Plant epidermal cells dedicate more than half of their lipid metabolism to the synthesis of cuticular lipids, which seal and protect the plant shoot. The cuticle is made up of a cutin polymer and waxes, diverse hydrophobic compounds including very-long-chain fatty acids and their derivatives. How such hydrophobic compounds are exported to the cuticle, especially through the hydrophilic plant cell wall, is not known. By performing a reverse genetic screen, we have identified LTPG, a glycosylphosphatidylinositol-anchored lipid transfer protein that is highly expressed in the epidermis during cuticle biosynthesis in Arabidopsis thaliana inflorescence stems. Mutant plant lines with decreased LTPG expression had reduced wax load on the stem surface, showing that LTPG is involved either directly or indirectly in cuticular lipid deposition. In vitro 2-p-toluidinonaphthalene-6-sulfonate assays showed that recombinant LTPG has the capacity to bind to this lipid probe. LTPG was primarily localized to the plasma membrane on all faces of stem epidermal cells in the growing regions of inflorescence stems where wax is actively secreted. These data suggest that LTPG may function as a component of the cuticular lipid export machinery.

Lign&#xE9;es de plantes mutantes dont l&#x26;#39;expression LTPG diminu&#xE9; avait r&#xE9;duit la charge de la cire sur la surface de la tige, montrant que LTPG est impliqu&#xE9; directement ou indirectement dans le d&#xE9;p&#xF4;t de lipides cuticulaires. Wie derartige hydrophoben Substanzen in die Kutikula, besonders durch die Zellwand hydrophile Pflanze exportiert werden, ist nicht bekannt. Wenn Sie einen umgekehrten genetischen Bildschirm ausf&uuml;hren, haben wir LTPG, ein Glykosylphosphatidyl verankerte Lipid-Transferprotein identifiziert, die in der Epidermis w&auml;hrend Kutikula-Biosynthese in Arabidopsis Thaliana Bl&uuml;tenstand Stengel hoch zum Ausdruck kommt. Eseguendo una schermata genetica inversa, abbiamo identificato LTPG, una proteina di trasferimento di glicosilfosfatidilinositolo ancorata lipido &egrave; altamente espresso nell&#39;epidermide durante la biosintesi di cuticola in Arabidopsis thaliana gambi infiorescenza. Linee di piante mutanti con diminuita espressione di LTPG avevano ridotto carico di cera sulla superficie del gambo, mostrando che LTPG &egrave; coinvolto direttamente o indirettamente nella deposizione di lipidi cuticolari. 2-P-toluidinonaphthalene-6-solfonato di saggi in vitro hanno dimostrato che LTPG ricombinante ha la capacit&agrave; di associare a questa sonda di lipidi. Acyl lipids in Arabidopsis and all other plants have a myriad of diverse functions. These include providing the core diffusion barrier of the membranes that separates cells and subcellular organelles. This function alone involves more than 10 membrane lipid classes, including the phospholipids, galactolipids, and sphingolipids, and within each class the variations in acyl chain composition expand the number of structures to several hundred possible molecular species. Acyl lipids in the form of triacylglycerol account for 35% of the weight of Arabidopsis seeds and represent their major form of carbon and energy storage. A layer of cutin and cuticular waxes that restricts the loss of water and provides protection from invasions by pathogens and other stresses covers the entire aerial surface of Arabidopsis. Similar functions are provided by suberin and its associated waxes that are localized in roots, seed coats, and abscission zones and are produced in response to wounding. This chapter focuses on the metabolic pathways that are associated with the biosynthesis and degradation of the acyl lipids mentioned above.
