Recientemente se han descrito varios m&eacute;todos para la activaci&oacute;n de neurodegeneraci&oacute;n apopt&oacute;tica extensa en el cerebro en desarrollo de mam&iacute;fero in vivo. Estos m&eacute;todos incluyen el tratamiento con f&aacute;rmacos que bloquean los receptores de glutamato NMDA, f&aacute;rmacos que promueven la neurotransmisi&oacute;n GABA(A), o tratamiento con etanol, que tiene propiedades GABAmimetic y antagonista NMDA. Un episodio &uacute;nico de intoxicaci&oacute;n inducido por cualquiera de estos agentes es suficiente para causar la neurodegeneraci&oacute;n generalizada a lo largo de muchas regiones del cerebro. Recentemente sono stati descritti diversi metodi per innescare neurodegenerazione apoptotica vasto nel cervello dei mammiferi in vivo in via di sviluppo. Questi metodi includono il trattamento con farmaci che bloccano i recettori NMDA glutammato, farmaci che promuovono la neurotrasmissione GABA(A), o il trattamento con etanolo, che ha sia antagonista NMDA e propriet&agrave; di GABAmimetic. Un episodio singolo intossicazione indotto da uno qualsiasi di questi agenti &egrave; sufficiente a causare la neurodegenerazione diffusa in molte regioni del cervello. Il processo di morte cellulare trapela rapidamente dalle fasi iniziali di ritardo entro alcune ore. Come i neuroni muoiono, diventano positivi TUNEL e spettacolo, da luce e da microscopia elettronica, tutte le caratteristiche morfologiche classiche del apoptosis. Nel presente studio, utilizzando metodi di immunocitochimica, documentiamo che intossicazione di etanolo di 7 giorni di et&agrave; infantili topi provoca un modello diffuso dell&#39;attivazione di caspase-3 corrispondente al modello di neurodegenerazione apoptotica che si verifica simultaneamente.

Recently several methods have been described for triggering extensive apoptotic neurodegeneration in the developing in vivo mammalian brain. These methods include treatment with drugs that block NMDA glutamate receptors, drugs that promote GABA(A) neurotransmission, or treatment with ethanol, which has both NMDA antagonist and GABAmimetic properties. A single intoxication episode induced by any of these agents is sufficient to cause widespread neurodegeneration throughout many brain regions. The cell death process transpires rapidly from early to late stages within several hours. As the neurons die, they become TUNEL positive and show, by both light and electron microscopy, all of the classical morphological characteristics of apoptosis. In the present study, using immunocytochemical methods, we document that ethanol intoxication of 7-day-old infant mice causes a widespread pattern of caspase-3 activation corresponding to the pattern of apoptotic neurodegeneration that is occurring simultaneously.

R&eacute;cemment, plusieurs m&eacute;thodes ont &eacute;t&eacute; d&eacute;crites pour d&eacute;clencher l&#39;apoptose vaste neurod&eacute;g&eacute;n&eacute;rescence dans le d&eacute;veloppement du cerveau chez les mammif&egrave;res in vivo. Nearly all vertebrates possess an olfactory organ but the vomeronasal organ is a synapomorphy for tetrapods. Nevertheless, it has been lost in several groups of tetrapods, including aquatic and marine animals. The present study examines the development of the olfactory and vomeronasal organs in two terrestrial anurans that exhibit different developmental modes. This study compares the development of the olfactory and vomeronasal organs in metamorphic anurans that exhibit an aquatic larva (Bufo americanus) and directly developing anurans that have eliminated the tadpole (Eleutherodactylus coqui). The olfactory epithelium in larval B. americanus is divided into dorsal and ventral branches in the rostral and mid-nasal regions. The larval olfactory pattern in E. coqui has been eliminated. Ontogeny of the olfactory system in E. coqui embryos starts to vary substantially from the larval pattern around the time of operculum development, the temporal period when the larval stage is hypothesized to have been eliminated. The nasal anatomy of the two frogs does not appear morphologically similar until the late stages of embryogenesis in E. coqui and the terminal portion of metamorphosis in B. americanus. Both species and their respective developing offspring, aquatic tadpoles and terrestrial egg/embryos, possess a vomeronasal organ. The vomeronasal organ develops at mid-embryogenesis in E. coqui and during the middle of the larval period in B. americanus, which is relatively late for neobatrachians. Development of the vomeronasal organ in both frogs is linked to the developmental pattern of the olfactory system. This study supports the hypothesis that the most recent common ancestor of tetrapods possessed a vomeronasal organ and was aquatic, and that the vomeronasal organ was retained in the Amphibia, but lost in some other groups of tetrapods, including aquatic and marine animals.

