Stress hydrique est une affection fr&eacute;quente d&#39;environnementale n&eacute;faste que s&eacute;rieusement affecte recadrer la productivit&eacute; dans le monde entier. En raison de la complexit&eacute; de la s&eacute;cheresse comme un signal de stress, d&eacute;crypter les m&eacute;canismes de tol&eacute;rance de la s&eacute;cheresse est rest&eacute; un enjeu majeur pour planter des biologistes. Pour d&eacute;velopper de nouvelles approches pour &eacute;tudier la tol&eacute;rance &agrave; la s&eacute;cheresse v&eacute;g&eacute;tale, nous cherch&eacute; des ph&eacute;notypes conf&eacute;r&eacute;s par le stress hydrique et identifi&eacute; l&#39;inhibition de la croissance des racines lat&eacute;rales par le stress de s&eacute;cheresse comme une r&eacute;ponse adaptative au stress. Cette r&eacute;ponse &agrave; la s&eacute;cheresse est en partie m&eacute;di&eacute;e par l&#39;acide abscissique phytohormone. &Eacute;crans g&eacute;n&eacute;tiques en utilisant Arabidopsis (Arabidopsis thaliana) ont &eacute;t&eacute; con&ccedil;us, et inhibition de la s&eacute;cheresse des mutants de croissance (dig) racine lat&eacute;rale avec r&eacute;ponses alt&eacute;r&eacute;es &agrave; la s&eacute;cheresse ou l&#39;acide abscissique dans le d&eacute;veloppement des racines lat&eacute;rales ont &eacute;t&eacute; isol&eacute;s. Creuser de caract&eacute;risation de ces mutants a r&eacute;v&eacute;l&eacute; qu&#39;ils pr&eacute;sentent &eacute;galement alt&eacute;r&eacute; la tol&eacute;rance au stress s&eacute;cheresse, indiquant que cette r&eacute;ponse de racine au stress hydrique est intimement li&eacute;e &agrave; l&#39;adaptation de la s&eacute;cheresse de l&#39;ensemble de la plante et peut &ecirc;tre utilis&eacute; comme un trait d&#39;acc&eacute;der le m&eacute;canisme de tol&eacute;rance s&eacute;cheresse insaisissable. &#x7531;&#x6839;&#x5E94;&#x5BF9;&#x5E72;&#x65F1;&#x80C1;&#x8FEB;&#x548C;&#x8131;&#x843D;&#x9178;&#x7684;&#x9057;&#x4F20;&#x5206;&#x6790;&#x5E72;&#x65F1;&#x5BB9;&#x5FCD;&#x51B3;&#x5B9A;&#x56E0;&#x7D20;&#x7684;&#x6807;&#x8BC6;&#x3002;

Drought stress is a common adverse environmental condition that seriously affects crop productivity worldwide. Due to the complexity of drought as a stress signal, deciphering drought tolerance mechanisms has remained a major challenge to plant biologists. To develop new approaches to study plant drought tolerance, we searched for phenotypes conferred by drought stress and identified the inhibition of lateral root development by drought stress as an adaptive response to the stress. This drought response is partly mediated by the phytohormone abscisic acid. Genetic screens using Arabidopsis (Arabidopsis thaliana) were devised, and drought inhibition of lateral root growth (dig) mutants with altered responses to drought or abscisic acid in lateral root development were isolated. Characterization of these dig mutants revealed that they also exhibit altered drought stress tolerance, indicating that this root response to drought stress is intimately linked to drought adaptation of the entire plant and can be used as a trait to access the elusive drought tolerance machinery. Our study also revealed that multiple mechanisms coexist and together contribute to whole-plant drought tolerance.

