Hepatitis C infection is associated with autoimmune disorders, such as the production of autoantibodies. Anti-LKM1 and anti-LC1, immunomarkers of type 2 autoimmune hepatitis, have been previously associated with a HCV infection. Anti-Soluble-Liver-Antigen autoantibodies (SLA) are specifically associated with type 1 and type 2 autoimmune hepatitis and more closely related to patients who relapse after steroid therapy. The recent molecular cloning of the soluble liver antigen provides the opportunity to develop more specific tests for the detection of antibodies against it. The aim of this work is to characterize anti-soluble-liver autoantibodies in sera from patients chronically infected by HCV. A recombinant cDNA from activated Jurkat cells coding for the full length tRNP(Ser)Sec/SLA antigen was obtained. ELISA, Western Blot and immunoprecipitation tests were developed and used to search for linear and conformational epitopes recognized by anti-SLA antibodies in sera from patients chronically infected by HCV. Anti-soluble liver antigen antibodies were found in sera from 10.4% of HCV-infected patients. The prevalence was significantly increased to 27% when anti-LKM1 was also present. Most anti-SLA reactivity was directed against conformational epitopes on the antigen. The means titers by ELISA were lower than those obtained in type 2 AIH. The result of autoantibody isotyping showed a subclass restriction to IgG1 and also IgG4. This study shows the presence of anti-SLA antibodies in approximately 10% of HCV infected patients. The prevalence of SLA autoantibodies in HCV infected patients increases when LKM1 autoantibodies are also present. The relationship between the prevalence of this characteristic autoimmune hepatitis autoantibody and the implication of an autoimmune phenomenon in the liver injury of patients chronically infected by HCV needs further investigation.

Anti-soluble liver antigen (SLA) antibodies in chronic HCV infection.

Infection par le virus de l&#39;h&eacute;patite C est associ&eacute;e &agrave; des maladies auto-immunes, comme la production d&#39;anticorps. Anti-LKM1 et anti-LC1, immunomarkers de l&#39;h&eacute;patite auto-immune de type 2, ont &eacute;t&eacute; pr&eacute;c&eacute;demment associ&eacute;s &agrave; une infection par le VHC. Hydrosolubles-foie-antig&egrave;ne anticorps (SLA) sont sp&eacute;cifiquement associ&eacute;es de type 1 et type 2 h&eacute;patite auto-immune et plus &eacute;troitement li&eacute;e aux patients qui rechutent apr&egrave;s corticoth&eacute;rapie. Le r&eacute;cent clonage mol&eacute;culaire de l&#39;antig&egrave;ne soluble du foie offre la possibilit&eacute; d&#39;&eacute;laborer des tests plus sp&eacute;cifiques pour la d&eacute;tection des anticorps dirig&eacute;s contre elle. L&#39;objectif de ce travail est de caract&eacute;riser les autoanticorps anti-hydrosolubles-foie dans le s&eacute;rum de patients chroniquement infect&eacute;s par le VHC. Un ADNc recombinant de cellules Jurkat activ&eacute;es codant pour l&#39;antig&egrave;ne de s/SLA tRNP (Ser) pleine longueur a &eacute;t&eacute; obtenu. Les tests ELISA, Western Blot et immunopr&eacute;cipitation ont &eacute;t&eacute; d&eacute;velopp&eacute;s et utilis&eacute;s pour la recherche lin&eacute;aires et conformationnelles des &eacute;pitopes reconnus par les anticorps de la SLA dans le s&eacute;rum de patients chroniquement infect&eacute;s par le VHC. Anticorps de l&#39;antig&egrave;ne du foie anti-soluble ont &eacute;t&eacute; trouv&eacute;s dans le s&eacute;rum de 10,4 % des patients infect&eacute;s par le VHC. La pr&eacute;valence augmentait de fa&ccedil;on significative &agrave; 27 % lorsque anti-LKM1, &eacute;tait &eacute;galement pr&eacute;sent. La plupart r&eacute;activit&eacute; SLA &eacute;tait dirig&eacute;e contre des &eacute;pitopes conformationnels sur l&#39;antig&egrave;ne. Les taux moyens par ELISA ont &eacute;t&eacute; inf&eacute;rieurs &agrave; ceux obtenus en type 2 HCA. Le r&eacute;sultat des auto-anticorps isotyping a montr&eacute; une restriction de la sous-classe IgG1 et IgG4. Cette &eacute;tude montre que la pr&eacute;sence d&#39;anticorps de la SLA dans environ 10 % du VHC les patients s&eacute;ropositifs. Test ELISA, Western Blot e immunoprecipitazione sono stati sviluppati e utilizzati per la ricerca di epitopi conformazionali e lineari riconosciuti dagli anticorpi anti-SLA in sieri di pazienti cronicamente infettati da HCV. Anticorpi anti-soluble antigene epatico sono stati trovati in sieri da 10,4% di pazienti affetti da HCV. La prevalenza &egrave; stata aumentata significativamente al 27% quando era presente anche anti-LKM1. Maggior reattivit&agrave; SLA era diretto contro gli epitopi conformazionali sull&#39;antigene. I titoli di mezzi da ELISA erano inferiori a quelli ottenuti nel tipo 2 AIH. Il risultato di autoanticorpi isotyping ha mostrato una restrizione della sottoclasse IgG1 e IgG4. Questo studio dimostra che la presenza di anticorpi anti-SLA in circa il 10% di HCV infetti pazienti. La prevalenza di autoanticorpi anti SLA in HCV infetti aumenta i pazienti quando sono presenti anche autoanticorpi anti LKM1. Il rapporto tra la prevalenza di autoanticorpi questa caratteristica epatite autoimmune e l&#39;implicazione di un fenomeno autoimmune le lesioni epatiche dei pazienti cronicamente infettati da HCV necessita di ulteriori indagini.