Casi todos los vertebrados poseen un &oacute;rgano olfativo, pero el &oacute;rgano vomeronasal es una Sinapomorfia de los tetr&aacute;podos. Sin embargo, se ha perdido en varios grupos de tetr&aacute;podos, incluidos los animales acu&aacute;ticos y marinos. El presente estudio examina el desarrollo de los &oacute;rganos olfativos y vomeronasal en dos anuros terrestres que presentan diferentes modos de desarrollo. Este estudio compara el desarrollo de los &oacute;rganos olfativos y vomeronasal en anuros metam&oacute;rficos que exhiben una larva acu&aacute;tica (Bufo americanus) y anuros directamente en desarrollo que han eliminado el renacuajo (Eleutherodactylus coqui). El epitelio olfativo en larvas B. americanus se divide en ramas dorsales y ventrales en las regiones rostrales y mid-nasal. El patr&oacute;n olfativo larval en E. coqui ha sido eliminado. Ontogenia del sistema olfativo en E. coqui embriones comienza a var&iacute;an notablemente con el patr&oacute;n de larvario en la &eacute;poca del desarrollo del op&eacute;rculo, el per&iacute;odo temporal cuando el estado larvario se presume haber sido eliminada. La anatom&iacute;a nasal de las dos ranas no aparece morfol&oacute;gicamente similar hasta las &uacute;ltimas etapas de la embriog&eacute;nesis en E. coqui y la porci&oacute;n terminal de la metamorfosis en B. americanus. Ambas especies y sus respectivas cr&iacute;as en desarrollo, renacuajos acu&aacute;ticos y terrestres huevos o embriones, poseen un &oacute;rgano vomeronasal. El &oacute;rgano vomeronasal se convierte en mid-embryogenesis en E. coqui y a mediados del per&iacute;odo larvario en B. americanus, que es relativamente tarde para Ranoides. Desarrollo del &oacute;rgano vomeronasal en ambas ranas est&aacute; relacionada con el patr&oacute;n del desarrollo del sistema olfativo. Este estudio apoya la hip&oacute;tesis de que el ancestro com&uacute;n m&aacute;s reciente de tetr&aacute;podos posey&oacute; un &oacute;rgano vomeronasal y acu&aacute;tico, y que el &oacute;rgano vomeronasal fue retenido en los anfibios, pero perdi&oacute; en algunos otros grupos de tetr&aacute;podos, incluidos los animales acu&aacute;ticos y marinos.

Embryonic development of the central serotonergic neurons in the directly developing frog, Eleutherodactylus coqui, was determined by using immunocytochemistry. The majority of anuran amphibians (frogs) possess a larval stage (tadpole) that undergoes metamorphosis, a dramatic post-embryonic event, whereby the tadpole transforms into the adult phenotype. Directly developing frogs have evolved a derived life-history mode where the tadpole stage has been deleted and embryos develop directly into the adult bauplan. Embryonic development in E. coqui is classified into 15 stages (TS 1-15; 1 = oviposition/15 = hatching). Serotonergic immunoreactivity was initially detected at TS 6 in the raphe nuclei in the developing rhombencephalon. At TS 7, immunopositive perikarya were observed in the paraventricular organ in the hypothalamus and reticular nuclei in the hindbrain. Development of the serotonergic system was steady and gradual during mid-embryogenesis. However, starting at TS 13 there was a substantial increase in the number of serotonergic neurons in the paraventricular, raphe, and reticular nuclei, a large increase in the number of varicose fibers, and a differentiation of the reticular nuclei in the hindbrain. Consequentially, E. coqui displayed a well-developed central serotonergic system prior to hatching (TS 15). In comparison, the serotonergic system in metamorphic frogs typically starts to develop earlier but the surge of development that transpires in this system occurs post-embryonically, during metamorphosis, and not in the latter stages of embryogenesis, as it does in E. coqui. Overall, the serotonergic development in E. coqui is similar to the other vertebrates.