Trockenstress ist ein gel&auml;ufiger nachteiligen &ouml;kologischen Zustand, dass ernsthaft beeintr&auml;chtigt Produktivit&auml;t weltweit zuschneiden. Aufgrund der Komplexit&auml;t der D&uuml;rre als Spannung Signal ist die Entschl&uuml;sselung D&uuml;rre Toleranz Mechanismen eine wichtige Herausforderung f&uuml;r Biologen Pflanzen geblieben. Um neue Ans&auml;tze zur Pflanze Trockenheitstoleranz studieren zu entwickeln, wir suchten Ph&auml;notypen, die durch Trockenstress &uuml;bertragen und die Hemmung der seitliche Wurzel Entwicklung von Trockenstress identifiziert, wie eine adaptive Reaktion auf Stress. Diese D&uuml;rre-Antwort ist teilweise durch die Phytohormone Abscisic S&auml;ure vermittelt. Genetische Schirme mit Arabidopsis (Arabidopsis Thaliana) wurden entwickelt und D&uuml;rre Hemmung der seitlichen Stamm Wachstum (Dig) Mutanten mit ver&auml;nderten Antworten auf Trockenheit oder Abscisic S&auml;ure in seitliche Wurzel Entwicklung wurden isoliert. Charakterisierung dieser Graben Mutants offenbart, dass sie auch zeigen ver&auml;ndert D&uuml;rre Stresstoleranz, kombinierbar und einsetzbar als Merkmal die schwer fassbaren D&uuml;rre Toleranz Maschine Zugriff auf darauf hinweist, dass diese Stamm-Antwort an Trockenstress D&uuml;rre Anpassung des gesamten eng verkn&uuml;pft ist. Unsere Studie zeigte auch, dass mehrere Mechanismen koexistieren und zusammen zur ganzen Pflanze Trockenheitstoleranz beitragen.

Siccit&agrave; lo stress &egrave; una condizione ambientale avversa comune che seriamente interessa Ritaglia produttivit&agrave; in tutto il mondo. A causa della complessit&agrave; della siccit&agrave; come un segnale di stress, decifrando i meccanismi di tolleranza alla siccit&agrave; &egrave; rimasta una grande sfida per piantare i biologi. Per sviluppare nuovi approcci per lo studio di tolleranza alla siccit&agrave; pianta, abbiamo cercato fenotipi conferiti da stress siccit&agrave; e identificato l&#39;inibizione dello sviluppo di radici laterali da stress siccit&agrave; come una risposta adattativa allo stress. Questa risposta di siccit&agrave; &egrave; parzialmente mediata dall&#39;acido abscissico phytohormone. Schermi genetici usando Arabidopsis (Arabidopsis thaliana) sono stati elaborati, e inibizione della siccit&agrave; di mutanti crescita (dig) radice laterale con alterata risposta alla siccit&agrave; o acido abscissico nello sviluppo di radici laterali sono stati isolati. Caratterizzazione di questi scavare mutanti ha rivelati che esibiscono anche alterata tolleranza allo stress siccit&agrave;, indicando che questa risposta radice a stress siccit&agrave; intimamente legata all&#39;adattamento alla siccit&agrave; dell&#39;intera pianta e pu&ograve; essere utilizzato come un tratto per accedere il macchinario di tolleranza di siccit&agrave; sfuggente. Il nostro studio ha anche rivelato che pi&ugrave; meccanismi coesistono e insieme contribuiscono alla tolleranza alla siccit&agrave; tutta la pianta.

