Substantial evidence indicates that the cyclo-oxygenase-2 (COX-2) inhibitor celecoxib, a widely prescribed anti-inflammatory agent, displays anti-tumour effect by sensitizing cancer cells to apoptosis. As part of our effort to understand the mechanism by which celecoxib mediates apoptosis in androgen-independent prostate cancer cells, we investigated its effect on intracellular calcium concentration ([Ca(2+)](i)). Digital ratiometric imaging analysis indicates that exposure of PC-3 cells to celecoxib stimulates an immediate [Ca(2+)](i) rise in a dose- and time-dependent manner. Kinetic data show that this Ca(2+) signal arises from internal Ca(2+) release in conjunction with external Ca(2+) influx. Examinations of the biochemical mechanism responsible for this Ca(2+) mobilization indicate that celecoxib blocks endoplasmic reticulum (ER) Ca(2+)-ATPases. Consequently, inhibition of this Ca(2+) reuptake mechanism results in Ca(2+) mobilization from ER stores followed by capacitative calcium entry, leading to [Ca(2+)](i) elevation. In view of the important role of Ca(2+) in apoptosis regulation, this Ca(2+) perturbation may represent part of the signalling mechanism that celecoxib uses to trigger rapid apoptotic death in cancer cells. This Ca(2+)-ATPase inhibitory activity is highly specific for celecoxib, and is not noted with other COX inhibitors tested, including aspirin, ibuprofen, naproxen, rofecoxib (Vioxx), DuP697 and NS398. Moreover, it is noteworthy that this activity is also observed in many other cell lines examined, including A7r5 smooth muscle cells, NIH 3T3 fibroblast cells and Jurkat T cells. Consequently, this Ca(2+)-perturbing effect may provide a plausible link with the reported toxicities of celecoxib such as increased cardiovascular risks in long-term anti-inflammatory therapy.

The genetic analysis of life span has revealed many interesting genes and pathways; however, our understanding of aging has been limited by the lack of a way to assay the aging process itself. Here we show that the tissues of aging worms have a characteristic appearance that is easy to recognize and quantify using Nomarski optics. We have used this assay to determine whether life-span mutations affect the rate of aging, to identify animals that age more rapidly than normal, and to infer the cause of death in C. elegans. Mutations that reduce insulin/IGF-1 signaling double the life span of C. elegans, and we find that tissue decline is slowed in these mutants. Thus this endocrine system appears to influence the rate at which tissues age. This effect extends even to the germline, which is the only mitotically active tissue in the adult. We find that Nomarski microscopy also allows a ready distinction between short-lived mutants that age more rapidly than normal and those that are simply sick, and we have identified an RNAi clone that confers a dramatic rapid-aging phenotype. This clone encodes the C. elegans heat-shock factor (HSF), a transcription factor that regulates the response to heat and oxidative stress. This suggests that heat-shock proteins, many of which act as chaperones, may function in normal animals to slow the rate of aging. Finally, we have identified a cause of death of C. elegans: namely, proliferating bacteria. This suggests that increased susceptibility to bacterial infections contributes to mortality in these animals, just as it does in humans.

To explore the role of mitochondrial activity in the aging process, we have lowered the activity of the electron transport chain and adenosine 5'-triphosphate (ATP) synthase with RNA interference (RNAi) in Caenorhabditis elegans. These perturbations reduced body size and behavioral rates and extended adult life-span. Restoring messenger RNA to near-normal levels during adulthood did not elevate ATP levels and did not correct any of these phenotypes. Conversely, inhibiting respiratory-chain components during adulthood only did not reset behavioral rates and did not affect life-span. Thus, the developing animal appears to contain a regulatory system that monitors mitochondrial activity early in life and, in response, establishes rates of respiration, behavior, and aging that persist during adulthood.

We find that HSF-1, like the transcription factor DAF-16, is required for daf-2-insulin/IGF-1 receptor mutations to extend life-span. Our findings suggest this is because HSF-1 and DAF-16 together activate expression of specific genes, including genes encoding small heat-shock proteins, which in turn promote longevity. The small heat-shock proteins also delay the onset of polyglutamine-expansion protein aggregation, suggesting that these proteins couple the normal aging process to this type of age-related disease.

