form at the end of larval development. We propose that similar feedback mechanisms might be generally used for coordinated growth, regeneration and homeostasis.

The seventeenth EMBO Conference on the Molecular and Developmental Biology of Drosophila took place in Kolymbari, Crete, between 20 and 26 June 2010. The conference covered a broad range of topics and much progress was made by combining two or more fields of study. Such combinations included quantitative approaches to cell and developmental biology, dissecting interrelations of physiology and development and integrated genomic analysis.

Die Konferenz behandelt eine Vielzahl von Themen und viel Fortschritte wurden durch die Kombination von zwei oder mehr Feldern Studie. Solche Kombinationen enthalten quantitative Ans&auml;tze zur Zell- und Entwicklungsbiologie, sezieren Wechselbeziehungen der Physiologie, der Entwicklung und der integrierte genomische Analyse.

Las c&#xE9;lulas madre y sus nichos son unidades que act&#xFA;an en conjunto para lograr la homeostasis del organismo adulto. C&#xF3;mo se forman las unidades tales y si los precursores de c&#xE9;lulas madre y el nicho podr&#xED;a ser coordinado ya durante la organog&#xE9;nesis, no se conocen. En moscas de la fruta, las c&#xE9;lulas germinales primordiales (PGC), los precursores de las c&#xE9;lulas germinales de la l&#xED;nea madre (GSCS) y precursores som&#xE1;ticas nichos se desarrollan dentro del ovario larval. Juntos forman las 16-20 unidades SGC del ovario adulto. Se demuestra que los receptores de ecdisona est&#xE1;n obligados a coordinar el desarrollo de nicho y los precursores de la SGC. En tercer instar temprano, los receptores de ecdisona reprimir la diferenciaci&#xF3;n precoz de los dos nichos y PGCs. Stem cells and their niches constitute units that act cooperatively to achieve adult body homeostasis. How such units form and whether stem cell and niche precursors might be coordinated already during organogenesis are unknown. In fruit flies, primordial germ cells (PGCs), the precursors of germ line stem cells (GSCs), and somatic niche precursors develop within the larval ovary. Together they form the 16-20 GSC units of the adult ovary. We show that ecdysone receptors are required to coordinate the development of niche and GSC precursors. At early third instar, ecdysone receptors repress precocious differentiation of both niches and PGCs. Early repression is required for correct morphogenesis of the ovary and for protecting future GSCs from differentiation. At mid-third instar, ecdysone signaling is required for niche formation. Finally, and concurrent with the initiation of wandering behavior, ecdysone signaling initiates PGC differentiation by allowing the expression of the differentiation gene bag of marbles in PGCs that are not protected by the newly formed niches. All the ovarian functions of ecdysone receptors are mediated through early repression, and late activation, of the ecdysone target gene broad. These results show that, similar to mammals, a brain-gland-gonad axis controls the initiation of oogenesis in insects. They further exemplify how a physiological cue coordinates the formation of a stem cell unit within an organ: it is required for niche establishment and to ensure that precursor cells to adult stem cells remain undifferentiated until the niches can accommodate them. Similar principles might govern the formation of additional stem cell units during organogenesis.

Coordinated regulation of niche and stem cell precursors by hormonal signaling.

Les cellules souches et leurs niches constituent des unit&#xE9;s qui agissent de concert pour atteindre l&#x26;#39;hom&#xE9;ostasie du corps adulte. Comment une telle forme d&#x26;#39;unit&#xE9;s et si les pr&#xE9;curseurs des cellules souches et de niche pourrait &#xEA;tre coordonn&#xE9; d&#xE9;j&#xE0; durant l&#x26;#39;organogen&#xE8;se ne sont pas connus. Dans les mouches des fruits, des cellules germinales primordiales (PGC), les pr&#xE9;curseurs de cellules germinales souches en ligne (CSS) et des pr&#xE9;curseurs de niche somatiques se d&#xE9;veloppent dans l&#x26;#39;ovaire des larves. Ensemble, ils forment des unit&#xE9;s de la CGC 16-20 de l&#x26;#39;ovaire adulte. Enfin, et parall&#xE8;lement &#xE0; l&#x26;#39;initiation de l&#x26;#39;errance comportement, ecdysone initie la diff&#xE9;renciation de signalisation PGC en permettant l&#x26;#39;expression de g&#xE8;ne de diff&#xE9;renciation du sac de billes dans PGC qui ne sont pas prot&#xE9;g&#xE9;s par les niches nouvellement form&#xE9;es. Toutes les fonctions ovariennes de r&#xE9;cepteurs ecdysone sont m&#xE9;di&#xE9;s par la r&#xE9;pression au d&#xE9;but, et l&#x26;#39;activation tardive, du g&#xE8;ne cible ecdysone large. Stammzellen und ihre Nischen bilden Einheiten, die kooperativ handeln, um K&ouml;rper-Hom&ouml;ostase zu erreichen. Fruchtfliegen, primordiale Keimzellen (PGCs), die Vorl&auml;ufer der Keim Linie Stammzellen (GSCs) und somatische Nische Vorstufen zu entwickeln, innerhalb der larvalen Fruchtknoten. Zusammen bilden sie die 16-20-GSC-Einheiten der Erwachsenen Eierstock. Wir zeigen, dass Ecdyson Rezeptoren sind verpflichtet, die Entwicklung der Nische und GSC Vorstufen zu koordinieren. Bei fr&uuml;hen dritten Instar unterdr&uuml;cken Ecdyson Rezeptoren fr&uuml;hreifen Differenzierung der Nischen und PGCs. Fr&uuml;he Repression ist f&uuml;r richtige Morphogenese des Ovars und zum Schutz der zuk&uuml;nftigen GSCs vor Differenzierung erforderlich. Bei 260 &ndash; 245 Instar ist das Ecdyson Signalisierung f&uuml;r Nische Bildung erforderlich. Schlie&szlig;lich, und gleichzeitig mit der Einleitung der Wanderschaft Verhalten, Ecdyson Signalisierung PGC Differenzierung dadurch, dass des Ausdrucks der Differenzierung gen Tasche mit Murmeln in PGCs, die nicht durch die neu gebildeten Nischen gesch&uuml;tzt sind initiiert. Cellule staminali e loro nicchie costituiscono unit&agrave; che agiscono in modo cooperativo per realizzare l&#39;omeostasi del corpo adulto. Insieme formano l&#39;unit&agrave; di GSC 16-20 dell&#39;ovaio adulto. Mostriamo che ecdisone recettori sono tenuti a coordinare lo sviluppo di nicchia e i precursori del GSC. All&#39;inizio terzo instar, recettori ecdisone reprimono precoce differenziazione di nicchie e di PGC. Repressione precoce &egrave; necessaria per la corretta morfogenesi dell&#39;ovaio e per proteggere il futuro artigiano dalla differenziazione.