Infecci&oacute;n de hepatitis C se asocia con trastornos autoinmunes, como la producci&oacute;n de autoanticuerpos. Anti-LKM1 y anti-LC1, control de la hepatitis autoinmune de tipo 2, se han asociado previamente a una infecci&oacute;n por el VHC. Los autoanticuerpos anti-Soluble-h&iacute;gado-ant&iacute;geno (SLA) est&aacute;n espec&iacute;ficamente asociados con el tipo 1 y tipo 2 hepatitis autoinmune y m&aacute;s estrechamente vinculado a los pacientes que recaen despu&eacute;s de terapia esteroide. La reciente clonaci&oacute;n molecular del ant&iacute;geno soluble h&iacute;gado proporciona la oportunidad de desarrollar pruebas m&aacute;s espec&iacute;ficas para la detecci&oacute;n de anticuerpos contra &eacute;l. El objetivo de este trabajo es caracterizar los autoanticuerpos anti-soluble-h&iacute;gado en sueros de pacientes cr&oacute;nicamente infectados por el VHC. Se obtuvo un cDNA recombinante de c&eacute;lulas Jurkat activadas de codificaci&oacute;n para el ant&iacute;geno de Sec/SLA Garc&iacute;a (Ser) de longitud total. Pruebas de ELISA y Western Blot inmunoprecipitaci&oacute;n fueron desarrollados y utilizados para buscar lineal y conformacional epitopos reconocidos por anticuerpos anti-SLA en sueros de pacientes cr&oacute;nicamente infectados por el VHC. Se encontraron anti-soluble ant&iacute;geno h&iacute;gado anticuerpos en sueros de 10,4% de los pacientes infectados por el VHC. La prevalencia aument&oacute; significativamente al 27% cuando anti-LKM1 tambi&eacute;n estaba presente. Reactividad de anti-SLA la mayor&iacute;a fue dirigida contra epitopos conformacionales en el ant&iacute;geno. Los t&iacute;tulos de medios por ELISA eran m&aacute;s bajos que los obtenidos en el tipo 2 de la AIH. El resultado de autoanticuerpo isotyping mostr&oacute; una restricci&oacute;n de la subclase IgG1 y tambi&eacute;n IgG4. Este estudio demuestra que la presencia de anticuerpos anti-SLA en aproximadamente el 10% del VHC pacientes infectados. La prevalencia de autoanticuerpos de SLA en VHC infectados aumenta de pacientes cuando LKM1 autoanticuerpos tambi&eacute;n est&aacute;n presentes. La relaci&oacute;n entre la prevalencia de este autoanticuerpo hepatitis autoinmune caracter&iacute;stico y la implicaci&oacute;n de un fen&oacute;meno autoinmune en la lesi&oacute;n hep&aacute;tica de pacientes cr&oacute;nicamente infectados por el VHC requiere m&aacute;s investigaci&oacute;n.