Most of our knowledge about the regulation of aging comes from mutants originally isolated for other phenotypes. To ask whether our current view of aging has been affected by selection bias, and to deepen our understanding of known longevity pathways, we screened a genomic Caenorhabditis elegans RNAi library for clones that extend lifespan. We identified 23 new longevity genes affecting signal transduction, the stress response, gene expression, and metabolism and assigned these genes to specific longevity pathways. Our most important findings are (i) that dietary restriction extends C. elegans' lifespan by down-regulating expression of key genes, including a gene required for methylation of many macromolecules, (ii) that integrin signaling is likely to play a general, evolutionarily conserved role in lifespan regulation, and (iii) that specific lipophilic hormones may influence lifespan in a DAF-16/FOXO-dependent fashion. Surprisingly, of the new genes that have conserved sequence domains, only one could not be associated with a known longevity pathway. Thus, our current view of the genetics of aging has probably not been distorted substantially by selection bias.

La mayor parte de nuestros conocimientos sobre la regulaci&#xF3;n del envejecimiento proviene de mutantes originalmente aislados de otros fenotipos. Para preguntar si nuestro punto de vista actual sobre el envejecimiento se ha visto afectada por el sesgo de selecci&#xF3;n, y de profundizar nuestra comprensi&#xF3;n de las v&#xED;as de la longevidad conocidos, hemos examinado un gen&#xF3;mica Caenorhabditis elegans biblioteca de RNAi para los clones que se extienden la vida &#xFA;til. Se identificaron 23 genes de la longevidad afecta a nuevos transducci&#xF3;n de se&#xF1;ales, la respuesta al estr&#xE9;s, la expresi&#xF3;n g&#xE9;nica y el metabolismo y se asignan estos genes de la longevidad de las v&#xED;as espec&#xED;ficas. Nuestros hallazgos m&#xE1;s importantes son: (i) que la restricci&#xF3;n diet&#xE9;tica prolonga la vida &#xFA;til de C. elegans &#x26;#39;por abajo de la regulaci&#xF3;n de la expresi&#xF3;n de genes clave, incluyendo un gen necesario para la metilaci&#xF3;n de las macromol&#xE9;culas muchos, (ii) que la se&#xF1;alizaci&#xF3;n de integrina es probable que desempe&#xF1;e un general, evolutivamente funci&#xF3;n conservada en el tiempo de vida regulaci&#xF3;n, y (iii) que las hormonas lipof&#xED;licas espec&#xED;ficas podr&#xE1;n influir en la vida &#xFA;til de una manera DAF-16/FOXO-dependent. One challenge in aging research concerns identifying physiological parameters or biomarkers that can reflect the physical health of an animal and predict its lifespan. In C. elegans, a model organism widely used in aging research, motor deficits develop in old worms. Here we employed machine vision to quantify worm locomotion behavior throughout lifespan. We confirm that aging worms undergo a progressive decline in motor activity, beginning in early life. Importantly, the rate of motor activity decline rather than the absolute motor activity in the early-to-mid life of individual worms in an isogenic population inversely correlates with their lifespan, and thus may serve as a lifespan predictor. Long-lived mutant strains with deficits in insulin/IGF-1 signaling or food intake display a reduction in the rate of motor activity decline, suggesting that this parameter might also be used for across-strain comparison of healthspan. Our work identifies an endogenous physiological parameter for lifespan prediction and healthspan comparison.