Sequ&iacute;a es una condici&oacute;n ambiental adversa com&uacute;n que seriamente afecta cultivos productividad en todo el mundo. Debido a la complejidad de la sequ&iacute;a como una se&ntilde;al de tensi&oacute;n, descifrar los mecanismos de tolerancia de sequ&iacute;a sigue siendo un desaf&iacute;o importante para los bi&oacute;logos de la planta. Para desarrollar nuevos enfoques para estudiar la planta tolerancia a la sequ&iacute;a, se ha buscado fenotipos conferidos por la sequ&iacute;a y se identificaron la inhibici&oacute;n del desarrollo de ra&iacute;ces laterales por sequ&iacute;a como una respuesta adaptativa al estr&eacute;s. Esta respuesta de sequ&iacute;a est&aacute; mediada en parte por el &aacute;cido absc&iacute;sico de fitohormonas. Pantallas gen&eacute;ticas con Arabidopsis (Arabidopsis thaliana) fueron ideadas e inhibici&oacute;n de sequ&iacute;a de mutantes de crecimiento (dig) de ra&iacute;ces laterales con respuestas alteradas a la sequ&iacute;a o el &aacute;cido absc&iacute;sico en desarrollo de ra&iacute;ces laterales fueron aislados. Caracterizaci&oacute;n de estos cavar mutantes revel&oacute; que tambi&eacute;n exhiben alterado tolerancia al estr&eacute;s sequ&iacute;a, indicando que esta respuesta de la ra&iacute;z a la sequ&iacute;a est&aacute; &iacute;ntimamente ligada a la adaptaci&oacute;n de la sequ&iacute;a de toda la planta y puede utilizarse como un rasgo para acceder a los mecanismos de tolerancia elusiva de la sequ&iacute;a. Nuestro estudio tambi&eacute;n revel&oacute; que varios mecanismos coexisten y juntos contribuyan a la tolerancia a la sequ&iacute;a de toda la planta.

Prote&iacute;nas de transferencia de l&iacute;pidos en las plantas se creen que son involucrados en muchos procesos de desarrollo y fisiolog&iacute;a de la c&eacute;lula. En este trabajo, un largometraje cDNA codificar una prote&iacute;na de transferencia de l&iacute;pidos novela, BcLTP designado fue aislado de Brassica chinensis. Al menos dos copias de BcLTP est&aacute;n presente en el genoma de B. chinensis, y sus transcripciones se acumulan preferentemente en &oacute;rganos de segundo a&ntilde;o, dando a entender su papel en la etapa de crecimiento reproductivo. La secuencia de 118 amino&aacute;cido deduce de un marco de lectura abierto de bp 354 (ORF) acciones caracter&iacute;sticas comunes con otros miembros de la familia de plantas LTPs. Un p&eacute;ptido se&ntilde;al putativos en N terminal fue probado para la funci&oacute;n de secreci&oacute;n por el sistema de trampa (YSST) de secuencia de se&ntilde;al de levadura y confirmado por localizaci&oacute;n vesicular y extracelular de la prote&iacute;na de fusi&oacute;n YFP. Se encontr&oacute; un sitio de uni&oacute;n de CaM altamente conservado en la terminal C y las propiedades de enlace con dos isoformas representantes de CaM, uno es AtCaM2 convergente, uno es SCaM5 divergentes, se determinaron por el recubrimiento de gel. Lipid transfer proteins in plants are believed to be involved in many processes of cell physiology and development. In this work, a full-length cDNA encoding a novel lipid transfer protein, designated BcLTP was isolated from Brassica chinensis. At least two copies of BcLTP are present in whole genome of B. chinensis, and its transcripts preferably accumulate in second-year organs, implying its role in reproductive growth stage. The 118 amino acid sequence deduced from a 354 bp open reading frame (ORF) shares common features with other members of plants LTPs family. A putative signal peptide at the N terminus was tested for secretion function by the yeast signal sequence trap (YSST) system, and further confirmed by vesicular and extracellular localization of YFP fusion protein. A highly conserved CaM binding site at C terminus was found and the binding properties with two representative CaM isoforms, one is convergent AtCaM2, one is divergent SCaM5, were determined by gel overlay. We found that convergent AtCaM2 prefer high concentration of Ca(2+) for binding BcLTP, while SCaM5 does not depend on Ca(2+ )concentration too much for binding BcLTP. The lipid binding feature of BcLTP was demonstrated using florescence-marked 1-pyrenedodecanoic acid, which can be enhanced by AtCaM2 in Ca(2+ )dependent manner and by SCaM5 in either presence or absence of Ca(2+). The collected data suggest that BcLTP may secrete and combine extracellular CaM isoforms, which in turn, facilitate lipid binding of BcLTP via Ca(2+) mediated signaling.