Anti-liver-kidney microsome type 1 (LKM1) autoantibodies directed against the cytochrome P450 2D6 (CYP2D6) are considered specific markers of type 2 autoimmune hepatitis, but are also found in 5% of sera from patients chronically infected by hepatitis C virus (HCV). Molecular mimicry between HCV proteins and CYP2D6 has been proposed to explain the emergence of these autoantibodies. Anti-LKM1 autoantibodies from hepatitis C-infected patients were affinity-purified against immobilized CYP2D6 protein and used to screen a phage display library. CYP2D6 conformational epitopes were identified using phage display analysis and the identification of statistically significant pairs (SSPs). Cross-reactivity between CYP2D6 and HCV protein candidates was tested by immunoprecipitation. Nineteen different clones were isolated, and their sequencing resulted in the mapping of a conformational epitope to the region of amino acids 254-288 of CYP2D6. Candidate HCV proteins for molecular mimicry included: core, E2, NS3 and NS5a. Affinity-purified autoantibodies from HCV+/LKM1+ patients immunoprecipitated either NS3, NS5a, or both, and these reactivities were specifically inhibited by immobilized CYP2D6. In conclusion, HCV+/LKM1+ sera recognize a specific conformational epitope on CYP2D6 between amino acids 254 to 288, the region that contains the major linear epitope in type 2 autoimmune hepatitis patients. Cross-reactivity due to molecular mimicry at the B-cell level was shown between the CYP2D6 and the HCV NS3 and NS5a proteins and could explain the presence of anti-LKM1 in patients chronically infected with HCV. Further investigation of the role played by this molecular mimicry in HCV-infected patients may lead to more specific strategies for diagnosis and treatment.

H&iacute;gado-ri&ntilde;&oacute;n microsoma tipo 1 (LKM1) los autoanticuerpos dirigidos contra el citocromo P450 2D6 (CYP2D6) son considerados marcadores espec&iacute;ficos de la hepatitis autoinmune tipo 2, pero tambi&eacute;n se encuentran en el 5% de los sueros de pacientes cr&oacute;nicamente infectados por virus de la hepatitis C (HCV). La m&iacute;mica molecular entre prote&iacute;nas HCV y CYP2D6 se ha propuesto para explicar la aparici&oacute;n de estos autoanticuerpos. Los autoanticuerpos anti-LKM1 de pacientes infectados con hepatitis C eran afinidad purificada contra la prote&iacute;na CYP2D6 inmovilizada y una biblioteca de phage display de la pantalla. CYP2D6 epitopos conformacionales fueron identificados mediante an&aacute;lisis de phage display y la identificaci&oacute;n de pares estad&iacute;sticamente significativas (SSPs). Reactividad cruzada entre candidatos de prote&iacute;na CYP2D6 y VHC fue probada por la inmunoprecipitaci&oacute;n. Susceptible cells are inoculated with serial logarithmic dilutions of samples in a 96-well plate. After viral growth, viral detection by IPA yields the infectious virus titer, expressed as "Tissue Culture Infectious Dose" (TCID50). This represents the dilution of a virus-containing sample at which half of a series of laboratory wells contain replicating virus. This technique is a reliable method for the titration of HCoV in biological samples (cells, tissues, or fluids).