Chez C. elegans, un organisme mod&#xE8;le largement utilis&#xE9; dans recherche sur le vieillissement, les d&#xE9;ficits moteurs se d&#xE9;velopper dans les vers anciens. Ici nous avons utilis&#xE9; la vision industrielle pour quantifier le comportement de locomotion ver tout au long de la dur&#xE9;e de vie. Nous confirmons que les vers de vieillissement subir un d&#xE9;clin progressif de l&#x26;#39;activit&#xE9; motrice, en commen&#xE7;ant t&#xF4;t dans la vie. Surtout, le taux de d&#xE9;clin de l&#x26;#39;activit&#xE9; motrice plut&#xF4;t que l&#x26;#39;activit&#xE9; motrice absolue dans le d&#xE9;but de la vie &#xE0; mi-parcours de chaque ver dans une population isog&#xE9;nique en corr&#xE9;lation inverse avec leur dur&#xE9;e de vie, et peut donc servir de pr&#xE9;dicteur dur&#xE9;e de vie. In Caenorhabditis Elegans, ein Modellorganismus, weit verbreitet in Altern-Forschung, entwickeln Motorische Defizite in alten W&uuml;rmer. Hier haben wir die industrielle Bildverarbeitung, Wurm Fortbewegung Verhalten in der gesamten Lebensdauer zu quantifizieren. Wir best&auml;tigen, dass Altern W&uuml;rmer eine progressive nachgibt in verst&auml;rkter motorischer Aktivit&auml;t, beginnend in den ersten Lebensjahren. Wichtig ist, die Rate der motorischen Aktivit&auml;t ablehnen, anstatt der absolute motorische Aktivit&auml;t im Leben Anfang bis Mitte der einzelne W&uuml;rmer in einer isogene Bev&ouml;lkerung umgekehrt korreliert mit ihrer Lebensdauer und kann somit als eine Lebensdauer-Predictor dienen. Langlebige mutierte St&auml;mme mit Defiziten in Insulin/IGF-1-Signalisierung oder Lebensmittel-Aufnahme zeigen eine Reduktion in der R&uuml;ckgang der motorischen Aktivit&auml;t, was darauf hindeutet, dass dieser Parameter auch ganz-Dehnungs-Vergleich von Healthspan verwendet werden k&ouml;nnte. Unsere Arbeit kennzeichnet einen endogenen physiologischen Parameter Lebensdauer Vorhersage und Healthspan Vergleich.

Uno de los retos del envejecimiento de investigaci&#xF3;n la identificaci&#xF3;n de las preocupaciones de par&#xE1;metros fisiol&#xF3;gicos o biomarcadores que pueden reflejar la salud f&#xED;sica de un animal y predecir su vida &#xFA;til. En C. elegans, un organismo modelo ampliamente utilizado en la investigaci&#xF3;n del envejecimiento, el d&#xE9;ficit motor se desarrollan en gusanos de edad. Aqu&#xED; se emplea la visi&#xF3;n artificial para cuantificar el comportamiento del gusano de locomoci&#xF3;n durante toda la vida. Confirmamos que el envejecimiento de los gusanos sometidos a una disminuci&#xF3;n progresiva de la actividad motora, a partir de la primera infancia. Es importante destacar que la tasa de disminuci&#xF3;n de la actividad motora en lugar de la actividad motora absoluta en la vida temprana a mediados de los gusanos individuales en una poblaci&#xF3;n isog&#xE9;nica inversamente correlacionada con su vida &#xFA;til, y por lo tanto puede servir como un predictor de vida &#xFA;til. Dietary restriction (DR) results in a robust increase in lifespan while maintaining the physiology of much younger animals in a wide range of species. Here, we examine the role of drr-2, a DR-responsive gene recently identified, in determining the longevity of Caenorhabditis elegans. Inhibition of drr-2 has been shown to increase longevity. However, the molecular mechanisms by which drr-2 influences longevity remain unknown. We report here that drr-2 encodes an ortholog of human eukaryotic translation initiation factor 4H (eIF4H), whose function is to mediate the initiation step of mRNA translation. The molecular function of DRR-2 is validated by the association of DRR-2 with polysomes and by the decreased rate of protein synthesis observed in drr-2 knockdown animals. Previous studies have also suggested that DR might trigger a regulated reduction in drr-2 expression to initiate its longevity response. By examining the effect of increasing drr-2 expression on DR animals, we find that drr-2 is essential for a large portion of the longevity response to DR. The nutrient-sensing target of rapamycin (TOR) pathway has been shown to mediate the longevity effects of DR in C. elegans. Results from our genetic analyses suggest that eIF4H/DRR-2 functions downstream of TOR, but in parallel to the S6K/PHA-4 pathway to mediate the lifespan effects of DR. Together, our findings reveal an important role for eIF4H/drr-2 in the TOR-mediated longevity responses to DR.