Plant recognition of pathogens leads to rapid activation of MPK3 and MPK6, two Arabidopsis mitogen-activated protein kinases (MAPKs), and their orthologs in other species. Here, we report that synthesis of camalexin, the major phytoalexin in Arabidopsis, is regulated by the MPK3/MPK6 cascade. Activation of MPK3/MPK6 by expression of active upstream MAPK kinase (MAPKK) or MAPKK kinase (MAPKKK) was sufficient to induce camalexin synthesis in the absence of pathogen attack. Induction of camalexin by Botrytis cinerea was preceded by MPK3/MPK6 activation, and compromised in mpk3 and mpk6 mutants. Genetic analysis placed the MPK3/MPK6 cascade upstream of PHYTOALEXIN DEFICIENT 2 (PAD2) and PAD3, but independent or downstream of PAD1 and PAD4. Camalexin induction after MPK3/MPK6 activation was preceded by rapid and coordinated up-regulation of multiple genes encoding enzymes in the tryptophan (Trp) biosynthetic pathway, in the conversion of Trp to indole-3-acetaldoxime (IAOx, a branch point between primary and secondary metabolism), and in the camalexin biosynthetic pathway downstream of IAOx. These results indicate that the MPK3/MPK6 cascade regulates camalexin synthesis through transcriptional regulation of the biosynthetic genes after pathogen infection.

Plants challenged by pathogens, especially necrotrophic fungi such as Botrytis cinerea, produce high levels of ethylene. At present, the signaling pathways underlying the induction of ethylene after pathogen infection are largely unknown. MPK6, an Arabidopsis stress-responsive mitogen-activated protein kinase (MAPK) was previously shown to regulate the stability of ACS2 and ACS6, two type I ACS isozymes (1-amino-cyclopropane-1-carboxylic acid synthase). Phosphorylation of ACS2 and ACS6 by MPK6 prevents rapid degradation of ACS2/ACS6 by the 26S proteasome pathway, resulting in an increase in cellular ACS activity and ethylene biosynthesis. Here, we show that MPK3, which shares high homology and common upstream MAPK kinases with MPK6, is also capable of phosphorylating ACS2 and ACS6. In the mpk3 mutant background, ethylene production in gain-of-function GVG-NtMEK2(DD) transgenic plants was compromised, suggesting that MPK6 and MPK3 function together to stabilize ACS2 and ACS6. Using a liquid-cultured seedling system, we found that B.Â cinerea-induced ethylene biosynthesis was greatly compromised in mpk3/mpk6 double mutant seedlings. In contrast, ethylene production decreased only slightly in the mpk6 single mutant and not at all in the mpk3 single mutant, demonstrating overlapping roles for these two highly homologous MAPKs in pathogen-induced ethylene induction. Consistent with the role of MPK3/MPK6 in the process, mutation of ACS2 and ACS6, two genes encoding downstream substrates of MPK3/MPK6, also reduced B.Â cinerea-induced ethylene production. The residual levels of ethylene induction in the acs2/acs6 double mutant suggest the involvement of additional ACS isoforms, possibly regulated by MAPK-independent pathway(s).

Mitogen-activated protein kinase 3 and 6 regulate Botrytis cinerea-induced ethylene production in Arabidopsis.