We have previously shown that the prototype OC43 strain of the common cold-associated human respiratory coronavirus has the capacity to infect human neuronal and glial cells and does persist in human brains. Moreover, it has neuroinvasive properties in susceptible BALB/c mice, where it leads to a chronic encephalitis with accompanying disabilities. Here, we show that mutations in the viral spike glycoprotein, reproducibly acquired during viral persistence in human neural cell cultures, led to a drastically modified virus-induced neuropathology in BALB/c mice, characterized by flaccid paralysis and demyelination. Even though infection by both mutated and wild-type viruses led to neuroinflammation, the modified neuropathogenesis induced by the mutated virus was associated with increased viral spread and significantly more CD4+ and CD8+ T-lymphocyte infiltration into the central nervous system, as well as significantly increased levels of the proinflammatory cytokine interleukin (IL)-6 and the chemokine CCL2 (monocyte chemoattractant protein [MCP]-1). Moreover, recombinant virus harboring the S glycoprotein mutations retained its neurotropism, productively infecting neurons. Therefore, interaction of a human respiratory coronavirus with the central nervous system may modulate virus and host factors resulting in a modified neuropathogenesis in genetically susceptible individuals.

Clonally expanded CD8(+) T lymphocytes are present in multiple sclerosis lesions, as well as in the cerebrospinal fluid of patients with multiple sclerosis. In experimental autoimmune encephalomyelitis, CD8(+) T lymphocytes are found in spinal cord and brainstem lesions. However, the exact phenotype of central nervous system-infiltrating CD8(+) T lymphocytes and the mechanism by which these cells cross the blood-brain barrier remain largely unknown. Using cerebrospinal fluid from patients with multiple sclerosis, spinal cord from experimental autoimmune encephalomyelitis and coronavirus-induced encephalitis, we demonstrate that central nervous system-infiltrating CD8(+) T lymphocytes are mostly of the effector memory phenotype (CD62L(-) CCR7(-) granzymeB(hi)). We further show that purified human effector memory CD8(+) T lymphocytes transmigrate more readily across blood-brain barrier-endothelial cells than non-effector memory CD8(+) T lymphocytes, and that blood-brain barrier endothelium promotes the selective recruitment of effector memory CD8(+) T lymphocytes. Furthermore, we provide evidence for the recruitment of interferon-Î³- and interleukin-17-secreting CD8(+) T lymphocytes by human and mouse blood-brain barrier endothelium. Finally, we show that in vitro migration of CD8(+) T lymphocytes across blood-brain barrier-endothelial cells is dependent on Î±4 integrin, but independent of intercellular adhesion molecule-1/leucocyte function-associated antigen-1, activated leucocyte cell adhesion molecule/CD6 and the chemokine monocyte chemotactic protein-1/CCL2. We also demonstrate that in vivo neutralization of very late antigen-4 restricts central nervous system infiltration of CD8(+) T lymphocytes in active immunization and adoptive transfer experimental autoimmune encephalomyelitis, and in coronavirus-induced encephalitis. Our study thus demonstrates an active role of the blood-brain barrier in the recruitment of effector memory CD8(+) T lymphocytes to the CNS compartment and defines Î±4 integrin as a major contributor of CD8(+) T lymphocyte entry into the brain.

Adem&#xE1;s, muestran que purifica la memoria humana efector CD8 (+) linfocitos T transmigran m&#xE1;s f&#xE1;cilmente a trav&#xE9;s de la sangre-cerebro endoteliales barrera de las c&#xE9;lulas que no efector de memoria CD8 (+) linfocitos T, y que el endotelio barrera sangre-cerebro promueve el reclutamiento selectivo de efector la memoria CD8 (+) linfocitos T. Nuestro estudio demuestra as&#xED; un papel activo de la barrera sangre-cerebro en la contrataci&#xF3;n de la memoria del efector CD8 (+) linfocitos T en el compartimiento del SNC y la integrina &#x3B1;4 define como un contribuyente importante de las c&#xE9;lulas CD8 (+) la entrada de los linfocitos T en el cerebro.
