Virus adeno-asociados (AAV) utilizan una variedad de receptores celulares o correceptores para poder entrar en las c&#xE9;lulas. Adeno-associated viruses (AAVs) use a variety of cellular receptors/coreceptors to gain entry into cells. A number of AAV serotypes are now available, and the cognate receptors/coreceptors for only a handful of those have been identified thus far. Of the 10 commonly used AAV serotypes, AAV3 is by far the least efficient in transducing cells in general. However, in our more recent studies, we observed that AAV3 vectors transduced human liver cancer cells remarkably well, which led to the hypothesis that AAV3 uses hepatocyte growth factor receptor (HGFR) as a cellular coreceptor for viral entry. AAV3 infection of human liver cancer cell lines was strongly inhibited by hepatocyte growth factor, HGFR-specific small interfering RNA, and anti-HGFR antibody, which corroborated this hypothesis. Glucocorticoids (GCs) are widely used to treat inflammatory diseases. However, they cause debilitating side effects, which limit the use of these compounds. In the past decade, many researchers have attempted to find so-called dissociated GCs that have separate distinct transactivation and transrepression activities. Anti-inflammation of GCs is a result of glucocorticoid receptor (GR)-mediated transactivation and transrepression in some tissues, similar to their side effects; therefore, the goal to discover a compound that has anti-inflammatory properties, but lacks the negative side effects seen with GCs, has yet to be achieved. In the present study, we introduce a plant-derived compound, ginsenoside Rg1, which possesses GC and estrogen-like activities. In this study, we show that Rg1 downmodulates LPS-induced proinflammatory cytokine release and inhibits NF-ÎºB nuclear translocation and DNA binding activity. The negative effects on NF-ÎºB activation are due to a decrease in IÎºB phosphorylation and protein stabilization. Furthermore, the inhibitory effect of Rg1 on NF-ÎºB is GR-dependent, as small interfering RNA knockdown of GR abrogated this function. Rg1 also displayed profound inhibitory effects on LPS-induced MAPK activation. Importantly, Rg1 did not impair proliferation or differentiation of mouse osteoblasts. Finally, we show that Rg1 can effectively inhibit acute and chronic inflammation in vivo, but it does not cause hyperglycemia or osteoporosis as seen with dexamethasone. These results suggest that ginsenoside Rg1 may serve as a novel anti-inflammatory agent and may exhibit a potential profile for therapeutic intervention in inflammatory diseases.

Ginsenoside Rg1, &#xC2DD;&#xBB3C; &#xAE30;&#xC6D0;&#xC758; &#xC18C;&#xC124; glucocorticoid &#xC218;&#xC6A9; &#xCCB4; &#xAE38; &#xD56D; &#xC81C; &#xAC10;&#xC18C; &#xBD80;&#xC791;&#xC6A9; glucocorticoid &#xD6A8;&#xB2A5;&#xC744; &#xC720;&#xC9C0;&#xD569;&#xB2C8;&#xB2E4;.

Glucocorticoides (GCs) son ampliamente utilizados para tratar enfermedades inflamatorias. Sin embargo, causan efectos secundarios debilitantes, que limitan el uso de estos compuestos. En la &uacute;ltima d&eacute;cada, muchos investigadores han intentado encontrar el supuesto GCs disociadas que tienen transactivaci&oacute;n distinta separada y Transrepresi&oacute;n actividades. Antiinflamatorio de GCs es un resultado del receptor de glucocorticoides (GR)-mediada de transactivaci&oacute;n y Transrepresi&oacute;n en algunos tejidos, similares a sus efectos secundarios; por lo tanto, el objetivo de descubrir un compuesto que tiene propiedades anti-inflamatorias, pero carece de los negativos efectos secundarios observados con GCs, todav&iacute;a tiene que lograrse. En el presente estudio, presentamos un origen vegetal compuesto, ginsenoside Rg1, que posee la GC y actividades similares al estr&oacute;geno. En este estudio demostramos que Rg1 downmodulates citoquinas proinflamatorias inducida por LPS liberar e inhibe la translocaci&oacute;n nuclear NF-&kappa;B y actividad obligatoria de ADN. Los efectos negativos en la activaci&oacute;n de NF-&kappa;B son debido a una disminuci&oacute;n en la fosforilaci&oacute;n de la I&kappa;B y la estabilizaci&oacute;n de la prote&iacute;na. Adem&aacute;s, el efecto inhibitorio de Rg1 sobre NF-&kappa;B es GR-dependiente, como ca&iacute;da de RNA interferente peque&ntilde;o de GR derog&oacute; esta funci&oacute;n. RG1 tambi&eacute;n muestra profundos efectos inhibitorios sobre activaci&oacute;n de MAPK inducida por LPS. Lo importante, Rg1 no afectar a proliferaci&oacute;n o diferenciaci&oacute;n de los osteoblastos de rat&oacute;n. Por &uacute;ltimo, os mostramos que Rg1 puede inhibir eficazmente la inflamaci&oacute;n aguda y cr&oacute;nica en vivo, pero no causa hiperglucemia o osteoporosis con dexametasona. Estos resultados sugieren ginsenoside Rg1 puede servir como un nuevo agente antiinflamatorio y puede presentar un perfil de potencial para la intervenci&oacute;n terap&eacute;utica en las enfermedades inflamatorias.

Ginsenoside Rg1, un agonista del receptor de glucocorticoides novela de origen vegetal, mantiene la eficacia de glucocorticoide con reducidos efectos secundarios.