En revanche, la production d&#39;&eacute;thyl&egrave;ne ne diminu&eacute; que l&eacute;g&egrave;rement dans le mutant simple mpk6 et pas du tout dans le mutant simple mpk3, d&eacute;montrant des r&ocirc;les qui se chevauchent pour ces deux MAPKs hautement homologues dans l&#39;induction de l&#39;&eacute;thyl&egrave;ne induite par l&#39;agent pathog&egrave;ne. Compatible avec le r&ocirc;le de MPK3/MPK6 dans le processus, la mutation de ACS2 et ACS6, deux g&egrave;nes codant pour les substrats en aval de MPK3/MPK6, a &eacute;galement r&eacute;duit la production de b. cinerea induite par l&#39;&eacute;thyl&egrave;ne. Pflanzen, die durch Krankheitserreger, vor allem Necrotrophic Pilzen wie Botrytis Cinerea, herausgefordert produzieren hohe Konzentrationen von Ethylen. Derzeit sind die Signalwege zugrunde liegen die Induktion von Ethylen nach Erreger Infektion weitgehend unbekannt. MPK6, ein Arabidopsis Spannung-responsive Mitogen-aktivierte Proteinkinase (MAPK) war zuvor gezeigt, dass die Stabilit&auml;t der ACS2 und ACS6 zu regulieren, zwei Typ I ACS Isoenzyme (1-amino-Cyclopropan-1-carboxylic Acid Synthase). Phosphorylierung von ACS2 und ACS6 durch MPK6 verhindert rasche Verschlechterung der ACS2/ACS6 von der 26S Proteasom Weg, was zu einer Zunahme der zellul&auml;ren ACS-Aktivit&auml;t und Ethylen-Biosynthese. Hier zeigen wir, dass MPK3, das hohe Homologie und gemeinsame upstream MAPK-Kinasen mit MPK6 wie auch PHOSPHORYLIEREND ACS2 und ACS6 kann. Im Hintergrund mutant mpk3 wurde Ethylen-Produktion in Gain of Function GVG-NtMEK2(DD) transgener Pflanzen kompromittiert, was darauf hindeutet, dass MPK6 und MPK3-Funktion zusammen, um ACS2 und ACS6 zu stabilisieren. Mit einer Fl&uuml;ssigkeit-kultivierte S&auml;mling-System fanden wir, dass B. Cinerea-induzierte Ethylen-Biosynthese in mpk3/mpk6 doppelt mutant S&auml;mlinge stark beeintr&auml;chtigt wurde. Ethylen-Produktion fiel hingegen nur leicht, in den einzelnen mpk6-Mutanten und &uuml;berhaupt nicht in den einzelnen mpk3-Mutanten, demonstrieren Sie &uuml;berlappende Rollen f&uuml;r diese zwei hochgradig homologe MAPKs in Ethylen Pathogen-induzierten Induktion. In &Uuml;bereinstimmung mit der Rolle der MPK3/MPK6 im Prozess, Mutation des ACS2 und ACS6, zwei Gene Kodierung nachgeschaltete Substrate der MPK3/MPK6, auch reduziert-B. Cinerea-induzierte Ethylen-Produktion. Die verbleibenden Ebenen der Ethylen-Induktion in der doppelten acs2/acs6-Mutant empfehlen die Einbeziehung der zus&auml;tzliche ACS-Isoformen, m&ouml;glicherweise durch MAPK-unabh&auml;ngige Pathway(s) geregelt.

Mitogen-aktivierte Proteinkinase 3 und 6 Regeln Botrytis Cinerea-induzierte Ethylen-Produktion in Arabidopsis.

Piante sfidati da agenti patogeni, soprattutto il parassita funghi come la Botrytis cinerea, producono alti livelli di etilene. Attualmente, le vie di segnalazione sottostante l&#39;induzione di etilene dopo l&#39;infezione del patogeno sono in gran parte sconosciute. MPK6, una Arabidopsis stress-responsive mitogene-attivato proteina chinasi (MAPK) precedentemente &egrave; stata indicata per regolare la stabilit&agrave; di ACS2 e ACS6, due di tipo isoenzimi ACS (1-ammino-ciclopropano-1-carbossilico acido sintasi). Fosforilazione di ACS2 e ACS6 da MPK6 impedisce la rapida degradazione del ACS2/ACS6 dal pathway del proteasoma 26S, risultanti in un aumento della biosintesi di attivit&agrave; e di etilene ACS cellulare. Mostriamo qui, che MPK3, che condivide elevata omologia e comuni a Monte chinasi MAPK con MPK6, &egrave; anche capace di fosforilante ACS2 e ACS6. I livelli residui di induzione di etilene in acs2/acs6 doppio mutante suggeriscono il coinvolgimento di ulteriori isoforme di ACS, eventualmente regolata da vie biochimiche MAPK-indipendente.