Ici, nous examinons le r&#xF4;le de la RRC-2, un g&#xE8;ne DR-sensible r&#xE9;cemment identifi&#xE9;, dans la d&#xE9;termination de la long&#xE9;vit&#xE9; de Caenorhabditis elegans. L&#x26;#39;inhibition de la RRC-2 a &#xE9;t&#xE9; montr&#xE9; pour augmenter la long&#xE9;vit&#xE9;. Des &#xE9;tudes ant&#xE9;rieures ont &#xE9;galement sugg&#xE9;r&#xE9; que la RD pourrait d&#xE9;clencher une r&#xE9;duction r&#xE9;glement&#xE9; en mati&#xE8;re de RRC-2 expression de lancer sa r&#xE9;ponse long&#xE9;vit&#xE9;. Hier untersuchen wir die Rolle der Drr-2, ein DR-responsive gen vor kurzem festgestellt, bei der Bestimmung der Langlebigkeit der Fadenwurm Caenorhabditis Elegans. Aqu&#xED;, examinamos el papel de la RRD-2, un gen de respuesta del DR-recientemente identificado, en la determinaci&#xF3;n de la longevidad de Caenorhabditis elegans. One goal of aging research is to develop interventions that combat age-related illnesses and slow aging. Although numerous mutations have been shown to achieve this in various model organisms, only a handful of chemicals have been identified to slow aging. Here, we report that celecoxib, a nonsteroidal anti-inflammatory drug widely used to treat pain and inflammation, extends Caenorhabditis elegans lifespan and delays the age-associated physiological changes, such as motor activity decline. Celecoxib also delays the progression of age-related proteotoxicity as well as tumor growth in C. elegans. Celecoxib was originally developed as a potent cyclooxygenase-2 (COX-2) inhibitor. However, the result from a structural-activity analysis demonstrated that the antiaging effect of celecoxib might be independent of its COX-2 inhibitory activity, as analogs of celecoxib that lack COX-2 inhibitory activity produce a similar effect on lifespan. Furthermore, we found that celecoxib acts directly on 3'-phosphoinositide-dependent kinase-1, a component of the insulin/IGF-1 signaling cascade to increase lifespan.

Le c&#xE9;l&#xE9;coxib a &#xE9;t&#xE9; initialement d&#xE9;velopp&#xE9;e comme un inhibiteur puissant de la cyclooxyg&#xE9;nase-2 (COX-2). Toutefois, le r&#xE9;sultat d&#x26;#39;une analyse structurelle-activit&#xE9; a d&#xE9;montr&#xE9; que l&#x26;#39;effet anti-&#xE2;ge du c&#xE9;l&#xE9;coxib peut &#xEA;tre ind&#xE9;pendant de son activit&#xE9; COX-2 inhibiteurs, comme des analogues de c&#xE9;l&#xE9;coxib que l&#x26;#39;activit&#xE9; inhibitrice de COX-2 manque produisent un effet similaire sur la dur&#xE9;e de vie. Hier berichten wir, dass Celecoxib, ein nicht-steroidale entz&Atilde;&frac14;ndungshemmende Arzneimittel zur Behandlung Schmerzen und Entz&uuml;ndungen, eingesetzt verl&auml;ngert Caenorhabditis Elegans Lebensdauer und verz&ouml;gert die Alters-assoziierten physiologischen &Auml;nderungen, z. B. R&uuml;ckgang der motorischen Aktivit&auml;t. Celecoxib verz&ouml;gert auch das Fortschreiten der altersbedingten Proteotoxicity sowie Tumorwachstum in C. Elegans. Celecoxib wurde urspr&uuml;nglich als ein potenter Cyclooxygenase-2 (COX-2)-Hemmer entwickelt. Jedoch nachgewiesen das Ergebnis aus einer Analyse der strukturellen-Aktivit&auml;t, dass die Anti-Aging Wirkung von Celecoxib unabh&auml;ngig von dessen COX-2-hemmende Aktivit&auml;t, sein k&ouml;nnte, als Analoga von Celecoxib, die COX-2-hemmende Aktivit&auml;t fehlt eine &auml;hnliche Wirkung auf Lebensdauer produzieren. Dar&uuml;ber hinaus fanden wir, dass Celecoxib Handlungen direkt am 3&#39;-Phosphoinositid-abh&auml;ngigen Kinase-1, eine Komponente der Insulin/IGF-1 signalisierende Kaskade, Lebensdauer zu erh&ouml;hen.