Die Mehrheit der Crocodiles Amphibien (Fr&ouml;sche) besitzen ein Larvenstadium (Kaulquappe), die Metamorphose, eine dramatische post-embryonic Ereignis erf&auml;hrt, wobei die Kaulquappe in der Erwachsenen-Ph&auml;notyp umwandelt. E. Coqui angezeigt konsequent, gut entwickelten zentrales serotonergen System vor dem Schl&uuml;pfen (TS 15). La mayor&iacute;a de los anfibios anuros (ranas) posee una etapa larval (renacuajos) que sufre metamorfosis, un suceso dram&aacute;tico posterior, por el que el renacuajo se transforma en el fenotipo adulto. Directamente ranas en desarrollo han evolucionado de un modo historia derivadas de la vida donde se ha eliminado la etapa de renacuajo y embriones convierten directamente en el bauplan adultos. Desarrollo embrionario en E. coqui se clasifica en 15 etapas (TS 1-15; 1 = oviposici&oacute;n/15 = eclosi&oacute;n). Serotonin&eacute;rgico inmunoreactividad fue detectado inicialmente en TS 6 en los n&uacute;cleos del rafe en el rombenc&eacute;falo en desarrollo. At the time of Golgi and Cajal's reception of the Nobel Prize in 1906 most scientists had accepted the notion that neurons are independent units. Although neuroscientists today still believe that neurons are independent anatomical units, functionally, it is thought that some sort of population coding occurs. Throughout this essay, we provide evidence that suggests that populations of neurons can code information through the synchronization of their responses. This synchronization occurs at several levels in the brain. Whereas spike synchrony refers to the correlation between spikes of different neurons' spike trains, oscillatory synchrony refers to the synchronization of oscillatory responses, generally among large groups of neurons. In the first section of this essay we describe the dependence of the brain's developmental processes on synchronous firing and how these processes form a brain that supports and is sensitive to synchronous spikes. Data are then presented that suggest that spike and oscillatory synchrony may serve as useful neural codes. Examples from sensory (auditory, olfactory and somatosensory), motor and higher cognitive (attention, memory) systems are then presented to illustrate potential roles for these synchronous codes in normal brain function. Results from these studies collectively suggest that spike synchrony in sensory and motor systems may provide detail information not available from changes in firing rate. Oscillatory synchrony, on the other hand, may be globally involved in the coordination of long-distance neuronal communication during higher cognitive processes. These concepts represent a dramatic shift in direction since the times of Golgi and Cajal.

Several studies have shown that neurons with similar response properties are arranged together in domains across primary visual cortex (V1). An orderly pattern of domains has been described for preferences to ocular dominance, orientation, and spatial frequency. Temporal frequency preference, another important attribute of the visual scene, also might be expected to map into different domains. Using optical imaging and a variety of quantitative methods, we examined how temporal frequency selectivity is mapped in V1 of the prosimian primate, bush baby (Otolemur garnetti). We found that unlike other attribute maps, selectivity for different temporal frequencies is arranged uniformly across V1 with no evidence of local clustering. Global tuning for temporal frequency, based on magnitude of response, showed a good match to previous tuning curves for single neurons. A peak response was found around 2.0 Hz, with smaller attenuation at lower temporal frequencies than at higher frequencies. We also examined whether the peak temporal frequency response differed between anatomical compartments defined by cytochrome oxidase (CO). No significant differences in the preference for temporal frequency were found between these CO compartments. Our findings show that key sensory attributes that are linked in perception can be organized in quite distinct ways in V1 of primates.

Mehrere Studien haben gezeigt, dass Neuronen mit &auml;hnlichen Reaktion Eigenschaften zusammen Dom&auml;nen &uuml;ber prim&auml;re Sehrinde (V1) angeordnet. Eine geordnete Muster der Domains wurde f&uuml;r die Pr&auml;ferenzen und okul&auml;re Dominanz, Orientierung und r&auml;umliche Frequenz beschrieben. Zeitlichen Frequenz bevorzugt, wird ein weiteres wichtiges Attribut der visuellen Szene, k&ouml;nnte auch rechnen, ordnen Sie in verschiedenen Dom&auml;nen. Mit optischen Bildgebung und eine Vielzahl von quantitativen Methoden, wir untersucht, wie zeitliche Frequenz-Selektivit&auml;t ist in V1 die Halbaffen Galagos, Bush Baby (Otolemur Garnetti) zugeordnet. Wir fanden, dass im Gegensatz zu anderen Karten Attribut Selektivit&auml;t f&uuml;r unterschiedliche zeitliche Frequenzen gleichm&auml;&szlig;ig &uuml;ber V1 ohne Beweise des lokalen clustering angeordnet ist. Globale tuning f&uuml;r zeitliche Frequenz, basierend auf der Gr&ouml;&szlig;e der Antwort, zeigte eine gute Partie zu fr&uuml;heren tuning Kurven f&uuml;r einzelne Neuronen. Eine Peak-ansprechen wurde rund 2,0 Hz, mit kleiner D&auml;mpfung bei niedrigeren zeitliche Frequenzen als bei h&ouml;heren Frequenzen gefunden. Wir pr&uuml;fen auch, ob unterschieden sich der zeitlichen H&ouml;hepunkt-Frequenzgang zwischen anatomische F&auml;cher durch Cytochrom-Oxidase (CO) definiert. Zwischen diese CO-F&auml;cher wurden keine signifikanten Unterschiede in der Pr&auml;ferenz f&uuml;r zeitliche Frequenz gefunden. Unsere Ergebnisse zeigen, dass sensorische Eigenschaften, die in der Wahrnehmung verkn&uuml;pft sind auf ganz unterschiedliche Weise in V1 von Primaten organisiert werden k&ouml;nnen.