Desafiado por pat&oacute;genos, especialmente hongos necr&oacute;trofos como Botrytis cinerea, las plantas producen altos niveles de etileno. En la actualidad, las v&iacute;as de se&ntilde;alizaci&oacute;n que subyace a la inducci&oacute;n de etileno despu&eacute;s de la infecci&oacute;n del pat&oacute;geno son en gran parte desconocidas. MPK6, un Arabidopsis sensible al estr&eacute;s-PROTEINA QUINASA mit&oacute;geno activada (MAPK) fue demostrado previamente para regular la estabilidad de ACS2 y ACS6, dos isoenzimas de la ACS (1-amino-ciclopropano-1-carboxylic acid synthase) de tipo I. Fosforilaci&oacute;n de ACS2 y ACS6 por MPK6 evita la r&aacute;pida degradaci&oacute;n de ACS2/ACS6 por la v&iacute;a del proteasoma 26S, resultando en un incremento en la bios&iacute;ntesis celular de actividad y etileno de ACS. A continuaci&oacute;n, os mostramos que MPK3, que comparte la elevada homolog&iacute;a y com&uacute;n quinasas MAPK por aguas arriba con MPK6, tambi&eacute;n es capaz de fosforilar ACS2 y ACS6. En el fondo mutante mpk3, producci&oacute;n de etileno en las plantas transg&eacute;nicas de ganancia de funci&oacute;n GVG-NtMEK2(DD) fue comprometida, sugiriendo esa funci&oacute;n MPK6 y MPK3 juntos para estabilizar ACS2 y ACS6. Usando un sistema de cultivo l&iacute;quido de la pl&aacute;ntula, encontramos que eso bios&iacute;ntesis de etileno B. cinerea-inducida fue enormemente comprometida en pl&aacute;ntulas de mutantes dobles mpk3/mpk6. En contraste, la producci&oacute;n de etileno disminuy&oacute; s&oacute;lo ligeramente en el mutante solo mpk6 y en absoluto en el mutante solo mpk3, demostrando funciones superpuestas para estas dos MAPKs altamente hom&oacute;logas en inducci&oacute;n de etileno inducida por el pat&oacute;geno. En concordancia con el papel de MPK3/MPK6 en el proceso, la mutaci&oacute;n de ACS2 y ACS6, dos genes que codifican sustratos aguas abajo de MPK3/MPK6, tambi&eacute;n reduce producci&oacute;n de etileno B. cinerea-inducida. Los niveles residuales de inducci&oacute;n de etileno en el acs2/acs6 doble mutante sugieren la implicaci&oacute;n de isoformas de ACS adicional, posiblemente regulado por MAPK independiente fiosol&oacute;gicos.

Sulfurtransferases (STRs) catalyze the transfer of a sulfur atom from a donor to a suitable acceptor molecule. The Arabidopsis thaliana genome encodes 20 putative STR proteins. The biological functions of most are unclear. We found that STR1 and STR2 play important roles in embryo/seed development. Mutation of STR1 alone resulted in a shrunken seed phenotype, although growth and development of vegetative and reproductive organs were not affected. The shrunken seed phenotype was associated with the delayed/arrested embryo development, in most cases, at the heart stage. The embryo defect of str1 mutant is not fully penetrant. Approximately 12.5% of embryos developed further and formed normal looking seeds. In severely shrunken seeds, no embryo could be identified after seed collection. Partially shrunken seeds that contained viable embryos could still germinate. However, cotyledons of the seedlings from such seeds were abnormal. An STR1-GUS fusion reporter revealed that the STR1 gene was universally expressed, with high levels of expression in specific tissues/organs including embryos. The incomplete penetrance of str1 embryo/seed phenotype is a result of functional STR2. Single str2 mutant had no phenotype. However, no str1(-/-)/str2(-/-) double mutant embryos were able to develop past the heart stage. Furthermore, STR2 is haplo-insufficient in str1 mutant background, and str1(-/-)/str2(+/-) embryos were 100% lethal. These data provide new insights into the biological functions of the ubiquitous sulfurtransferase in Arabidopsis embryogenesis and seed development.