Celecoxib erweitert C. Elegans Lebensdauer &uuml;ber Hemmung Insulin-&auml;hnliche aber nicht Cyclooxygenase-2-Aktivit&auml;t zu signalisieren.

Un obiettivo di ricerca di invecchiamento &egrave; quello di sviluppare interventi che combattere le malattie legate all&#39;et&agrave; e rallentano l&#39;invecchiamento. Celecoxib &egrave; stato originariamente sviluppato come un inibitore potente cyclooxygenase-2 (COX-2). Tuttavia, il risultato da un&#39;analisi strutturale-attivit&agrave; hanno dimostrato che l&#39;effetto antiaging di celecoxib potrebbe essere indipendente dalla sua attivit&agrave; inibitoria COX-2, come analoghi di celecoxib che la mancanza di attivit&agrave; inibitoria COX-2 producono un effetto simile sulla durata della vita. In c. elegans, studi genetici suggeriscono che fattore di trascrizione di scossa di calore HSF-1 &egrave; richiesto per IIS modulare la longevit&agrave;. Qui, segnaliamo che l&#39;attivit&agrave; di HSF-1 &egrave; regolata da IIS. Il presente regolamento si verifica un passaggio precoce di HSF-1 attivazione tramite due regolatori HSF-1, 1-DDL e DDL-2. Inibizione del DDL-1/2 aumenta la longevit&agrave; e thermotolerance in maniera hsf-1-dipendente. Extended longevity is often correlated with increased resistance against various stressors. Insulin/IGF-1-like signaling (IIS) is known to have a conserved role in aging and cellular mechanisms against stress. In C.Â elegans, genetic studies suggest that heat-shock transcription factor HSF-1 is required for IIS to modulate longevity. Here, we report that the activity of HSF-1 is regulated by IIS. This regulation occurs at an early step of HSF-1 activation via two HSF-1 regulators, DDL-1 and DDL-2. Inhibition of DDL-1/2 increases longevity and thermotolerance in an hsf-1-dependent manner. Furthermore, biochemical analyses suggest that DDL-1/2 negatively regulate HSF-1 activity by forming a protein complex with HSF-1. The formation of this complex (DHIC) is affected by the phosphorylation status of DDL-1. Both the formation of DHIC and the phosphorylation of DDL-1 are controlled by IIS. Our findings point to DDL-1/2 as a link between IIS and the HSF-1 pathway.

La long&#xE9;vit&#xE9; &#xE9;tendue est souvent corr&#xE9;l&#xE9;e &#xE0; une r&#xE9;sistance accrue contre les divers facteurs de stress. Insulin/IGF-1-like signalisation (IIS) est connu pour avoir conserv&#xE9; un r&#xF4;le dans les m&#xE9;canismes de vieillissement cellulaire et contre le stress. Chez C. elegans, les &#xE9;tudes g&#xE9;n&#xE9;tiques sugg&#xE8;rent que le facteur de transcription de choc thermique HSF-1 est requis pour IIS afin de moduler la long&#xE9;vit&#xE9;. Ici, nous d&#xE9;clarons que l&#x26;#39;activit&#xE9; de HSF-1 est r&#xE9;gul&#xE9;e par IIS. Ce r&#xE8;glement intervient &#xE0; une &#xE9;tape pr&#xE9;coce de HSF-1 activation par deux HSF-1 r&#xE9;gulateurs, DDL-1 et DDL-2.