Diversi studi hanno dimostrato che i neuroni con propriet&agrave; simili di risposta sono organizzati insieme in domini attraverso la corteccia visiva primaria (V1). Un modello ordinato dei domini &egrave; stato descritto per le preferenze di dominanza oculare, orientamento e frequenza spaziale. Preferenza frequenza temporale, un altro attributo importante della scena visiva, potrebbe essere previsto anche per mappare in domini diversi. Utilizzando optical che Imaging e una variet&agrave; di metodi quantitativi, abbiamo esaminato come temporale selettivit&agrave; di frequenza &egrave; mappato in V1 il primate prosimian, bush baby (Otolemur garnetti). Abbiamo trovato che a differenza di altre mappe di attributo, selettivit&agrave; per diverse frequenze temporali &egrave; organizzato in modo uniforme attraverso V1 con nessuna evidenza di clustering locale. Ottimizzazione globale per frequenza temporale, basato sulla grandezza della risposta, ha mostrato una buona corrispondenza al precedente curve tuning per singoli neuroni. Una risposta di picco &egrave; stata trovata circa 2.0 Hz, con minore attenuazione a bassa frequenza temporale rispetto alle frequenze pi&ugrave; alte. Abbiamo anche esaminato se la risposta in frequenza temporale picco differiva tra compartimenti anatomici definiti dal citocromo ossidasi (CO). Tra questi compartimenti CO sono state trovate differenze significative nella preferenza per frequenza temporale. I nostri risultati mostrano che chiavi attributi sensoriali che sono collegati nella percezione possono essere organizzati in modi distinti V1 dei primati.

Coincident spikes have been implicated in vision-related processes such as feature binding, gain modulation, and long-distance communication. The source of these spike-time correlations is unknown. Although several studies have proposed that cortical spikes are correlated based on stimulus structure, others have suggested that spike-time correlations reflect ongoing cortical activity present even in the absence of a coherent visual stimulus. To examine this issue, we collected single-unit recordings from primary visual cortex (V1) of the anesthetized and paralyzed prosimian bush baby using a 100-electrode array. Spike-time correlations for pairs of cells were compared under three conditions: a moving grating at the cells' preferred orientation, an equiluminant blank screen, and a dark condition with eyes covered. The amplitudes, lags, and widths of cross-correlation histograms (CCHs) were strongly correlated between these conditions although for the blank stimulus and dark condition, the CCHs were broader with peaks lower in amplitude. In both preferred stimulus and blank conditions, the CCH amplitudes were greater when the cells within the pair had overlapping receptive fields and preferred similar orientations rather than nonoverlapping receptive fields and different orientations. These data suggest that spike-time correlations present in evoked activity are generated by mechanisms common to those operating in spontaneous conditions.

Recentemente, la misura di T (2)-intensit&agrave; di segnale ponderata (T2SI) &egrave; stato indicato per essere utile nell&#39;identificazione di neoplasie cerebellare bassa qualit&agrave;. L&#39;obiettivo di questo studio era di valutare se i risultati oggettivi su queste sequenze MRI attendibilmente correlato con l&#39;esame istopatologico sottostante.

Risultati sulla risonanza magnetica cerebrale pre-operatoria predicono istopatologia in bambini con neoplasie cerebellare.

La mayor&iacute;a de pacientes con Neoplasias Cerebelosas Puerto astrocitomas piloc&iacute;ticos (PAs), meduloblastomas y ependimomas. Conocimiento de una probabilidad preoperatoria de histopatolog&iacute;a en este grupo de pacientes tiene el potencial para influir en muchos aspectos de la atenci&oacute;n. Estudios anteriores han demostrado hiperintensidad en difusi&oacute;n-cargado proyecci&oacute;n de imagen para correlacionar con meduloblastomas. Recientemente, medici&oacute;n de T (2)-intensidad de la se&ntilde;al ponderada (T2SI) ha demostrado ser &uacute;til en la identificaci&oacute;n de calidad inferior Neoplasias Cerebelosas. El objetivo de este estudio fue evaluar si los resultados objetivos en estas secuencias de MRI confiablemente correlacionan con la histopatolog&iacute;a subyacente.