Nous avons trouv&eacute; que la STR1 et STR2 jouent un r&ocirc;le important dans le d&eacute;veloppement de l&#39;embryon/graine. Mutation de STR1 seul a entra&icirc;n&eacute; un ph&eacute;notype de graines ratatin&eacute;es, bien que la croissance et le d&eacute;veloppement des organes v&eacute;g&eacute;tatifs et reproducteurs ne sont pas affect&eacute;es. Le ph&eacute;notype de graines ratatin&eacute;es &eacute;tait associ&eacute; avec le d&eacute;veloppement de l&#39;embryon retard&eacute;/arr&ecirc;t&eacute;, dans la plupart des cas, au stade cordiforme. Le d&eacute;faut d&#39;embryon de str1 mutant n&#39;est pas enti&egrave;rement par ressuage. Graines partiellement ratatin&eacute;es qui contenaient des embryons viables pourraient encore germer. Toutefois, les cotyl&eacute;dons des plantules de ces graines &eacute;taient anormaux. Un journaliste de fusion STR1-GUS a r&eacute;v&eacute;l&eacute; que le g&egrave;ne STR1 &eacute;tait universellement exprim&eacute;, avec des niveaux &eacute;lev&eacute;s d&#39;expression dans les tissus/organes sp&eacute;cifiques, y compris les embryons. La p&eacute;n&eacute;trance incompl&egrave;te du ph&eacute;notype embryon/graine de str1 est le r&eacute;sultat de STR2 fonctionnelle. Str2 seul mutant n&#39;avait aucun ph&eacute;notype. Wir fanden, dass STR1 und STR2 eine wichtige Rolle in der Entwicklung des Embryos/Seed spielen. Mutation von STR1 allein f&uuml;hrte zu einem geschrumpften Saatgut-Ph&auml;notyp, obwohl Wachstum und Entwicklung der vegetativen und reproduktiven Organe nicht betroffen waren. Der geschrumpfte Saatgut-Ph&auml;notyp war die Entwicklung verz&ouml;gert/verhaftet Embryos in den meisten F&auml;llen im Herzen Stadium zugeordnet. Der Embryo-Mangel von str1 Mutante ist nicht vollst&auml;ndig Eindringpr&uuml;fung. Ca. 12,5 % der Embryonen weiterentwickelt und normale aussehende Samen gebildet. In stark geschrumpften Samen konnte kein Embryo nach Saatgut Collection identifiziert werden. Teilweise geschrumpfte Samen, die lebensf&auml;hige Embryonen enthalten k&ouml;nnte noch Keimen. Allerdings waren die Keimbl&auml;tter der S&auml;mlinge aus dem Saatgut abnorme. Ein STR1: GUS-Fusion-Reporter ergab, dass das Gen STR1 universell ausgedr&uuml;ckt wurde, mit einem hohen Grad des Ausdrucks in bestimmte Gewebe/Organe, einschlie&szlig;lich Embryonen. Der unvollst&auml;ndige Penetranz str1 Embryo/Seed Ph&auml;notyp resultiert aus funktionalen STR2. Einzelne str2 Mutant hatte keine Ph&auml;notyp. Keine str1(-/-)/str2(-/-)-Doppel-mutant-Embryonen konnten sich jedoch &uuml;ber die Herz-Stadium zu entwickeln. Dar&uuml;ber hinaus STR2 ist Haplo-unzureichende in str1 mutant Hintergrund und str1(-/-)/str2(+/-) Embryonen wurden 100 % t&ouml;dlich. Diese Daten erlauben neue Einblicke in die biologischen Funktionen von der allgegenw&auml;rtigen schwefelhaltige in Arabidopsis der Embryogenese und Saatgut.

Sulfurtransferases (STRs) catalizzano il trasferimento di un atomo di zolfo da un donatore ad un molecola accettore idoneo. Il genoma di Arabidopsis thaliana codifica putative STR 20 proteine. Le funzioni biologiche della maggior parte non sono chiare. Abbiamo trovato che STR1 e STR2 giocano un ruolo importante nello sviluppo dell&#39;embrione/seme. Mutazione di STR1 da solo ha portato un fenotipo seme rattrappito, anche se la crescita e lo sviluppo degli organi vegetativi e riproduttivi non sono stati colpiti. Il fenotipo di seme rattrappito &egrave; stato associato con lo sviluppo di embrione ritardata/arrestato, nella maggior parte dei casi, nella fase di cuore. Il difetto di embrione di str1 mutante non &egrave; completamente penetrante. Circa il 12,5% degli embrioni ulteriormente sviluppato e formato normale semi cercando. In semi gravemente rattrappiti, nessun embrione potrebbe essere identificato dopo la raccolta del seme. Potrebbero ancora germinare i semi parzialmente rattrappiti che contenevano embrioni vitali. Tuttavia, i cotiledoni dei semenzali da tali semi erano anormali. Un reporter di fusione STR1-GUS ha rivelato che il gene STR1 universalmente &egrave; stata espressa, con alti livelli di espressione in tessuti/organi specifici, tra cui gli embrioni. La penetranza incompleta del fenotipo dell&#39;embrione/seme str1 &egrave; un risultato di STR2 funzionale. Str2 singolo mutante non aveva nessun fenotipo. Sulfurtransferases (STRs) catalizan a la transferencia de un &aacute;tomo de azufre de un donante a una mol&eacute;cula de aceptor adecuado. El genoma de Arabidopsis thaliana codifica 20 prote&iacute;nas putativas de STR. Las funciones biol&oacute;gicas de la mayor&iacute;a no son claros. Encontramos que cadena1 y cadena2 desempe&ntilde;an papeles importantes en el desarrollo del embri&oacute;n/semilla. Mutaci&oacute;n de STR1 solo dio lugar a un fenotipo semilla encogida, aunque el crecimiento y desarrollo de &oacute;rganos vegetativos y reproductivos no fueron afectados. Plant sensing of invading pathogens triggers massive metabolic reprogramming, including the induction of secondary antimicrobial compounds known as phytoalexins. We recently reported that MPK3 and MPK6, two pathogen-responsive mitogen-activated protein kinases, play essential roles in the induction of camalexin, the major phytoalexin in Arabidopsis thaliana. In search of the transcription factors downstream of MPK3/MPK6, we found that WRKY33 is required for MPK3/MPK6-induced camalexin biosynthesis. In wrky33 mutants, both gain-of-function MPK3/MPK6- and pathogen-induced camalexin production are compromised, which is associated with the loss of camalexin biosynthetic gene activation. WRKY33 is a pathogen-inducible transcription factor, whose expression is regulated by the MPK3/MPK6 cascade. Chromatin immunoprecipitation assays reveal that WRKY33 binds to its own promoter in vivo, suggesting a potential positive feedback regulatory loop. Furthermore, WRKY33 is a substrate of MPK3/MPK6. Mutation of MPK3/MPK6 phosphorylation sites in WRKY33 compromises its ability to complement the camalexin induction in the wrky33 mutant. Using a phospho-protein mobility shift assay, we demonstrate that WRKY33 is phosphorylated by MPK3/MPK6 in vivo in response to Botrytis cinerea infection. Based on these data, we conclude that WRKY33 functions downstream of MPK3/MPK6 in reprogramming the expression of camalexin biosynthetic genes, which drives the metabolic flow to camalexin production in Arabidopsis challenged by pathogens.

WRKY33 &egrave; un fattore di trascrizione inducibili patogeno, cui espressione &egrave; regolata dalla cascata della MPK3/MPK6. Nos inform&oacute; recientemente que MPK3 y MPK6, dos quinasas de prote&iacute;nas activadas por mit&oacute;genos sensible al pat&oacute;geno, un papel esencial en la inducci&oacute;n de camalexin, la fitoalexina principal en Arabidopsis thaliana.
